薯蓣皂甙元的极谱催化波及其应用 总被引:4,自引:0,他引:4
采用线性扫描极谱技术,研究了薯蓣皂甙元(C27H42O3,简称皂素)的极谱催化波及应用。结果表明,在pH=5.3±0.5的BR缓冲溶液、0.1mol/LKCl和8×10-3mol/LH2O2的底液中,皂素产生极谱催化波,其一阶导数波的峰电位在-1.05V(vs.SCE)处,峰电流与皂素浓度在1.0×10-8~8.0×10-8mol/L范围内有良好的线性关系,线性回归方程:i′P=7.322×108c+3.919(μA/s),相关系数r=0.9984,检出限为1.0×10-8mol/L;该波为不可逆吸附、平行催化波。建立了测定皂素的新分析方法,并测定了黄姜中皂素的含量。 相似文献

to electrode reaction as oxidized the free radical from one electron reduction of the Δ1,4-3 keto group of prednisone to regenerate the original keto group. The catalytic wave can be used for the determination of
prednisone with the help of conventional polarographic equipment, such as linear-potential scan polarograph. In 0.12 mol l−1 HAc-0.08 mol l−1 NaAc-0.014 mol l−1 K2S2O8 (pH 4.6) supporting electrolyte, the second-order derivative peak current of the catalytic wave was rectilinear to prednisone
concentration in the range of 3.2 × 10−7∼1.6 × 10−5 mol l−1. The detection limit was 8.0 × 10−8 mol l−1.

基于陈皮甙在KIO3存在下产生的极谱催化波,拟定了测定陈皮甙的新方法。在 0.55 mol/L HAc-0.01mol/L NaAc(pH 4.00±0.05)-1.0×10~(-3)mol/L KIO3缓冲溶液中,陈皮甙催化波的峰电位为-1.35 V(vs,SCE),它的一阶导数峰峰电流与陈皮甙浓度在2.6 × 10~(-7)~2.6 × 10~(-6)mol/L范围内呈线性关系(r=0.9970,n=6)。本方法比现有的电化学方法灵敏度高一个数量级。可用于直接测定中草药贯众水提液中陈皮甙含量。 相似文献

应用线性扫描极谱法(LSV)、循环伏安法(CV),在1.0 mol/L HAc-NaAc(pH 4.1)含溶解氧的支持电解质中,对肉桂酸的电化学行为进行了研究。在该支持电解质中于-1.50 V(vs.SCE)处发现肉桂酸产生一灵敏的催化极谱还原峰,其峰电流与肉桂酸的浓度在2.5×10-6~7.5×10-4mol/L范围内呈良好的线性关系(r=0.999 1);检出限为8.0×10-7mol/L,对8.0×10-5mol/L的肉桂酸溶液进行11次平行测定,RSD为1.4%。并采用循环伏安法对肉桂酸的电极反应机理进行了探讨,结果表明该电极反应过程为不可逆过程。该方法已用于测定脉络宁口服液中的肉桂酸。 相似文献

ABSTRACT A polarographic catalytic wave of human serum albumin (HSA) in the presence of KIO3 was reported. In 0.1 M NaAc~HAc buffer (pH4.7) solution, a reduction wave of HSA with peak potential ?0.60 V (vs., Ag/AgCl) resulted from the reduction of five disulfide linkages to sulfhydryl group. In the presence of KIO3, HSA yielded a polarographic catalytic wave at the original potential due to reduction and regeneration of these disulfide linkages. The catalytic wave can be used to determine trace of HSA. In the 0.1 M HAc~NaAc (pH4.7±0.2) ~1×10?3 M KIO3 solution, the peak current was linearly proportional to HSA concentration in the range of 1.5×10?7~7.5×10?7 M. The catalytic wave improved two orders of magnitude in sensitivity compared with corresponding reduction wave. 相似文献

