This study investigates the comprehensive organosulphur compounds and evaluation of antioxidant activities in Allium hookeri, called hooker chive. The non-volatile and volatile organosulphur compounds were determined by high-performance liquid chromatography-photodiode array detector (HPLC-PDA) and gas chromatography-mass spectrometry (GC-MS), respectively. Among 11 non-volatileorganosulphur compounds, methiin and cycloalliin were major compounds. A total of 42 volatile compounds were identified and allyl methyl sulphides and dimethyl sulphides were primarily volatiles. The total phenols ranged from 19.32 to 71.19 mg gallic acid equivalents (GAE)/g. The antioxidant capacities expressed IC50 values ranged from 9.59 to 38.54 mg/mL for DPPH radical scavenging activity and from 0.39 to 2.36 for ferric reducing antioxidant power assay. The results were proposed as useful tools for evaluating the nutraceutical of hooker chive.  相似文献

Abstract

Rhaponticum acaule (L.) DC. is a medicinal plant commonly used for the treatment of some illnesses such as gastrointestinal infections. In this work, we report the composition of different parts of this plant on phenolic compounds, their quantification, and antioxidant activity. The obtained results reported that methanolic extracts of the three parts studied revealed high phenolic contents. For flavonoid contents, the highest contents were reported in organic extracts of leaf part. In addition, results obtained from the study of the antioxidant activity showed that methanolic extract of root presented the highest activity, in DPPH? scavenging ability test with an IC50 of 0.31?±?0.04?mg/mL and in FRAP test with an EC50 of 1.06?±?0.02?mg/mL. The RP-HPLC-PDA analysis revealed the presence of five phenolic acids (sinapic, caffeic, chlorogenic, ferulic and syringic acids), one flavanone (naringenin), one flavonol (rutin) and vanillin.  相似文献

The effect of gamma irradiation on the content of total phenolic compounds, especially quercetin (Q), in onion (Allium cepa L.) skin was investigated. Onion skin extracts contained two predominant flavonoid compounds, Q and quercetin-4′-glucoside (Q4′G). After 10 kGy gamma irradiation, the yield of Q in the extracts increased significantly from 36.8 to 153.9 μg/ml of the extract, and the Q4′G content decreased slightly from 165.0 to 134.1 μg/ml. In addition, the total phenolic compound content also increased after irradiation at 10 kGy, from 228.0 μg/g of fresh weight to 346.6 μg/g; negligible changes (237.1–256.7 μg/g) occurred at doses of up to 5 kGy. As we expected, radical-scavenging activity was enhanced remarkably (by 88.8%) in the 10 kGy irradiated sample. A dose-dependent increase in the peak intensity of the electron paramagnetic resonance (EPR) spectra was observed in all irradiated samples, with a maximum increase at 10 kGy. The intensity relative to that of the control was 0.15, and it increased to 1.10 in 10 kGy irradiated samples. The optimum gamma irradiation dose, which is sufficient to break the chemical or physical bonds and release soluble phenols of low molecular weight in onion skin, is about 10 kGy.  相似文献

The present study was carried out to determine some biochemical characteristics, in particular the total polyphenol content and the free radical scavenging activity, of the extracts recovered from bulbs and aerial parts (these last often considered as by-products) of two landraces of A. ampeloprasum var. holmense cultivated in Southern Italy. For the first time, the capacity of the extracts of these landraces to inhibit the formation of biofilm of different Gram-negative and Gram-positive bacteria and to affect the metabolism of the cells present within the bacterial biofilm was evaluated. All extracts exhibited an amount of total polyphenols not lower than 2.86 mg/g of dried product and revealed a noteworthy antioxidant activity, with EC50 values not exceeding 4.95 mg. In both cases, the aerial parts extracts were more effective than the bulb extracts, which also showed a minor amount of total polyphenols. The extracts inhibited mainly the adhesive capability of Pseudomonas aeruginosa and Staphylococcus aureus, by 95.78% and 85.01%, respectively. The extracts demonstrated to inhibit also the metabolism of the bacterial cells reaching levels up to 90%. Finally, as assessed by the assays performed on the 24-h preformed biofilms, all the extracts were also capable to cause a reduction in bacterial biomass and to affect their metabolism.  相似文献

