Organogels that are self‐assembled from simple peptide molecules are an interesting class of nano‐ and mesoscale soft matter with simplicity and functionality. Investigating the precise roles of the organic solvents and their effects on stabilization of the formed organogel is an important topic for the development of low‐molecular‐weight gelators. We report the structural transition of an organogel self‐assembled from a single dipeptide building block, diphenylalanine (L ‐Phe‐L ‐Phe, FF), in toluene into a flower‐like microcrystal merely by introducing ethanol as a co‐solvent; this provides deeper insights into the phase transition between mesostable gels and thermodynamically stable microcrystals. Multiple characterization techniques were used to reveal the transitions. The results indicate that there are different molecular‐packing modes formed in the gels and in the microcrystals. Further studies show that the co‐solvent, ethanol, which has a higher polarity than toluene, might be involved in the formation of hydrogen bonds during molecular self‐assembly of the dipeptide in mixed solvents, thus leading to the transition of organogels into microcrystals. The structural transformation modulated by the co‐solvent might have a potential implication in controllable molecular self‐assembly.  相似文献

The incorporation of the β‐amino acid residues into specific positions in the strands and β‐turn segments of peptide hairpins is being systematically explored. The presence of an additional torsion variable about the C(α) C(β) bond (θ) enhances the conformational repertoire in β‐residues. The conformational analysis of three designed peptide hairpins composed of α/β‐hybrid segments is described: Boc‐Leu‐Val‐Val‐DPro‐β Phe ‐Leu‐Val‐Val‐OMe ( 1 ), Boc‐Leu‐Val‐β Val ‐DPro‐Gly‐β Leu ‐Val‐Val‐OMe ( 2 ), and Boc‐Leu‐Val‐β Phe ‐Val‐DPro‐Gly‐Leu‐β Phe ‐Val‐Val‐OMe ( 3 ). 500‐MHz 1H‐NMR Analysis supports a preponderance of β‐hairpin conformation in solution for all three peptides, with critical cross‐strand NOEs providing evidence for the proposed structures. The crystal structure of peptide 2 reveals a β‐hairpin conformation with two β‐residues occupying facing, non‐H‐bonded positions in antiparallel β‐strands. Notably, βVal(3) adopts a gauche conformation about the C(α) C(β) bond (θ=+65°) without disturbing cross‐strand H‐bonding. The crystal structure of 2 , together with previously published crystal structures of peptides 3 and Boc‐β Phe ‐β Phe ‐DPro‐Gly‐β Phe ‐β Phe ‐OMe, provide an opportunity to visualize the packing of peptide sheets with local ‘polar segments' formed as a consequence of reversal peptide‐bond orientation. The available structural evidence for hairpins suggests that β‐residues can be accommodated into nucleating turn segments and into both the H‐bonding and non‐H‐bonding positions on the strands.  相似文献

Cyclic D,L ‐α‐peptides are able to self‐assemble to nanotubes, although the inherent reason of the stability of this kind of nanotube as well as the intrinsic driving force of self‐assembly of the cyclic D ,L ‐α‐peptides still remain elusive. In this work, using several computational approaches, we investigated the structural and energy characteristics of a series of cyclo[(‐L ‐Phe‐D ‐Ala‐)4] and cyclo[(‐L ‐Ala‐D ‐Ala‐)4] oligomers. The results reveal that the thermodynamic stability, cooperativity, and self‐assembly patterns of cyclic D ,L ‐α‐peptide nanotubes are mainly determined by the interactions between cross‐strand side chains instead of those between backbones. For cyclo[(‐L ‐Phe‐D ‐Ala‐)4] oligomers, the steric interaction between cross‐strand side chains, especially the electrostatic repulsion between the phenyls in Phe residues, brings anticooperative effect into parallel stacking mode, which is responsible for the preference of self‐assembling nanotube in antiparallel vs. parallel stacking orientation. Based on our results, a novel self‐assembling mechanism is put forward—it is the L ‐L antiparallel dimer of cyclo[(‐L ‐Phe‐D ‐Ala‐)4], instead of the commonly presumed monomer, that acts as the basic building block in self assembly. Understanding the roles of various parameters in orchestrating the preferential chiral molecular organization in supramolecular self‐assembly processes is of great significance in designing novel molecular functional systems. Cyclic dipeptide (CDP) chiral auxiliary‐functionalized naphthalenediimides (NCDPs 1 – 6 ) have been prepared and their chiral self‐assembly properties have been investigated. Detailed photophysical and circular dichroism (CD) studies have unveiled the crucial role of the solvent in the chiral aggregation of these NCDPs. NCDPs 1 – 3 form supramolecular helical assemblies and exhibit remarkable chiroptical switching behaviour (M‐ to P‐type) depending on the solvent composition of HFIP and DMSO. The strong influence of solvent composition on the supramolecular chirality of NCDPs has been further corroborated by concentration and solid‐state thin‐film CD studies. The chiroptical switching between supramolecular aggregates of opposite helicity (M and P) has been found to be reversible, and can be achieved through cycles of solvent removal and redissolution in solvent mixtures of specific composition. The control molecular systems (NCDPs 4 – 6 ), with an achiral or D ‐isomer second amino acid in the CDP auxiliary, did not show chiral aggregation properties. The substantial roles of hydrogen bonding and π–π interactions in the assembly of the NCDPs have been validated through nuclear magnetic resonance (NMR), photophysical, and computational studies. Quantum chemical calculations at the ab initio, semiempirical, and density functional theory levels have been performed on model systems to understand the stabilities of the right (P‐) and left (M‐) handed helical supramolecular assemblies and the nature of the intermolecular interactions. This study emphasizes the role of CDP chiral auxiliaries on the solvent‐induced helical assembly and reversible chiroptical switching of naphthalenediimides.  相似文献