Zebrafish (ZF; Danio rerio) larvae have become a popular in vivo model in drug metabolism studies. Here, we investigated the metabolism of methyl 2-[1-(4-fluorobutyl)-1H-indazole-3-carboxamido]-3,3-dimethylbutanoate (4F-MDMB-BINACA) in ZF larvae after direct administration of the cannabinoid via microinjection, and we visualized the spatial distributions of the parent compound and its metabolites by mass spectrometry imaging (MSI). Furthermore, using genetically modified ZF larvae, the role of cannabinoid receptor type 1 (CB1) and type 2 (CB2) on drug metabolism was studied. Receptor-deficient ZF mutant larvae were created using morpholino oligonucleotides (MOs), and CB2-deficiency had a critical impact on liver development of ZF larva, leading to a significant reduction of liver size. A similar phenotype was observed when treating wild-type ZF larvae with 4F-MDMB-BINACA. Thus, we reasoned that the cannabinoid-induced impaired liver development might also influence its metabolic function. Studying the metabolism of two synthetic cannabinoids, 4F-MDMB-BINACA and methyl 2-(1-(5-fluoropentyl)-1H-pyrrolo[2,3-b]pyridine-3-carboxamido)-3,3-dimethylbutanoate (7′N-5F-ADB), revealed important insights into the in vivo metabolism of these compounds and the role of cannabinoid receptor binding. 相似文献

Neutrophils, also known as polymorphonuclear leukocytes (PMN), are the most common type of white blood cells, comprising about 50-70% of all white blood cells. In the event of inflammatory processes, neutrophils display increased mobility, tissue influx ability, prolonged life span, and an increased phagocytic capacity, constituting the initial participants in the cellular defense of the organism. One of the most important defense systems of neutrophils corresponds to their ability to mediate a strong oxidative burst through the formation of reactive oxygen species (ROS) and reactive nitrogen species (RNS). While oxidative burst is important for the elimination of invading microorganisms, the overproduction of ROS and RNS or the impairment of endogenous antioxidant defenses may result to detrimental effects to the host. The nature and the extent of ROS and RNS production by neutrophils in response to different stimuli is, consequently, a matter of extensive research, with scientific reports showing an enormous variability on the detection methodologies employed. This review attempts to provide a critical assessment of the most common approaches to identify and quantify reactive species formed during the neutrophils’ oxidative burst. The detection mechanisms and performance, as well as advantages and limitations of the different methodologies, are scrutinized, focusing on the use of fluorimetric, chemiluminometric and colorimetric probes. 相似文献

A set of substituted 2-hydroxy-6(7)-methoxynaphthazarins and 7(8)-hydroxypyranonaphthazarins were synthesized. The IR spectra of 2-hydroxy-6-methoxynaphthazarins and 7-hydroxypyranonaphthazarins, on the one hand, and of 2-hydroxy-7-methoxynaphthazarins and 8-hydroxypyranonaphthazarins, on the other hand, have the characteristic nonoverlapping intervals of stretching vibration frequencies of the -hydroxy groups. In recent years, original hybrid assemblies composed of a particle core surrounded by a lipid shell emerged as promising entities for various biotechnological applications. Their broadened bio-potentialities, ranging from model membrane systems or biomolecule screening supports, to substance delivery reservoirs or therapeutic vectors, are furthered by their versatility of composition due to the possible wide variation in the particle nature and size, as well as in the lipid formulation. The synthesis, the characteristics, and the uses of these Lipid/Particle assemblies encountered in the literature so far are reviewed, and classified according to the spherical core size in order to highlight general trends. Moreover, several criteria are particularly discussed: i) the interactions involved between the particles and the lipids, and implicitly the assembly elaboration mechanism, ii) the most suited techniques for an accurate characterization of the entities from structural and physicochemical points of view, and iii) the remarkable properties of the solid-supported lipid membrane obtained. 相似文献

Vanadate and vanadium compounds exist in many environmental, biological and clinical matrices, and despite the need only limited progress has been made on the analysis of vanadium compounds. The vanadium coordination chemistry of different oxidation states is known, and the result of the characterization and speciation analysis depends on the subsequent chemistry and the methods of analysis. Many studies have used a range of methods for the characterization and determination of metal ions in a variety of materials. One successful technique is high performance liquid chromatography (HPLC) that has been used mainly for measuring total vanadium level and metal speciation. Some cases have been reported where complexes of different oxidation states of vanadium have been separated by HPLC. Specifically reversed phase (RP) HPLC has frequently been used for the measurement of vanadium. Other HPLC methods such as normal phase, anion-exchange, cation-exchange, size exclusion and other RP-HPLC modes such as, ion-pair and micellar have been used to separate selected vanadium compounds. We will present a review that summarizes and critically analyzes the reported methods for analysis of vanadium salts and vanadium compounds in different sample matrices. We will compare various HPLC methods and modes including sample preparation, chelating reagents, mobile phase and detection methods. The comparison will allow us to identify the best analytical HPLC method and mode for measuring vanadium levels and what information such methods provide with regard to speciation and quantitation of the vanadium compounds. 相似文献

