Abstract Trillium govanianum Wall. ex D. Don (Melanthiaceae alt. Trilliaceae), is native to the Himalayas. The present study, for the first time, was undertaken to explore the antimicrobial potential, to determine the minimum inhibitory concentration (MIC) values of the methanol extract of the roots of Trillium govanianum and its solid phase extraction (SPE) fractions by using resazurin microtiter assay (REMA) against Gram positive and Gram negative bacterial registered strains and to carry out phytochemical analysis. The remarkable amount of gallic acid equivalent phenolic and quercetin equivalent flavonoid content was manifested by MeOH extract (20.27?±?3.03?mg GAE/g DW and 9.25?±?0.50?mg QE/g DW respectively). The GC/MS analysis revealed the presence saturated and unsaturated components. Considerable level of antibacterial potential against Gram-positive bacteria (MIC: 2.5-0.009?mg/mL) than against Gram-negative bacteria (MIC: 2.5-0.165?mg/mL) were observed. The use of microtiter plates has the advantage of lower cost, fast and quantitative results. 相似文献

Asparagus adscendens Roxb. (Asparagaceae), is native to the Himalayas. The present study, for the first time, was undertaken to explore the antimicrobial potential, to determine the minimum inhibitory concentration (MIC) values of the methanol extract of the roots of A. adscendens and its solid-phase extraction (SPE) fractions using resazurin microtitre assay against Gram-positive and negative bacterial-registered strains and to carry out HPLC-photodiode array analysis of the SPE fractions. The methanol extract and all SPE exhibited considerable level of antibacterial potential against Gram-positive bacteria (MIC: 2.5–0.009 mg/mL) than against Gram-negative bacteria (MIC: 1.25–2.5 mg/mL). The use of microtitre plates has the advantage of lower cost, fast and quantitative results. Like other Asparagus species, the presence of phenolic compounds in all SPE fractions was evident in the HPLC-PDA data. 相似文献

Phosphorylation events catalyzed by protein kinases represent one of the most prevalent as well as important regulatory posttranslational modifications, and dysregulation of protein kinases is associated with the pathogenesis of different diseases. Therefore, interest in developing potent small molecule kinase inhibitors has increased enormously within the last two decades. A critical step in the development of new inhibitors is cell-free in vitro testing with the intention to determine comparable parameters like the commonly used IC50 value. However, values described in the literature are often biased as experimental setups used for determination of kinase activity lack comparability due to different readout parameters, insufficient normalization or the sheer number of experimental approaches. Here, we would like to hold a brief for highly sensitive, radioactive-based in vitro kinase assays especially suitable for kinases exhibiting autophosphorylation activity. Therefore, we demonstrate a systematic workflow for complementing and validating results from high-throughput screening as well as increasing the comparability of enzyme-specific inhibitor parameters for radiometric as well as non-radiometric assays. Using members of the CK1 family of serine/threonine-specific protein kinases and established CK1-specific inhibitors as examples, we clearly demonstrate the power of our proposed workflow, which has the potential to support the generation of more comparable data for biological characterization of kinase inhibitors. 相似文献

By optimizing the extraction and analytical conditions, a reliable and precise HPLC method coupled with photodiode array detection (HPLC–DAD) has been developed for the identification and quantification of three major aconitine-type alkaloids (aconitine, mesaconitine, hypaconitine) in the roots of Aconitum carmichaelii Debeaux. The qualitative analysis of the plant material was carried out by LC-APCI-MSn. By means of this method, 26 lipo-alkaloids were also identified from the roots of A. carmichaelii. The effect of processing on aconitine-type alkaloids, lipo-alkaloids and pure aconitine was studied. As part of our investigation, two lipo-alkaloids, 14-benzoylaconine-8-palmitate and 14-benzoylaconine-8-linoleate were produced semisynthetically. The COX-1, COX-2 and LTB4 formation inhibitory activity of aconite root extracts and different types of diterpene alkaloids and the toxicity of lipo-alkaloids were also investigated. 相似文献

