 A new sensitive method exploiting solid-phase spectrophotometry is proposed for the determination of cobalt in pharmaceutical preparations. The chromogenic reagent 1-(2-thiazolylazo)-2-naphthol (TAN) was immobilized on C18 bonded silica loaded into a home-made cell with 1.5 mm of optical path for cobalt determination. Cobalt(II) reacts with TAN on C18 material, at pH 6.0–7.5, to give a coloured complex which has maximum absorption at 572 nm. In this way, the sample was passed through the cell and Co(II) ions were quantitatively retained on the solid-phase. After the direct measurement of light-absorption in the solid phase, only the cobalt was eluted with 0.1 mol L−1 hydrochloric acid. The cell was washed with water and then another sample solution could be passed through the cell. The procedure allowed the determination of cobalt in the range of 10–160 μg L−1 with coefficient of variation of 4.7% (n=10) and apparent molar absorptivity of 2.62 × 106 L mol−1 cm−1 using sample volume of 3-mL. Summary.  A new simple, rapid, sensitive, and selective method is proposed for the microdetermination of mercury. Mercury(II) forms insoluble complexes with 2,3-dichloro-6-(2-hydroxy-3,5-dinitrophenylazo)-quinoxaline (1), 2,3-dichloro-6-(5-amino-3-carboxy-2-hydroxy-phenylazo)-quinoxaline (2), 2,3-dichloro-6-(2,7-dihydroxynaphth-1-ylazo)-quinoxaline (3), and 2,3-dichloro-6-(3-carboxy-2-hydroxy-naphth-1-ylazo)-quinoxaline (4) in aqueous acidic medium; the complexes can be made soluble by the action of an anionic surfactant. The solution of the pink coloured compounds is stable for at least 24 h. Beer’s law is obeyed over the concentration range from 0.1 to 2.8 μg · cm−3 of mercury. For a more accurate analysis, Ringbom optimum concentration ranges were found to be 0.25–2.5 μg · cm−3. The molar absorpitivity, Sandell sensitivity, and relative standard deviations were also calculated. A slight interference from Pd2+ and Cd2+ is exhibited by the first three ligands, whereas the last one is only negligibly affected by these metal ions. Strong interference from Ag(I) is evident for all ligands, whereas alkali, alkaline earth, and other transition metals tested posed negligible interference. 15 μg · cm−3 of Cd2+ and Pd2+ or 10 μg · cm−3 of Ag+ can be tolerated if 1.0 mg of potassium bromide and 2.0 mg of citrate as masking agents are added for the determination of 1.5 μg · cm−3 of mercury(II). The method was applied to the determination of methyl- and ethylmercury chloride and the analysis of environmental water samples. Received August 7, 2000. An anodic stripping voltammetric procedure for the determination of Cu(II) at an in situ-plated stannum film electrode (SnFE) was described. The results indicated that the SnFE had an attractive electroanalytical performance, with two distinct voltammetric stripping signals for copper and stannum, and showed the superior advantage for the determination of copper compared with the bismuth film electrode. Several experimental parameters were optimized. The SnFE exhibited highly linear behavior in the concentration range from 1.0 to 100.0 μg L−1 of Cu(II) (r = 0.994) with the detection limit of 0.61 μg L−1 (S/N = 3), and the relative standard deviation for a solution containing 40.0 μg L−1 Cu(II) was 2.2% (n = 8). The procedure has been successfully applied for the determination of Cu(II) in lake water sample.  相似文献

