Abstract— Kinetic studies of the hematoporphyrin–sensitized photooxidation of l -tryptophan and tryptamine at pH 10 in either homogeneous aqueous solutions or in aqueous dispersions of Triton X–100 and cetyltrimethylammonium bromide micelles indicate that the indole substrates are attacked via a mixed type I (electron transfer from triplet dye)/type II (1O2-involving) mechanism. Both reactive intermediates, generated by micelle-solubilized hematoporphyrin, can diffuse to attack substrate molecules located in either the bulk aqueous phase or a different micelle. In particular, incorporation of the substrate into a micelle has only minor effects on its reactivity toward1O2, although the 1O2—indole interaction appears to be more efficient in cationic micelles owing to a favourable orientation of the target with respect to the attacking species. On the other hand, the electron transfer from triplet porphyrin to a micellized substrate is virtually non-operative when the latter is located in an anionic micelle, whereas in neutral or cationic micelles, the efficiency of the process is again controlled by the substrate orientation. Studies of tryptamine photooxidation sensitized by meso-tetra-(4-sulfonato-phenyl) porphine in the presence of sodium dodecylsulphate micelles lend further support to the abovementioned conclusions.  相似文献

Deactivation of excited pyrene incorporated to cationic (cetyltrimethylammonium chloride), anionic (sodium dodecyl sulfate) and neutral (Triton X-100) micelles by tryptophan has been investigated over a wide pH range. Data obtained allow an estimation of the tryptophan association to the micelles, of the tryptophan apparent p K in the micellar solutions, and of the dynamics of tryptophan incorporation to the micellar pseudophase. In particular, the data obtained at pH 7 allow an estimation of the effect of the micellar charge upon the binding capacity of the tryptophan zwitterion.  相似文献

Abstract— The 1,3-diphenylisobenzofuran oxidation by singlet oxygen was investigated in aqueous dode-cyltrimethylammonium chloride micellar solutions using pyrene as photosensitizer. It was found that indole and tryptophan markedly enhanced the furan oxidation in micellar solutions. This enhancement effect strongly depended on the ethanol content in micellar solutions; enhancement effect below about 0.3 mol fraction of ethanol and inhibition effect above this mole fraction. In the former ethanol range, a quenching of pyrene fluorescence by indole or tryptophan and a difference spectrum of furan between micellar solutions in the presence and absence of indole or tryptophan appeared strongly, but they were weakened in the latter ethanol range. The interactions between indole or tryptophan and pyrene and/or furan might be responsible for the enhancement effect observed. Discussion was made in connection with the micelle structure.  相似文献

Abstract— The efficiencies of the excitation–energy transfer from tyrosine to tryptophan residues in eight globular proteins in the native and denatured states are obtained by studying the wavelength dependence of the fluorescence quantum yield. The measurements are made over a wide wavelength range using a computer-controlled spectrophotometer which can measure the fluorescence and absorbance simultaneously in one sample solution (Wada et al. , 1980). The values of the energy transfer efficiencies ranged from 0.17 ± 0.12 to 0.69 ± 0.06 in the native state and from -0.04 ± 0.09 to 0.12 ± 0.06 in the denatured state. These values are considerably lower than the values reported by Kronman and Holmes (1971); in particular, an almost complete absence of energy transfer for the denatured state is shown.  相似文献

Abstract— The electron paramagnetic resonance (EPR) of the triplet excited state of solutions of a series of porphyrins and aromatic amino-acids has been studied at 77°K. It has been shown that for some of the compounds it is possible to observe EPR for transitions with δ M =±1, ±2.
It has been shown that photoreactions in solid solutions of porphyrins and aromatic amino-acids proceed through the triplet excited state.
Reactions of photosensitized deamination of aliphatic amino-acids in solid aqueous solutions at 77°K by aromatic compounds have been studied.  相似文献

There has been considerable interest in the photochemistry of tryptophan in connection with ultraviolet inactivation of enzymes. Earlier flash photolysis work has demonstrated that the hydrated electron (e-aq) is an initial product in the irradiation of indole derivatives, accompanied by a longer-lived transient absorption near 500 nm attributed to an aromatic radical species[1–5]. Similar transients were observed in a recent flash photolysis study of lysozyme[6] in which it was proposed that inactivation is a consequence of electron ejection from 1 to 2 essential tryptophan residues in the active center. However, there has been uncertainty concerning the tryptophan radical structure and its relationship to the triplet state and radical spectra reported for tryptophan photolysis in low-temperature rigid media. This note reports a flash photolysis investigation of L-tryptophan (Trp) and 1-Methyl-L-tryptophan (1-MeTrp) undertaken to clarify these points. Abstract— An excimer of 1-cyanonaphthalene is produced in homogeneous organic solvents and in micelle containing detergent solutions. From solvent effects in homogeneous solution it is concluded that the excimer is relatively polar (dipole moment ∼ 4D). From a comparison with emission from that observed in homogeneous solvents, it is concluded that 1-cyanonaphthalene is solubilized mainly in the Stern Layer of sodium dodecyl sulfate (SDS) and hexadecyltrimethyl ammonium bromide (HDTBr) micelles. The influence of variation of detergent concentration on the excimer to monomer emission ratio and the influence of NaCl concentration on the excimer to monomer emission ratio have been determined. Excimer formation is shown to be a convenient method for determination of the Kraft point of SDS solutions.  相似文献

