In this study, three kinds of L ‐lactide‐based copolymers, poly(lactide‐co‐glycolide) (PLGA), poly(lactide‐co‐p‐dioxanone) (PLDON) and poly(lactide‐co‐caprolactone) (PLC), were synthesized by the copolymerization of L ‐lactide (L) with glycolide (G), or p‐dioxanone (DON) or ε‐caprolactone (CL), respectively. The copolymers were easily soluble in common organic solvents. The compositions of the copolymers were determined by 1H‐NMR. Thermal/mechanical and shape‐memory properties of the copolymers with different comonomers were compared. Moreover, the effect of the chain flexibility of the comonomers on thermal/mechanical and shape‐memory properties of the copolymers were investigated. The copolymers with appropriate lactyl content showed good shape‐memory properties where both the shape fixity rate (Rf)and the shape recovery rate (Rr) could exceed 95%. It was found that the comonomers with different flexible molecular chain have different effects on their thermal/mechanical and shape‐memory properties. Among them, PLGA has the highest mechanical strength and recovery rate while PLC copolymer has high recovery rate when the lactyl content exceeded 85% and the lowest transition temperature (Ttrans). A series of copolylactones was synthesized by ring‐opening copolymerization of glycolide, L ‐lactide and ?‐caprolactone, using stannous octoate as catalyst. The in vitro degradation behaviors of them were studied and data demonstrated different degradation rates which mainly depended on the compositions. Investigation of the 5‐fluorouracil (5‐Fu) release from these copolylactones revealed that the composition, degradation rate and the morphology of the polymeric matrix played an important role on the drug release kinetics. A sustained 5‐Fu release without initial time lag was obtained from random poly(lactide‐co‐glycolide‐co‐caprolactone) (r‐PGLC) drug carrier, and it differed from the cases of polylactide (PLA) or random poly(lactide‐co‐glycolide) (PLGA), which usually showed an initial time lag or biphasic drug release behavior. It was due to the low glass transition temperature (T g) of the r‐PGLC and the drug would diffuse faster in rubbery state under the experimental temperature. Furthermore, a significant change in the drug release behavior of r‐PGLC was observed when the temperatures were changed around the T g of the drug carrier, which implied that the drug release behavior could be regulated by adjusting the morphology of the drug carrier. Environmental switches may be fabricated for the controlled release of pharmaceutical drug using a thermally responsive polymer with the intrinsic chemical and physical nature of stimuli‐sensitive smart materials. Particularly, much attention has been paid to the biomedical applications of poly(N‐isopropyl acrylamide) (PNIPAAm) because of its unique reversible transition at a specific lower critical solution temperature (LCST).Thermally sensitive block copolymers, poly(N‐isopropyl acrylamide‐b‐poly(L ‐lactide‐co‐glycolide) (PNIPAAm‐b‐PLGA), and polyethylene glycol‐poly (lactide‐co‐glycolide) (PEG‐PLGA) triblock copolymers with different compositions and length of PLGA block were synthesized via ring‐opening polymerization of lactide and glycolide in the presence of OH‐terminated PNIPAAm or PEG. The composition and structure of the polymer were determined by NMR and FTIR. The effect of important factors, such as ionic strength, pH, and polymer concentration on the phase transition behavior of temperature‐sensitive polymers, were investigated by cloud point measurements. The resulting thermosensitive polymers were used for the entrapment of a narcotic antagonist drug, naltrexone, as the model drug. The loading efficiency and drug release behavior of naltrexone‐loaded hydrogels were investigated. The naltrexone loaded thermosensitive polymers were able to sustain the release of naltrexone for different periods of time, depending on the polymer composition, and concentration. In vitro release studies showed that these thermosensitive polymers are able to deliver naltrexone in biologically active forms at a controlled rate for 3–8 weeks. Novel poly(lactide‐co‐glycolide) (PLGA)/polycaprolactone (PCL) ABA block copolymers were synthesized by bulk copolymerization of glycolide and lactide with PCL diols prepolymer using stannous octoate as catalyst. The resulting copolymers were characterized by various analytical techniques including gel permeation chromatography, IR, 1H nuclear magnetic resonance, differential scanning calorimeter and X‐ray diffractometry. Mechanical properties and hydrophilicity of the copolymers were also studied. Data showed that the copolymers