Allium schoenoprasum L. (family Amaryllidaceae), commonly known as chives has great culinary value besides being used as ethnomedicine. This review emphasises on phytochemistry and pharmacological activities of A. schoenoprasum, and discusses the future opportunities for systematic investigations. Scientific evaluation of chives validates its traditional claims and demonstrates diverse pharmacological potential including an anti-inflammatory, anticancer, antioxidant, anthelmintic and antihypertensive. Though phytochemical studies revealed the presence of sulphur and phenolic compounds, flavonoids, saponin and steroidal glycosides yet methodical research to identify bioactive compounds is required. This review confirms the medicinal importance of A. schoenoprasum and could stimulate future research on its unexplored aspects, especially identification of bioactive compounds and related mechanisms and safety, which might develop it as a drug.  相似文献

The hypoglycemic and antioxidants activities of the methanol extract of Lepidium sativum seeds was tested in alloxan-induced diabetic male rats. Thirty male albino rats weighing 190–200 g were divided into five groups as follows: negative control, positive control and three diabetic groups treated with three concentrations of L. sativum methanol extract for four weeks. Induction of hyperglycemia in the positive control group increased blood glucose, glycated hemoglobin A1c, immunoglobulins, liver enzyme, lipid peroxide and kidney function, total cholesterol, low-density lipoproteins, very low-density lipoproteins and decreased antioxidants and high-density lipoproteins compared with the negative control. Furthermore, pancreas tissues showed pathological changes compared with the negative control. Treating the diabetic rats with L. sativum methanol extract decreased blood sugar and restored all biochemical and histological changes to the normal. It could be concluded that L. sativum methanol extract succeeded in controlling diabetes, increasing antioxidants and ameliorating lipid profile.  相似文献

The influence of natural drying (ND), hot-air drying (HD), vacuum drying (VD), infrared drying (ID) and freeze drying (FD) on bioactive compounds and bioactivities of Isodon rubescens (Hemsl.) was investigated in this study. The results showed that different drying methods resulted in the differences in bioactive compositions’ content, antioxidant and antibacterial activities of extracts from I. rubescens. FD sample possessed the highest content of total phenolics, total flavonoids and several main phenolic compounds, as well as the stronger antioxidant and antibacterial activities, followed by ND, HD and VD, the lowest for ID samples. For this reason, freeze drying would seem to be more advisable for the drying I. rubescens, and future studies could focus on the quality evaluation and optimising various drying parameters.  相似文献

Abstract

The purpose of this study was to conduct a comparative evaluation of some biological properties of methanol and water extracts of leaves of five Juniperus taxa growing in Turkey: J. communis L. var. communis (Jcc), J. communis L. var. saxatilis Pall. (Jcs), J. drupacea Labill. (Jd), J. oxycedrus L. subsp. oxycedrus (Joo), J. oxycedrus L. subsp. macrocarpa (Sibth. & Sm.) Ball. (Jom). The antioxidant properties were examined in vitro; both in the DPPH and in the reducing power tests, Joo methanol extract resulted the most active (IC50?=?0.09?±?0.01mg/mL and ASE/mL?=?2.56?±?0.06). In the TBA assay, Jcs methanol extract exhibited the highest activity (IC50?=?4.39?±?0.47?μg/mL). The extracts displayed bacteriostatic activity against Staphylococcus aureus, and Jd methanol extract resulted the most effective (MIC?=?19.53?μg/mL); no effect on the S. aureus biofilm formation was observed. The extracts resulted non-toxic in the Artemia salina lethality bioassay. Finally, the phenolic profile of the methanol extracts was characterized by HPLC-PDA/ESI-MS.  相似文献