New poly(dimethylsiloxane)‐based polymer organogelators with L ‐lysine derivatives were synthesized on the basis of synthetically simple procedure, and their organogelation abilities were investigated. These polymer organogelators have a good organogelation ability and form organogels in many organic solvents. In the organogels, polymer gelators constructed a mesoporous structure with a pore size of about 1 μm formed by entanglement of the self‐assembled nanofibers. The L ‐lysine derivatives in the polymer gelators functioned as a gelation‐causing segment and the organogelation was induced by self‐assembly of the L ‐lysine segments through a hydrogen bonding interaction. © 2006 Wiley Periodicals, Inc. The self‐assembly of head‐tail type block copolymers composed of polyamidoamine dendron head block and poly(L ‐lysine) (PLL) tail block was studied using a light scattering technique and transmission electron microscopy. A PLL tail block in a head‐tail type block copolymer exhibits a coil‐to‐helix transition as a result of the change in solvent quality from water to methanol. When the PLL tail block takes a helical conformation in high methanol content, the resulting head‐tail type block copolymer has a defined three‐dimensional structure like that of a protein molecule. Self‐assemblies of such block copolymers having a totally fixed molecular shape spontaneously form polymersome‐like self‐assemblies with an extremely narrow size distribution through converging to a thermodynamically stable assembling state. © 2009 Wiley Periodicals, Inc. Hierarchical self‐assembly of disubstituted ferrocene (Fc)–peptide conjugates that possess Gly‐Val‐Phe and Gly‐Val‐Phe‐Phe peptide substituents leads to the formation of nano‐ and micro‐sized assemblies. Hydrogen‐bonding and hydrophobic interactions provide directionality to the assembly patterns. The self‐assembling behavior of these compounds was studied in solution by using 1H NMR and circular dichroism (CD) spectroscopies. In the solid state, attenuated total reflectance (ATR) FTIR spectroscopy, single‐crystal X‐ray diffraction (XRD), powder X‐ray diffraction (PXRD), and scanning electron microscopy (SEM) methods were used. Spontaneous self‐assembly of Fc–peptides through intra‐ and intermolecular hydrogen‐bonding interactions induces supramolecular assemblies, which further associate and give rise to fibers, large fibrous crystals, and twisted ropes. In the case of Fc[CO‐Gly‐Val‐Phe‐OMe]2 ( 1 ), molecules initially interact to form pleated sheets that undergo association into long fibers that form bundles and rectangular crystalline cuboids. Molecular offsets and defects, such as screw dislocations and solvent effects that occur during crystal growth, induce the formation of helical arrangements, ultimately leading to large twisted ropes. By contrast, the Fc–tetrapeptide conjugate Fc[CO‐Gly‐Val‐Phe‐Phe‐OMe]2 ( 2 ) forms a network of nanofibers at the supramolecular level, presumably due to the additional hydrogen‐bonding and hydrophobic interactions that stem from the additional Phe residues.  相似文献