Abstract— The photoreduction of oxonine, thionine and selenine with the reducing agent allylthiourea was investigated by flash photolysis. The oxonine triplet state was produced by triplet-triplet energy transfer with 9,10-dibromoanthracene as donor. For all three dyes the rate constant of the electron transfer is considerably higher for the acid triplet form than that of the corresponding reaction of the basic triplet form. It is shown that the higher reactivity of the acid triplet can be related to its higher reduction potential which is available from the difference of the pK values of triplet and semiquinone of the dye.  相似文献

The photooxidation of N,N -diethylhydroxylamine (DEHA) by Rose Bengal (RB) has been investigated in micellar and nonmicellar aqueous solutions. We measured the quantum yield of oxygen consumption forming H2O2 and monitored two intermediates, the superoxide and diethylnitroxide radicals. When the pH was vaned, the quantum yield of oxidation remained constant for 6 < pH < 10.5, decreased in acidic pH, and increased considerably in NaOH solution; these changes could be attributed to the protonation and dissociation processes of the >N-OH moiety of DEHA. The formation of diethylnitroxide radical was enhanced by superoxide dismutase or strong alkaline solution. Around neutral pH, the oxidation proceeded mainly via electron transfer from DEHA to the RB triplet ( k q = 107 M -1 s-1) with little 1O2 participation ( kq < 105 M -1 s-1). However, when RB was incorporated into micelles in alkaline solution, the contribution of the singlet oxygen pathway increased at the expense of electron transfer, which was inhibited by the less polar micellar environment. Dark autoxidation of DEHA was accelerated by heavy metal impurities and increased very strongly in NaOH solution.  相似文献

采用电子吸收光谱和光谱-电化学方法研究了中位-四(邻硝基苯基)四苯并卟啉的Fe(Ⅲ)配合物与抗坏血酸在DMF溶液中的均相电子转移反应的动力学和机理。结果表明此电子转移反应来源于抗坏血酸与铁卟啉中心铁离子的轴向配位作用,并将一个电子转移至铁离子。反应速度对铁卟啉和抗坏血酸均为一级,并与抗坏血酸的离解有关。  相似文献

Abstract— Tryptophan has been flash photolysed in deoxygenated and air equilibrated solutions. The kinetics of the subsequent reactions of the tryptophyl radical and the hydrated electron have been elucidated. Oxygen has been found to reduce the primary photoionization yield, indicating a triplet state precursor, but no evidence has been found for a biphotonic process. Appreciable differences in the photoionization quantum yields have been found for tryptophan, alanyl tryptophan and tryptophyl alanine.  相似文献

Abstract— To investigate the existence and energy position of a photoionization threshold. tryptophan (Trp) has been photoionized in desecrated neutral aqueous or alcoholic solution under monochromatic light of variable frequency, in presence of N2O to scavenge the photoelectron.
Present findings and some literature data converge to show the existence of a threshold for the one photon ionization process. This threshold is located at 4.5 ± 0.1 eV and 4.85 ± 0.1 cV for Trp in aqueous and ethanol solutions. respectively, which corresponds to a lowering with respect to the gas phase ionization potential of 3.4 and 3.0 eV.
The photoionization quantum yields for Trp is found about 4 times greater at 250 nm than at Λcx= 265 nm, where φe-4M=0.080±0.025. In such spectral range. at most one photoelectron out of 4–5 escaping geminate recombination would lead to Trp photodegradation in acrated solutions.
These results also point out that the neutral radical Trp. which has been previously observed for Λcx > 275 nm, i.e. below the ionization threshold energy—would not necessarily derive from Trp + deprotonation or cation-electron dissociative recombination. Similarly, the opening of the indole ring with formylkynurenine (FK) formation which is observed under aerobic conditions and Λcx >, 280 nm would not imply an electron attachment on O2 but reactions such as Trp +3O2 or Trp*+3O2 or else

Abstract— The influence of nucleotides or polynucleotides on the photophysics and the photochemistry of tryptophan (Trp) derivatives has been investigated in aqueous solutions using the 265 nm laser flash photolysis technique. In solutions containing mixtures of N -acetyltryptophanamide and uridine monophosphate (UMP) or mercurated dUMP, the Trp triplet and the hydrated electron (eaq) are quenched at almost diffusion controlled rates by the nucleotides leading to uracil reduction. Lysyl-tryptophyl-α-lysine (Lys-Trp-Lys) forms stable complexes in solution with normal or mercurated poly(uridylic acid) [poly(U)]. In the Poly(rU)-Lys-Trp-Lys complex the Trp triplet state is completely quenched, whereas the Trp triplet formation quantum yield is enhanced in complexes with mercurated poly(U). In this last case, the 'heavy atom effect' is characterized by a shortening of the Trp triplet lifetime in agreement with low temperature experiments. Our results also show that photoionization of Trp does occur in the complexed state with both polymers. The eaq lifetime is however longer with the complexed than with the free peptide.  相似文献
