Abstract— The formation of pyrimidine dimers on ultraviolet irradiation of TMV-RNA in water is demonstrated in the region from 254 nm to 302 nm. No dimer is present in either unirradiated E. coli ribosomal RNA or TMV-RNA. Dimer formation was also examined in TMV-RNA irradiated in the presence of 5times10-6 M proflavin, in high salt, on dry ice, and in 90% methanol. No correlation of pyrimidine dimers with any biologically defined lesion is presently possible and it is suggested that dimer may not be involved in the inactivation of this material.  相似文献

The wavelength dependence of the formation of two types of DNA damage, single-strand breaks and base changes, was investigated in the UV region from 150 nm to 254 nm using superhelical closed circular (form I) colicin El DNA with synchrotron radiation. Single-strand breaks were measured by agarose gel electrophoresis as a direct conversion of form I DNA to form II DNA (open circular). Base damages were defined as sensitive sites to a crude extract of endonuclease from Micrococcus luteus. They also were estimated using the same conversion, from form I to form II after the DNA was treated with endonuclease. The fluence-effect relationship could be fitted by a simple exponential function for both types of damage. Action spectra were constructed based on the reciprocal of the 37% fluence. The action spectrum for strand breaks increased rather monotonically over three decades from 254 nm to 150 nm in a logarithmic scale, while that for base damages showed a breaking point at 190 nm, being relatively flat above 190 nm. The characteristics of the action spectra are compared with the absorption spectra of the DNA and its main chain moiety calculated on the basis of data on calf thymus DNA and synthetic polynucleotides. Our main conclusions are (1) that the majority of single-strand breaks were induced by the absorption of photon in the sugar-phosphate group in the vacuum-UV region and (2) that the base changes were induced equally well by absorption in the vacuum-UV and in the far-UV region.  相似文献

Abstract— The induction of DNA single-strand breaks in normal human fibroblasts exposed to monochromatic wavelengths from 240–546 nm was measured by the alkaline elution assay. The cells were irradiated at 1°C to prevent both repair of induced breaks and formation of enzymatically induced breaks through excision repair. The cultures were also washed with and irradiated while suspended in phosphate buffered saline to prevent the formation of DNA damaging photoproducts from medium components. The action spectrum for DNA strand breakage was found to exhibit one peak at 265 nm, consistent with DNA absorption, and a second peak at 450 nm. The normalized action spectrum in the visible is similar to the normalized absorption spectrum for riboflavin, a known photosensitizing agent, implicating this molecule as the absorbing chromophore.  相似文献

Abstract— When mammalian cells were exposed to visible-fluorescent light or near-UV light in the medium containing riboflavin and L-tryptophan, single-strand breaks appeared in their DNA. This did not occur if either riboflavin or tryptophan was omitted from the medium. The same effect was observed when cells were added to the pre-irradiated medium, indicating that a stable photoproduct was responsible. The induced DNA lesions were shown to be equally repairable in both excision proficient and defective (xeroderma pigmentosum) human cell lines. The active photoproduct formed was shown to be hydrogen peroxide. The possible relationship between these results and the near-UV induced killing of mammalian cells is discussed.  相似文献

Irradiation of DNA with 193 nm light results in monophotonic photoionization, with the formation of a base radical cation and a hydrated electron (φP1 = 0.048–0.065). Although >50% of the photoionization events initially occur at guanine in DNA, migration of the “hole” from the other bases to guanine occurs to yield predominantly its radical cation or its deprotonated form. From sequence analysis, the data reveal that 193 nm light induces single strand breaks (ssb) in double-stranded DNA preferential 3’ to a guanine residue. However, it has previously been reported that 193 nm light yields very low yields of ssb (<2% of the yield of eaq). The distribution of these ssb at guanine is nonrandom, showing a dependence on the neighboring base moiety. The efficiency of ssb formation at nonguanine sites is estimated to be at least one order of magnitude lower. The preferred cleavage at guanine is consistent with migration and localization of the electron loss center at guanine. It is argued that singlet oxygen and the photoionized phosphate group of the sugar moiety are not major precursors to ssb. At present, the mechanisms of strand breakage are not known although a guanine radical or one of its products remain potential precursors.  相似文献