presented a part‐regular structure, containing both PCL crystalline and amorphous PLGA domains. In this study, polymeric (MW 50 000) and oligomeric (MW 2000) poly (lactic acid) (PLA), both with and without end‐group deuterium exchange, were analysed using static secondary ion mass spectrometry (SSIMS) to investigate the contribution of end‐group‐derived secondary ions to the SSIMS spectra. By monitoring the SSIMS intensities between the non‐deuterated and deuterated PLA, it is evident that the only significant end‐group‐derived secondary ions are [nM + H]+ (n > 1) and C4H9O2+. The gentle‐SIMS (G‐SIMS) methodology was employed to establish that deuterated fragments were produced through low energy processes and were not the result of substantial rearrangements. It was noted that end‐group‐derived secondary ions had higher G‐SIMS intensities for oligomeric PLA than polymeric PLA, showing that these secondary ions are simple fragment products that are not the result of rearrangement or degraded product ions. A new hyperbranched poly (amine‐ester)‐poly (lactide‐co‐glycolide) copolymer (HPAE‐co‐PLGA) was synthesized by ring‐opening polymerization of D , L ‐lactide (DLLA) glycolid and branched poly (amine‐ester) (HPAE‐OHs) with Sn(Oct)2 as catalyst. The chemical structures of copolymers were determined by FT‐IR, 1H‐NMR(13C NMR), TGA and their molecular weights were determined by gel permeation chromatography (GPC). Paclitaxel‐loaded copolymer nanoparticles were prepared by the nanoprecipitation method. Their physicochemical characteristics, e.g. morphology and nanoparticles size distribution were then evaluated by means of fluorescence spectroscopy, environmental scanning electron microscopy (ESEM), and dynamic light scattering (DLS). Paclitaxel‐loaded nanoparticles assumed a spherical shape and have unimodal size distribution. It was found that the chemical composition of the nanoparticles was a key factor in controlling nanoparticles size, drug‐loading content, and drug release behavior. As the molar ratio of DL ‐lactide/glycolide to HPAE increased, the nanoparticles size and drug‐loading content increased, and the drug release rate decreased. The antitumor activity of the paclitaxel‐loaded HPAE‐co‐PLGA nanoparticles against human liver cancer H7402 cells was evaluated by 3‐(4, 5‐dimethylthiazol‐2‐yl)‐2,5‐diphenyl tetrazolium bromide (MTT) method. Poly(D,L‐lactide‐co‐glycolide) 50:50 (PLGA)/graphene oxide (GO) nanocomposite films were prepared with various GO weight fractions. A significant enhancement of mechanical properties of the PLGA/GO nanocomposite films was obtained with GO weight fractions. The incorporation of only 5 wt% of GO resulted in an ~2.5‐fold and ~4.7‐fold increase in the tensile strength and Young's modulus of PLGA, respectively. The thermomechanical behaviors of composite films were investigated by dynamic mechanical analysis. Results indicated that the values of Tg and storage moduli of the PLGA/GO composites were higher than those of the pristine PLGA. The improvement in oxygen barrier properties of composites was presumably attributed to the filler effect of the randomly dispersed GO throughout the PLGA matrix. In this work, we also studied in vitro biodegradation behavior. PLGA/GO composite films were hydrolyzed at 37°C for periods up to 49 days. Poly(lactide‐co‐glycolide) (PLGA) copolymers are a kind of biocompatible and biodegradable materials being widely used in tissue engineering. However, phase separation had not been reported successfully in fabricating these amorphous polymers into nanofibrous matrix, although this technique had shown advantages over electrospinning in producing a nanofiber network. In this study, tetrahydrofuran (THF)/H2O solvent pairs were found suitable solvents to induce the formation of uniform PLGA gel at selected gelation temperatures. The results indicated that fine nanofibrous structures with fiber diameter around 40–60 nm could be obtained following the steps of gel formation, solvent extraction, and freeze‐drying, by controlling the concentration of PLGA/THF/H2O solution, THF/H2O ratio, and gelation temperature. Biodegradable polyesters were synthesized via an emulsion polymerization in supercritical carbon dioxide (SC‐CO2). Copolymers of lactide and glycolide were synthesized in SC‐CO2 with stannous octoate as the ring‐opening catalyst and a fluorocarbon polymer surfactant as an emulsifying agent. The conversion of lactide and glycolide was monitored with respect to the reaction time and temperature with 1H NMR spectroscopy. The conversion of glycolide surpassed 99% within 72 h for an SC‐CO2 phase maintained at 200 bar and 70 °C. Under