A new spectrofluorimetric method has been developed and validated for the quantification of ceftriaxone in bulk powder, pharmaceutical formulations and spiked human plasma. The developed method is reproducible, accurate, sensitive and cost effective. In this method, ceftriaxone was converted into a fluorescent compound by reacting with 0.8 M ethyl acetoacetate and 25% formaldehyde in a buffered medium (pH = 4.2) at 90 °C. The excitation and emission wavelengths of the fluorescent reaction product are 316 nm and 388 nm respectively. Optimization of the experimental conditions affecting the condensation reaction were carefully carried out and the optimum experimental conditions were incorporated in the procedure. The developed method has a broad linear range (0.2–20 μg mL−1) with a correlation coefficient of 0.9992. The limit of detection (LOD) and limit of quantification (LOQ) was found to be 1.94 × 10−2 μg mL−1 and 6.47 × 10−2 μg mL−1 respectively. The common excipients and co-administered drugs were investigated for their interferences effect in the assay. The developed method was validated statistically through recovery studies and successfully applied to ceftriaxone determination in bulk powder, pharmaceutical formulations and spiked human plasma samples. The percent recoveries were found to be in the range of 99.04–100.26% for bulk powder, 98.88–99.92% for pharmaceutical formulations and 94.22–98.48% for spiked human plasma. The results were verified by comparing with reference literature HPLC method and were found in good agreement.  相似文献

A rapid, simple and highly sensitive first derivative synchronous fluorometric method has been developed for the simultaneous analysis of binary mixture of sulpiride (SUL) and mebeverine hydrochloride (MEB). The method is based upon measurement of the synchronous fluorescence intensity of these drugs at ∆λ = 100 nm in water. The different experimental parameters affecting the fluorescence of the two drugs were carefully studied and optimized. The fluorescence-concentration plots were rectilinear over the range of 0.05–1 μg/mL and 0.2–3.2 μg/mL for SUL and MEB respectively with lower detection limits (LOD) of 0.006 and 0.01 μg/mL and quantification limits (LOQ) of 0.0.02 and 0.05 μg/mL for SUL and MEB, respectively. The proposed method was successfully applied for the determination of the two compounds in synthetic mixtures and in commercial tablets. The high sensitivity attained by the proposed method allowed the determination of both of SUL and MEB metabolite (veratic acid) in real human plasma samples applying second derivative synchronous fluorometric technique. The mean% recoveries (n = 3) for both MEB metabolite (veratic acid) and SUL were 99.82 ± 2.53 and 98.84 ± 6.20 for spiked human plasma respectively, while for real human plasma, the mean% recoveries (n = 3) were 91.49 ± 4.25 and 91.36 ± 8.46 respectively.  相似文献

A sensitive, simple and selective spectrofluorimetric method was developed for the determination of oxamniquine (OXM) in pharmaceutical formulations and biological fluids. The method is based on the reaction between the drug and 1-dimethylaminonaphthalene-5-sulphonyl chloride (dansyl chloride) in presence of 0.5 M sodium carbonate (pH 10) to yield a highly fluorescent derivative that is measured at 445 nm after excitation at 335 nm. The different experimental parameters affecting the development and stability of the reaction product were carefully studied and optimized. The fluorescence concentration plot was rectilinear over the range of 0.02–0.2 μg ml−1 with a lower detection limit (LOD) of 0.007 μg ml−1 and limit of quantitation (LOQ) of 0.02 μg ml−1. The proposed method was successfully applied to the analysis of commercial capsules. The results obtained were in good agreement with those obtained using the official spectrophotometric method. Furthermore, the method was applied for the determination of oxamniquine in spiked human plasma, the mean % recovery (n = 4) is 97.77 ± 1.19. A sensitive, simple and selective spectrofluorimetric method was developed for the determination of Lamotrigine (LMT) in pharmaceutical formulations and biological fluids. The method is based on reaction of LMT with o-phthalaldehyde in presence of 2-mercaptoethanol in borate buffer of pH 9.8 to yield a highly fluorescent derivative that is measured at 448 nm after excitation at 337 nm. The different experimental parameters affecting the development and stability of the reaction product were carefully studied and optimized. The fluorescence-concentration plot was rectilinear over the range of 0.1–1.0 μg ml−1 with lower limit of detection (LOD) 0.02 μg ml−1 and limit of quantification (LOQ) 0.06 μg ml−1 respectively. The proposed method was successfully applied to the the analysis of commercial tablets. Statistical comparison of the results obtained by the proposed and reference method revealed no significant difference in the performance of the two methods regarding the accuracy and precision respectively. The proposed method was further extended to the in-vitro and in-vivo determination of the drug in spiked and real human plasma. The mean percentage recoveries in spiked and real human plasma (n = 3) were 95.78 ± 1.37 and 90.93 ± 2.34 respectively. Interference arising from co-administered drugs was also studied. A proposal for the reaction pathway with o-phthalaldehyde was postulated.  相似文献