This study reports the results of Koc and soil DT50 experiments used for the definition of the leaching potential through the GUS Index of five metabolites: endosulfan sulphate (ES) from endosulfan, 2,6-dichlorobenzoic acid (DBA) from dichlobenil, 3,4-dichloroaniline (DCA) from propanil, 2,4-dichlorophenol (DCP) from 2,4-D and 4-chloro-2-methylphenol (CMP) from MCPA. The Koc values for all the compounds were lower than 300, indicating high potential mobility in soil. Soil DT50 values were around 24 days for DBA and ES, and <3 days for the other three metabolites. The leaching potential expressed by the GUS Index was high for DBA and ES (>2.8) and low to negligible for CMP, DCA and DCP (<0.32). In short, the intrinsic characteristics of DBA and ES are typical of leachers, but their significance as groundwater contaminants should be further surveyed through leaching in soil columns treated with their parent compounds. 相似文献

Fatty acid amides are a diverse family of underappreciated, biologically occurring lipids. Herein, the methods for the chemical synthesis and subsequent characterization of specific members of the fatty acid amide family are described. The synthetically prepared fatty acid amides and those obtained commercially are used as standards for the characterization and quantification of the fatty acid amides produced by biological systems, a fatty acid amidome. The fatty acid amidomes from mouse N18TG2 cells, sheep choroid plexus cells, Drosophila melanogaster, Bombyx mori, Apis mellifera, and Tribolium castaneum are presented. 相似文献

[Fe(N2H4)2(CH3COO)2] was synthesised and characterized for the first time by chemical analysis, magnetic measurements, electronic and IR spectral studies. Its thermal reactivity was ascertained by thermogravimetric (TG) and derivative thermogravimetric (DTG) techniques and it has been concluded that unlike some other metal carboxylate hydrazinates, it does not show any autocatalytic behaviour. The decomposition was also subjected to kinetic analysis using the equations of Coats-Redfern and Horowitz-Metzger by the method of weighted least-squares. 相似文献

In this work, we obtained the first selenium-containing chlorin with a chalcogen atom in exlocycle E. It was shown that the spectral properties were preserved in the target compound and the stability increased at two different pH values, in comparison with the starting purpurin-18. The derivatives have sufficiently high fluorescence and singlet oxygen quantum yields. The photoinduced cytotoxicity of sulfur- and selenium-anhydrides of chlorin p6 studied for the first time in vitro on the S37 cell line was found to be two times higher that of purpurin-18 and purpurinimide studied previously. Moreover, the dark cytotoxicity increased four-fold in comparison with the latter compounds. Apparently, the increase in the dark cytotoxicity is due to the interaction of the pigments studied with sulfur- and selenium-containing endogenous intracellular compounds. Intracellular distributions of thioanhydride and selenoanhydride chlorin p6 in S37 cells were shown in cytoplasm by diffusion distribution. The intracellular concentration of the sulfur derivative turned out to be higher and, as a consequence, its photoinduced cytotoxicity was higher as well. 相似文献

Vascular wilt caused by F. oxysporum (FOX) is one of the main limitations of producing several agricultural products worldwide, causing economic losses between 40% and 100%. Various methods have been developed to control this phytopathogen, such as the cultural, biological, and chemical controls, the latter being the most widely used in the agricultural sector. The treatment of this fungus through systemic fungicides, although practical, brings problems because the agrochemical agents used have shown mutagenic effects on the fungus, increasing the pathogen’s resistance. The design and the synthesis of novel synthetic antifungal agents used against FOX have been broadly studied in recent years. This review article presents a compendium of the synthetic methodologies during the last ten years as promissory, which can be used to afford novel and potential agrochemical agents. The revision is addressed from the structural core of the most active synthetic compounds against FOX. The synthetic methodologies implemented strategies based on cyclo condensation reactions, radical cyclization, electrocyclic closures, and carbon–carbon couplings by metal–organic catalysis. This revision contributes significantly to the organic chemistry, supplying novel alternatives for the use of more effective agrochemical agents against F. oxysporum. 相似文献

