Ferula persica wild (Apiaceae) is a perennial herb indigenous to Iran. It has been used in folk medicine for treatment of diabetes, lowering of blood pressure and for antispasmodic, carminative, laxative and expectorant effects in central Iran. Dried ground roots of F. persica (150 g) were extracted sequentially with n-hexane, dichloromethane and methanol (MeOH), 500 ml each, using a Soxhlet apparatus. The n-hexane extract of the roots (3 g) was subjected to vacuum liquid chromatography on silica gel, eluting with solvent mixtures of increasing polarity: 100% n-hexane–ethyl acetate (EtOAc), to yield a number of fractions, Fraction 4 (80% EtOAc in n-hexane) was further analysed by preparative TLC (mobile phase was 12% acetone in chloroform) to yield a coumarin ester (10.1 mg, Rf = 0.31, blue florescent). The structure of the isolated compound was elucidated by spectroscopic means. The compound is 7-O-(4,8,12 -trihydroxy-4,8,12-trimethyl-tridecanoyl)-coumarin, named, ferulone C as a new natural product.  相似文献

A thorough investigation of the ethyl acetate soluble fraction from a methanol extract of the resin of Ferula foetida has resulted in the isolation of a new sesquiterpene coumarin namely epi‐conferdione ( 1 ), along with four known compounds, colladonin ( 2 ), karatavicinol ( 3 ), 8‐acetoxy‐5‐hydroxyumbelliprenin ( 4 ), and asacoumarin ( 5 ). These structures were elucidated on the basis of extensive spectroscopic techniques, including 1D and 2D NMR spectroscopy. The absolute configuration of the new compound was established by circular dichroism (CD).  相似文献

Chemical investigation of the oleo-gum resin of Ferula assa-foetida L. from Yasuj region led to the identification of five sesquiterpene coumarins namely, conferone (1), badrakemin (2), feslol (3), isosamarcandin (4) and samarcandin (5). The compounds were characterised by detailed 1D (1H and 13C) and 2D NMR (HMBC, HSQC and NOESY) experiments. Among these compounds, conferone (1) and isosamarcandin (4) are being newly reported in F. assa-foetida L.  相似文献

One new coumarin derivative (1) and two known compounds, quercetin (2) and glyceraldehyde (3) have been isolated from the whole plants of Euphorbia wallichii. Their structures were elucidated by means of extensive spectroscopic analysis (NMR and ESI-MS) and by comparison with data reported in the literature. A novel optimised isolation method, TLC-bioautography, was evaluated and utilised in this research. Antibacterial compounds which were isolated from the dichloromethane extract of Ferula ferulioides (Steud.) Korovin were detected by means of the method. Their structures were elucidated by extensive spectral and chemical methods. Their antibacterial activities against drug-resistant Staphylococcus aureus (S. aureus) strains were evaluated with broth microdilution method, and the results proved that TLC-bioautography was an effective and highly efficient way to screen natural compounds from plant extracts against drug-resistant strains.  相似文献

Ferula orientalis (Apiaceae) is a well-known perennial herb growing wild in Iran used in traditional medicine. To perform phytochemical studies, dried ground roots of F. orientalis were sequentially Soxhlet-extracted using n-hexane, dichloromethane and methanol. A combination of vacuum liquid chromatography and preparative thin-layer chromatographic analyses were performed to isolate coumarin esters. The structures of the isolated compounds were elucidated by spectroscopic means, and in vitro free-radical-scavenging property was determined by the DPPH assay. Two new coumarin esters, 7-O-(4,8,12,16-tetrahydroxy-4,8,12,16-tetramethyl-heptadecanoyl)-coumarinand 7-O-(4-hydroxy-4,8,12-trimethyl-trideca-7,11-dienoyl)-coumarin, named ferulone A and ferulone B, respectively, were isolated from the n-hexane extract of the roots of F. orientalis. Both compounds showed a low level of free-radical-scavenging property with the RC50 values of 0.252 and 0.556 mg/mL for compounds 1 and 2, respectively, as opposed to that of the positive control (quercetin) 0.004 mg/mL. This is the first report on the purification of coumarin esters from the genus Ferula.  相似文献

