Morphologies of poly(ε‐caprolactone)‐b‐poly(ethylene glycol)‐b‐poly(ε‐caprolactone) (PCL‐PEG‐PCL) triblock copolymer self‐assemblies in the diluted solution and in gel were studied by atomic force microscopy (AFM). The copolymer self‐assembled into wormlike aggregates, of uniform diameter, in water. The wormlike aggregates arranged in order to form separate clusters in the diluted copolymer solution; at a higher copolymer concentration, the clusters became bigger and bigger, and packed together to form gel. Novel poly(ε‐caprolactone)‐b‐poly(ethylene glycol)‐b‐poly(ε‐caprolactone) (PCL‐PEG‐PCL) bearing pendant hydrophobic γ‐(carbamic acid benzyl ester) groups (PECB) and hydrophiphilic amino groups (PECN) were synthesized based on the functionalized comonomer γ‐(carbamic acid benzyl ester)‐ε‐caprolactone (CABCL). The thermal gelation behavior of the amphiphilic copolymer aqueous solutions was examined. The phase transition behavior could be finely tuned via the pendant groups, and an abnormal phenomenon occurred that the sol–gel transition temperature shifted to a higher temperature for PECB whereas a lower temperature for PECN. The micelles percolation was adopted to clarify the hydrogel mechanism, and the effect of the pendant groups on the micellization was further investigated in detail. The results demonstrated that the introduction of γ‐(carbamic acid benzyl ester) pendant groups significantly decreased the crystallinity of the copolymer micelles whereas amino pendant groups made the micelles easy to aggregate. Thus, the thermal gelation of PEG/PCL aqueous solution could be finely tuned by the pendant groups, and the pendant groups modified PEG/PCL hydrogels are expected to have great potential biomedical application. © 2016 Wiley Periodicals, Inc. Block copolymers of ethylene oxide and ε‐caprolactone were synthesized by microwave‐assisted polymerization of ε‐caprolactone with polyethylene glycol monomethyl ethers as initiator. The samples thus obtained were characterized by two‐dimensional liquid chromatography with liquid chromatography at critical conditions as the first and liquid exclusion adsorption chromatography as the second dimension. A full baseline separation of all oligomers could be achieved in both dimensions. 相似文献

Star-shaped poly(ethylene glycol) (PEG)-poly(?-caprolactone) (?-PCL) block copolymers were synthesized via a ring-opening polymerization. Nanoparticles prepared by the precipitation/solvent evaporation technique exhibit a core-shell structure. The hydrolytic degradation of 3-arm PEG-PCL copolymeric nanoparticles was studied by dynamic light scattering (DLS), nuclear magnetic resonance (NMR) and gel permeation chromatography (GPC). It was found that copolymers with shorter PCL block length degraded faster. The sizes of nanoparticles fluctuated during the initial degradation period, and then increased slightly before finally dropping off. The degradation mainly occurred at CL-CL linkages firstly then at the EO-CL linkages. The CL/EO molar ratios and the molecular weights of copolymers decreased as degradation time and a zero-order degradation behavior was observed. 相似文献

Ring opening polymerization of ε‐caprolactone was realized in the presence of monomethoxy poly(ethylene glycol) with Mn = 1000 and 2000, using Zn(La)2 as catalyst. The resulting PCL‐PEG diblock copolymers with CL/EO repeat unit molar ratios from 0.2 to 3.0 were characterized by using DSC, WAXD, SEC, and 1H NMR. The crystal phase of PCL blocks exist in all polymers, and the crystallization ability of PCL blocks increases with CL/EO ratio. PEG blocks are able to crystallize for copolymers with CL/EO below 1.0 only. Melt crystallization results were analyzed with Avrami equation. The Averami exponent n is around 3.0 in most cases, in agreement with heterogeneous nucleation with three dimensional growth. The morphology of the crystals was observed by using POM. Rod‐like crystals were found to grow in 1, 3 or 2, 4 quadrants for samples with low molecular weights. In the case of a copolymer with Mn,PEG = 2000 and Mn,PCL = 800, PEG blocks could crystallize and grow on PCL crystals after PCL finished to form rod‐like crystals, leading to formation of poorly or well structured spherulites. The