Biomimetic colloidal particles are promising agents for biosensing, but current technologies fall far short of Nature's capabilities for sensing, assessing, and responding to stimuli. Phospholipid-containing cell membranes are capable of binding and responding to an enormous variety of biomolecules by virtue of membrane organization and the presence of receptor proteins. By tuning the composition and functionalization of simulated membranes, soft colloids such as droplets and bubbles can be designed to respond to various stimuli. Moreover, because lipid monolayers can surround almost any hydrophobic phase, the interior of the colloid can be selected to provide a sensitive readout, for example in the form of optical microscopy or acoustic detection. In this work, we review some advances made by our group and others in the formulation of lipid-coated particles with different internal phases such as fluorocarbons, hydrocarbons, or liquid crystals. In some cases, binding or displacement of stabilizing lipids gives rise to conformational changes or disruptions in local membrane geometry, which can be amplified by the interior phase. In other cases, multivalent analytes can promote aggregation or even membrane fusion, which can be utilized for an optical or acoustic readout. By highlighting a few recent examples, we hope to show that lipid monolayers represent a versatile biosensing platform that can react to and detect biomolecules by leveraging the unique capabilities of phospholipid membranes. 相似文献

The protonation equilibria of 2-amino-N-(2-oxo-2-(2-(pyridin-2-yl)ethyl amino)ethyl)acetamide ([H2(556)–N]) and the complexation of this ligand with Cu(II) Ca(II), Zn(II) and Ni(II) have been studied by glass electrode potentiometry and UV–visible spectrophotometry. From pH ∼2.00–11.00, five models for Cu(II) with the following complexes; MLH, ML, MLH−1, MLH−2 and MLH−3 were generated and observed to describe the experimental data equally well as far as the statistical criteria were concerned. The MLH−2 complex predominates at physiological pH in all five models, while the MLH−1 complex species exists only at low concentration in two models. The coordination in the MLH−2 complex suggested the involvement of one amino, two deprotonated peptides and one pyridyl nitrogen atoms. Molecular mechanics (MM) calculations confirmed the MLH−2 complex as the most stable species. Speciation calculations, using a blood plasma model, predicted that the Cu(II)–[H2(556)–N] complex is able to mobilize Cu(II). Octanol/water partition of CuLH−2 showed that 30% of the complex went into the octanol phase, hence promoting percutaneous absorption of copper. The complex is a poor mimic of native copper–zinc superoxide dismutase. 相似文献

The purpose of this study was to illustrate the mechanism of “enzyme inactivation and toxicity reduction” of Fructus Tribuli (FT) after being heating processed. Ultra-high performance liquid chromatography coupled with triple quadrupole tandem mass spectrometry (UHPLC-MS/MS) was used to quantitatively analyze the contents of four steroidal saponins in crude Fructus Tribuli (CFT) and stir-fried Fructus Tribuli (SFT) under different storage times at room temperature. The enzyme activity of β-D-glucosidase in CFT and SFT were determined and calculated by ultraviolet–visible spectrometry (UV-VIS spectrometry). In addition, the enzyme hydrolysates of FOT and tribuluside A were qualitatively analyzed by ultra-high-performance liquid chromatography coupled with quadruple-time-of-flight mass spectrometry (UHPLC-Q-TOF/MS). The hepatorenal toxicity of spirostanol saponins in FT were further confirmed by in vivo and in vitro experiment. This study confirmed that “enzyme inactivation and toxicity reduction” was one of the reasons why the stir-frying can reduce hepatorenal toxicity of FT, and further enriched the exploration on the mechanism of processing toxicity reduction. 相似文献
