A simple and highly sensitive method called thermal desorption (TD)-gas chromatography-mass spectrometry (GC-MS), which is used for the determination of trace amounts of 4-nonylphenol (NP) and 4-tert.-octylphenol (OP) in water samples, is described. NP and OP in samples are extracted from water samples and concentrated by the stir bar sorptive extraction (SBSE) technique. A stir bar coated with polydimethylsiloxane (PDMS) is added to a 2.0 ml water sample and stirring is carried out for 60 min at room temperature (25 °C) in a headspace vial. Then the extract is high sensitively analyzed by TD-GC-MS without any derivatization step. The optimum SBSE conditions are realized at an extraction time of 60 min. The detection limits are 0.02 ng ml−1 for NP and 0.002 ng ml−1 for OP. The method shows good linearity over the concentration range of 0.1-10 ng ml−1 for NP and 0.01-10 ng ml−1 for OP, and the correlation coefficients are higher than 0.999. The average recoveries of NP and OP are higher than 97% (R.S.D.: 3.6-6.2%) with correction using the added surrogate standards, 4-(1-methyl) octylphenol-d5 and deuterium 4-tert.-octylphenol. This simple, accurate, sensitive and selective analytical method may be used in the determination of trace amounts of NP and OP in tap and river water samples.  相似文献

Extraction methods were developed for quantification of the xenoestrogens 4-tert.-octylphenol (tOP) and bisphenol A (BPA) in water and in liver and muscle tissue from the rainbow trout (Oncorhynchus mykiss). The extraction of tOP and BPA from tissue samples was carried out using microwave-assisted solvent extraction (MASE) followed by solid-phase extraction (SPE). Water samples were extracted using only SPE. For the quantification of tOP and BPA, liquid chromatography mass spectrometry (LC-MS) equipped with an atmospheric pressure chemical ionisation interface (APCI) was applied. The combined methods for tissue extraction allow the use of small sample amounts of liver or muscle (typically 1 g), low volumes of solvent (20 ml), and short extraction times (25 min). Limits of quantification of tOP in tissue samples were found to be approximately 10 ng/g in muscle and 50 ng/g in liver (both based on 1 g of fresh tissue). The corresponding values for BPA were approximately 50 ng/g in both muscle and liver tissue. In water, the limit of quantification for tOP and BPA was approximately 0.1 microg/l (based on 100 ml sample size).  相似文献

A method for the simultaneous measurement of trace amounts of phenolic xenoestrogens, such as 2,4-dichlorophenol (2,4-DCP), 4-tert-butyl-phenol (BP), 4-tert-octylphenol (OP), 4-nonylphenol (NP), pentachlorophenol (PCP) and bisphenol A (BPA), in water samples was developed using stir bar sorptive extraction (SBSE) with in situ derivatization followed by thermal desorption (TD)-gas chromatography-mass spectrometry (GC-MS) analysis. The conditions for derivatization with acetic acid anhydride were investigated. A polydimethylsiloxane (PDMS)-coated stir bar and derivatization reagents were added to 10 ml of water sample and stirring was commenced for 10-180 min at room temperature (25 degrees C) in a headspace vial. Then, the extract was analyzed by TD-GC-MS. The optimum time for SBSE with in situ derivatization was 90 min. The detection limits of 2,4-DCP, BP, OP, NP, PCP and BPA were 2, 1, 0.5, 5, 2 and 2 pg ml(-1), respectively. The method showed good linearity over the concentration ranges of 10, 5, 2, 20, 10 and 10-1000 pg ml(-1) for 2,4-DCP, BP, OP, NP, PCP and BPA, respectively, and the correlation coefficients were higher than 0.99. The average recoveries of those compounds in river water samples were equal to or higher than 93.9% (R.S.D. <7.2%) with correction using the added surrogate standards. This simple, accurate, sensitive and selective method can be used in the determination of trace amounts of phenolic xenoestrogens in river water samples.  相似文献