The chloroform fraction of methanol (MeOH) extract of the aerial parts of Launaea mucronata was in vitro investigated for cytotoxicity against HCT116, HepG2 and MCF-7 cell lines, and resulted with IC50 = 20.0, 18.6 and 14.30 μg/mL, respectively. The chloroform fraction of the MeOH extract was subjected to further fractionation, which led to the isolation of two new coumarin compounds (6-isobutyl coumarin and 6-isobutyl-7-methyl- coumarin). The structures of the new compounds were elucidated by high field 1D and 2D NMR and ESI-MS spectroscopies. Low polar fractions revealed the isolation of other known triterpene compounds which were identified according to its spectral data and comparison with the literature. New coumarin compounds show high cytotoxicity against MCF-7, HCT116 and HepG2 cell lines.  相似文献

Twelve angular oxazole-fused coumarin derivatives were designed, synthesised and characterised by 1H NMR, 13C NMR and HRMS. The structure of compound 4a was further confirmed by X-ray single-crystal diffraction. The bioassay experiment results indicated that compounds 4f and 4l have high antifungal activity on the mycelium growth of 4 plant disease fungi. Especially, compound 4l has a stronger antifungal activity compare to the commercial fungicide, Carbendazim. The herbicidal activity experiment showed that 4a and 4b can significantly inhibit the taproot and caulis development of Chenopodium album seedling and have better activities than the commercial herbicide, Acetochlor.  相似文献

Ferulasinkins A–D (1–4), four new norlignans, were isolated from the resins of Ferula sinkiangensis, a medicinal plant of the Apiaceae family. All of them were obtained as racemic mixtures, chiral HPLC was used to produce their (+)- and (−)-antipodes. The structures of these new compounds, including their absolute configurations, were elucidated by spectroscopic and computational methods. This isolation provides new insight into the chemical profiling of F. sinkiangensis resins beyond the well-investigated structure types such as sesquiterpene coumarins and disulfides. Compounds 2a and 3a were found to significantly inhibit the invasion and migration of triple-negative breast cancer (TNBC) cell lines via CCK-8 assay. On the other hand, the wound-healing assay also demonstrated that compounds 4a and 4b could promote the proliferation of human umbilical vein endothelial cells (HUVECs). Notably, the promoting effects of 4a and 4b were observed as more significant versus a positive control using basic fibroblast growth factor (bFGF).  相似文献

2-Hydrazino-4-hydroxy-5H- [1] -benzopyrano- [ 4,3-d ] -pyrim-idin-5-one (3) was prepared via condensation of 2 with hy-drazine hydrate. Treatment of 3 with methylene chloride, ethyl chloroformate, ethyl chloroacetate and benzaldehyde yielded the corresponding 2 - ( substituted ) hydrazino - 4 -hydro-xy-5H-[1]-benzopyrano-[4, 3-d]-pyrimidin-5-one (4, 5, 6, and 10), followed by cyclization of 4, 5 and 6 with dimethyl fonnamide and fused sodium acetate under reflux, while compound 10 was cyclized with bromine and sodium acetate in acetic acid. Compound 3 reacted with β-(toloyl) acrylic acid, ethyl α-cyano-p-methoxytinnamate, diethyl mal-onate and acetyl chloride affording the corresponding 2-(substituted) hydrazino-4-hydroxy-5H-[1]-benzopyrano-[4, 3-d]-pyrimidin-5-one (12, 13, 14, 15 and 16).  相似文献

Abstract

Two new compounds, versicolones A and B (1 and 2), and three known pyrone derivatives (3?5) were isolated from the insect-associated fungus Aspergillus versicolor. Their structures were elucidated through a combination of HRESIMS and NMR spectroscopic analysis. Versicolone A (1) was revealed as a coumarin derivative with the rare 5-alkyl side chain substitution. Compound 5 exhibited significant antioxidant activity with EC50 value of 8.0?μM in the ABTS (2,2-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid) assay, which was more than 2-fold potency of the positive control trolox.  相似文献