This contribution describes the synthesis and ring‐opening (co)polymerization of several L ‐lysine N‐carboxyanhydrides (NCAs) that contain labile protective groups at the ?‐NH2 position. Four of the following L ‐lysine NCAs were investigated: N?‐trifluoroacetyl‐L ‐lysine N‐carboxyanhydride, N?‐(tert‐butoxycarbonyl)‐L ‐lysine N‐carboxyanhydride, N?‐(9‐fluorenylmethoxycarbonyl)‐L ‐lysine N‐carboxyanhydride, and N?‐(6‐nitroveratryloxycarbonyl)‐L ‐lysine N‐carboxyanhydride. In contrast to the harsh conditions that are required for acidolysis of benzyl carbamate moieties, which are usually used to protect the ?‐NH2 position of L ‐lysine during NCA polymerization, the protective groups of the L ‐lysine NCAs presented here can be removed under mildly acidic or basic conditions or by photolysis. As a consequence, these monomers may allow access to novel peptide hybrid materials that cannot be prepared from ?‐benzyloxycarbonyl‐L ‐lysine N‐carboxyanhydride (Z‐Lys NCA) because of side reactions that accompany the removal of the Z groups. By copolymerization of these L ‐lysine NCAs with labile protective groups, either with each other or with γ‐benzyl‐L ‐glutamate N‐carboxyanhydride or Z‐Lys NCA, orthogonally side‐chain‐protected copolypeptides with number‐average degrees of polymerization ≤20 were obtained. Such copolypeptides, which contain different side‐chain protective groups that can be removed independently, are interesting for the synthesis of complex polypeptide architectures or can be used as scaffolds for the preparation of synthetic antigens or protein mimetics. © 2003 Wiley Periodicals, Inc. Although the role of intermolecular aromatic π–π interactions in the self‐assembly of di‐l ‐phenylalanine (l ‐Phe‐l ‐Phe, FF), a peptide that is known for hierarchical structure, is well established, the influence of intramolecular π–π interactions on the morphology of the self‐assembled structure of FF has not been studied. Herein, the role of intramolecular aromatic π–π interactions is investigated for FF and analogous alanine (Ala)‐containing dipeptides, namely, l ‐Phe‐l ‐Ala (FA) and l ‐Ala‐l ‐Phe (AF). The results reveal that these dipeptides not only form self‐assemblies, but also exhibit remarkable differences in structural morphology. The morphological differences between FF and the analogues indicate the importance of intramolecular π–π interactions, and the structural difference between FA and AF demonstrates the crucial role of the nature of intramolecular side‐chain interactions (aromatic–aliphatic or aliphatic–aromatic), in addition to intermolecular interactions, in deciding the final morphology of the self‐assembled structure. The current results emphasise that intramolecular aromatic π–π interaction may not be essential to induce self‐assembly in smaller peptides, and π (aromatic)–alkyl or alkyl–π (aromatic) interactions may be sufficient. This work also illustrates the versatility of aromatic and a combination of aromatic and aliphatic residues in dipeptides in the formation of structurally diverse self‐assembled structures.  相似文献

α‐Methyl‐L ‐proline is an α‐substituted analog of proline that has been previously employed to constrain prolyl peptide bonds in a trans conformation. Here, we revisit the cistrans prolyl peptide bond equilibrium in derivatives of α‐methyl‐L ‐proline, such as N‐Boc‐protected α‐methyl‐L ‐proline and the hexapeptide H‐Ala‐Tyr‐αMePro‐Tyr‐Asp‐Val‐OH. In Boc‐α‐methyl‐L ‐proline, we found that both cis and trans conformers were populated, whereas, in the short peptide, only the trans conformer was detected. The energy barrier for the cistrans isomerization in Boc‐α‐methyl‐L ‐proline was determined by line‐shape analysis of NMR spectra obtained at different temperatures and found to be 1.24 kcal/mol (at 298 K) higher than the corresponding value for Boc‐L ‐proline. These findings further illuminate the conformationally constraining properties of α‐methyl‐L ‐proline.  相似文献