spherulite growth rate (G) was determined at different crystallization temperatures (Tc) ranging from 9 to 49 °C. All the copolymers present a steady G decrease with increasing crystallization temperature due to lower undercooling. A series of amphiphilic triblock copolymers, methoxy poly(ethylene glycol)‐b‐poly(octadecanoic anhydride)‐b‐methoxy poly(ethylene glycol) (mPEG‐b‐POA‐b‐mPEG), were prepared via melt polycondensation of methoxy poly(ethylene glycol) (mPEG) and poly(octadecanoic anhydride) (POA). mPEG‐b‐POA‐b‐mPEG were characterized by FTIR, 1H‐NMR, GPC, DSC, and XRD. Drug‐loaded mPEG‐b‐POA‐b‐mPEG nanoparticles (NPs) with spherical morphology and narrow size polydispersity index were prepared by nanoprecipitation technique with paclitaxel as the model drug. In vitro release behaviors of drug‐loaded NPs present that the biphasic process and the release mechanism of each phase are zero order drug releases. According to this study, mPEG‐b‐POA‐b‐mPEG NPs could serve as suitable delivery agents for paclitaxel and other hydrophobic drugs. Copyright © 2009 John Wiley & Sons, Ltd. 相似文献

Nontoxic and biodegradable poly(?‐caprolactone)‐b‐poly(ethylene glycol)‐b‐poly(?‐caprolactone) triblock copolymers were synthesized by the solution polymerization of ?‐caprolactone in the presence of poly(ethylene glycol). The chemical structure of the resulting triblock copolymer was characterized with 1H NMR and gel permeation chromatography. In aqueous solutions of the triblock copolymers, the micellization and sol–gel‐transition behaviors were investigated. The experimental results showed that the unimer‐to‐micelle transition did occur. In a sol–gel‐transition phase diagram obtained by the vial‐tilting method, the boundary curve shifted to the left, and the gel regions expanded with the increasing molecular weight of the poly(?‐caprolactone) block. In addition, the hydrodynamic diameters of the micelles were almost independent of the investigated temperature (25–55 °C). The atomic force microscopy results showed that spherical micelles formed at the copolymer concentration of 2.5 × 10?4 g/mL, whereas necklace‐like and worm‐like shapes were adopted when the concentration was 0.25 g/mL, which was high enough to form a gel. © 2006 Wiley Periodicals, Inc. Hydroxyl‐terminated poly(butadiene) (HTPB; Mn = 2100 g mol−1) was capped with 30 and 60 wt % of ɛ‐caprolactone to reach amphiphilic triblock copolymers in form of capped poly(butadiene) CPB. The former (CPB30; Mn = 3300 g/mol) is amorphous with a glass temperature of −56 °C. CPB60 (Mn = 4000 g mol−1) is semi‐crystalline with a melting point of 50 °C and a glass transition at −47 °C. The CPBs, HTPB and polycaprolactone diol (Mn = 2000 g mol−1) were used as soft segment components in the preparation of polyurethane elastomers (PUE), using a 1/1 mixture of an MDI prepolymer and uretonimine modified MDI, and hard phase components in form of 1,3‐propane diol, 1,4‐butane diol, and 1,5‐pentane diol. CPB‐based elastomers with 1,4 butane diol (8 wt %) show hard domains as fringed aggregates with a better connection to the continuous phase than the HTPB‐based PUE. The soft segment glass transition temperature (Tg) is at −28 °C for HTPB‐based PUE and at −43 °C for those of CPB. Per‐2,3‐acetyl‐β‐cyclodextrin with seven primary hydroxyl groups was synthesized by selective modification and used as multifunctional initiator for the ring‐opening polymerization of ε‐caprolactone (CL). Well‐defined β‐cyclodextrin‐centered seven‐arm star poly(ε‐caprolactone)s (CDSPCLs) with narrow molecular weight distributions (≤1.15) have been successfully prepared in the presence of Sn(Oct)2 at 120 °C. The molecular weight of CDSPCLs was characterized by end group 1H NMR analyses and size‐exclusion chromatography (SEC), which could be well controlled by the molar ratio of the monomer to the initiator. Furthermore, amphiphilic seven‐arm star poly(ε‐caprolactone‐b‐ethylene glycol)s (CDSPCL‐b‐PEGs) were synthesized by the coupling reaction of CDSPCLs with carboxyl‐terminated mPEGs. 