Abstract— Single-strand breaks are produced in the phosphodiester backbone of ultraviolet-light-irradiated 5–bromodeoxyuridine-containing DNA (BU-DNA) after treatment with alkali. No radiation dependent breakage is observed in thymine-containing DNA (thy-DNA). The relative yields of breaks terminated by 5'-hydroxyl and 5'-phosphate groups was determined by measuring the rate of phosphorylation achieved with polynucleotide kinase in BU-DNA single strands before and after treatment with alkaline phosphatase. The ratio of 5'-phosphate to 5' hydroxyl groups ranged from 2.3 to 2.9 in different experiments. When cysteamine was present during irradiation no new end groups were produced.
In order to identify the nucleoside(s) at the 5'-termini, phosphate groups were removed with alkaline phosphatase and the 5'-hydroxyl groups were phosphorylated with polynucleotide kinase. Electrophoresis of enzymatic digests showed a single 32P-labeled component migrating more rapidly than any of the four usual 5'-mononucleotides. Upon column chromatography this component resolved into a major peak coincident with 5'-dUMP and a lesser unidentified constituent. No 5'-dBUM32P was observed among these nucleotides.  相似文献

The effect of pretreatment with vitamin E on cytotoxicity, DNA single strand breaks, and chromosomal aberrations as well as on mutation induced by ultraviolet-B light (UV-B) was investigated in Chinese hamster V-79 cells. Cellular pretreatment with non-toxic levels of 25 microM alpha-tocopherol succinate (vitamin E) for 24 h prior to exposure resulted in a 10-fold increase in cellular levels of alpha-tocopherol. Using a colony-forming assay, this pretreatment decreased the cytotoxicity of UV-B light. However, alkaline elution assays demonstrated that pretreatment with vitamin E did not affect the number of DNA single strand breaks caused by UV-B light. In addition, UV-B exposure produced a dose-dependent induction of chromosomal aberrations and mutations at the HGPRT locus, and neither of these actions of UV-B was influenced by pretreatment with the vitamin. These results suggest that vitamin E protects cells from UV-B-induced cytotoxicity, possibly through its ability to scavenge free radicals. The results also suggest that the extent of genotoxicity induced by UV-B light may not correlate directly with the cytotoxic action of this wavelength region in sunlight.  相似文献

Intracellular properties of three photosensitizers relevant to photodynamic cancer therapy were compared using cultured human NHIK 3025 cells. When taken up in the cells, the hydrophilic photosensitizer aluminum phthalocyanine tetra sulfonate required about 10 times more quanta of light absorbed per cell to kill 90% of the cells than did the hydrophobic dyes Photofrin II and tetra(3-hydroxyphenyl)porphyrin. In spite of this, the phthalocyanine molecule was the most efficient dye per quantum of excitation light, since the extinction coefficient of the phthalocyanine is more than 10 times higher than that of the two hydrophobic photosensitizers at therapeutic wavelengths. The two hydrophobic dyes had significantly higher fluorescence quantum yields when taken up by cells than when bound to human plasma or human serum albumin, whereas the opposite was true for the hydrophilic phthalocyanine dye--suggesting intracellular aggregation. Finally, the potential genetic toxicities of the drugs in the form of DNA strand breaks were compared. The aluminum phthalocyanine tetra sulfonate photosensitized more DNA strand breaks than did Photofrin II and tetra(3-hydroxyphenyl)porphyrin when compared at the same level of cell survival.  相似文献

Abstract— DNA single-strand breaks were produced in uvrA and uvrB strains of E. coli K-12 after UV (254 nm) irradiation. These breaks appear to be produced both directly by photochemical events, and by a temperature-dependent process. Cyclobutane-type pyrimidine dimers are probably not the photoproducts that lead to the temperature-dependent breaks, since photoreactivation had no detectable effect on the final yield of breaks. The DNA strand breaks appear to be repairable by a process that requires DNA polymerase I and polynucleotide ligase, but not the recA, recB, recF, lexA 101 or uvrD gene products. We hypothesize that these temperature-dependent breaks occur either as a result of breakdown of a thermolabile photoproduct, or as the initial endonucleolytic event of a uvrA , uvrB -independent excision repair process that acts on a UV photoproduct other than the cyclobutane-type pyrimidine dimer.  相似文献