Chlrosulfuron, a classical sulfonylurea herbicide that exhibits good safety for wheat but causes a certain degree of damage to subsequent corn in a wheat–corn rotation mode, has been suspended field application in China since 2014. Our previous study found that diethylamino-substituted chlorsulfuron derivatives accelerated the degradation rate in soil. In order to obtain sulfonylurea herbicides with good crop safety for both wheat and corn, while maintaining high herbicidal activities, a series of pyrimidine- and triazine-based diethylamino-substituted chlorsulfuron derivatives (W102–W111) were systematically evaluated. The structures of the synthesized compounds were confirmed with 1H NMR, 13C NMR, and HRMS. The preliminary biological assay results indicate that the 4,6-disubstituted pyrimidine and triazine derivatives could maintain high herbicidal activity. It was found that the synthesized compounds could accelerate degradation rates, both in acidic and alkaline soil. Especially, in alkaline soil, the degradation rate of the target compounds accelerated more than 22-fold compared to chlorsulfuron. Moreover, most chlorsulfuron analogs exhibited good crop safety for both wheat and corn at high dosages. This study provided a reference for the further design of new sulfonylurea herbicides with high herbicidal activity, fast degradation rates, and high crop safety. 相似文献

A series of osmium double perovskite oxides, Ln2LiOsO6 (Ln=La, Pr, Nd, Sm), has been prepared as single crystals from acidic molten hydroxide. All four oxides crystallize in the monoclinic space group P21/n (Glazer tilt system #10, a−a−b+), forming a 1:1 ordered rock salt lattice of Li+ and Os5+ cations. Magnetic susceptibility measurements show that these compounds are antiferromagnetic at low temperature with ordering temperatures of 39, 35, 23, and 32 K for Ln2LiOsO6 (Ln=La, Pr, Nd, Sm), respectively. 相似文献

Subsolidus phase equilibria and crystal chemistry were studied for the La2O3-MgO-TiO2 system and for the ternary sections LaMg1/2Ti1/2O3-CaTiO3-La2O3 and LaMg1/2Ti1/2O3-CaTiO3-La0.833Mg0.25Ti0.75O3 in the quaternary La2O3-CaO-MgO-TiO2 system. Dielectric properties (relative permittivity and temperature coefficient of resonant frequency, τf) were measured at 5-10 GHz and mapped onto the phase equilibria relations to reveal the compositions of temperature-stable (τf=0) compounds and mixtures. Phase equilibria relations were obtained by X-ray powder diffraction analysis of approximately 80 specimens prepared by solid-state reactions in air at ∼1450°C. Six ternary phases were found to form in the La2O3-MgO-TiO2 system, including the three previously reported compounds LaMg1/2Ti1/2O3, La5Mg0.5Ti3.5O15, and “La6MgTi4O18”; and the new phases La10MgTi9O34, La9Mg0.5Ti8.5O31, and a perovskite-type solid solution (1−x)LaMg1/2Ti1/2O3-xLa2/3TiO3 (0?x?0.5). The phase previously reported as “La6MgTi4O18” was found to form off-composition, apparently as a point compound, at La6Mg0.913Ti4.04O18. Indexed experimental X-ray powder diffraction patterns are given for LaMg1/2Ti1/2O3, La5Mg0.5Ti3.5O15, La6Mg0.913Ti4.04O18, La10MgTi9O34, and La9Mg0.5Ti8.5O31. LaMg1/2Ti1/2O3 exhibits a slightly distorted perovskite structure with ordered B-cations (P21/n; a=5.5608(2) Å, b=5.5749(3) Å, c=7.8610(5) Å, β=90.034(4)°). La5Mg0.5Ti3.5O15 (Pm1; a=5.5639(1), c=10.9928(5) Å) and La6Mg0.913Ti4.04O18 (R3m; a=5.5665(1), c=39.7354(9) Å) are n=5 and n=6 members, respectively, of the (111) perovskite-slab series AnBn−1O3n. The new phases La10MgTi9O34 (a=5.5411(2), b=31.3039(9), c=3.9167(1) Å) and La9Mg0.5Ti8.5O31 (a=5.5431(2), b=57.055(1), c=3.9123(1) Å) are n=5 and n=4.5 members, respectively, of the (110) perovskite-slab series AnBnO3n+2, which exhibit orthorhombic subcells; electron diffraction revealed monoclinic superlattices with doubled c-parameters for both compounds. Extensive perovskite-type solid solutions form in the ternary sections LaMg1/2Ti1/2O3-CaTiO3-La2O3 and LaMg1/2Ti1/2O3-CaTiO3-La0.833Mg0.25Ti0.75O3. The La2O3-MgO-TiO2 system contains two regions of temperature-stable (τf=0) compositions. The quaternary La2O3-CaO-MgO-TiO2 system contains an extensive single-phase perovskite-type volume through which passes a surface of temperature-stable compositions with permittivities projected to be in the 40-50 range. Traces of this surface occur as lines of τf=0 perovskite-type phases in the ternary sections LaMg1/2Ti1/2O3-CaTiO3-La2O3 and LaMg1/2Ti1/2O3-CaTiO3-La0.833Mg0.25Ti0.75O3. 相似文献