 Simple, rapid, sensitive and selective methods for the determination of Cr(III) and W(VI) with flavonol derivatives in the presence of surface-active agents are proposed. In the pH ranges 3.4–4.2 and 1.9–2.5, the molar absorptivities of Cr(III)-morin-emulsifier S (EFA) and W(VI)-morin-polyvinylpyrrolidone (PVP) systems are 1.13×105 and 2.13×104 L mol−1 cm−1 at 435 and 415 nm, respectively. The Cr(III)-quercetin-PVP and W(VI)-quercetin-cetylpyridinium bromide (CPB) systems are formed in the pH ranges 4–4.6 and 2.2–2.8 with molar absorptivities 1.02×105 and 9.02×104 L. mol−1 cm−1 at 441 and 419 nm, respectively. The linear dynamic ranges for the determination of Cr(III) and W(VI) with morin in the presence of EFA and PVP are 0.03–0.46 and 0.71–8.1 μg mL−1, respectively. The corresponding ranges with quercetin are 0.04–0.54 and 0.14–2.1 μg mL−1 of Cr(III) and W(VI), respectively. The r.s.d (n = 10) for the determination of 0.25 and 3.7 μg mL−1 of Cr(III) and W(VI) with morin and their detection limits are 0.88 and 0.99% and 0.016 and 0.63 μg mL−1, respectively. Using quercetin, the r.s.d (n = 10) for 0.22 and 1.2 μg mL−1 of Cr(III) and W(VI) and their detection limits are 0.92 and 0.91% and 0.015 and 0.08 μg mL−1, respectively. The critical evaluation of the proposed methods is performed by statistical analysis of the experimental data. The proposed methods are applied to determine Cr in steel, non-ferrous alloys, wastewater and mud filtrate and to the determination of W in steel. Received March 8, 1999.  A novel catalytic procedure for zirconium was proposed based on Zr(IV) catalyzed oxidation of gallocyanine by hydrogen peroxide in hexamethylene tetramine-hydrochloric acid buffer medium. The calibration graph is linear for 0–110 ngċml−1, and the detection limit is 0.4 ngċml−1 Zr(IV). Most foreign ions do not interfere with the determination, except for Cu2+, Fe3+ and Cr(VI). The interferences of Cu2+ and Fe3+ could be eliminated by masking with EDTA and mannitol, and that of Cr(VI) by reducing to Cr(III) with ascorbic acid. The typical features of this procedure are that it is sensitive for zirconium, and the determination could be carried out at room temperature. It had been used to the determination of zirconium in zirconium bronze, simulated samples and a certified reference material. The recoveries were 98.6 ∼ 102%, and relative standard deviations (R.S.D.) were 0.9 ∼ 1.5%, respectively. Received September 12, 1999. Revision April 10, 2000.  相似文献

 Two simple, fast, accurate and precise methods for the determination of six phenothiazines and a number of their pharmaceutical formulations are described. The titrimetric method involves the oxidation of the drugs by metavanadate in sulphuric acid medium and titration of vanadium(IV) formed, with cerium(IV) using ferroin indicator and acetone as catalyst. In spectrophotometry, vanadium(IV) formed was reacted with ferriin and the resulting ferroin measured at 510 nm. Phenothiazines in the ranges 5–100 mg and 2.5–25.0 μg mL−1 can be determined by titrimetry and spectrophotometry, respectively, with detection limits of 0.96–2.05 mg and 0.0359–0.0565 μg mL−1, respectively. Both methods were applied successfully to the determination of the studied drugs in pharmaceutical preparations. The reliability of the assays was established by parallel determination by the official methods of British Pharmacopoeia and the results being statistically evaluated. Received September 26, 2000. An “off–on” rhodamine-based fluorescence probe for the selective signaling of Cr(III) has been designed by exploiting the guest-induced structure transform mechanism. This system shows a sharp Cr(III)-selective fluorescence enhancement response in 100% aqueous system under physiological pH value and possesses high selectivity against the background of environmentally and biologically relevant metal ions including Cr(VI), Al(III), Fe(III), Cd(II), Co(II), Cu(II), Ni(II), Zn(II), Mg(II), Ba(II), Pb(II), Na(I), and K(I). Under optimum conditions, the fluorescence intensity enhancement of this system is linearly proportional to Cr(III) concentration from 5.0 × 10−8 to 7.0 × 10−6 mol L−1 with a detection limit of 1.6 × 10−8 mol L−1.  相似文献