1H NMR and SEC analyses confirmed the expected star block structures. Differential scanning calorimetry analyses suggested that the melting temperature (Tm), the crystallization temperature (Tc), and the crystallinity degree (Xc) of CDSPCLs all increased with the increasing of the molecular weight, and were lower than that of the linear poly(ε‐caprolactone). As for CDSPCL‐b‐PEGs, the Tc and Tm of the PCL blocks were significantly influenced by the PEG segments in the copolymers. Moreover, these amphiphilic star block copolymers could self‐assemble into spherical micelles with the particle size ranging from 10 to 40 nm. pH‐responsive methoxy poly(ethylene glycol)‐b‐poly(ε‐caprolactone) bearing pendant carboxyl groups mPEG‐b‐P(2‐CCL‐co‐6‐CCL) was synthesized based on our newly monomer benzyloxycarbonylmethly functionalized ε‐caprolactone. Their structure was confirmed by 1H NMR, 13C NMR, and Fourier transform infrared spectrum spectra. In addition, SEC results indicated that the copolymers had a relatively narrow polydispersity. WXRD and DSC demonstrated that the introduction of carboxymethyl groups had significant effect on the crystallinity of the copolymers. Furthermore, the solution behavior of mPEG‐b‐P(2‐CCL‐co‐6‐CCL) has been studied by various methods. The results indicated that mPEG‐b‐P(2‐CCL‐co‐6‐CCL) had a rich pH‐responsive behavior and the micelles could be formed by pH induction, and the mPEG‐b‐P(2‐CCL‐co‐6‐CCL) could existed as unimers, micelles or large aggregates in different pH range accordingly. The mechanism of which was supposed to depend on the counteraction between the hydrophobic interaction from PCL and the ionization of the carboxyl groups along the polymer chain. In this study, in situ polyurethane (PU) bionanocomposites of poly(ethylene glycol) (PEG)/poly(ε‐caprolactone) (PCL) polyols, bare cellulose nanocrystals (CNCs) and PCL‐grafted CNCs (G‐CNC) were synthesized with different contents of CNCs as cross‐linking agent to control the extent of phase separation. The effect of confining the chains between CNCs through urethane linkages and presence of PCL grafts on phase and crystallization behavior was evaluated. Crystallization and chemical networking were controlled to tune the shape fixity (SF) and recovery (SR) of the specimens, resulting in a SF of 100% for linear and PU nanocomposites of G‐CNC (0.5% and 1%) samples. The PU nanocomposite of G‐CNC (0.5%) was selected as the optimum sample with the highest SR of 100%. The effect of surface hydrophobicity on cellular behavior of Human Foreskin Fibroblast (as a normal cell) and HepG2 (as a cancerous cell) cells was evaluated. Cell adhesion analysis of the prepared samples indicated two different behaviors possibly due to the difference in the epigenetic nature of the cells and cellular integrin‐ based bonds showing a great potential for a variety of tissue engineering applications. 相似文献

Concentration dependent morphology of 3‐armed poly(ethylene glycol)‐b‐poly(ε‐caprolactone) copolymer aggregates in aqueous system was investigated by atomic force microscopy (AFM). The AFM results show that, at a low concentration, 4 × 10?5 g/mL, spherical micelles occur, and unmicellized molecules are not distributed homogeneously in the copolymer aqueous solution. Unequal outspread clusters composed of wormlike aggregates are formed at a moderate copolymer concentration, 4 × 10?4 g/mL, those wormlike aggregates are orderly packed in the clusters. At a high concentration of 0.05 g/mL, the copolymer aqueous system is indeed a gel at room temperature, outspread clusters of wormlike aggregates join together to forma network structure. © 2008 Wiley Periodicals, Inc. Triblock copolymers of poly(styrenesulfonate)‐b‐poly(ethylene glycol)‐b‐poly(styrenesulfonate) with narrow molecular weight distribution (Mw/Mn = 1.28–1.40) and well‐defined structure have been synthesized in aqueous solution at 70 °C via reversible addition‐fragmentation chain transfer polymerization. Poly(ethylene glycol) (PEG) capped with 4‐cyanopentanoic acid dithiobenzoate end groups was used as the macro chain transfer agent (PEG macro‐CTA) for sole monomer sodium 4‐styrenesulfonate. The reaction was controllable and displayed living polymerization characteristics and the triblock copolymer had designed molecular weight. The reaction rate depended strongly on the CTA and initiator concentration ratio [CTA]0/[ACPA]0: an increase in [CTA]0/[ACPA]0 from 1.0 to 5.0 