A sensitive method for the simultaneous determination of phenolic xenoestrogens such as bisphenol A, 2,4-dichlorophenol, 4-tert,-butylphenol, 4-n2-pentylphenol, 4-n-hexylphenol, 4-n-heptylphenol, 4-octylphenol, 4-nonylphenol was developed using reversed-phase LC and coulometric-array detection. Stepwise gradient elution with phosphoric acid in water-acetonitrile was used. The calibration curves were linear in the range of 5.0 (or 10.0)-1000 ng ml(-1) with correlation coefficients of 0.9978-0.9999, the limits of detection were 0.01-0.02 ng ml(-1). Sample clean-up was performed by solid-phase extraction (SPE) using 3M Empore extraction disks. Three commercial sorbents, C18, SDB-XD (styrene-divinylbenzene polymer) and SDB-RPS (sulfonated styrene-divinylbenzene polymer) were compared. The highest recoveries were obtained with SDB-RPS. They were above 70% with a relative standard deviation of less than 15%. The proposed method was applied to the determination of phenolic xenoestrogens in various water samples.  相似文献

Alkylphenols can affect human health because they disrupt the endocrine system. In this study, an analytical method for determining trace amounts of 4-nonylphenol (NP) and 4-octylphenol (OP) in human blood samples was developed. Reversed-phase HPLC with multi-electrode electrochemical coulometric-array detection was used for the determination of NP and OP in plasma and serum samples prepared with a solid-phase extraction method. The separation was achieved using an isocratic mobile phase of 0.7% phosphoric acid-acetonitrile with a C18 reversed phase column. The detection limits of NP and OP were 1.0 and 0.5 ng ml-1, respectively. The recoveries of NP and OP added to human plasma samples were above 70.0% with a relative standard deviation of less than 15.5%. The method was found to be applicable to the determination of NP and OP in various human blood samples such as serum and plasma.  相似文献

The combined gas chromatographic determination of a number of hydroxyl-group containing endocrine disruptors, including 4-octylphenol, 4-nonylphenol, 2,4-dichlorophenol, pentachlorophenol, 4-tert.-butylbenzoic acid, bisphenol-A, 17beta-estradiol and 17alpha-ethynylestradiol, was investigated. Derivatization, required for sensitive determination of these compounds, was carried out using N-methyl-N-(tert.-butyldimethyltrifluoroacetamide). A number of parameters affecting the derivatization reaction, like temperature, time, matrix, solvent, and amount of reagent were studied in detail. Quantitative yields were obtained for real-life extracts after optimization, but the hormones were only mono-substituted. Both solid-phase extraction (SPE) and liquid-liquid extraction were studied as extraction methods, with emphasis on SPE material and effect of pH. Recoveries and RSD for analysis of surface water samples were 58-106 and 6-16% (n=4), respectively, when using SPE, and 109-117 and 6-14% (n=6) when using liquid-liquid extraction. The method developed allows routine analysis of surface water for traces of endocrine disruptors. The limits of detection of were 4-6 ng/l but higher for the hormones.  相似文献

An analytical method based on gas chromatography-mass spectrometry (GC-MS) has been developed to simultaneously determine selected acidic and neutral pharmaceuticals and endocrine-disrupting substances in surface and tap water. Solid-phase extraction (SPE) with Oasis HLB cartridges is followed by derivatization of the target analytes in the eluted extract. Derivatization was systematically optimized by employing a factorial experimental design. More specifically a central composite design was applied to search for the optimal conditions of the derivatization process and it was demonstrated that N-methyl-N-tert-butyl-dimethysilyl-trifluoroacetamide (MTBSTFA) had a better overall performance compared to N,O-bis(trimethylsilyl) trifluoroacetamide (BSTFA). The influence of solvent ratios and elution volumes while using SPE were also studied using a factorial design. The method was developed successfully for most of the selected compounds [i.e. ibuprofen, salicylic acid, gemfibrozil, naproxen, triclosan, propranolol, diclofenac, carbamazepine, 4-octylphenol (OP), 4-nonylphenol (NP), nonylphenol-monoethoxylate (NP1EO), nonylphenoxyacetic acid (NP1EC), estrone (E1), and 17alpha-ethinyloestradiol (EE2)]. Relative recoveries for spiked river and tap water ranged from 47 to 130% and 60-109%, respectively. Typical limits of detection were less than 5 ng/L in tap water and less than 10 ng/L in river water. Twelve target compounds were detected in river and tap water samples using the developed method. This method is currently used in bench-scale drinking water treatment studies.  相似文献