Both nitrazepam and flunitrazepam have been determined by high-performance liquid chromatography dual electrode detection (LC-DED) in the reductive–reductive mode, using a carbon fibre veil electrode (CFVE) in conjugation with a glassy carbon electrode. Initial studies were made to optimise the chromatographic conditions. These were found to be 45% acetonitrile-55% acetate buffer (50 mM, pH 4.1) at a flow rate of 1.0 ml/min, employing a Hypersil C18, 5 μm, 250 mm × 4.6 mm column. Cyclic voltammetric studies performed to ascertain the redox behaviour of nitrazepam and flunitrazepam at a CFVE in the optimised mobile phase. Studies showed that similar voltammetric behaviour was obtained to that report at Hg or glassy carbon based electrodes. Further studies were then carried out to identify the optimum conditions required for the LC-DED determination of nitrazepam and flunitrazepam in beverage samples. Hydrodynamic voltammetry was used to optimise the applied potential at the generator and detector cells; these were identified to be −2.40 and −0.25 V, respectively. A linear range of 2.0 to 100 μg ml−1, with a detection limit of 20 ng ml−1 was obtained. A convenient and rapid method for the determination of both nitrazepam and flunitrazepam in beverage sample was developed. Following a simple sample extraction procedure, extracts were examined using the optimised LC-DED procedure. An average percentage recovery of 95.5% (%CV = 4.5%) for nitrazepam and 78.0% (%CV = 8.8%) was achieved for a sample of “Pepsi Max” spiked at 1.0 μg ml−1 nitrazepam, 1.47 μg ml−1 flunitrazepam.  A simple, rapid, accurate and sensitive spectrophotometric method for the determination of norfloxacin (NRF), ofloxacin (OFL) and ciprofloxacin (CPF) is described. This method is based on the formation of an ion pair with sudan III in aqueous-acetone medium [40% (v/v) acetone]. The coloured products are measured at 567, 565 and 566 nm for NRF, OFL and CPF, respectively. The optimization of various experimental conditions is described. Beer’s law is obeyed in the range 0.4–12.0, 0.4–8.8 and 0.4–10.4  ;μg mL−1 of NRF, OFL and CPF, respectively. For more accurate results, Ringbom optimum concentration ranges were 0.8–11.2, 0.6–8.5 and 0.8–10.0 μg mL−1, respectively. The results obtained showed good recoveries of ±1.2, ±1.5 and ±1.7% with relative standard deviations of 0.67, 0.83 and 1.08% for NRF, OFL, and CPF, respectively. The molar absorptivity and Sandell sensitivity were also calculated. Applications of the proposed method to representative pharmaceutical formulations are successfully presented.  A sensitive catalytic method is developed for the spectrophotometric determination of oxalic acid. It is based on the catalytic action of oxalic acid on a new indicator reaction – the oxidation of Bromophenol Blue by dichromate in dilute sulfuric acid medium. The reaction rate is monitored spectrophotometrically by measuring the absorbance at 600 nm after quenching the reaction with sodium hydroxide. A calibration graph from 0.1 to 8.0 μg mL−1 of oxalic acid and a detection limit of 0.04 μg mL−1 was obtained. The applicability of this method was demonstrated by the determination of oxalic acid in water extracts from vegetables such as spinach, mushrooms and fresh kidney beans. Received October 18, 1999.  A simple, rapid, selective, sensitive and economical method has been developed for the simultaneous determination of trace amounts of palladium and nickel in aqueous methanolic medium using 2-(2-thiazolylazo)-5-dimethylam inobenzoic acid as an analytical reagent by first derivative spectrophotometr y. Palladium is determined by measuring base to peak distance at λ=695.0 nm while nickel is estimated by zero crossing method in the mixture. The linearity is maintained between 0.12–1.75 μg mL−1 for palladium and 0.07–1.60 μg mL−1 for nickel in the pH range 2.8–7.2 and 3.4–8.8 respectively. Seven replicate determinations of 1.0 μ g mL−1 of palladium and 0.8 μg mL−1 of nickel in a mixture give a mean signal height of 0.391 for Pd and 0.541 for Ni with relative standard deviations of 0.9% and 1.2%, respectively. The sensitivity of the proposed method is 0.391 (dA/dλ)/(μg mL−1) for palladium and 0.685 (dA/dλ)/(μg mL−1) for nickel. A new chemiluminescence (CL) method combined with flow injection technique is described for the determination of Cr(III) and total Cr. It is found that a strong CL signal is generated from the reaction of Cr(III), lucigenin and KIO4 in alkaline condition. The determination of total Cr is performed by pre-reduction of Cr(VI) to Cr(III) by using H2SO3. The CL intensity is linearly related to the concentration of Cr in the range 4.0 × 10−10–1.0 × 10−6 g mL−1. The detection limit (3s b) is 1 × 10−10 g mL−1 Cr and the relative standard deviation is 1.9% (5.0 × 10−8 g mL−1 of Cr(III) solution, n = 11). The method was applied to the determination of Cr(III) and total Cr in water samples and compared satisfactorily with the official method.  相似文献

