A series of polyamido amine (PAMAM) dendrimers (Generations 2, 3, 4, and 6) fully functionalized at their periphery with first‐ and second‐generation poly (phenylenevinylene) (PPV) dendrons have been efficiently prepared. MALDI‐TOF mass spectrometry proved to be particularly useful for the characterization of the new hybrid dendrimers as well as for the estimation of the average number of PPV dendrons attached to the surface. The optical absorption and emission properties of these systems were studied. The materials display extremely high molar extinction coefficients and emit blue light with only slightly lower fluorescence quantum yields than the corresponding free dendrons. Self‐quenching interactions between PPV units were not observed in THF. However, the luminescence properties underwent a dramatic change when toluene was used as the solvent. The lower polarity of toluene caused shrinkage of the PAMAM structure and brought the PPV chromophores closer together, leading to self‐quenching interactions and excimer formation. © 2009 Wiley Periodicals, Inc. Azide-functionalized PAMAM dendrons containing an azidopropylamine focal point were synthesized by the divergent method and applied for the construction of symmetric PAMAM-like dendrimers containing 1,2,3-triazole rings as connectors via stitching with two different multi-terminal alkynes. Mannose‐functionalized and ethoxyethanol‐functionalized poly(amido)amine dendrimers bound multiple vanadate‐substituted polyoxotungstate Wells–Dawson‐type polyoxometalates (POMs). Dendrimers incorporating 10–30 POMs were characterized with NMR, transmission electron microscopy, and matrix‐assisted laser desorption/ionization time‐of‐flight mass spectrometry techniques. The number of metal clusters per dendrimer molecule varied according to the dendrimer generation and the nature of the surface functional groups. This work deals with the covalent coupling of azide‐functionalized polymeric nanoparticles as a reactive core and amino‐terminated PAMAM dendrons as a reactive shell. The nanoassemblies thereby obtained could be modified after the dendronization step by grafting an alkynyl Bodipy dye on the unreacted azide moieties. Only a few steps are required to attain nanoassemblies that could mimic dendrimers of high generation with sizes of nano‐objects beyond those of dendrimers. The structure of the nanoassemblies are composed of a polystyrene core, an inner shell including the Bodipy dyes along with the internal branches of the PAMAM, and the terminal amino groups from the outer shell. The dendritic shell acts as a protective layer that prevents NP from aggregation in a surfactant free aqueous solution. The nano‐objects display absorption and emission maxima above 500 nm with brightness that are the same order of magnitude than Qdots. © 2015 Wiley Periodicals, Inc. J. Polym. Sci., Part A: Polym. Amphiphilic poly(amidoamine) (PAMAM) dendrimers consisting of a hydrophilic dendrimer core and hydrophobic aromatic dansyl or 1‐(naphthalenyl)‐2‐phenyldiazene (NPD) shells have been synthesized. These amphiphilic dendrimers from the zero generation to the third generation self‐assemble into vesicular aggregates in water. The self‐assembly behavior of these dendrimers strongly depends on their generations. The generation dependence has been further investigated by an exploration of their electrochemical properties. Poly(amidoamine)(PAMAM) dendrimers with a cinnamoyl shell were prepared by reacting full generation PAMAM dendrimers (G=3.0) with 2‐chloroethanol and cinnamoyl chloride, which resulted in densely packed polymerizable unsaturated groups on the periphery. The cinnamoyl shell of the dendrimers dimerized when irradiated under a UV light by using 5‐nitroacenaphthylene as an initiator in dilute dimethylformamide (DMF). FTIR, 1H NMR, UV‐Vis, SEC, and a viscosity test certified that the photocycloaddition of the cinnamoyl shell of the dendrimers took place within the molecules with the disappearance of double bond signals in the FTIR. 1H NMR spectra as well as the intrinsic viscosity and polydispersity value of the products both before and after irradiation showed no difference. It was further found that the cinnamoyl shell‐modified dendrimers possessed fluorescence property, and the fluorescence intensity became stronger when the shell was photocyclized under UV‐ irradiation. © 2000 John Wiley & Sons, Inc. This work reports a new electrochemical monitoring platform for sensitive detection of Cu2+ coupling click chemistry with nanogold‐functionalized PAMAM dendrimer (AuNP‐PAMAM). The system involved an alkyne‐modified carbon electrode and an azide‐functionalized AuNP‐PAMAM. Initially, the added Cu2+ was reduced to Cu+ by the ascorbate, and then the azide‐modified AuNP‐PAMAM was covalently conjugated to the electrode via Cu+‐catalyzed azide‐alkyne click reaction. The carried AuNPs accompanying PAMAM dendrimer could be directly monitored by stripping voltammetry after acidic pretreatment. By introduction of high‐loading PAMAM dendrimer with gold nanoparticles, as low as 2.8 pM Cu2+ (ppt) could be detected, which was 125‐fold lower than that of gold nanoparticle‐based labeling strategy. The method exhibited high specificity toward target Cu2+ against other potentially interfering ions, and was applicable for monitoring Cu2+ in drinking water with satisfactory results.  相似文献

