This paper presents an HPLC-MS method for the fully automated determination of a group of naphthalene monosulphonates in environmental water samples. The analytical procedure consisted of on-line ion-pair solid-phase extraction using a PLRP-S precolumn and ion-pair LC separation with triethylamine as ion-pair reagent in both cases. A mass spectrometric detector, coupled to LC through an electrospray interface and operated in negative ion mode, was used. Diagnostic ions usually corresponded to [SO3]- and/or [M-SO2H]- together with [M-H] and/or [M-2H+Na]-. The method was applied to the trace determination of several sulphonates present in tap water, seawater and water from the Ebro river. The analytes were determined at a concentration level between 0.05 and 1 microg l(-1) under selected ion monitoring acquisition by preconcentrating just 15 ml of sample. Naphthalene-1-sulphonate and naphthalene-2-sulphonate were identified and quantified in one of the samples of seawater.  相似文献

Anatoxin-a is a toxin produced from cyanobacterial blooms in freshwaters. In order to determine trace anatoxin-a in freshwaters, an automated on-line derivatization procedure with fluorenyl methylchloroformate using liquid chromatography-electrospray ionization mass spectrometry was developed. Anatoxin-a was extracted using solid-phase extraction with adequate recovery (75.7+/-7.2%, n=6) at 20 ng/l. The limits of quantification and detection were calculated to be 15.2 ng/l and 2.1 ng/l, respectively, using selected ion monitoring.  相似文献

A highly selective imprinted amino-functionalized silica gel sorbent was prepared by combining a surface molecular imprinting technique with a sol-gel process for on-line solid-phase extraction-HPLC determination of trace pentachlorophenol (PCP) in water samples. The PCP-imprinted amino-functionalized silica sorbent was characterized by FT-IR, SEM, nitrogen adsorption and the static adsorption experiments. The imprinted functionalized silica gel sorbent exhibited high selectivity and offered a fast kinetics for the adsorption and desorption of PCP. The prepared sorbent was shown to be promising for on-line solid-phase extraction for HPLC determination of trace levels of PCP in environmental samples. With a sample loading flow rate of 5 ml min(-1) for 2 min, an enhancement factor of 670 and a detection limit (S/N = 3) of 6 ng l(-1) were achieved at a sample throughput of five samples h(-1). The precision (RSD) for nine replicate on-line sorbent extractions of 10 microgl(-1) PCP was 3.8%. The sorbent also offered good linearity (r = 0.9997) for on-line solid-phase extraction of trace levels of PCP. The method was applied to the determination of PCP in local lake water, river water and wastewater samples.  相似文献

Seven benzene- and naphthalenesulfonates (3-nitrobenzenesulfonate, 4-methylbenzenesulfonate, 1-hydroxy-4-naphthalenesulfonate, 1-amino-7-naphthalenesulfonate, 4-chlorobenzenesulfonate, 1-naphthalenesulfonate and 2-naphthalenesulfonate) were studied. A rapid method for quantifying aromatic sulfonated compounds from waste water samples was developed. This method consists in on-line in-field sampling and monitoring based on ion-pair solid-phase extraction with PLRP-S sorbent, using the new programmable field extraction system and ion-pair liquid chromatography with UV diode-array and electrospray mass spectrometry. Limits of detection for the studied compounds, using the SIM acquisition mode, ranged from 0.01 to 0.33 ng ml(-1). The influence of the aqueous matrix on the on-line SPE was checked by spiking ground and waste waters. Recoveries varied from 70 to 99% when 10 ml of water sample were enriched. The method was applied to the analysis of some environmental sewage samples. This study confirmed that high concentration levels of aromatic sulfonated compounds can be found in sewage samples. In addition, the stability of the seven studied sulfonated benzene and naphthalene compounds was investigated using on-line polymeric SPE pre-columns, based on the styrene-divinylbenzene polymer PLRP-S. Different storage conditions were tested to carry out the stability survey, which included storage at room temperature, at 4 degrees C and at -20 degrees C, during a period of up to 2 weeks. This study showed that the stability of aromatic sulfonic acids on disposable on-line SPE polymeric pre-columns is related to temperature and that the target compounds are more stable at lower temperatures.  相似文献