the same conditions, lactide conversion reached 65% after 72 h of polymerization. Unpolymerized monomer was removed after the reaction by extraction with an SC‐CO2 mobile phase. The molecular weights of all the copolymers were measured by gel permeation chromatography. Weight‐average molecular weights (Mw) ranged between 2500 and 30,200 g/mol and polydispersity indices ranged from 1.4 to 2.3 for polymerization times of 6 and 48 h, respectively. Although the molecular weight increased significantly during the first 48 h of reaction, there was no significant difference in the Mw for polymerization times of 48 and 72 h. Emulsion polymerization within the benign solvent SC‐CO2 demonstrated improved conversion and molecular weight versus polymers synthesized without surfactant. The emulsion polymerization of lactide and glycolide copolymers in SC‐CO2 is proposed as a novel production technique for high‐purity, biodegradable polymers. © 2001 John Wiley & Sons, Inc. In this study, porous poly(lactide‐co‐glycolide) (PLGA) microparticles with low mass density and large particle size were developed for chronic obstructive pulmonary disease treatment using anticholinergic drug (tiotropium). The porous PLGA microparticles were prepared by the water‐in‐oil‐in‐water (W1/O/W2) multi‐emulsion method using PLGA polymer and ammonium bicarbonate (as a porogen). Herein, soluble starch was incorporated in porous PLGA microparticles for long‐term tiotropium release. In vitro drug release studies determined that the rapid release of tiotropium from porous PLGA microparticles was reduced because of the high viscosity of the incorporated starch. Tiotropium release from porous PLGA microparticles continued up to 3 days. Furthermore, the inhaled microparticles showed longer drug residence in in vivo lung epithelium. Using a mutual solvent technique, blend films of poly[(alaine ethyl ester)0.62(glycine ethyl ester)0.38]phosphazene/poly(lactide‐co‐glycolide) (PAGP/PLGA blend) were prepared at different conditions including weight ratios, solvents, environmental humidity, film thickness, and substrates. The morphology and properties of blend films were characterized by scanning electron microscope (SEM), energy dispersive spectrometer (EDX), X‐ray photoelectron spectrometry (XPS), and solvent selective etching. Compared with dichloromethane and tetrahydrofuran (THF), chloroform was the better solvent to form miscible PAGP/PLGA blend films at relatively anhydrous atmosphere. However, in the humid atmosphere, the hexagonal arrangement of holes appeared on the surface of PAGP/PLGA blend films due to the ordered array of water droplets. A sandwich‐liked structure was formed with the hydrophilic PAGP component at the top and bottom, while the PLGA component in the middle. In addition, the surface morphology of PAGP/PLGA blend films was also influenced by the film thickness and the property of the substrate. Copyright © 2010 John Wiley & Sons, Ltd.  相似文献

Static SIMS (SSIMS) is a surface analytical technique capable of providing molecular chemical information from solids. A major barrier to the wider take‐up of the technique is the complexity associated with the interpretation of SSIMS spectra. Quality of the interpretation depends on the expertise of analysts and making references to the limited mass spectral libraries. For many materials, there are no SSIMS library spectra. A new library‐independent method, G‐SIMS, is capable of facilitating the interpretation of SSIMS data. G‐SIMS spectra contain parent fragments, which are formed without substantial degradation or rearrangements, and highlight molecular fragments, which are directly related to the surface. In our study, G‐SIMS has been tested on medically relevant biodegradable polyester series, including poly (glycolic acid) (PGA), poly‐l‐(lactic acid) (PLA), poly‐β‐(hydroxybutyrate) (PHB) and poly‐ε‐(caprolactone) (PCL). The polyester series chosen here have closely related structures, which allow us to explore the capabilities of G‐SIMS. The G‐SIMS spectra have facilitated the identification of different polyesters by exhibiting mainly characteristic ions, representative of the polymers' molecular structures. The results also indicated that for the chosen polyester series, the larger the repeating monomer structures, the smaller the maximum number of repeat units were seen in the G‐SIMS spectra. The G‐SIMS spectra for the homologous polyester series have provided an insight into the fragmentation mechanisms as a function of repeating monomer molecular weights and structures. The understanding of the simultaneous transport and chain‐scission phenomena involved in the