In an extensive search for bioactive compounds from plant sources, the quantitative and qualitative characterisation of the compounds present in Cynoglossum cheirifolium extracts was studied. Total phenolic and flavonoid contents were determined by spectrophotometric techniques. In vitro antioxidant and radical scavenging profiling was determined through DPPH? scavenging activity and Ferric reducing antioxidant power (FRAP). Our study showed that leaves produce more phenolic compounds, followed by flowering aerial part. The butanolic fraction obtained from leaves extract exhibited the highest total phenolics (78.65 ± 3.58 mg GAE/g DW) and flavonoids (22.15 ± 4.66 mg CE/g DW). In contrast, this fraction displayed the highest DPPH? scavenging ability with IC50 values of 0.06 ± 0.003 mg/mL. The RP-HPLC-PDA analysis of phenolic compounds from leaves of C. cheirifolium lets to identify: rosmarinic acid, ferulic acid, caffeic acid, p-coumaric acid, syringic acid, sinapic acid and rutin. The obtained results indicate that this plant represent a valuable source of natural antioxidants.  相似文献

In the present paper, for the first time, we are interested to separate and identify some bioactive fractions isolated from the roots of a Saharan plant Anabasis articulata, which is widely used in traditional medicine against cancer. The crude methanolic extract of the roots was fractionated on column chromatography, and eluted with dichloromethane/methanol each time with increasing polarity of methanol; 17 fractions were separated. One of these fractions named F12 showed more antioxidant activity to scavenge DPPH free radical with percentage inhibition of 95.29%. F12 was separated by thin-layer chromatography (TLC) to give 12 compounds. A second preparative TLC of compound 2, which has antioxidative activity of 74.92%, provided the three phenolic acids M1, M2 and M3, analysed by high-performance liquid chromatography and UV–visible spectrophotometry.  相似文献

Allium cepa L. is a highly consumed garden crop rich in biologically active phenolic and organosulfur compounds. This study aimed to assess the in vitro bioaccessibility and anti-inflammatory effect of a chemically characterized A. cepa extract rich in quercetin and its derivatives. Different varieties of A. cepa were studied; based on the highest total phenolic content, the “Golden” variety was selected. Its extracts, obtained from the tunicate bulb, tunic, and bulb, were subjected to determination of quercetin and its derivatives with LC-MS analysis and based on the highest total quercetin content, the tunic extract was utilized for further experiments. The extraction method was optimized through a design of experiment (DoE) method via full factorial design, which showed that 40% ethanol and 1 g tunic/20 mL solvent are the best extraction conditions. HPLC analysis of the optimized tunic extract identified 14 flavonols, including 10 quercetin derivatives. As far as in vitro bioaccessibility was concerned, the increases in some quercetin derivatives following the gastro-duodenal digestion process support the bioaccessibility of these bioactive compounds. Moreover, the extract significantly inhibited the production of PGE2 in stimulated J774 cell lines, while no effects of the tunic extract were observed against the release of IL-1β, TNF-α, and nitrites. The study provided insights into the optimized extraction conditions to obtain an A. cepa tunic extract rich in bioavailable quercetin derivatives with significant anti-inflammatory effects against PGE2.  相似文献

Ruta chalepensis L. (Rutaceae) leaf extract was investigated for its chemical profile and antioxidant and hypoglycaemic properties. The antioxidant effects were investigated by 2,2-diphenyl-1-picrylhydrazyl (DPPH), β-carotene bleaching, and metal chelating activity assays. The carbohydrate-hydrolysing enzymes inhibition assay was used to test the hypoglycaemic potential. R. chalepensis showed a high content of hesperidin and rutin with values of 591.9 and 266.7 mg/g dry extract, respectively. The extract exhibited a promising protection of lipid peroxidation (IC50 value of 16.9 μg/mL) and inhibited both α-amylase and α-glucosidase enzymes in a concentration-dependent manner. The highest activity was found against α-amylase (IC50 value of 69.0 μg/mL). The obtained results support the use of R. chalepensis leaves as healthy food ingredients.  相似文献