A high-performance liquid chromatographic (HPLC) method with ultraviolet detection at 278 nm is presented for the determination of 5,10-dideaza-5,6,7,8-tetrahydrofolic acid in plasma. Sample pretreatment was achieved by using cation-exchange solid-phase extraction columns with methotrexate as internal standard. Chromatographic separation was based on ion-pair HPLC with 1-octanesulphonic acid as the ion-pairing compound. The detection limit was 10 ng/ml using an 500-microliters sample volume. The assay was linear from the detection limit up to 5000 ng/ml with good reproducibility. The applicability of the assay was demonstrated in a study in the rat.  相似文献

The ion-pair solid-phase extraction (SPE) of 4-alkylphenols followed by derivatization with pentafluoropyridine is demonstrated. Under alkaline conditions, the 4-alkylphenols could be efficiently adsorbed on a C18 SPE cartridge conditioned with an ion-pair reagent, tetra-n-hexylammonium bromide. The ion pairs, ammonium phenolates, formed on the C18 solid phase, were eluted with a solvent containing the derivatizing reagent, pentafluoropyridine, and completely derivatized during the elution. After optimization of the adsorption and derivatization, we established a method for the determination of the 4-alkylphenols in water samples. The method showed good linearity between 20 and 1000 ng (200-10,000 ng for nonylphenol). By processing 20-ml samples, the method detection limits (MDL) were in the range of 5.2-8.9 ng/l for the 4-alkylphenols (76 ng/l for nonylphenol). To evaluate its applicability to a real aqueous matrix, several river water samples were analyzed.  相似文献

A sensitive method is described for the simultaneous determination of glycyrrhizin and glycyrrhetic acid in human plasma. Quantitation is by high-performance liquid chromatography using ion-pair chromatography on a reversed-phase column. Variable-wavelength ultraviolet detection is employed. For the extraction of both compounds from plasma, a new solid-phase ion-pair extraction procedure using octadecylsilane columns was developed. Because of the strong forces involved in the protein binding of glycyrrhizin, quantitative extraction of this compound from plasma was possible only after an alkaline pH shift. A considerable improvement in selectivity and sensitivity was obtained by automated column switching involving on-line preseparation of the solid-phase extract on a short precolumn and chromatographic analysis of a heart-cut from the precolumn eluate. The limit of detection of both glycyrrhizin and glycyrrhetic acid was 0.1 mg/l in 0.5 ml of plasma. From a preliminary study in human volunteers, it was concluded that glycyrrhetic acid rather than glycyrrhizin is preferred in a study in human volunteers to assess the zero effect level of glycyrrhizin.  相似文献

We have established a robust, fully automated analytical method for the analysis of fluvoxamine in rat plasma using a column-switching ion-pair high-performance chromatography system. The plasma sample was injected onto a precolumn packed with Shim-pack MAYI-ODS (50 microm), where the drug was automatically purified and enriched by on-line solid-phase extraction. After elution of the plasma proteins, the analyte was back-flushed from the precolumn and then separated isocratically on a reversed-phase C18 column (L-column ODS) with a mobile phase (acetonitrile-0.1% phosphoric acid, 36:64, v/v) containing 2 mM sodium 1-octanesulfonate. The analyte was monitored by a UV detector at a wavelength of 254 nm. The calibration line for fluvoxamine showed good linearity in the range of 5-5000 ng/mL (r > 0.999) with the limit of quantification of 5 ng/mL (RSD = 6.51%). Accuracy ranged from -2.94 to 4.82%, and the within- and between-day precision of the assay was better than 8% across the calibration range. The analytical sensitivity and accuracy of this assay is suitable for characterization of the pharmacokinetics of orally-administered fluvoxamine in rats.  相似文献

An analytical procedure based on headspace solid-phase microextraction (SPME) followed by gas chromatography coupled to mass spectrometry in the electron impact mode has been developed for the determination of low-molecular-mass sulfides and disulfides in wastewater. Parameters affecting to the extraction of these volatile alkyl sulfides (VASs) with the SPME, such as the extraction temperature, sample volume, pH and the NaCl addition to the matrix, have been optimised using a polydimethylsiloxane-Carboxen fibre. The linear dynamic range was close to three orders of magnitude for all the studied compounds. Detection limits of 4 ng l(-1) for dimethyl sulfide, 0.7 ng l(-1) for ethylmethyl sulfide, 5 ng l(-1) for diethyl sulfide and 1 ng l(-1) for dimethyl disulfide were achieved, with a relative standard deviation between 4 and 6%. The developed analytical methodology was applied to determine those VASs in different wastewaters.  相似文献