The solubility of 1,4-naphthoquinone, plumbagin, lawsone, and juglone in supercritical carbon dioxide was determined spectroscopically at 40°C, and in the pressure range 8–18 MPa. Their solubilities at 12 MPa were between 0.3 and 10 g L−1. Plumbagin from Plumbago scandens L. roots was extracted at 40°C and 20 MPa. The extracted plumbagin mass fraction was up to 0.2% in fresh roots but down to about 0.006% in aged roots. n-Hexane and chloroform extraction of such aged roots indicates that the older and dryer the roots are, the stronger they bind plumbagin. Reversed-phase HPLC indicated a relatively pure plumbagin extract with supercritical carbon dioxide.

Background: The [99mTc][Tc(N)(PNP)] system, where PNP is a bisphosphinoamine, is an interesting platform for the development of tumor ‘receptor-specific’ agents. Here, we compared the reactivity and impact of three [Tc(N)(PNP)] frameworks on the stability, receptor targeting properties, biodistribution, and metabolism of the corresponding [99mTc][Tc(N)(PNP)]-tagged cRGDfK peptide to determine the best performing agent and to select the framework useful for the preparation of [99mTc][Tc(N)(PNP)]-housing molecular targeting agents. Methods: cRGDfK pentapeptide was conjugated to Cys and labeled with each [Tc(N)(PNP)] framework. Radioconjugates were assessed for their lipophilicity, stability, in vitro and in vivo targeting properties, and performance. Results: All compounds were equally synthetically accessible and easy to purify (RCY ≥ 95%). The main influences of the synthon on the targeting peptide were observed in in vitro cell binding and in vivo. Conclusions: The variation in the substituents on the phosphorus atoms of the PNP enables a fine tuning of the biological features of the radioconjugates. ws[99mTc][Tc(N)(PNP3OH)]– and [99mTc][Tc(N)(PNP3)]– are better performing synthons in terms of labeling efficiency and in vivo performance than the [99mTc][Tc(N)(PNP43)] framework and are therefore more suitable for further radiopharmaceutical purposes. Furthermore, the good labeling properties of the ws[99mTc][Tc(N)(PNP3OH)]– framework can be exploited to extend this technology to the labeling of temperature-sensitive biomolecules suitable for SPECT imaging. 相似文献

The equilibrium structure, electronic structure, and formation heat of Pu3M (PuM3) (M=Ga, In, Sn, and Ge) compounds with AuCu3 structure have been calculated using full potential linear augmented plane wave (FPLAPW) method within generalized-gradient approximation (GGA) including spin-orbit coupling (SOC) and spin polarization (SP). The calculated lattice parameters are in good agreement with the experimental values. Density of state analysis shows hybridization effects between Pu and M are governed by the competitions depending on the M amount: Pu 6d-Pu 5f, M p-Pu 6d, and M sp-M sp interactions. Electronegativity difference and electronic hybridization effect are two important factors to influence the formation heat and stability of Pu3M (PuM3) compounds. The larger is the electronegativity difference and the lower is M s-band or p-band center relative to the Fermi energy, the more negative is the formation heat and the more stable are Pu3M (PuM3) compounds. 相似文献

By using a simultaneous thermogravimetry (TG) and differential thermal analyzer (DTA), the hydration processes of the pure
C3S and with the addition of Ca(NO3)2 was followed. The peak temperature was determined and kinetic analysis on one of the hydration products, calcium hydroxide,
was performed. Results show that the use of Ca(NO3)2 increased the activation energy value of calcium hydroxide formed which is one aspect of the accelerating properties of Ca(NO3)2 while there is no sign of hydration for the addition of sucrose which proved its retarding property. It was also shown that
the activation energy increased when the hydrated pastes aged.