hydrolysis of bulk‐degrading polymers requires the experimental separation of chain cleavage and water diffusion. The hydrolytic chain cleavage of poly(rac‐lactide) rac‐(PLA) and poly[(rac‐lactide)‐co‐glycolide] (PLGA) is analysed on the basis of monolayer degradation experiments combined with an improved data reduction procedure. Different, partly contradictory models of the hydrolytic degradation and erosion mechanism of PLA and PLGA, namely random chain scission and chain‐end scission, are discussed in the literature. The instantaneous linear area reduction observed for the polymer Langmuir films indicates a chain‐end scission mechanism. As monolayers of end‐capped and non‐end‐capped polymers degrade with exactly the same rate, the observed differences in the degradation kinetics of bulk samples do clearly result from differences in the water penetration into these polymers. A pronounced ‘auto‐inhibition’ effect is observed for the polymers degraded at initially high pH of the aqueous subphase in the absence of buffers. Copyright © 2007 John Wiley & Sons, Ltd.  相似文献

This paper reports the feasibility to prepare a biodegradable material stable to γ‐irradiation by grafting of poly(lactide‐co‐glycolide) with pyrogallic acid (PLGA‐g‐PA) and ferulic acid (PLGA‐g‐FA) in mild condition. Only the grafting procedure with PA did not modify molecular weight (Mw) of the starting polymer and PLGA‐g‐PA showed antioxidant properties. The polymer degradation in pH 7.4 phosphate buffer saline (PBS) was mainly governed by a random chain scission mechanism according to a first‐order reaction. The irradiation at the dose of 25 kGy caused only a very slight decrease of Mw and the degradation patterns of the non‐irradiated and irradiated material were superimposable. PLGA‐g‐PA resulted a promising material to develop biodegradable drug delivery systems which would be sterilizable in the final container. Copyright © 2010 John Wiley & Sons, Ltd.  相似文献

Comb‐like amphiphilic poly(poly((lactic acid‐co‐glycolic acid)‐block‐poly(ethylene glycol)) methacrylate (poly((PLGA‐b‐PEG)MA)) copolymers were synthesized by radical polymerization. (PLGA‐b‐PEG)MA macromonomer was prepared by ring‐opening bulk polymerization of DL ‐lactide and glycolide using purified poly(ethylene glycol) monomethacrylate (PEGMA) as an initiator. (PLGA‐b‐PEG)MA macromonomer was copolymerized with PEGMA and/or acrylic acid (AA) by radical polymerization to produce comb‐like amphiphilic block copolymers. The molecular weight and chemical structure were investigated by GPC and 1H NMR. Poly((PLGA‐b‐PEG)MA) copolymer aqueous solutions showed gel–sol transition behavior with increasing temperature, and gel‐to‐sol transition temperature decreased as the compositions of the hydrophilic PEGMA and AA increased. The gel‐to‐sol transition temperature of the terpolymers of the poly((PLGA‐b‐PEG)MA‐co‐PEGMA‐co‐AA) also decreased when the pH was increased. The effective micelle diameter obtained from dynamic light scattering increased with increasing temperature and with increasing pH. The critical micelle concentration increased as the composition of the hydrophilic monomer component, PEGMA and AA, were increased. The spherical shape of the hyperbranched polymers in aqueous environment was observed by atomic force microscopy. © 2008 Wiley Periodicals, Inc. In this work, a series of PLGA‐PEG diblock copolymers were synthesized by ring‐opening polymerization of L‐lactide and glycolide using mPEG as macroinitiator and stannous octoate as catalyst. Spherical micelles were obtained from the various copolymers by using co‐solvent evaporation method. The biocompatibility of micelles was evaluated with the aim of assessing their potential in the development of drug delivery systems. Various aspects of biocompatibility were considered, including MTT assay, agar diffusion test, release of cytokines, hemolytic test, dynamic clotting time, protein adsorption in vitro, and zebrafish embryonic compatibility in vivo. The combined results revealed that the micelles present good cytocompatibility and hemocompatibility in vitro. Moreover, the cumulative effects of micelles throughout embryos developing stages have no toxicity in vivo. It is thus concluded that micelles prepared from PLGA‐PEG copolymers present good biocompatibility as potential drug carrier.  相似文献