The use of ozonation for the purification of drinking water can lead to the formation of bromate. The US Environmental Protection Agency and the European Directive for human drinking water has lowered the regulatory level for bromate down to 10 μg l−1, such that methods must be developed for monitoring the formation of bromate, particularly in on-site situations. In the present work we report a fluorometric method for the determination of bromate based on the reaction with carbostyril-124, a compound that shows florescence mainly at pH values above 4 and, when bromated, generates a non-fluorescent product. The reaction can thus be used as an indirect method for determination of the ion. The proposed method, which uses the flow injection (FI) technique, allows online application and kinetic control of the variables affecting the process, together with shorter reaction times, and it provides maximum sensitivity and selectivity. Under optimum conditions, it is possible to determine the analyte within the 4–200 μg l−1 range, with a limit of detection of 0.9 μg l−1 and a relative standard deviation (n = 12, [] = 5 and 30 μg l−1) of 3.2% and 2.6% respectively. The determination rate was ten samples per hour.  相似文献

A sensitive, rapid, and specific assay has been developed for the simultaneous determination of acetylsalicylic acid and caffeine in commercial tablets based on their natural fluorescence. The mixture of these drugs was resolved by first derivative synchronous fluorimetric technique using two scans. At Δλ=106 nm, using first derivative synchronous scanning, only acetylsalicylic acid yields a detectable signal at 316 nm (peak to zero method) which is unaffected by caffeine. At Δλ=30 nm, the signal of caffeine at 288 nm (peak to zero method) is not affected by acetylsalicylic acid. The range of application is between 0.021 and 41.62 μg ml−1 (correlation coefficient, R=0.9995) for acetylsalicylic acid and between 0.4486 and 44.86 μg ml−1 (correlation coefficient, R=0.99786) for caffeine. The recovery range of 98.40–102% for acetylsalicylic acid and 90–100.5% for caffeine from their synthetic mixture was reported. Overall recovery of both compounds about 97–99% for acetylsalicylic acid and 97–98% for caffeine was obtained from real sample analysis. The detection limits are 0.0013 μg ml−1 and 0.0306 μg ml−1 for acetylsalicylic acid and caffeine, respectively. The relative standard deviation (n=10) for 20 μg ml−1 of acetylsalicylic acid is 2.75% and for 2.2 μg ml−1of caffeine is 1.7%.  相似文献

It is found that silver nanoparticles (AgNPs) can further enhance the fluorescence intensity of curcumin (CU) - cetyltrimethylammonium bromide (CTAB) – nucleic acids and improve its anti-photobleaching activity. Under optimum conditions, the enhanced fluorescence intensity is proportion to the concentration of nucleic acids in the range of 2.0 × 10−8–1.0 × 10−6 g mL−1 for fish sperm DNA (fsDNA), 2.0 × 10−8–1.0 × 10−6 g mL−1 for calf thymus DNA (ctDNA), 1.0 × 10−8–1.0 × 10−6 g mL−1 for yeast RNA (yRNA), and their detection limits (S/N = 3) are 8.0 ng mL−1, 10.5 ng mL−1 and 5.8 ng mL−1, respectively. This method is used for determining the concentration of DNA in actual sample with satisfactory results. The interaction mechanism is also studied.  相似文献