 Necessary conditions were established for simultaneous nickel and cobalt determination in environmental samples, such as oak wood and soil, based on cathodic adsorptive stripping voltammetry. Ni(II) and Co(II), complexed with dimethylglyoxime, were determined using a hanging mercury drop electrode. Optimum conditions were found to be: accumulation time 90 s, accumulation potential −0.80 V vs. SCE, supporting electrolyte 0.2 mol dm−3 ammonia-ammonium chloride buffer (pH = 9.4) + 0.05 mol dm−3 NaNO2 and dimethylglyoxime 2 × 10−4 mol dm−3. A linear current-concentration relationship was observed up to 7.51×10 −7 mol dm−3 for Ni(II) and 7.0 × 10−7 mol dm−3 for Co(II). Excess amounts of zinc(II) interfering with cobalt peaks were masked by complexation with EDTA. Wood and soils were mineralized by applying a microwave digestion system, using the mixtures H2O2 + HNO3 or HNO3 + HF, respectively. The developed procedure was tested by analysing international reference materials (BCR 62 Olive Leaves and GBW 08302 Tibet Soil). A mid-infrared enzymatic assay for label-free monitoring of the enzymatic reaction of fructose-1,6-bisphosphatase with fructose 1,6-bisphosphate has been proposed. The whole procedure was done in an automated way operating in the stopped flow mode by incorporating a temperature-controlled flow cell in a sequential injection manifold. Fourier transform infrared difference spectra were evaluated for kinetic parameters, like the Michaelis–Menten constant (K M) of the enzyme and V max of the reaction. The obtained K M of the reaction was 14 ± 3 g L−1 (41 μM). Furthermore, inhibition by adenosine 5′-monophosphate (AMP) was evaluated, and the K MApp value was determined to be 12 ± 2 g L−1 (35 μM) for 7.5 and 15 μM AMP, respectively, with V max decreasing from 0.1 ± 0.03 to 0.05 ± 0.01 g L−1 min−1. Therefore, AMP exerted a non-competitive inhibition.  相似文献

 A direct chemiluminescence method for the determination of DNA has been developed. The method is based on the chemiluminescence light produced by the reaction of DNA, Ru(bipy)3 2+(bipy= 2,2′-bipydyl) and Ce(IV) in sulfuric acid medium. Under the optimum conditions, the response is linearly proportional to the concentration of DNA between 7.0×10−7 and 5.0×10−5g ml−1. A new method allowing the simultaneous determination of arsenic(V), selenium(IV) and selenium(VI) using miniaturised isotachophoresis has been developed. The method uses 0.02 M nitric acid buffered to pH 5.5 with histidine as the leading electrolyte. Using a miniaturised poly(methyl methacrylate) chip device with an integrated conductivity detector, separations of model samples and an industrial process stream sample were achieved. Limits of detection were calculated to be 0.85 mg L−1 for arsenic(V), 0.95 mg L−1 for selenium(IV) and 1.0 mg L−1 for selenium(VI). A method for the analysis of arsenic(III), using a glycolic acid based leading electrolyte to eliminate carbonate interference is also presented.  相似文献