The design and synthesis of β‐peptides from new C‐linked carbo‐β‐amino acids (β‐Caa) presented here, provides an opportunity to understand the impact of carbohydrate side chains on the formation and stability of helical structures. The β‐amino acids, Boc‐(S)‐β‐Caa(g)‐OMe 1 and Boc‐(R)‐β‐Caa(g)‐OMe 2 , having a D ‐galactopyranoside side chain were prepared from D ‐galactose. Similarly, the homo C‐linked carbo‐β‐amino acids (β‐hCaa); Boc‐(S)‐β‐hCaa(x)‐OMe 3 and Boc‐(R)‐β‐hCaa(x)‐OMe 4 , were prepared from D ‐glucose. The peptides derived from the above monomers were investigated by NMR, CD, and MD studies. The β‐peptides, especially the shorter ones obtained from the epimeric (at the amine stereocenter Cβ) 1 and 2 by the concept of alternating chirality, showed a much smaller propensity to form 10/12‐helices. This substantial destabilization of the helix could be attributed to the bulkier D ‐galactopyranoside side chain. Our efforts to prepare peptides with alternating 3 and 4 were unsuccessful. However, the β‐peptides derived from alternating geometrically heterochiral (at Cβ) 4 and Boc‐(R)‐β‐Caa(x)‐OMe 5 (D ‐xylose side chain) display robust right‐handed 10/12‐helices, while the mixed peptides with alternating 4 and Boc‐β‐hGly‐OMe 6 (β‐homoglycine), resulted in left‐handed β‐helices. These observations show a distinct influence of the side chains on helix formation as well as their stability.  相似文献

Two new series of Boc‐N‐α,δ‐/δ,α‐ and β,δ‐/δ,β‐hybrid peptides containing repeats of L ‐Ala‐δ5‐Caa/δ5‐Caa‐L ‐Ala and β3‐Caa‐δ5‐Caa/δ5‐Caa‐β3‐Caa (L ‐Ala = L ‐alanine, Caa = C‐linked carbo amino acid derived from D ‐xylose) have been differentiated by both positive and negative ion electrospray ionization (ESI) ion trap tandem mass spectrometry (MS/MS). MSn spectra of protonated isomeric peptides produce characteristic fragmentation involving the peptide backbone, the Boc‐group, and the side chain. The dipeptide positional isomers are differentiated by the collision‐induced dissociation (CID) of the protonated peptides. The loss of 2‐methylprop‐1‐ene is more pronounced for Boc‐NH‐L ‐Ala‐δ‐Caa‐OCH3 (1), whereas it is totally absent for its positional isomer Boc‐NH‐δ‐Caa‐L ‐Ala‐OCH3 (7), instead it shows significant loss of t‐butanol. On the other hand, second isomeric pair shows significant loss of t‐butanol and loss of acetone for Boc‐NH‐δ‐Caa‐β‐Caa‐OCH3 (18), whereas these are insignificant for its positional isomer Boc‐NH‐β‐Caa‐δ‐Caa‐OCH3 (13). The tetra‐ and hexapeptide positional isomers also show significant differences in MS2 and MS3 CID spectra. It is observed that ‘b’ ions are abundant when oxazolone structures are formed through five‐membered cyclic transition state and cyclization process for larger ‘b’ ions led to its insignificant abundance. However, b1+ ion is formed in case of δ,α‐dipeptide that may have a six‐membered substituted piperidone ion structure. Furthermore, ESI negative ion MS/MS has also been found to be useful for differentiating these isomeric peptide acids. Thus, the results of MS/MS of pairs of di‐, tetra‐, and hexapeptide positional isomers provide peptide sequencing information and distinguish the positional isomers. The self‐assembly and gelation behavior of a series of mono‐ and disubstituted ferrocene (Fc)–peptide conjugates as a function of ferrocene conformation and amino acid chirality are described. The results reveal that ferrocene–peptide conjugates self‐assemble into organogels by controlling the conformation of the central ferrocene core, through inter‐ versus intramolecular hydrogen bonding in the attached peptide chain(s). The chirality controlled assembling studies showed that two monosubstituted Fc conjugates FcCO–L FL FL A‐OMe and FcCO–L FL FD A‐OMe form gels with nanofibrillar network structures, whereas the other two diastereomers FcCO–D FL FL A‐OMe and FcCO–L FD FL A‐OMe exclusively produced straight nanorods and non‐interconnected small fibers, respectively. This suggests the potential tuning of gelation behavior and nanoscale morphology by altering the chirality of constituted amino acids. The current study confirms the profound effect of diastereomerism and no influence of enantiomers on gelation. Correspondingly, the diastereomeric and enantiomeric Fc[CO‐FFA‐OMe]2 were constructed for the study of chirality‐organized structures.  相似文献