A specific and sensitive method using high-performance liquid chromatography-tandem mass spectrometry (LC-MS-MS) equipped with automatic on-line solid-phase extraction device for the quantitative measurement of anabolic hormone residues, 4-androstene-3,17-dione, testosterone and dihydrotestosterone in cell culture medium was developed. Steroid content in cell culture medium was determined directly without an additional sample preparation step. Separation of analytes from polar endogenous compounds was carried out on an automatic column-switching device coupled with a C4-alkyl-diol silica restricted-access solid-phase extraction column. The lipophilic fraction containing anabolic hormone residues were back-flushed on to a conventional C-18 reversed-phase column for the final chromatography. The analyte was ionized in an ElectroSpray interface under positive ion mode before entering a quadrupole mass analyzer. The lowest points of calibration curves were 0.05 ng ml(-1) for 4-androstene-3,17-dione and testosterone, and 1 ng ml(-1) dihydrotestosterone, respectively. A comparison with results from radioimmunoassay (RIA) is also presented.  相似文献

A procedure was developed for the analysis of estrogens in environmental water and effluents. Samples were extracted by passing through polymer-impregnated solid-phase extraction discs or C18 cartridges, followed by gas chromatography-negative chemical ionisation mass spectrometry of the pentafluorobenzoyl derivatives. The derivatives were stable and gave diagnostic negative molecular ions as the base peak for each of the major estrogens studied. The absolute recovery of estrogens spiked into clean groundwater using the disc procedure was 84-116% at the 10 ng l(-1) level (calculation not based on use of internal standards). Using doubly deuterated estradiol as internal standard added prior to extraction, the % relative standard deviation of estrogen extraction and analysis in spiked groundwater at the 10 ng(-1) level was 2.6-9.8%. Detection limits were 0.2 ng l(-1) or below for the major estrogens, based on a 2.5 litre sample. The most abundant estrogen was estrone, with concentrations over the range 6.4-29 ng l(-1) in effluents, and 0.2 to 17 ng l(-1) in water from the River Thames.  相似文献

Abstract

A reversed-phase HPLC post-column ion-pair extraction system was developed for the analysis of carboxylic acid drugs and their salts using α-(3,4-dimethoxyphenyl)-4′ -trimethylammonium-methylcinnamonitrile methosulfate (DTM) as a new fluorescent ion-pair reagent. The on-line post-column extraction system was optimized with respect to the reagent concentration, extraction coil length and internal diameter, ionic strength of mobile phase, extraction solvent, and the membrane phase separator. Sodium salicylate, ketoprofen, ibuprofen, probenecid, and valproic acid were used as model compounds to evaluate the ion-pair extraction system. The method was applied to pharmaceutical dosage forms containing ketoprofen and valproic acid. Other acidic compounds evaluated using the ion-pair reagent showed that lipophilic acids produced more extractable ion-pairs and higher sensitivities than hydrophilic acids.  相似文献

A new sorbent, maleic acid grafted polytetrafluoroethylene fiber (MA-PTFE), was prepared and evaluated for on-line solid-phase extraction coupled with inductively coupled plasma mass spectrometry (ICP-MS) for fast, selective, and sensitive determination of (ultra)trace rare earth elements (REEs) in environmental samples. The REEs in aqueous samples at pH = 3.0 were selectively extracted onto a microcolumn packed with the MA-PTFE fiber, and the adsorbed REEs were subsequently eluted on-line with 0.9 mol l(-1) HNO3 for ICP-MS determination. The new sorbent extraction system allows effective preconcentration and separation of the REEs from the major matrix constituents of alkali and alkali earth elements, particularly their separation from barium that produces considerable isobaric interferences of 134Ba16O1H+, 135Ba16O+, 136Ba16O1H+, and 137Ba16O+ on 151Eu+ and 153Eu+. With the use of a sample loading flow rate of 7.4 ml min(-1) for 120 s preconcentration, enhancement factors of 69-97 and detection limits (3s) of 1-20 pg l(-1) were achieved at a sample throughput of 22 samples h(-1). The precision (RSD) for 16 replicate determinations of 50 ng l(-1) of REEs was 0.5-1.1%. The developed method was successfully applied to the determination of (ultra)trace REEs in sediment, soil, and seawater samples.  相似文献