Aliphatic polyesters bearing pendant alkyne groups were successfully prepared by step‐growth polymerization of different building blocks such as adipic acid and succinic acid in combination with an acetylene‐based diol, 2‐methyl‐2‐propargyl‐1,3‐propanediol, besides 1,4‐butanediol and ethylene glycol. It was demonstrated that the alkyne groups survive the high reaction temperatures (200 °C) in the presence of a radical inhibitor. The alkyne loading has been tuned by the ratio of the different monomers used, up to 25 mol % of alkyne groups. Subsequently, the alkyne groups have been reacted with azides by the copper‐catalyzed Huisgen 1,3‐dipolar cycloaddition reaction, a popular type of “click” chemistry. “Click” reactions have been performed quantitatively in the presence of benzyl azide and azide‐terminated poly(ethylene glycol), yielding brush copolymers in the latter case. Kinetic investigations about this click reaction have been performed by means of on‐line Fourier transform mid‐infrared spectroscopy, which was reported for the first time in the field of the click chemistry research. A whole range of functionalized polyesters, based on poly(ethylene succinate) and poly(butylene adipate), is available, the properties of which can be tailored by choosing the appropriate azide compound. © 2008 Wiley Periodicals, Inc. Hyperbranched poly(amido amine) (h‐PAMAM) was synthesized from different feed ratios of diethylene triamine and methyl acrylate by the simple one‐pot and commercial synthesis method. Reaction procedures and products were intensively studied by FTIR, inherent viscosity and fluorescence techniques. The ill‐structured h‐PAMAM shared similar chemical and physical properties with well defined poly(amido amine) (PAMAM) dendrimers in generation 2 or 3. Its strong fluorescence properties were influenced by pH values, solvents, concentrations, terminal groups and other factors.  相似文献

Here we redesigned the branches of polyamidoamine (PAMAM) dendrimers by moving the amide carbonyl group on the other side of the amide nitrogen atom, transforming the β-alaninyl-amidoethylamine branch, which easily undergoes retro-Michael reactions and renders PAMAMs intrinsically unstable, into a more stable glycyl-amidopropylamine branch. The resulting inverse PAMAM (i-PAMAM) dendrimers have the same carbon framework as PAMAMs and only differ by the position of the carbonyl group. In contrast to PAMAMs which are prepared in solution and are difficult to purify, we synthesize i-PAMAMs using high-temperature solid-phase peptide synthesis by iterative coupling and deprotection of the commercially available N,N-bis(N′-Fmoc-3-aminopropyl)glycine and purify them preparative reverse phase HPLC. Our i-PAMAM dendrimers show no detectable degradation over time. We demonstrate this new class of dendrimers with the synthesis of antimicrobial dendrimers with potent yet non-membrane disruptive activities against both Gram-negative and Gram-positive bacteria.  相似文献

A straightforward strategy for the synthesis and functionalization of polyurethanes (PUs) via the use of alkyne‐functionalized polytetrahydrofuran (PTHF) diols is described. The alkyne groups have been introduced into the PTHF chains by the cationic ring‐opening copolymerization of tetrahydrofuran and glycidyl propargyl ether. These PTHF prepolymers were combined with 1,4‐butanediol and hexamethylene diisocyanate for the synthesis of linear PUs with latent functionalization sites. The polyether segments of the PUs have then been coupled with several types of functionalized azides by the copper‐catalyzed azide‐alkyne “click” chemistry, for example with phosphonium containing azides for their antibacterial properties. © 2011 Wiley Periodicals, Inc. Fabrication and functionalization of hydrogels from well‐defined dendron‐polymer‐dendron conjugates is accomplished using sequential radical thiol‐ene “click” reactions. The dendron‐polymer conjugates were synthesized using an azide‐alkyne “click” reaction of alkene‐containing polyester dendrons bearing an alkyne group at their focal point with linear poly(ethylene glycol)‐bisazides. Thiol‐ene “click” reaction was used for crosslinking these alkene functionalized dendron‐polymer conjugates using a tetrathiol‐based crosslinker to provide clear and transparent hydrogels. Hydrogels with residual alkene groups at crosslinking sites were obtained by tuning the alkene‐thiol stoichiometry. The residual alkene groups allow efficient postfunctionalization of these hydrogel matrices with thiol‐containing molecules via a subsequent radical thiol‐ene reaction. The photochemical nature of radical thiol‐ene reaction was exploited to fabricate micropatterned hydrogels. Tunability of functionalization of these hydrogels, by varying dendron generation and polymer chain length was demonstrated by conjugation of a thiol‐containing fluorescent dye. © 2015 Wiley Periodicals, Inc. A step-by-step synthesis/purification (CC, HILIC, HPLC) of poly(amidoamine) PAMAM dendrimers was performed. MALDI-TOF MS in the linear and reflectron mode was used to analyze the purified samples and byproduct samples of G0-G5 generations of the dendrimers up to the mass of 35 000 Da. DHB/fucose was found to give the best resolution, causing the least fragmentation of the samples. The precise mass number for the ideally branched dendrimers and their “structural errors” was obtained. The profile of the structural errors was established.  相似文献