The NCN‐pincer Pd‐complex‐bound norvalines Boc‐D /L ‐[PdCl(dpb)]Nva‐OMe ( 1 ) were synthesized in multigram quantities. The molecular structure and absolute configuration of 1 were unequivocally determined by single‐crystal X‐ray structure analysis. The robustness of 1 under acidic/basic conditions provides a wide range of N‐/C‐terminus convertibility based on the related synthetic transformations. Installation of a variety of functional groups into the N‐/C‐terminus of 1 was readily carried out through N‐Boc‐ or C‐methyl ester deprotection and subsequent condensations with carboxylic acids, R1COOH, or amines, R2NH2, to give the corresponding N‐/C‐functionalized norvalines R1‐D /L ‐[PdCl(dpb)]Nva‐R2 2 – 9 . The dipeptide bearing two Pd units 10 was successfully synthesized through the condensation of C‐free 1 with N‐free 1 . The robustness of these Pd‐bound norvalines was adequately demonstrated by the preservation of the optical purity and Pd unit during the synthetic transformations. The lipophilic Pd‐bound norvalines L ‐ 2 , Boc‐L ‐[PdCl(dpb)]Nva‐NH‐n‐C11H23, and L ‐ 4 , n‐C4H9CO‐L ‐[PdCl(dpb)]Nva‐NH‐n‐C11H23, self‐assembled in aromatic solvents to afford supramolecular gels. The assembled structures in a thermodynamically stable single crystal of L ‐ 2 and kinetically stable supramolecular aggregates of L ‐ 2 were precisely elucidated by cryo‐TEM, WAX, SAXS, UV/Vis, IR analyses, and single‐crystal X‐ray crystallography. An antiparallel β‐sheet‐type aggregate consisting of an infinite one‐dimensional hydrogen‐bonding network of amide groups and π‐stacking of PdCl(dpb) moieties was observed in the supramolecular gel fiber of L ‐ 2 , even though discrete dimers are assembled through hydrogen bonding in the thermodynamically stable single crystal of L ‐ 2 . The disparate DSC profiles of the single crystal and xerogel of L ‐ 2 indicate different thermodynamics of the molecular assembly process.  相似文献

Dipeptide diphenylalanine has attracted significant research interests because of its ability to self‐assemble into various nanostructures such as nanotubes, nanowires, and nanoribbons. In this article, we present the synthesis and self‐assembly of a novel diphenylalanine‐based homopolymer and block/random copolymers by the reversible addition–fragmentation chain transfer (RAFT) polymerization of an acrylamide having a dipeptide moiety. The RAFT polymerization of N‐acryloyl‐l ,l ‐diphenylalanine (A‐Phe‐Phe‐OH) afforded novel amino acid‐based polymers with predetermined molecular weights and relatively narrow‐molecular weight distributions. The hierarchical self‐assembled structures of poly(A‐Phe‐Phe‐OH), which involve nanorods, larger nanofiber‐like microcrystals, and fiber bundles, were characterized by atomic force microscopy (AFM), transmission electron microscopy, scanning electron microscopy, and dynamic light scattering measurements. The circular dichroic measurements of poly(A‐Phe‐Phe‐OH) revealed its characteristic chiroptical property, which is affected by the nature of the solvents and the addition of urea and salts via hydrophobic, hydrogen bonding, and electrostatic interactions. Thermo‐ and pH‐responsive block and random copolymers composed of A‐Phe‐Phe‐OH and N‐isopropylacrylamide were synthesized by RAFT polymerization, and the thermoresponsive properties and assembled structures of the resulting copolymers were investigated by AFM and turbidity measurements. © 2019 Wiley Periodicals, Inc. This paper describes the peculiar co‐assembly supramolecular polymerization behavior of triphenylamine trisamide derivatives with d ‐alanine ( T‐ala ) or glycine ( T‐gly ) moieties. Concentration and temperature‐dependent circular dichroism (CD) spectroscopy revealed that the heating curves of co‐assemblies obtained at various molar ratios of T‐ala to T‐gly exhibited two distinct transition temperatures. The first transition was due to the transformation from coiled helical bundles to single helical fibers without handedness. The second was due to a change from typical elongation to nucleation. These phenomena were confirmed by solvent‐dependent decoiling of coiled helical structures and concentration‐dependent morphological analysis. The two transitioning temperatures were dependent on the concentration of T‐ala in the co‐assemblies, suggesting that T‐ala concentration plays an important role in the formation of coiled helical bundles. Our study demonstrated the first observation of two distinct transition temperatures in supramolecular polymers.  相似文献