slowed down the polymerization rate and improved the molecular weight distribution with a prolonged induction time. To create a novel vector for specifically delivering anticancer therapy to solid tumors, we used diafiltration to synthesize pH‐sensitive polymeric micelles. The micelles, formed from a tetrablock copolymer [poly(ethylene glycol)‐b‐poly(L ‐histidine)‐b‐poly(L ‐lactic acid)‐b‐poly(ethylene glycol)] consisted of a hydrophobic poly(L ‐histidine) (polyHis) and poly(L ‐lactic acid) (PLA) core and a hydrophilic poly(ethylene glycol) (PEG) shell, in which we encapsulated the model anticancer drug doxorubicin (DOX). The robust micelles exhibited a critical micellar concentration (CMC) of 2.1–3.5 µg/ml and an average size of 65–80 nm pH 7.4. Importantly, they showed a pH‐dependent micellar destabilization, due to the concurrent ionization of the polyHis and the rigidity of the PLA in the micellar core. In particular, the molecular weight of PLA block affected the ionization of the micellar core. Depending on the molecular weight of the PLA block, the micelles triggering released DOX at pH 6.8 (i.e. cancer acidic pH) or pH 6.4 (i.e. endosomal pH), making this system a useful tool for specifically treating solid cancers or delivering cytoplasmic cargo in vivo. A series of novel types of three‐armed poly(trans‐4‐hydroxy‐N‐benzyloxycarbonyl‐L ‐proline)‐block‐poly(ε‐caprolactone) (PHpr‐b‐PCL) copolymers were successfully synthesized via melt block copolymerization of trans‐4‐hydroxy‐N‐benzyloxycarbonyl‐L ‐proline (N‐CBz‐Hpr) and ε‐caprolactone (ε‐CL) with a trifunctional initiator trimethylolpropane (TMP) and stannous octoate (SnOct2) as a catalyst. For the homopolycondensation of N‐CBz‐Hpr with TMP initiator and SnOct2 catalyst, the number‐average molecular weight (Mn) of prepolymer increases from 530 to 3540 g mol?1 with the molar ratio of monomer to initiator (3–30), and the molecular weight distribution (Mw/Mn) is between 1.25 to 1.32. These three‐armed prepolymer PHpr were subsequently block copolymerized with ε‐caprolactone (ε‐CL) in the presence of SnOct2 as a catalyst. The Mn of the copolymer increased from 2240 to 18,840 g mol?1 with the molar ratio (0–60) of ε‐CL to PHpr. These products were characterized by differential scanning calorimetry (DSC), 1H NMR, and gel permeation chromatography. According to DSC, the glass‐transition temperature (Tg) of the three‐armed polymers depended on the molar ratio of monomer/initiator that were added. A biodegradable diblock copolymer of poly(ϵ‐caprolactone) (PCL) and poly(L ‐lactide) (PLLA) was synthesized and characterized. The inclusion compound (IC) of this copolymer with α‐cyclodextrin (α‐CD) was formed and characterized. Wide‐angle X‐ray diffraction showed that in the IC crystals α‐CDs were packed in the channel mode, which isolated and restricted the individual guest copolymer chains to highly extended conformation. Solid‐state 13C NMR techniques were used to investigate the morphology and dynamics of both the bulk and α‐CD‐IC isolated PCL‐b‐PLLA chains. The conformation of the PCL blocks isolated within the α‐CD cavities was similar to the crystalline conformation of PCL blocks in the bulk copolymer. Spin–lattice relaxation time (T1C) measurements revealed a dramatic difference in the mobilities of the semicrystalline bulk copolymer chains and those isolated in the α‐CD‐IC channels. Carbon‐observed proton spin–lattice relaxation in the rotating frame measurements (T1ρH) showed that the bulk copolymer was phase‐separated, while, in the IC, exchange of proton magnetization through spin‐diffusion between the isolated guest polymer chains and the host α‐CD was not complete. The two‐dimensional solid‐state heteronuclear correlation (HetCor) method was also employed to monitor proton communication in these samples. Intrablock exchange of proton magnetization was observed in both the bulk semicrystalline and IC copolymer samples at short mixing times; however, even at the longest mixing time, interblock proton communication was not observed in either sample. In spite of the physical closeness between the isolated included guest chains and the host α‐CD molecules, efficient proton spin diffusion was not observed between them in the IC. © 