A novel poly(phthalazine ether sulfone ketone) (PPESK) film prepared by immersion precipitation technique was coated on stir bars for sorptive extraction. Scanning electron micrographs showed that the coating has a denser porous surface (about 1 microm in thickness) with a sponge-like sublayer, and the thickness of the coating was 250 microm. The PPESK coated stir bar has high thermostability (290 degrees C) and long lifetime (50 times). The extraction properties of this stir bar were evaluated for the extraction of both polar and semi-polar analytes, including organochlorine compounds and organophosphorus pesticides. The PPESK stir bar was proved to show higher affinity towards polar compounds than that of PDMS coated stir bar and higher sample load compared with corresponding PPESK fiber. It was applied to the determination of organochlorine compounds in seawater samples and organophosphorus pesticides in juices by gas chromatographic analysis. The effect of sample matrix was evaluated at optimized condition of extraction temperature, extraction time and salt concentration. Limits of detection were in the range of 0.05-2.53 ng L(-1) for organochlorine compounds in seawater samples using electron capture detector (ECD), with precisions of less than 11% RSD. Limits of detection for organophosphorus pesticides were in the range of 0.17-2.25 ng L(-1) and 2.47-10.3 ng L(-1) in grape and peach juice, respectively, using thermionic specified detector (TSD), with precisions of less than 12% RSD and 20% RSD, respectively.  相似文献

This study describes the development of a new method for determining p-hydroxybenzoic esters (parabens) in house dust. This optimised method was based on the pressurised hot water extraction (PHWE) of house dust, followed by the acetylation of the extracted parabens, stir bar sorptive extraction (SBSE) with a polydimethylsiloxane stir bar, and finally analysis using thermal desorption-gas chromatography-mass spectrometry (TD-GC-MS). The combination of SBSE and PHWE allows the analytes to be preconcentrated and extracted from the aqueous extract in a single step with minimal manipulation of the sample. Furthermore the in situ acetylation of parabens prior to SBSE improved their extraction efficiency and their GC-MS signal. The method showed recoveries of between 40 and 80%, good linearity, repeatability and reproducibility (<10% RSD, at 100 ng g(-1), n=5), low limits of detection (from 1.0 ng g(-1) for propyl paraben to 2.1 ng g(-1) for methyl paraben) and quantification (from 3.3 ng g(-1) for propyl paraben to 8.5 ng g(-1) for methyl paraben). The proposed method was applied to the analysis of house dust samples. All the target parabens were found in the samples. Methyl and propyl parabens were the most abundant, with concentrations up to 2440 ng g(-1) and 910 ng g(-1), respectively. The high levels of parabens found in the samples confirm the importance of determining organic contaminants in indoor environments.  相似文献

We report a method for determining 4-nonylphenol (NP) and 4-tert-octylphenol (OP) levels in human urine samples by column-switching liquid chromatography-electrospray mass spectrometry after enzymatic deglucuronidation. The method involves enzymatic deconjugation by β-glucuronidase and correction by the stable isotopically labeled internal standard, 4-(1-methyl)octylphenol-d5. The compounds were separated by reversed-phase chromatography with a C18 column, and detected by selected ion monitoring in the negative mode. After adding an internal standard to urine samples, a direct analysis was carried out. The average recoveries of OP and NP were above 85.0% with correction using the added internal standard. The quantitation limit in the urine samples was 0.3 ng ml−1. The method enables the precise determination of standards and may be applied to the detection of trace amounts of OP and NP in human urine samples.  相似文献

A novel stir bar coated with molecularly imprinted polymer (MIP) as selective extraction phase for sorptive extraction of triazine herbicides was developed. The stir bar was prepared by chemically bonding the MIP to the glass bar to improve its stability. A homogeneous and porous structure was observed on the stir bar surface. Extraction performance shows that the MIP-coated stir bar has stronger affinity to the template molecule terbuthylazine as compared with that of the reference stir bar without addition of template. Owning to the shape and structural compatibility, the obtained stir bar also demonstrated specific selectivity to the structural related-compounds of nine triazines, and thus can be applied to simultaneous determination of these compounds from complex samples coupled with high performance liquid chromatography. Four complex samples with different matrix, including rice, apple, lettuce and soil were used to evaluate this proposed method. The limits of detection obtained are in the range of 0.04-0.12 μg L−1, and the recoveries for the spiked rice, apple, lettuce and soil samples were 80.8-107.7%, 80.6-107.8%, 72.0-109.8% and 89.0-114.8% with RSD from 1.2 to 7.9%, respectively. Moreover, this MIP-coated stir bar was firm, durable and can be prepared simply and reproducibly. The developed coating method would be useful to prepare a range of selective stir bars in order to extend the applicability of stir bar sorptive extraction (SBSE) in complex sample analysis.  相似文献