Abstract— The production of UV-induced thymine dimers and their fate upon post-irradiation incubation in the dark was studied in DNA of the intact water plants Wolffia microscopica and Spirodela polyrhiza. The results demonstrate production of thymine dimers, and the ability of the plant cells to remove the dimers from their DNA. The rate of removal is rapid during the first few h of post-irradiation incubation in the dark. It continues at a slower rate for the next 24–48 h, at which time it is essentially complete. The disappearance of thymine dimers in light or in the dark is analogous to the well-known processes in other biological systems, namely, photoreactivation and dark excision.  相似文献

Abstract— DNA from Escherichia coli was irradiated at 254 nm in the presence of silver in order to preferentially enhance the rate of formation of pyrimidine-dimer damage over nondimer damage. The irradiated DNA was treated with formaldehyde in order to measure the unwinding velocity of the defects associated with the pyrimidine dimers. This velocity was found to be 0.18 base pairs/min per pyrimidine dimer, which is nearly 8 times less than that found for a double-strand break (1.37 base pairs/min) obtained by use of sheared DNA whose size was determined by electron microscopy. The rate of reaction of the DNA with formaldehyde varied linearly with the pyrimidine dimer concentration and showed no inflection due to clustering. Treatment of irradiated DNA with UV endonuclease enhanced the formaldehyde reaction by ? 7-fold, consistent with the conversion of a dimer into the faster-reacting defect associated with a single-strand break. These results indicate that the distribution of dimers in DNA is random and not clustered, and that previous interpretations of clustering were based on the false assumption that dimer and chain break defects unwind with similar velocities when treated with formaldehyde.  相似文献

Abstract— A Xenon-chloride excimer laser emitting energy at 308 nm was used to induce single-strand breaks (SSBs, frank breaks plus alkali-labile lesions as assayed by alkaline sucrose sedimentation techniques) in purified DNA from Bacillus subtilis . A fluence response study and a peak pulse intensity study were performed. At a pulse energy of 0.1 mJ/pulse, the radiation induced SSBs in a linear fashion (91 SSB/108 Da per MJ/m2) to a maximum exprimental fluence of 1.28 MJ/m2. The pulse intensity study showed that there were no significant changes in DNA breakage (105 SSB/108 Da) between 2.93 times 109 and 5.86 times 1011 W/m2 (0.11 and 22.0 mJ/pulse) at a constant total fluence of 1.1 MJ/m2 (27000 mJ dose). This study has verified and extended previous work by quantifying the yield of SSBs induced in DNA by this laser radiation.  相似文献

Abstract— –The repair of u.v. damaged DNA in HeLa cells can be detected using the alkaline sucrose gradient technique. As a result of pyrimidine dimer excision single strand breaks are produced in DNA of irradiated cells. Rejoining of these breaks occurs during an 8 hr post-irradiation incubation period and is prevented by hydroxyurea and acriflavine. The inhibition of repair by hydroxyurea can be reversed by a mixture of all 4 deoxyribonucleosides at a concentration that does not reverse the inhibition of total DNA synthesis.  相似文献

A covalently closed, circular, supercoiled plasmid was exposed to singlet oxygen by a separated-surface sensitizer. For each exposure, the quantity of single oxygen entering the DNA target solution was estimated by its oxidation of histidine. After singlet oxygen exposure, some DNA samples were treated to disclose occult lesions. Agarose gel electrophoresis was then used to resolve the unrelaxed supercoils from the relaxed circular and linear species, and all bands were quantitated fluorometrically. Exposure of supercoiled plasmid DNA to singlet oxygen induced frank DNA strand breaks, alkali-labile sites (pH 12.5, 90 degrees C, 30 min), and piperidine-labile sites (0.4 M, 60 degrees C, 30 min), all in a dose-dependent manner. Yields of alkali-labile and piperidine-labile sites ranged from one to four times the frank strand break yield. Replacement of buffered H2O by buffered D2O as the DNA solvent for singlet oxygen exposures increased DNA lesion yields by a factor of 2.6 (averaged over lesion classes). Our data for the detection of frank strand breaks is at variance with published results from studies in which singlet oxygen was derived from a thermolabile endoperoxide dissolved in the DNA solution.  相似文献