This article deals with the preparation of novel co‐oligoethers constituted with 1,3‐propanediol (PDO) and isosorbide units and, prepared according to two different melt processes, without any solvent in the presence of acid catalyst: co‐etherification of PDO and isosorbide (process A) and, trans‐etherification between polytrimethylene ether glycol (PTEG) and isosorbide (process B). Complementary analytical methods: D and 2D 1H NMR and gas chromatography analysis, coupled with FID and MS‐MALDI‐TOF mass spectrometry, were performed to precisely define the microstructure of the final products. In particular, one can observe that two mechanisms involve during the reaction: etherification and trans‐etherification where isosorbide reacts decreasing the molar mass of polymers chains. This led to oligomers having isosorbide units at each extremity and little inner isosorbide units. Computational calculations have been performed in parallel, and the data well duplicate the experimental results. Finally, it was shown that these new telechelic oligoethers have higher compatibility to water and higher Tg level and thermal stability than PTEG homopolymer. Precise nano‐ and microscale control of the architecture of biodegradable biomaterials is desirable for several biotechnological applications such as drug delivery, diagnostics, and medical imaging. Herein, we combine electrohydrodynamic co‐jetting and highly specific surface modification (via Huisgen 1,3‐dipolar cycloaddition) to prepare particles and fibers with spatioselective surface modification. We first prepared biphasic particles and fibers from commercial poly(lactide‐co‐glycolide) copolymers via electrohydrodynamic co‐jetting of two organic solutions loaded with fluorescent macromolecules and acetylene‐modified PLGA derivatives. (i) Spatially controlled reaction of poly[lactide‐co‐(propargyl glycolide)] with O‐(2‐aminoethyl)‐O′‐(2‐azidoethyl)heptaethylene glycol and (ii) subsequent conversion of the newly introduced amino groups with fluorescence probes resulted in particles and fibers with surface modification of one hemisphere only.

In this study, biodegradable shape‐memory polymers—polylactide‐co‐poly(glycolide‐co‐caprolactone) multiblock (PLAGC) copolymers—were synthesized by the coupling reaction of both macrodiols of polylactide (PLLA‐diol) and poly(glycolide‐co‐caprolactone) (PGC‐diol) in the presence of 1,6‐hexanediisocyanate as coupling agent. The copolymers formed were found to be thermoplastic and easily soluble in common solvents. The compositions of the copolymers were determined by 1H‐NMR and the influences of segment lengths and contents of both macrodiols on the properties of the PLAGC copolymers were investigated. It was found that the copolymers had adjustable mechanical properties which depended on contents and segment lengths of both macrodiols. The copolymers showed such good shape‐memory properties that the strain fixity rate (Rf) and the strain recovery rate (Rr) exceed 90%. By means of adjusting the compositions of the copolymers, PLAGC copolymers with transition temperatures around 45°C could be obtained. The degradation rate determination showed that the PLAGC copolymers have fast degradation rates, the mechanical strengths of the PLAGC copolymers would be completely lost within 1–2 months depending on molecular weights and contents of the both segments of PLLA and PGC. Diblock copolymers consisting of methoxy poly(ethylene glycol) (MPEG) and poly(?‐caprolactone) (PCL), poly(δ‐valerolactone) (PVL), poly(L ‐lactic acid) (PLLA), or poly(lactic‐co‐glycolic acid) (PLGA) as biodegradable polyesters were prepared to examine the phase transition of diblock copolymer solutions. MPEG–PCL and MPEG–PVL diblock copolymers and MPEG–PLLA and MPEG–PLGA diblock copolymers were synthesized by the ring‐opening polymerization of ?‐caprolactone or δ‐valerolactone in the presence of HCl · Et2O as a monomer activator at room temperature and by the ring‐opening polymerization of L ‐lactide or a mixture of L ‐lactide and glycolide in the presence of stannous octoate at 130 °C, respectively. The synthesized diblock copolymers were characterized with 1H NMR, IR, and gel permeation chromatography. The phase transitions for diblock copolymer aqueous solutions of various concentrations were explored according to the temperature variation. The diblock copolymer solutions exhibited the phase transition from gel to sol with increasing temperature. As the polyester block length of the diblock copolymers increased, the gel‐to‐sol transition moved to a lower concentration region. The gel‐to‐sol transition showed a dependence on the length of the polyester block segment. According to X‐ray diffraction and differential scanning calorimetry thermal studies, the gel‐to‐sol transition of the diblock copolymer solutions depended on their degrees of crystallinity because water could easily diffuse into amorphous polymers in comparison with polymers with a crystalline structure.