Monomer of 7-methacryloyloxy-4-methylcoumarin(MAOMC) was synthesized and characterized by FTIR, 1H-NMR and 13C-NMR spectroscopy. Copolymers of MAOMC with butoxyethylmethacrylate(BOEMA) at different feed compositions were prepared by free radical solution polymerization at(70 ± 1) °C in ethylmethylketone(EMK) using benzyl peroxide(BPO) as an initiator. The copolymers were characterized by FTIR and 1H-NMR spectroscopy. Thermogravimetric analysis(TGA) and differential scanning calorimetry(DSC) of the copolymers showed moderate thermal stability and higher Tg values. Gel permeation chromatography(GPC) was used to find out the molecular weights of the different copolymers. Antibacterial activities of the copolymers were also investigated against the selected pathogenic bacteria's. The antibacterial activity of the copolymer increases as the MAOMC content increases in the copolymer. This shows that coumarin moiety plays a very important role in the antibacterial activity.  相似文献

Ferula communis is a showy herbaceous plant typical of the Mediterranean area where it is used as a traditional medicine. The plant is a source of bioactive compounds such as daucane sesquiterpenes and prenylated coumarins. In Italy, most of phytochemical studies focused on Sardinian populations where poisonous and nonpoisonous chemotypes were found, while investigations on peninsular populations are scarce. In this work, we report the chemical characterisation of the essential oils obtained from different parts of F. communis growing in central Italy. The chemical profiles of the plant parts, as detected by GC-FID and GC-MS, were different from each other and from those reported in insular populations. Notably, α-pinene (10.5%), γ-terpinene (7.6%) and hedycariol (8.4%) were the major volatile constituents in flowers; α-pinene (55.9%), β-pinene (16.8%) and myrcene (5.9%) in fruits; β-eudesmol (12.1%), α-eudesmol (12.1%) and hedycariol (10.3%) in leaves; (E)-β-farnesene (9.5%), β-cubebene (8.2%) and (E)-caryophyllene (7.2%) in roots. The volatile profiles detected did not allow to classify the investigated central Italy population into the poisonous and nonpoisonous chemotypes previously described in Sardinia.  相似文献

Four new metabolites (1-4) have been isolated from the organic extract of the seagrass Cymodocea nodosa, collected at the coastal area of Porto Germeno, in Attica Greece. Compounds 1 and 2 belong to the structural class of diarylheptanoids, which have been found only once before in marine organisms [Kontiza, I.; Vagias, C.; Jakupovic, J.; Moreau, D.; Roussakis, C.; Roussis, V. Tetrahedron Lett.2005, 46, 2845-2847]. Compound 3 is a new meroterpenoid, while compound 4, to the best of our knowledge, is the first briarane diterpene isolated from seaweeds, and only the second analog of this class with a tricyclic skeleton. Furthermore metabolite 4 is the first brominated briarane diterpene. The structures and the relative stereochemistry of the new natural products were established by spectral data analyses. The new metabolites were submitted for evaluation of their antibacterial activity against multidrug-resistant (MDR) pathogens including methicillin-resistant (MRSA) strains of Staphylococcus aureus, as well as the rapidly growing mycobacteria, Mycobacterium phlei, Mycobacterium smegmatis, and Mycobacterium fortuitum.  相似文献

Fucoidan is a fucose-rich sulfated polysaccharide with attractive therapeutic potential due to a variety of biological activities, including antioxidant action. Fucoidan is typically found in the cell wall of marine brown algae, but extra-algal sources have also been discovered. In the present work, for the first time we extracted a water soluble fucoidan fraction from the roots of the terrestrial shrub Ferula hermonis. This fucoidan fraction was termed FUFe, and contained fucose, glucose, sulfate, smaller amounts of monosaccharides such as galactose and mannose, and a minor quantity of proteins. FUFe structural features were investigated by FTIR, 1H NMR and 13C NMR spectroscopy. The antioxidant property of FUFe was measured by DPPH, ABTS and FRAP assays, which revealed a high radical scavenging capacity that was confirmed in in vitro cellular models. In hepatic and endothelial cells, 50 μg/mL FUFe could reduce ROS production induced by intracellular lipid accumulation. Moreover, in hepatic cells FUFe exhibited a significant antisteatotic action, being able to reduce intracellular triglyceride content and to regulate the expression of key genes of hepatic lipid metabolism. Altogether, our results candidate FUFe as a possible bioactive compound against fatty liver disease and related vascular damage.  相似文献