A sensitive method based on pressurized liquid extraction (PLE) and liquid chromatography-electrospray ionization mass spectrometry (LC-ESI-MS) has been developed for the determination in cereal samples of seven endocrine-disrupting compounds: bisphenol A (BPA), 4-tert-butylbenzoic acid (BBA), 4-nonylphenol (NP), 4-tert-butylphenol (t-BP), 2,4-dichlorophenol (DCP), 2,4,5-trichlorophenol (TCP) and pentachlorophenol (PCP). For the PLE procedure, methanol was selected as the extraction solvent. An experimental design approach was applied to optimize other PLE parameters. The recoveries achieved for the all seven compounds were in the 81-104% range, with relative standard deviations of 4-9%. An additional preconcentration step, based on solid-phase extraction (SPE), after the PLE step proved to be a successful way for obtaining a more sensitive method. The detection limits achieved in corn breakfast cereals were in the 0.003-0.013 microg g(-1) range, except for BPA, with a detection limit of 0.043 microg g(-1), for a sample size of 2.5 g. These values are similar to or even lower than currently legislated limits for pesticides in cereals and cereal-based foodstuffs. We also investigated possible contamination during the experimental process by the target compounds released from purified water, plastics, syringes, peristaltic pump tubes, glassware and other laboratory materials in contact with the samples along the analytical process.  相似文献

An organic-inorganic hybrid titania-hydroxy-terminated silicone oil (titania-OH-TSO) stir bar coating was prepared by sol-gel method. The extraction performance of titania-OH-TSO coated stir bar was evaluated and compared with poly(dimethysiloxane) (PDMS), poly(dimethysiloxane)-divinylbenzene (PDMS-DVB), poly(dimethysiloxane)-β-cyclodextrin (PDMS-β-CD) and C(18) coated stir bar with five polar drugs of abuse including amphetamine (PA), methamphetamine (MA), 3,4-methylenedioxyamphetamine (MDA), 3,4-methylenedioxymethamphetamine (MDMA) and ketamine (Ke) as the model analytes. The experimental results revealed that the titania-OH-TSO coated stir bar exhibited highly pH-resistant ability, good preparation reproducibility, superior selectivity and high extraction efficiency for the target compounds. Based on this fact, a new method of titania-OH-TSO coated stir bar sorptive extraction (SBSE) combined with high performance liquid chromatography (HPLC)-ultraviolet visible (UV) detection was developed for the analysis of five drugs of abuse in urine samples. The factors affecting the extraction efficiency of SBSE such as sample pH, desorption solvent, sample volume, extraction time, desorption time, stirring rate and ionic strength were investigated and the optimal extraction conditions were established. Under the optimized conditions, the limits of detection (LODs) for titania-OH-TSO coated SBSE-HPLC-UV determination of five polar drugs of abuse were in the range of 2.3-9.1 μg/L with relative standard deviations (RSDs) ranging from 7.3 to 8.9% (c=300 μg/L, n=6), and all of the target compounds exhibited good linearity over a concentration range of 30-3000 μg/L. The developed method was applied to the determination of amphetamines and Ke in urine samples of drug abusers with satisfactory results.  相似文献