The sorption behavior of a newly synthesized silica gel sorbent with thioetheric sites (STS) towards microgram levels of Au(III), Pt(IV) and Pd(II) was studied. Au(III) is quantitatively (>95%) sorbed in the pH region of 1–9. The sorption of Pt(IV) starts at pH 1 and does not exceed 25% in the entire pH region examined. The sorption of Pd(II) starts at pH 7 and reaches 80% at pH 9. The sorption of Au(III) on STS at pH 1 is not affected by milligram amounts of Ni(II), Zn(II), Fe(III), Cu(II), Pb(II), Cd(II) or Co(II). Au(III) is quantitatively eluted with a 5% aqueous solution of thiourea. The adsorption capacity of STS towards Au(III) is 195 mg g−1. The detection limit (DL) of Au(III) (3σ, n = 9) is 25 ng mL−1. The RSD at a level of 10 × DL is about 2%. Solid-phase extraction of trace amounts of Au(III) on the STS sorbent, followed by its flame AAS determination in the eluate was applied to the determination of gold in geological samples. The results obtained for the gold content in the samples were in good agreement with those of the ICP-AES analysis.  相似文献

Neopterin is a valuable biomarker of cellular immunity associated with various pathological situations such as viral and bacterial infections, autoimmune, cardiovascular, neurodegenerative and malignant disorders. To produce specific antibodies against neopterin for a rapid multi-biomarker-based diagnosis, a novel hapten derivative was synthesized and attached to carrier proteins. The thoroughly characterized conjugates were used for immunization of BALB/c mice and rabbits. The produced monoclonal antibody reached in both direct and indirect enzyme-linked immunosorbent assay (ELISA) format LoD of 0.18 and 0.45 μg L−1, respectively, and was a superior immunoreagent for further biosensor developments with regard to assay sensitivity and material availability. The best polyclonal antibody was somewhat more sensitive in direct ELISA with LoD of 0.05 μg L−1. The optimized ELISA method was evaluated with blood samples collected from patients with renal insufficiency, patients with sepsis, patients without confirmed clinical diagnosis, and healthy volunteers. In plasma samples, neopterin concentrations ranging from 3.2 to 103 μg L−1 could be determined with the monoclonal ELISA whereas twofold lower results were obtained with the polyclonal ELISA. A satisfactory correlation of results was found between the polyclonal ELISA and IBL Neopterin ELISA kit within the concentration range 0.5–16 μg L−1 (R = 0.874; n = 40), and slightly lower correlation was found for monoclonal-based ELISA (R = 0.819; n = 40). These data show that the generated antibodies may be used as functional analytical reagents for the integration into multianalyte biochip detection systems.  相似文献

Aqueous solutions of indium(III) undergo 1e reductions by Sm(II) (E0-1.55 V) and by Yb(II) (−1.05 V), but not by U(III) (−0.52 V). The facile and irreversible reduction, by In+ aq, of Ru(NH3)6 3+ (E0+0.067 V) at reagent concentrations near 10−3 M implies a potential more negative than −0.23 V for In(II,I) and, in conjunction with the known value of −0.44 V for In(III,I), a complementary potential less negative than −0.65 V for In(III,II). These observations, taken together, support le indium potentials EII,I o−0.2 V and EIII,II 0−0.6 V.  相似文献

A sensitive and non chromatographic analytical procedure for the separation of inorganic selenium species in natural water has been performed. A combination of APDC coprecipitation and determination by an absolute thin layer Energy dispersive X-ray fluorescence spectrometry method was used. The influence of various analytical parameters such as element concentration, oxidation states and pH on the recoveries of Se (IV) was examined. The presence of organic matter and bicarbonate anions, typical components in Cuban groundwater samples, was also tested. Negligible matrix effects were observed. At pH 4 a 100% recovery was found for Se (IV). The coprecipitation recovery of the oxidized selenium species (Se (VI)) was null for the selected concentration range (5–100 μg L−1). When the Se (VI) was reduced by heating the solution with 4 mol L−1 HCl, quantitative recovery was also obtained. The determination of total selenium was conducted by the application of the oxidation–reduction process and the analytical procedure for Se (IV). Se (VI) content was calculated as the difference between total selenium and Se (IV). The detection limit was 0.13 μg L−1. The relative standard deviation was lower than 3.5% for 5 μg L−1 of Se (IV). The trueness of the method was verified by using standardized hydride generation-atomic absorption spectrometry technique. The results obtained using the EDXRF technique were in good agreement with the ones determined by HG-AAS. The proposed method was applied to the determination of Se (IV) in surface water and groundwater samples.  相似文献