In order to find novel antibacterial agents with superior antibacterial activity and overcoming multidrug resistance,a series of pleuromutilin derivatives with novel C(14) side chain were synthesized and evaluated for their in vitro antibacterial activities.The results of antibacterial acticities indicated that most of the derivatives showed potent activities against Gram-positive organisms.In particular,compound lOd exhibited the most potent inhibitory activity compared with pleuromutilin and linezoid,emerged as potential molecule for further investigation.  相似文献

Coumarins are omnipresent in several plants and exhibit a plethora of pharmacological properties, making them an important scaffold in organic synthesis. Naturally, the chemistry of this motif has attracted ever-increasing attention, among which palladium-catalyzed coupling reactions are the most prevalent one. Numerous useful, easy, and concise syntheses and reactions have been achieved using palladium-catalyzed coupling reactions. <正>A new methacrylamide monomer,hexylamine methacrylamide(MAHA),was synthesized and used in polymerizations.The homopolymer of MAHA and its copolymers were synthesized by free radical polymerization techniques with N-isopropyl acrylamide(NIPAAm) in two different compositions.The quaternization of the homopolymer and copolymers were carried out using 1-bromopropane.The copolymers with NIPAAm and a low MAHA content showed temperature-responsive behavior in an aqueous environment.The lower critical solution temperatures(LCSTs) of these polymers varied between 32℃and 44℃.The LCSTs of quaternized copolymers were higher than those of neutral copolymers because they were more hydrophilic.The obtained homopolymers and copolymers were tested for antibacterial activities against S.aureus and E.coli.The quaternized water-soluble copolymers showed antibacterial activities against S.aureus.The quaternization resulted in the synthesis of both antibacterial and temperature-responsive copolymers.  相似文献

Four natural chalcones bearing hydroxyisoprenyl or prenyl groups, named Paratocarpin E (2), Xanthoangelol D (3), Angusticornin A (4) and Kanzonol C (5), were prepared by employing the Claisen-Schmidt condensation as the key step. In an attempt to investigate the effect of the hydroxyisoprenyl group on biological activity, two of their derivatives were also prepared for antibacterial activity research. The synthesized compounds were investigated for their expected antibacterial activities against Gram positive bacteria (Bacillus subtilis, Staphylococcus aureus) as well as Gram negative bacteria (Escherichia coli, Pseudomonas aeruginosa). Paratocarpin E (2) was found to be the most potent against two Gram positive bacteria while the majority of the remaining compounds showed promising activity as well. However, all of the compounds were inactive against both Gram-negative bacteria.  相似文献

Synthesis of the antibacterial emodin was improved using Friedel-Crafts acylation as a key step leading to 37% overall yield. In addition, 21 analogues were synthesized by structural modification of the hydroxyl and methyl groups, as well as the aromatic ring of emodin. Antibacterial activity against methicillin-resistant Staphylococcus aureus (MRSA) and cytotoxicity against noncancerous Vero cells were evaluated. A structure-activity relationship (SAR) study indicated that the hydroxyl groups and the methyl group in the emodin skeleton were crucial for anti-MRSA activity. Furthermore, the presence of an iodine atom or ethylamino group on the aromatic ring enhanced the anti-MRSA activity with higher selectivity indices, while derivatives containing bromine, chlorine atoms or quaternary ammonium salt were as active as emodin. The quaternary ammonium group on the aromatic ring also led to non-cytotoxicity against Vero cells.  相似文献