In this paper, a novel molecularly imprinted polymer (MIP) coated stir bar with ractopamine as template by glass capillary filling with magnetic core as substrate was prepared reproducibly. The ractopamine MIP coating was homogeneous and porous with the average thickness of 20.6 μm. The extraction apparatus for the stir bar was improved to avoid coating loss. The MIP-coated stir bar showed better extraction capacity and good selectivity than that of non-imprinted polymer (NIP) coated stir bar to ractopamine and its analogues. The extraction capacities of ractopamine, isoxsuprine, clenbuterol and fenoterol for MIP-coated stir bar were 3.3, 3.1, 2.8 and 2.4 times as much as that of the NIP coated stir bar, respectively. The MIP-coated stir bars could be used at least 40 times without apparent damage and kept in dried air for 8 months without reduce of extraction ability. A method for the determination of β2-agonists in complex samples by MIP-coated stir bar sorptive extraction coupled with high-performance liquid chromatography (HPLC) was developed. The linear ranges were 0.5–40 μg/L for ractopamine and 1.0–40 μg/L for isoxsuprine and clenbuterol. The detection limits were within the range of 0.10–0.21 μg/L. The method was successfully applied to the analysis of β2-agonists in spiked pork, liver and feed samples with the recoveries of 83.7–92.3%, 80.5–90.2% and 73.6–86.2%, respectively. The RSDs was within 2.9–8.1%. The method is very suitable for the determination of trace β2-agonists in pork, liver and feed samples.  相似文献

建立了固相萃取-高效液相色谱-电喷雾质谱分析海水中辛基酚、壬基酚、辛基酚聚氧乙烯醚和壬基酚聚氧乙烯醚的方法。海水样品经C18固相萃取柱富集净化后,以甲醇-水为流动相,在Hypersil GOLD色谱柱上分离,电喷雾质谱在选择离子监测模式下分析目标化合物,采用外标法定量。结果表明,4种化合物的平均加标回收率为59.6%~104.4%,相对标准偏差(RSD, n=3)为1.0%~13.5%;仪器检出限为0.08~3 μg/L。将本方法用于大连海岸6个采样点海水中辛基酚、壬基酚、辛基酚聚氧乙烯醚和壬基酚聚氧乙烯醚的检测发现,样品中壬基酚和壬基酚聚氧乙烯醚均有检出,且油港和海港附近海水中的含量较高。  相似文献

The commonly used solid-phase extraction (SPE) was compared with the recently developed stir bar sorptive extraction (SBSE) to determine EPA-PAHs from precipitation water samples. SPE was performed using C18 PAH-filled cartridges; magnetic stirring rods 10 mm long and coated with 0.5 mm (24 µL) polydimethylsiloxane were used for SBSE. Determination was performed by high performance liquid chromatography and fluorescence detection. The investigations were performed at a concentration level of 30 ng/L, adjusted to actual PAH concentration levels of precipitation water. The recoveries ranged between 54% (SBSE) and 20% (SPE) respectively and about 100%, while the standard deviation ( n = 5) varied in the range of 4.7-13.5% (SBSE) and 3.6-16.9% (SPE) except for the more volatile compounds acenaphthene and fluorene. The detection limits calculated were between 0.4 and 5.0 ng/L. Both techniques were applied to study the PAH contents of precipitation water of several polluted sites in the city of Halle (Saxony-Anhalt, Germany). The advantages and disadvantages of the methods used are discussed.  相似文献

Stir bar sorptive extraction is a powerful technique for the extraction and analysis of organic compounds in aqueous matrices. Carbonyl compounds are ubiquitous components in rainwater, however, it is a major challenge to accurately identify and sensitively quantify carbonyls from rainwater due to the complex matrix. A stir bar sorptive extraction technique was developed to efficiently extract carbonyls from aqueous samples following chemical derivatization by O‐(2,3,4,5,6‐pentafluorobenzyl) hydroxylamine hydrochloride. Several commercial stir bars in two sizes were used to simultaneously measure 29 carbonyls in aqueous samples with detection by gas chromatography with mass spectrometry. A 100 mL aqueous sample was extracted by stir bars and the analytes on stir bars were desorbed into a 2 mL solvent solution in an ultrasonic bath. The preconcentration Coefficient for different carbonyls varied between 30 and 45 times. The limits of detection of stir bar sorptive extraction with gas chromatography mass spectrometry for carbonyls (10–30 ng/L) were improved by ten times compared with other methods such as gas chromatography with electron capture detection and stir bar sorptive extraction with high‐performance liquid chromatography and mass spectrometry. The technique was used to determine carbonyls in rainwater samples collected in York, UK, and 20 carbonyl species were quantified including glyoxal, methylglyoxal, isobutenal, 2‐hydroxy ethanal.  相似文献