In this contribution, a versatile approach for the synthesis of functionalized particles for drug delivery is presented, using two nonaggressive standardized procedures. The first procedure considered is the functionalization of an azido‐terminated α‐norbornenyl poly(ethylene oxide) (PEO) macromonomer with an alkyne‐containing active molecule via the copper catalyzed azide alkyne cycloaddition, click type reaction. The functionalized macromonomer is then polymerized by Ring‐Opening Metathesis Polymerization (ROMP) in dispersion to form functionalized particles. The second procedure consists in synthesizing particles by ROMP in dispersed media of norbornene with azido‐terminated α‐norbornenyl PEO macromonomer. The ROMP was initiated by the first generation Grubbs catalyst. Such functionalized core‐shell particles have stealthy properties due to their PEO shell and can be viewed as universal nanocarriers on which any alkyne‐modified active molecule can be grafted by click chemistry. © 2012 Wiley Periodicals, Inc. A range of block copolymers (BCs) consisting of a linear poly(methyl methacrylate) (PMMA) block linked to an aliphatic polyester dendron functionalized with azobenzene moieties have been synthesized by sequential atom transfer radical polymerization (ATRP) and Click Chemistry. Two alkyne‐functionalized PMMA homopolymers with different molecular weights were obtained by ATRP and coupled to generations 2 to 4 of azodendrons bearing an azide group at the focal points. In the case of the azodendron with the highest generation number, the length of the flexible spacer attaching the cyanoazobenzene units to the dendron has also been modified. The coupling of both blocks and purity of BCs were checked by gel permeation chromatography, nuclear magnetic resonance, and infrared spectroscopy. The thermal transitions and liquid crystalline behavior of the BCs were investigated by differential scanning calorimetry and polarized‐light optical microscopy. A morphological study was carried out by transmission electron microscopy, using samples annealed at 115 °C. Photo‐induced anisotropy was induced in thin films of these materials after annealed at 115 °C. The highest stable birefringence values were obtained for the BCs bearing 8 and 16 azobenzene units in the dendritic block. © 2010 Wiley Periodicals, Inc. The synthesis and characterization of four families of anionic carbosilane dendrimers bearing carboxylate, phosphonate, naphthylsulfonate, and sulfate terminal groups prepared by cycloaddition of azide–alkyne catalyzed by copper (CuAAC) are presented here. For the preparation of these anionic carbosilane dendrimers, two strategies starting from azide‐terminated carbosilane dendrimers were followed: (i) click coupling of neutral alkynes followed by derivatization into anionic moieties or (ii) click coupling of anionic alkynes. Both strategies require different reaction conditions in order to accommodate the different substrate polarities. These anionic dendrimers, in general, do not present cell toxicity in vitro until concentration up to 20 µM. Therefore, they can be used in inhibition experiments in concentrations below this limit. We have observed that dendrimers bearing phosphonate groups possess poor anti‐HIV capabilities in vitro in PBMCs, while carboxylate dendrimers can reduce HIV infection levels moderately. On the other hand, sulfate and naphthylsulfonate dendrimers are powerful anti‐HIV agents and their antiviral activity is generation and concentration dependent. © 2014 Wiley Periodicals, Inc. J. A facile click chemistry method of immobilizing surface‐functionalized polymer vesicles on casted polymeric PAN substrates is described. Microporous PAN membranes were subjected to hydrochloric acid hydrolysis to obtain surface carboxylates. The carboxylic groups were activated with EDC/NHS‐solution and were then reacted with propargylamine to introduce alkyne groups for CuAAC reactions. The alkyne functionality of the modified membrane surface was verified by reaction with an azide functional click dye both before and after the immobilization of azide‐functionalized ABA vesicles. The efficient postfunctionalization of the membrane with alkyne allowed quantitative coverage of the membrane surface with a polymersome monolayer, as confirmed by immobilization of polymerzomes loaded with a fluorescent dye. Polymersome monolayers immobilized on alkyne functionalized PAN‐membranes were characterized by cryo‐SEM and monolayers were confirmed by atom force microscopy.