A new anthracene-based fluorescent PET sensor 1 with a tridentate ionophore of amide/β-amino alcohol displays very good selectivity and sensitivity for Fe3+ (K a = 1.6 × 103 M−1) and Hg2+ (K a = 2.1 × 103 M−1) in CH3CN–H2O (3:7, v/v) with detection limit of 1 μM. More fluorescence enhancement was observed when 1 selectively detected Fe3+ or Hg2+ in CH3CN and its detection limit was up to 0.03 μM.  相似文献

Two simple, accurate and highly sensitive spectrofluorometric methods were developed for the determination of ethamsylate (ETM). Method I is based on measuring the native fluorescence of ethamsylate in water at 354 nm after excitation at 302 nm. The calibration plot was rectilinear over the range of 0.05–1 μg/mL for ETM with limits of detection and quantitation of 7.9 and 26 ng/mL, respectively. Method II involved synchronous and first derivative synchronous fluorometric methods for the simultaneous determination of ethamsylate (ETM) and hydroquinone (HQ) which is considered as an impurity and/or acidic degradation product. The synchronous fluorescence of both the drug and its impurity were measured in methanol at Δ λ of 40 nm. The peak amplitudes (1D) were estimated at 293.85 or 334.17 nm for ETM and at 309.05 nm for HQ. Good linearity was obtained for ETM over the ranges 0.1–1.4 μg/mL and 0.1–1.0 μg/mL at 293.85 and 334.17 nm, respectively. For HQ, the calibration plot was rectilinear over the range of 0.01–0.14 μg/mL at 309.05 nm. Limits of detection were 20, 2.01 ng/mL and limits of quantitation were 60, 6.7 ng/mL for ETM and HQ by method II, respectively. Both methods were successfully applied to commercial ampoules and tablets. The results were in good agreement with those obtained by the reference method. Method I was utilized to study the stability of ETM and its degradation kinetics using peroxide. The apparent first-order rate constant, half-life times and activation energy of the degradation process were calculated. Method I was further extended to the in-vitro and in-vivo determination of ETM in spiked and real plasma samples. The mean% recoveries were 99.57 ± 3.85 and 89.39 ± 5.93 for spiked and real human plasma, respectively.  相似文献

A simple, sensitive and rapid spectrofluorometric method for determination of methocarbamol in pharmaceutical formulations and spiked human plasma has been developed. The proposed method is based on the measurement of the native fluorescence of methocarbamol in methanol at 313 nm after excitation at 277 nm. The relative fluorescence intensity-concentration plot was rectilinear over the range of 0.05–2.0 μg/mL, with good correlation (r = 0.9999), limit of detection of 0.007 μg/ mL and a lower limit of quantification of 0.022 μg/ mL. The described method was successfully applied for the determination of methocarbamol in its tablets without interference from co-formulated drugs, such as aspirin, diclofenac, paracetamol and ibuprofen, The results obtained were in good agreement with those obtained using the official method (USP 30).The high sensitivity of the method allowed the determination of the studied drug in spiked human plasma with average percentage recovery of 99.42 ± 3.84.  相似文献

It was first found that 4-(2-pyridylazo)-resorcinol (PAR) can combine with lysozyme forming a polymolecular complex, with the molar ratio of 1:12 (lysozyme/PAR) in the medium of acetate buffer (pH 6.0). The fluorescence spectra suggested that PAR molecules may produce self-aggregation, and it was suggested that there are interactions between lysozyme and PAR possibly mainly on the cooperation of hydrophobic, electrostatic actions and chelate bond. The coordination complex of lysozyme-PAR can cause an enhanced resonance light scattering (RLS) signal with the scattering peak located at 521 nm. The increment of RLS intensity was linear with the concentration of lysozyme over the range from 1 ng mL-1 to 30.0 μg mL-1, with the limit of detection of 0.3 ng mL−1 (3σ, n = 5). Effect of experimental conditions, including temperature, pH, concentration of PAR, and the ionic strength on RLS were also tested. The proposed RLS method was successfully applied to analyze lysozyme in hen egg white, human saliva and tear samples without any special pretreatment. Compared with other methods, the proposed procedure is of high sensitivity and selectivity.  相似文献