Welsh onion (Allium fistulosum L.) is usually used to enhance the flavor characteristics of various foods. Volatile compounds in Welsh onions, including sulfur-containing compounds, may vary during heat process and storage. Accordingly, the changes in the volatile compounds in Welsh onions, subjected to heat and antioxidant (ascorbic acid and glutathione) treatments during storage, are investigated in the present study. The majority of sulfur-containing compounds in Welsh onions showed significant differences between the untreated Welsh onions and heated Welsh onions. During the heating of the Welsh onions, some sulfur-containing compounds, such as 2-methylthiirane, 1-(methyldisulfanyl)prop-1-ene, 1-[[(E)-prop-1-enyl]disulfanyl]propane, 1-(propyltrisulfanyl)propane, 1-[[(E)-prop-1-enyl]trisulfanyl]propane, and (methyltetrasulfanyl)methane, showed significant differences between the untreated and heated Welsh onions (p < 0.05). In addition, partial least square discriminant analysis (PLS-DA) was applied to discriminate the heated Welsh onion samples added with different antioxidants. The heated Welsh onion samples added with ascorbic acid was mainly associated with 2-phenylacetaldehyde, acetic acid, methylsulfanylmethane, prop-2-ene-1-thiol, undecan-2-one, and (2E,4E)-deca-2,4-dienal. Moreover, the key volatile compounds in the heated Welsh onion samples added with glutathione were 3-ethylthiophene, 1-(methyldisulfanyl)-1-methylsulfanylpropane, 1-methylsulfanylpentane, 2-prop-2-enylsulfanylpropane, and 1-propan-2-ylsulfanylbutane.  相似文献

The aim of this study was to evaluate the involvement of nanoparticles prepared from Allium cepa L. as anti-inflammatory agents. In the present study, we identified nanoparticles from Allium cepa L. using the ultracentrifugation exosome purification method. The nanoparticles were referred to as 17,000× g and 200,000× g precipitates, and they contained quercetins, proteins, lipids, and small-sized RNA. The nanoparticles inhibited nitric oxide production from lipopolysaccharide (LPS)-stimulated RAW264 cells without cytotoxic properties. Cellular incorporation was confirmed by laser microscopic observation after PKH26 staining. The inhibition of caveolae-dependent endocytosis and macropinocytosis significantly prevented the incorporation of the nanoparticles but had no effect on the inhibition of nitric oxide in RAW264 cells. Collectively, the identified nanoparticles were capable of inhibiting the LPS response via extracellular mechanisms. Taken together, the way of consuming Allium cepa L. without collapsing the nanoparticles is expected to provide an efficient anti-inflammatory effect.  相似文献

In our study, Allium subhirsutum L. (AS) was investigated to assess its phenolic profile and bioactive molecules including flavonoids and organosulfur compounds. The antioxidant potential of AS and wound healing activity were addressed using skin wound healing and oxidative stress and inflammation marker estimation in rat models. Phytochemical and antiradical activities of AS extract (ASE) and oil (ASO) were studied. The rats were randomly assigned to four groups: group I served as a control and was treated with simple ointment base, group II was treated with ASE ointment, group III was treated with ASO ointment and group IV (reference group; Ref) was treated with a reference drug “Cytolcentella® cream”. Phytochemical screening showed that total phenols (215 ± 3.5 mg GAE/g) and flavonoids (172.4 ± 3.1 mg QE/g) were higher in the ASO than the ASE group. The results of the antioxidant properties showed that ASO exhibited the highest DPPH free radical scavenging potential (IC50 = 0.136 ± 0.07 mg/mL), FRAP test (IC50 = 0.013 ± 0.006 mg/mL), ABTS test (IC50 = 0.52 ± 0.03 mg/mL) and total antioxidant capacity (IC50 = 0.34 ± 0.06 mg/mL). In the wound healing study, topical application of ASO performed the fastest wound-repairing process estimated by a chromatic study, percentage wound closure, fibrinogen level and oxidative damage status, as compared to ASE, the Cytolcentella reference drug and the untreated rats. The use of AS extract and oil were also associated with the attenuation of oxidative stress damage in the wound-healing treated rats. Overall, the results provided that AS, particularly ASO, has a potential medicinal value to act as effective skin wound healing agent.  相似文献