An automatic method for determining diuron, irgarol 1051, folpet and dichlofluanid in seawater samples have been developed. This method is based on the on-line coupling of solid-phase extraction (SPE) with a highly crosslinked polymeric sorbent, LiChrolut EN, to liquid chromatography followed by atmospheric pressure chemical ionization (APCI) and mass spectrometry. The operational parameters affecting the APCI interface have been studied in both positive and negative ionization modes. The use of LiChrolut EN in the SPE produced recoveries of over 85% for all the compounds when 100 ml of seawater sample was preconcentrated. Calibration was carried out in both ionization modes and in full-scan and selected-ion monitoring (SIM). The method allowed all the analytes to be detected at 5 ng l(-1) in SIM acquisition mode except folpet, which, because of its low response, could only be detected at 250 ng l(-1). The method was used to analyse water samples taken from five different marina and fishing ports along the coast of Tarragona, Catalonia (Spain), over a 5-month period. Diuron and irgarol 1051 were detected and quantified in most samples at concentration levels ranging from 27 to 420 ng l(-1) for diuron and from 15 to 511 ng l(-1) for irgarol 1051.  相似文献

A bioanalytical method is described that allows the determination of a number of beta-endorphin-related peptides. The method is based on the application of fluorescence detection after high-performance liquid chromatography followed by post-column derivatization with o-phthaldialdehyde. Concentrations exceeding 10-25 ng/ml could be determined by using conventional fluorescence detection, whereas lower concentrations demand the use of laser-induced fluorescence detection. The sample pretreatment includes the use of on-line gel permeation, on-line solid-phase isolation and heart cutting of a peak from reversed-phase gradient elution. The sample pretreatment procedure does not discriminate between the dodecapeptide des-enkaphalin-gamma-endorphin (DE gamma E) and its metabolites in order to obtain similar recoveries for all components. The final chromatographic phase system is based on ion-pair formation, which permits the separation of DE gamma E from its metabolites and degradation products. The optimized procedure allows the determination of these peptides in plasma at concentration levels down to about 1 ng/ml, demanding a sample volume of 1 ml.  相似文献

This work presents a modified method to analyze selected pharmaceutical residues (clofibric acid, ibuprofen, carbamazepine, naproxen, ketoprofen and diclofenac) in water samples. Various solid-phase extraction cartridges were investigated. The newly developed Oasis HLB (polystyrene-divinylbenzene-N-vinyl pyrrolidone terpolymer) solid-phase extraction (SPE) cartridge provides the optimal sample extraction results. The analytes were then identified and quantitatively determined by gas chromatography-mass spectrometry (GC-MS) via on-line derivatization in the injection-port using a large-volume (10 microl) sample injection with tetrabutylammonium (TBA) salts. This injection-port derivatization technique provides sensitivity, fast and reproducible results for pharmaceutical residues analysis. Mass spectra of butylated derivatives and tentative fragmentation profiles are proposed. Molecular ions and some characteristic ions were used as the quantitation ions to obtain maximum detection sensitivity and specificity. The quantitation limits of these compounds ranged from 1.0 to 8.0 ng/l in 500 ml tap water samples. Recovery of these residues in spiked various water samples ranged from 50 to 108% while RSD ranged from 1 to 10%. The selected analytes were detected in concentrations of 30 to 420 ng/l in wastewater treatment plant effluent and river water samples.  相似文献

An on-line solid-phase extraction (SPE) protocol using the cigarette filter as sorbent coupled with high-performance liquid chromatography (HPLC) was developed for simultaneous determination of trace naphthalene (NAPH), phenanthrene (PHEN), anthracene (ANT), fluoranthene (FLU), benzo(b)fluoranthene (BbF), benzo(k)fluoranthene (BkF), benzo(a)pyrene (BaP), and benzo(ghi)perylene (BghiP) in water samples. To on-line interface solid-phase extraction to HPLC, a preconcentration column packed with the cigarette filter was used to replace a conventional sample loop on the injector valve of the HPLC for on-line solid-phase extraction. The sample solution was loaded and the analytes were then preconcentrated onto the preconcentration column. The collected analytes were subsequently eluted with a mobile phase of methanol-water (95:5). HPLC with a photodiode array detector was used for their separation and detection. The detection limits (S/N = 3) for preconcentrating 42 mL of sample solution ranged from 0.9 to 58.6 ng L(-1) at a sample throughput of 2 samples h(-1). The enhancement factors were in the range of 409-1710. The developed method was applied to the determination of trace NAPH, PHEN, ANT, FLU, BbF, BkF, BaP and BghiP in local river water samples. The recoveries of PAHs spiked in real water samples ranged from 87 to 115%. The precisions for nine replicate measurements of a standard mixture (NAPH: 4.0 microg L(-1), PHEN: 0.40 microg L(-1), ANT: 0.40 microg L(-1), FLU: 2.0 microg L(-1), BbF: 1.6 microg L(-1), BkF: 2.0 microg L(-1), BaP: 2.0 microg L(-1), BghiP: 1.7 microg L(-1)) were in the range of 1.2-5.1%.  相似文献