The N‐terminal nonapeptide domain of the fungal nonribosomal peptide antibiotics cephaibol A and cephaibol C (AcPheAib4LeuIvaGly‐ Aib) is reported to adopt a right‐handed helical conformation in the crystalline state. However, this conformation is at odds with the left‐handed helicity observed in solution in related synthetic oligomers capped with Ac‐L ‐PheAib4 fragments. We report the synthesis of four diastereoisomers of the cephaibol N‐terminal nonapeptide, and show by NMR and CD spectroscopy that the peptide containing the chiral amino acids Phe and Leu in the naturally occurring relative configuration exists in solution as an interconverting mixture of helical screw‐sense conformers. In contrast, the nonapeptide containing the unnatural relative configuration at Phe and Leu adopts a single, stable helical screw‐sense, which is left handed when the N‐terminal Phe residue is L and right‐handed when the N‐terminal Phe residue is D .  相似文献

The known solid‐state structure (Fig. 1, top) of cyclo(β‐HAla)4 was used to model the structure of the title compound 1 as a prospective somatostatin mimic (Fig. 1, bottom). The synthesis started with the N‐protected natural amino acids Boc‐Phe‐OH, Boc‐Trp‐OH, Boc‐Lys(2‐Cl‐Z)‐OH, and Boc‐Thr(OBn)‐OH, which were homologated to the corresponding β‐amino‐acid derivatives (Scheme 1) and coupled to the β‐tetrapeptide Boc‐β‐HTrp‐β‐HPhe‐β‐HThr(OBn)‐β‐HLys(2‐Cl‐Z)‐OMe ( 16 ); the (N‐Me)‐β‐HThr‐(N‐Me)‐β‐HPhe analog 17 was also prepared. C‐ and N‐terminal deprotection and cyclization through the pentafluorophenyl ester gave the insoluble β‐tetrapeptide with protected Thr and Lys side chains ( 18 ). Solubilization and debenzylation could only be effected in LiCl‐containing THF (ca. 10% yield; with ca. 55% recovery). HPLC Purification provided a sample of the title compound 1 , the structure of which, as determined by NMR‐spectroscopy (Fig. 2, left) was drastically different from the `theoretical' model (Fig. 1). There is a transannular H‐bond dividing the macrocyclic 16‐membered ring, thus forming a ten‐ and a twelve‐membered H‐bonded ring, the former mimicking, or actually being superimposable on, an α‐peptidic so‐called β‐turn. Still, the four side chains occupy equatorial positions on the ring, as planned, albeit with somewhat different geometry as compared to the `original'. The cycloβ‐tetrapeptide has micromolar affinities to the human somatostatin receptors (hsst 1 – 5). Thus, we have demonstrated for the first time that it is possible to mimic a natural peptide hormone with a small β‐peptide. Furthermore, we have discovered a simple way to construct the ubiquitous β‐turn motif with β‐peptides (which are known to be stable to mammalian peptidases).  相似文献

Single‐crystal X‐ray diffraction studies of two terminally protected tetrapeptides Boc‐Ile‐Aib‐Val‐m‐ABA‐OMe ( I ) and Boc‐Ile‐Aib‐Phe‐m‐ABA‐OMe ( II ) (Aib=α‐aminoisobutyric acid; m‐ABA=meta‐aminobenzoic acid) reveal that they form continuous H‐bonded helices through the association of double‐bend (type III and I) building blocks. NMR Studies support the existence of the double‐bend (type III and I) structures of the peptides in solution also. Field emission scanning electron‐microscopic (FE‐SEM) and high‐resolution transmission electron‐microscopic (HR‐TEM) images of the peptides exhibit amyloid‐like fibrils in the solid state. The Congo red‐stained fibrils of peptide I and II , observed between crossed polarizers, show green‐gold birefringence, a characteristic of amyloid fibrils.  相似文献

Diastereoselective C3‐elongation processes of N‐Boc‐, N‐Z‐, N‐Bn‐N‐Boc‐, and N‐Bn‐N‐Z‐L ‐alaninals (Boc=tBuOCO, Z=PhCH2OCO, Bn=PhCH2) using various allyl reagents, such as allyl bromide in the presence of Zn/aqueous NH4Cl solution, of SnCl2⋅2 H2O/NaI or of Mg/CuCl2⋅2 H2O, as well as allyltrichlorosilane, are described. A substantially different influence of the N‐protecting groups replacing either one or two amino protons was observed, allowing the selective synthesis of either the syn‐ or anti‐diastereoisomer as a major product.  相似文献