The formation of a complex between ketoconazole and β-cyclodextrin was followed by spectrofluorimety. The inclusion of ketoconazole in β-cyclodextrin cavity enhanced the native fluorescence of the drug. The stoichiometry of the complex was 1:1 β-cyclodextrin to ketoconazole and the stability constant of the complex (log K f) was determined to be 4.3 ± 0.01 at pH = 7.9 and 3.7 ± 0.04 at pH = 2.6. A sensitive spectrofluorimetric method for the detection of ketoconazole is presented. At optimized experimental conditions, a linear relationship between the fluorescence intensity of the solution and concentration of ketoconazole is observed in the range of 0.01–10 μg ml−1 (5 × 10−8 M–1.88 × 10−5 M). The method was applied to the detection of ketoconazole in pharmaceutical products and the results were satisfactory in comparison to the official method (relative error = 2.8% and standard deviation = 0.06 for tablets of ketoconazole). The recovery of ketoconazole from a blood serum sample, determined by the proposed method, was 97.1 ± 2.4%.  相似文献

When 1.00 mol l−1 I is used as ion perturber, rhodamine 6G (Rh 6G) can emit strong and stable room temperature phosphorescence (RTP) on filter paper substrate in KHC8H4O4–HCl buffer solution (pH = 3.50), heated at 70 °C for 10 min. NaIO4 can oxidize Rh 6G, which makes the RTP signal quench. Terbutaline sulfate (TBS) can inhibit NaIO4 from oxidizing Rh 6G, which makes the RTP signal of Rh 6G enhance sharply. The content of TBS is linear correlation to ΔIp of the system. Based on the facts above, a new inhibition solid substrate-room temperature phosphorimetry (SS-RTP) for the determination of trace TBS has been established. The linear range of this method is 0.0104–2.08 pg spot−1 (corresponding concentration: 0.026–5.2 ng ml−1, with a sample volume of 0.4 μl) with a detection limit (L.D.) of 2.6 fg spot−1 (corresponding concentration: 6.5 × 10−12 g ml−1), and the regression equation of working curve is ΔIp = 2.040 + 54.54 mTBS (pg spot−1), n = 6, correlation coefficient is 0.9994. For the samples containing 0.0104 pg spot−1 and 2.08 pg spot−1 TBS, the relative standard deviation (RSD) are 3.8% and 2.3% (n = 8), respectively, indicating good precision. This method has been applied to determination of trace TBS in the practical samples with satisfactory results. The reaction mechanism of NaIO4 oxidizing Rh 6G to inhibit SS-RTP for the determination of trace TBS is also discussed.  相似文献

In pH 1.8 ∼ 2.8 weak acid medium, polyvinylpyrrolidone (PVP) and Eosin Y reacted to form complex that could result in Eosin Y (EY) fluorescence quenching. The maximum quenching wavelength was at 542 nm. The fluorescence quenching (ΔF) was proportional to the concentration of polyvinylpyrrolidone in a certain range. The linear range, the correlation coefficient and the detection limit were 0.33 ∼ 2.0 μg•mL−1, 0.9994 and 99.6 ng•mL−1, respectively. The influences of the coexistence substances were tested and the results showed that the method had good selectivity. Therefore, a new method based on fluorescence quenching of eosin Y by PVP for the determination of trace PVP was developed. The method was sensitive, simple and rapid, which was applied to the determination of trace PVP in the beer with satisfactory results. The reaction mechanism was also discussed.  相似文献

A fluorophotometric method for the determination of hydrogen peroxide (H2O2) using fluorescin was developed. This method was based on the oxidative reaction of fluorescin, a colorless, non-fluorescent lactoid fluorescein, by H2O2 to give highly fluorescein fluorescence emission. In the determination of H2O2, the calibration curve exhibited linearity over the H2O2 concentration range of 1.5–310 ng mL−1 at an emission wavelength of 525 nm with an excitation of 500 nm and with relative standard deviations (n = 6) of 2.51%, 2.48%, and 1.31% for 3.1 ng mL−1, 30.8 ng mL−1, and for 308 ng mL−1 of H2O2, respectively. The detection limit for H2O2 was 1.9 ng mL−1 six blank determinations was performed (ρ = 6). This proposed method was applied to detection of other reactive oxygen species and nitrogen species (ROS/RNS) such as singlet oxygen (1O2), hydroxyl radical (OH), peroxynitrite (ONOO) etc., and it was possible to detect them with a high sensitivity. In addition, this proposed method was applied to the recovery tests of H2O2 in calf serum, human saliva, rain water, and wheat noodles; the results were satisfactory.  相似文献