This study describes a simple and sensitive method for determining the alkylphenolic compounds, 4-tert-octylphenol (4-t-OP), 4-nonylphenol isomers (4-NPs), and their monoethoxylates (4-t-OP1EO and 4-NP1EOs), in fresh fruits and vegetables. The method involves extracting a sample by a modified Nielson-Kryger steam distillation extraction using n-hexane for 1 h. The alkylphenolic compounds were identified and quantitated by gas chromatography-mass spectrometry (GC-MS) in selected ion monitoring (SIM) mode. Various pH values and amounts of NaCl added to the sample solution were evaluated as extraction conditions. The quantitation limit of this method was less than 0.2 ng/g in 10 g (fresh weight) of sample. Recovery of alkylphenolic compounds in spiked samples exceeded 64% while R.S.D. ranged from 1.0 to 9.8%. Alkylphenolic residues were detected in fresh fruits and vegetables at concentrations of 4-NPs and 4-t-OP from n.d. to 16 ng/g and from n.d. to 4.8 ng/g (fresh weight), respectively. NP1EO and OP1EO were always below the quantitation limit.  相似文献

A simple and fast sample preparation method for the determination of nonylphenol (NP) and octylphenol (OP) in aqueous samples by simultaneous derivatization and dispersive liquid–liquid microextraction (DLLME) was investigated using gas chromatography–mass spectrometry (GC/MS). In this method, a combined dispersant/derivatization catalyst (methanol/pyridine mixture) was firstly added to an aqueous sample, following which a derivatization reagent/extraction solvent (methyl chloroformate/chloroform) was rapidly injected to combine in situ derivatization and extraction in a single step. After centrifuging, the sedimented phase containing the analytes was injected into the GC port by autosampler for analysis. Several parameters, such as extraction solvent, dispersant solvent, amount of derivatization reagent, derivatization and extraction time, pH, and ionic strength were optimized to obtain higher sensitivity for the detection of NP and OP. Under the optimized conditions, good linearity was observed in the range of 0.1–1000 μg L−1 and 0.01–100 μg L−1 with the limits of detection (LOD) of 0.03 μg L−1 and 0.002 μg L−1 for NP and OP, respectively. Water samples collected from the Pearl River were analyzed with the proposed method, the concentrations of NP and OP were found to be 2.40 ± 0.16 μg L−1 and 0.037 ± 0.001 μg L−1, respectively. The relative recoveries of the water samples spiked with different concentrations of NP and OP were in the range of 88.3–106.7%. Compared with SPME and SPE, the proposed method can be successfully applied to the rapid and convenient determination of NP and OP in aqueous samples.  相似文献

A new monolithic coating based on vinylpyrrolidone‐ethylene glycol dimethacrylate polymer was introduced for stir bar sorptive extraction. The polymerization step was performed using different contents of monomer, cross‐linker and porogenic solvent, and the best formulation was selected. The quality of the prepared vinylpyrrolidone‐ethylene glycol dimethacrylate stir bars was satisfactory, demonstrating good repeatability within batch (relative standard deviation < 3.5%) and acceptable reproducibility between batches (relative standard deviation < 6.0%). The prepared stir bar was utilized in combination with ultrasound‐assisted liquid desorption, followed by high‐performance liquid chromatography with ultraviolet detection for the simultaneous determination of diazepam and nordazepam in human plasma samples. To optimize the extraction step, a three‐level, four‐factor, three‐block Box–Behnken design was applied. Under the optimum conditions, the analytical performance of the proposed method displayed excellent linear dynamic ranges for diazepam (36–1200 ng/mL) and nordazepam (25–1200 ng/mL), with correlation coefficients of 0.9986 and 0.9968 and detection limits of 12 and 10 ng/mL, respectively. The intra‐ and interday recovery ranged from 93 to 106%, and the relative standard deviations were less than 6%. Finally, the proposed method was successfully applied to the analysis of diazepam and nordazepam at their therapeutic levels in human plasma. The novelty of this study is the improved polarity of the stir bar coating and its application for the simultaneous extraction of diazepam and its active metabolite, nordazepam in human plasma sample. The method was more rapid than previously reported stir bar sorptive extraction techniques based on monolithic coatings, and exhibited lower detection limits in comparison with similar methods for the determination of diazepam and nordazepam in biological fluids.  相似文献