A flow-injection on-line preconcentration-electrothermal atomic absorption spectrometric (ETAAS) method coupled with a coprecipitation method has been developed for the determination of lead in seawater. The combination of two preconcentration procedures, coprecipitation with iron(II) hydroxide and solid-phase extraction with a lead-selective resin, Pb-Spec, allowed the determination of lead at the ng kg(-1) level. Lead in 250 g of a sample solution was collected by coprecipitation with 10 mg of iron. The precipitate was dissolved in 25 ml of 1 mol l(-1) nitric acid; then, a 4-ml aliquot of the sample solution was introduced into the flow-injection system to preconcentrate and separate lead from iron on a Pb.Spec microcolumn. The sorbed lead was eluted with a 1.0 x 10(-4) mol l(-1) EDTA solution. The 30-microl portion of the eluate corresponding to the highest analyte concentration zone was injected into a graphite furnace. The overall enhancement factor was about 200 for 250 g of the sample. The average and standard deviation of ten blank values obtained were 1.7 ng and 0.38 ng, respectively. The recovery was 93.7 +/- 5.0% for seawater spiked with 20 ng kg(-1) lead. The proposed method is applicable to the analysis of seawater for lead at slightly higher levels.  相似文献

A fully automated analyzer for methylated L-arginine metabolites [N,N-dimethyl-L-arginine (ADMA), N-methylarginine (NMMA) and N,N'-dimethyl-L-arginine (SDMA)] by high-performance liquid chromatography with post-column fluorescence derivatization was developed. This system consists of an on-line extraction, a separation on a reversed phase ion-pair chromatograph, a post-column derivatization by o-phthaladehyde (OPA) and thiol reaction, and fluorescence detection. NMMA, ADMA and SDMA were separated in 40 min with isocratic elution by a combination of octanoate and cyclohexane carboxylate as ion-pair reagents. The eluate was monitored at 450 nm with excitation at 337 nm. The calibration curves for NMMA, ADMA and SDMA showed linearity over the range from 0.05 micromol l(-1) (0.5 pmol on column) to 5.0 micromol l(-1) (50 pmol on column). This method does not require any time-consuming pre-treatment and requires only 10 microl of plasma sample for assay.  相似文献

A common sample preparation procedure capable of efficiently concentrating various groups of pesticides, taking advantage of universal detectors like the mass spectrometer or combined techniques of group selective detectors like gas chromatography-electron capture detection (ECD)/thermionic specific detection (TSD), is desirable in environmental analysis. Six solid-phase microextraction fibres available for analysis of semi-volatiles (7, 30 and 100 microm poly(dimethylsiloxane) (PDMS), 85 microm polyacrylate, 60 microm PDMS-divinylbenzene (PDMS-DVB) and 65 microm Carbowax-DVB) were evaluated and the 60 microm PDMS-DVB was selected for the simultaneous extraction of 34 compounds, included in the organochlorine (OCPs), organophosphorous (OPPs), pyrethroid and triazine pesticide groups. All parameters affecting the extraction efficiency from water samples, namely fibre coating, sample agitation, pH and ionic strength, extraction temperature and time, were optimised. The analytical procedure involves solid-phase microextraction extraction, gas chromatographic separation and subsequent ECD and TSD via a post-column splitter adjusted to a split ratio of 1:10, respectively. Detection limits in the range of 1-10 ng l(-1) for OCPs, 1-30 ng l(-1) for OPPs, 20-30 ng l(-1) for pyrethroids and 8-50 ng l(-1) for triazines are easily attainable with the optimised procedure. The method validated for ground and drinking waters has low cost of implementation and operation although it requires careful maintenance.  相似文献