Abstract— UVA irradiation of human lymphocytes induces DNA strand breaks and a portion of these breaks are closed at a slower rate than X-ray induced DNA strand breaks and the strand breaks generated during repair of UVC induced DNA lesions. In addition, the yield of DNA strand breaks in lymphocytes pretreated with UVA radiation and given a subsequent exposure with UVC radiation is higher and shows a slower decrease with increasing repair time in comparison with the expected yield based on additivity between UVA and UVC induced DNA strand breaks. This indicates that UVA delays the closure of the intermediate strand breaks formed in the repair process of UVC induced DNA lesions.  相似文献

Abstract— Recent studies have shown that ultraviolet (UV) irradiation induces a systemic effect which enhances subsequent tumor induction by benzo[a]pyrene in a manner which is dependent on the dose of benzo[a]pyrene. The present study was designed to test whether UV-B irradiation renders mice susceptible to subcutaneous or intravenous injection of a regressor tumor induced by benzo[a]pyrene. The sources of UV-B irradiation were banks of 6 Westinghouse FS-40 sunlamps, situated 20 cm above the mouse cages. Female BALB/cAnNHsd received five 30-min dorsal UV-B radiation treatments per week for 12 weeks, resulting in a total dose of approx. 6.4 × 105 J m-2. Two to seven days after termination of UV treatments, syngeneic regressor tumor cells (BP2) induced by benzo[a]pyrene were injected subcutaneously or intravenously into irradiated mice and unirradiated controls. By 38 days post subcutaneous implantation, 24/30 and 3/30 BP2 implants were detectable in the irradiated and unirradiated mice, respectively. Ultraviolet irradiated mice were also unable to reject lung colonies resulting from intravenous administration of BP2 cells, although they were rejected by unirradiated mice. The mean number of lung colonies per mouse was 16- to 35-fold greater in UV irradiated mice than in unirradiated controls, at 14 to 17 days post injection. Thus, UV irradiation rendered mice, with no known exposure to benzo[a]pyrene, susceptible to a subcutaneous or intravenous injection of a regressor tumor induced by benzo[a]pyrene.  相似文献

Abstract— The formation of free radicals in keratin and related proteins by exposure to u.v. light has been examined by electron spin resonance spectroscopy. From a comparison of the spectra obtained and the effects of different wavelengths and the stabilities of the radicals produced, it has been shown that several species of free radicals are produced by the u.v. irradiation of keratin. However it has been possible to identify only those free radicals associated with the cystine residues. The free radicals produced in keratin by exposure to shorter wavelength irradiation (below 3250 Å) were found to be quite different to those produced at longer wavelengths.  相似文献

Abstract— The induction of single-strand breaks (alkali-labile bonds plus frank breaks) in the DNA of Bacillus subtilis irradiated in vivo by monochromatic UV light at wavelengths from 254 to 434 nm was measured. The spectrum consists of a major far-UV (below 320 nm) component and a minor near-UV shoulder. A mutant deficient in DNA polymerase I accumulates breaks caused by near-UV (above 320 nm) wavelengths faster than the wild-type strain proficient in polymerase I. Measurable breaks in extracted DNA are induced at a higher frequency than those induced in vivo. Anoxia, glycerol, and diazobicyclo (2.2.2.) octane inhibit break formation in extracted DNA. Alkali-labile bonds induced by 365-nm UV radiation are largely (78%) covalent bond chain breaks, the remainder consists of true alkali-labile bonds, probably apurinic and apyrimidinic sites.  相似文献