A rapid, simple and highly sensitive first derivative synchronous spectrofluorimetric method was developed for the simultaneous analysis of a binary mixture of labetalol HCl (LBT) and furosemide (FUR) without prior separation. The method was based upon measuring the first derivative of synchronous fluorescence spectra of the two drugs at Δλ =130 nm in aqueous ethanol (55% V/V). The different experimental parameters affecting the synchronous fluorescence of the studied drugs were carefully studied and optimized. The first derivative amplitude-concentration plots were rectilinear over the range of 0.10 to 1.00 μg/mL and 0.05–0.50 μg/mL with lower detection limits of 0.0149 and 7×10−3 μg/mL and quantification limits of 0.045 and 0.021 μg/mL for LBT and FUR, respectively. The proposed method was successfully applied for the determination of the studied drugs in synthetic mixtures. The results obtained were in good agreement with those obtained by the reference methods.  相似文献

A simple, sensitive and accurate method has been developed for spectrofluorimetric determination of cefixime in pure form and pharmaceutical preparations. The method is based on the reaction of cefixime with 2-cyanoacetamide in the presence of 21% ammonia at 100 °C. The fluorescent reaction product showed maximum fluorescence intensity at λ 378 nm after excitation at λ 330 nm. The factors affecting the derivatization reaction were carefully studied and optimized. The fluorescence intensity versus concentration plot was rectilinear over the range of 0.02 to 4 μg mL−1 with correlation coefficient of 0.99036. The limit of detection (LOD) and limit of quantification (LOQ) was found to be 2.95 ng mL−1 and 9.84 ng mL−1, respectively. The proposed method was validated statistically and through recovery studies. The method was successfully applied for the determination of cefixime in pure and dosage form with percent recoveries from 98.117% to 100.38%. The results obtained from the proposed method have been compared with the official HPLC method and good agreement was found between them.  相似文献

A new, simple and sensitive spectrofluorimetric method has been developed for the determination of oseltamivir phosphate (OSP) in capsules. The method is based on the reaction between oseltamivir and fluorescamine in borate buffer solution of pH 8.50 to give highly fluorescent derivatives that are measured at 483 nm using an excitation wavelength of 381. The different experimental parameters effecting the development and stability of the reaction product were carefully studied and optimized. The fluorescence intensity concentration plot is rectilinear over the range 50–450 ng mL−1 with a lower detection limit (LOD) of 1.219 ng mL−1 and limit of quantitation (LOQ) of 4.064 ng mL−1. Selectivity was validated by subjecting stock solution of OSP to acidic, basic, oxidative, and thermal degradation. No interference was observed from excipients present in formulations. The developed method was successfully applied to determination of the drug in capsules. The mean % recovery (n = 6) was 100.08. The results obtained were in good agreement with those obtained using a reported spectrophotometric method.  相似文献

The method is based on measuring the native fluorescence of ZPS in acetate buffer of pH 4.5 at 398 nm after excitation at 315 nm. The fluorescence-concentration plot was rectilinear over the range of 0.05–0.80 μg mL−1 with a lower detection limit (LOD) of 6.0 ng mL−1 and quantification limit (LOQ) of 20.0 ng mL−1. The method was fully validated and successfully applied to the determination of ZPS in its capsules with average percentage recovery of 99.7 ± 1.4. The method was extended to stability study of ZPS. The drug was exposed to acidic, alkaline, oxidative and photolytic degradation according to ICH guidelines. Moreover, the method was utilized to investigate the kinetics of the alkaline, acidic and oxidative degradation of the drug. A proposal for the degradation pathways was postulated.  相似文献