2005 Wiley Periodicals, Inc. A series of amphiphilic triblock copolymers, poly[oligo(ethylene glycol) methacrylate]x‐block‐poly(ε‐caprolactone)‐block‐poly[oligo(ethylene glycol) methacrylate]x, POEGMACo(x), were synthesized. Formation of hydrophobic domains as cores of the micelles was studied by fluorescence spectroscopy. The critical micelle concentrations in aqueous solution were found to be in the range of circa 10?6 M. A novel methodology by modulated temperature differential scanning calorimetry was developed to determine critical micelle temperature. A significant concentration dependence of cmt was found. Dynamic light scattering measurements showed a bidispersed size distribution. The micelles showed reversible dispersion/aggregation in response to temperature cycles with lower critical solution temperature between 75 and 85 °C. The interplay of the two hydrophobic and one thermoresponsive macromolecular chains offers the chance to more complex morphologies. © 2010 Wiley Periodicals, Inc. A series of novel amphiphilic brush‐dendritic‐linear poly[poly(ethylene glycol) methyl ether methacrylate]‐b‐polyamidoamine‐b‐poly(ε‐caprolactone) copolymers (PPEGMEMA‐b‐Dm‐b‐PCL) (m = 1, 2, and 3: the generation number of dendron) were synthesized by the combination techniques of click chemistry, atom transfer radical polymerization (ATRP), and ring‐opening polymerization (ROP). The brush‐dendritic copolymers bearing hydrophilic brush PPEGMEMA and hydrophobic dendron polyamidoamine protected by the tert‐butoxycarbonyl (Boc) groups [Dm‐(Boc) (m = 1, 2, and 3)] were for the first time prepared by ATRP of poly(ethylene glycol) methyl ether methacrylate monomer (PEGMEMA) initiated with the dendron initiator, which was prepared from 2′‐azidoethyl‐2‐bromoisobutyrate (AEBIB) and Dm‐(Boc) terminated with a clickable alkyne by click chemistry. Then, the brush‐dendritic copolymers with primary amine groups (PPEGMEMA‐b‐Dm) were obtained from the removal of the protected Boc groups of the brush‐dendritic copolymers in the presence of trifluoroacetic acid. The brush‐dendritic‐linear PPEGMEMA‐b‐Dm‐b‐PCL copolymers were synthesized from ROP of ε‐caprolactone monomer using PPEGMEMA‐b‐Dm as the macroinitiators and stannous octoate as catalyst in toluene at 130 °C. To the best of our knowledge, this is the first report that integrates hydrophilic brush polymer PPEGMEMA with hydrophobic polyamidoamine (PAMAM) dendron and PCL to form amphiphilic brush‐dendritic‐linear copolymers. We report the coating of maghemite (γ‐Fe2O3) nanoparticles with poly(ε‐caprolactone) (PCL) through a covalent grafting to technique. ω‐Hydroxy‐PCL was first synthesized by the ring‐opening polymerization of ε‐caprolactone with aluminum isopropoxide and benzyl alcohol as a catalytic system. The hydroxy end groups of PCL were then derivatized with 3‐isocyanatopropyltriethoxysilane in the presence of tetraoctyltin. The triethoxysilane‐functionalized PCL macromolecules were finally allowed to react on the surface of maghemite nanoparticles. The composite nanoparticles were characterized by diffuse reflectance infrared Fourier transform (DRIFT) spectroscopy, thermal gravimetric analysis (TGA) and differential scanning calorimetry (DSC). Effects of the polymer molar mass and concentration on the amount of polymer grafted to the surface were investigated. Typical grafting densities up to 3 μmol of polymer chains per m2 of maghemite surface were obtained with this grafting to technique. © 2004 Wiley Periodicals, Inc. The ability to predict the in vivo performance of multiblock‐copolymer‐based biomaterials is crucial for their applicability in the biomedical field. In this work, XPS analysis of PCL‐PEG copolymers was carried out, as well as morphological and wettability evaluations by SEM and CA measurements, respectively. XPS analysis on films equilibrated in PBS demonstrated a further enrichment in the PEG component on the surface. Copolymer films obtained by casting using different solvents showed a dependence in segregation according to the solvent employed. Cell adhesion tests demonstrated the importance of copolymer segregation and rearrangement in a wet environment, with a dependence of these phenomena on the copolymer molecular weight.