Inula graveolens (L.) Desf. is an annual aromatic herb which has various uses on alternative medicine in many region of the world. In this study, antioxidant activities of ethanol and water extracts of the plant leaves were determined by in vitro DPPH method and phenolic composition of the plant sample was determined by LC-MS/MS analysis. The results showed that chlorogenic acid, quinic acid, hyperoside, protocatechuic acid and quercetin were the major phenolic compounds among the 27 standard compounds. The significant antioxidant capacity of the plant might be related with the high abundance of phenolic compounds.  相似文献

Cellulose microfibril orientation in plant cell walls changes during cell expansion and development. The cellulose microfibril orientation in the abaxial epidermis of onion scales was studied by atomic force microscopy (AFM) and sum frequency generation (SFG) vibrational spectroscopy. Onion epidermal cells in all scales are elongated along the onion bulb axis. AFM images showed that cellulose microfibrils exposed at the innermost surface of the abaxial epidermis are oriented perpendicular to the bulb axis in the outer scales and more dispersed in the inner scales of onion bulb. SFG analyses can determine the orientation of cellulose microfibrils averaged over the entire thickness of the cell wall. We found that the average orientation of cellulose microfibrils inside onion abaxial epidermal cell walls as revealed by SFG is similar to the orientation observed at the innermost cell wall surface by AFM. The capability to determine the average orientation of cellulose microfibrils in intact cell walls will be useful to study how cellulose microfibril orientation is related to biomechanical properties and the growth mechanism of plant cell walls.  相似文献

Aralia elata (Miq.) Seem is a wild medicinal and culinary plant. The newly grown shoots are edible, and the stem and root bark are traditional materials used for medicinal extraction. However, few studies have examined the quality and biological activity of the fruit. In this study, we compared the nutritional and phytochemical composition and antioxidant activity of A. elata fruit (AF) picked from five regions (Harbin, Benxi, Raohe, Jiaohe, and Mudanjiang) in Northeast China. AF picked from Harbin had the highest saponin, anthocyanin, and flavonoid contents. UHPLC-MS/MS analyses showed that araloside A was one of the main saponin monomers in AF. The highest arloside A concentration was detected in fruits picked from Harbin and the lowest concentration was detected in fruits picked from Benxi. The relative reactive oxygen species production in cells after pretreatment with AFs from Harbin (50 μg/mL) was 0.78 ± 0.01, second only to those from Raohe. AFs (50 μg/mL) from Harbin had the highest catalase activity (43.61 ± 0.44 U/mg protein), followed by that of the AF collected from Jiaohe, Benxi, Mudanjiang and Raohe. The malondialdehyde levels of the AFs (50 μg/mL) obtained from Benxi, Raohe, Jiaohe, and Mudanjiang were significantly higher than those of the AF obtained from Harbin (2.67 ± 0.15 nmol/mg protein). Compared with AFs from Benxi, Raohe, Jiaohe, and Mudanjiang, the strongest H2O2 scavenging ability (144.95 ± 2.89 mmol TE/100 g dm) was observed for AF from Harbin. The ferric reducing antioxidant power of AFs ranged from 7.70 to 4.52 mmol TE/100 g dm in the order of Harbin > Mudanjiang > Raohe > Jiaohe > Benxi. The results provided support for the selection of the best quality AF and the expanded cultivation of A. elata.  相似文献

Metabolite profile, antioxidant and antinociceptive activities of Syringa vulgaris bark and leaf methanolic extracts were investigated. By means of HPLC-DAD-ESI-TOF and HPLC-DAD-ESI-MS/MS, a total of 33 phenolics were identified, including 15 secoiridoids, 6 phenylpropanoids, 3 flavonoids, 3 lignans and 6 low molecular weight phenols. Validated quantitative analysis show that syringin (2.52%) and rutin (1.13%) are the main phenolic compounds in bark and leaf, respectively. Notable radical scavenging and antinociceptive activities of the bark and leaf extracts were confirmed by in vitro DPPH and ABTS●+ assays and by in vivo hot-plate method in mice, respectively. Our results could lay the scientific basic of future clinical perspectives of lilac bark and leaf.  相似文献
