Miniaturized solid-phase extraction (SPE) has been developed and successfully employed for the determination of organic species in water samples by liquid chromatography (LC). The method is based on the concept of a microscale extraction technique using a fused-silica capillary column for gas chromatography (GC), so-called in-tube solid-phase microextraction (SPME). The extraction conditions, such as the extraction time and flow-rate for the extraction and desorption process, were investigated as well as the effect of the internal structure of the extraction capillary on the efficiency. By inserting a stainless steel wire into the extraction capillary to reduce the internal volume of the capillary with the same surface area of the coating, an improved extraction and pre-concentration effects were obtained. Further pre-concentration was accomplished by the extraction device with a novel fiber-in-tube configuration. The direct coupling of the extraction method with a LC system has made it possible to determine low levels of phthalates in water samples without high consumption of organic solvents. The system developed must have potential applications for the analysis of environmental and biological samples in aqueous sample matrices.  相似文献

It has been indicated that hydrophilic solid powder to which aqueous solution of a novel dehydrocondensing reagent DMT-MM is adsorbed becomes a simple solid-phase dehydrocondensing reagent of low cost. Reaction in a liquid--liquid biphasic system on the surface of a solid phase with a large area was accelerated by suspending this powder in a dichloromethane solution of a carboxylic acid and an amine to be condensed. The reaction was rapid with a high yield despite the heterogeneity of the system. Like general solid-phase reagents, a hydrophobic carboxamide alone could be isolated at a relatively high purity only by filtration of the resulting suspension of reaction mixture.  相似文献

In this study, a novel and simple dual-phase solvent system for the improvement of extraction capability of magnetic molecularly imprinted polymer (MIP) beads in aqueous sample was proposed. The method integrated MIP extraction and micro-liquid-liquid extraction (micro-LLE) into only one step. A magnetic MIP beads using atrazine as template was synthesized, and was applied to aqueous media by adding micro-volume of n-hexane to form a co-extraction system. The magnetic MIP beads preferred to suspend in the organic phase, which shielded them from the disturbance of water molecule. The target analytes in the water sample was extracted into the organic phase by micro-LLE and then further bound to the solid-phase of magnetic MIP beads. The beads specificity was significantly improved with the imprinting efficiency of template increasing from 0.5 to 4.4, as compared with that in pure aqueous media. The extraction capacity, equilibration process and cross-selectivity of the MIP dual-phase solvent extraction system were investigated.The proposed method coupled with high-performance liquid chromatography was applied to the analysis of atrazine, simazine, propazine, simetryn, prometryne, ametryn and terbutryn in complicated sample such as tomato, strawberry juice and milk. The method is selective, sensitive and low organic solvent-consuming, and has potential to broaden the range of MIP application in biological and environmental sample.  相似文献

This paper describes an improved method for the extraction and determination of three steroids, oestrone, 17beta-oestradiol, and the synthetic contraceptive steroid 17alpha-ethinyloestradiol in aqueous matrices. Samples of wastewater and environmental water were spiked with internal standards, comprising isotopically labelled analogues of the steroids to be determined. The samples were extracted using solid-phase extraction disks and the extracts were then derivatized to form tert.-butyldimethylsilyl derivatives. The derivatised steroids were determined in the final extracts by GC-MS or GC-MS-MS allowing an operational detection limit for each steroid in effluent samples of 1 ng l(-1).  相似文献

This study developed an acid hydrolysis coupled to a solid-phase microextraction method employing a semi-microcolumn liquid chromatography system, instead of enzyme hydrolysis with solid-phase extraction for the pretreatment of human urine samples, to detect urinary 1-hydroxypyrene (1-OHP). The complete separation and detection of urinary 1-hydroxyprene was performed using a high-performance liquid-chromatography fluorescence detection system with an analytical C(18) semi-microcolumn, 60% (v/v) aqueous acetonitrile elution, and a lambda(ex/em) = 348/388 nm pair detection wavelength. Calibration graphs were linear with very good correlation coefficients (r = 0.9997), and the detection limit was 1.0 ng/L. These important parameters for acid hydrolysis and solid-phase microextraction were investigated. The total recovery was above 83% in acid hydrolysis with solid-phase microextraction. The proposed method provided a relatively simple, convenient, and practical procedure to determine the level of urinary 1-hydroxypyrene in biological samples, and was successfully applied to detect the urine of students.  相似文献

Headspace microextraction procedures such as solid-phase microextraction (SPME) and single drop microextraction (SDME) or liquid-phase microextraction (LPME) are increasingly used for the extraction of environmental organic pollutants from a variety of aqueous, viscous, semisolid and solid environmental and biological matrices. In this article, recent analytical applications of these methodologies when used as an isolation and trace enrichment step prior to the analysis of organic pollutants (pesticides, polycyclic aromatic hydrocarbons, polychlorinated compounds, organotin compounds, phenolic derivatives, aromatic amines, phthalates, etc.) by gas and liquid chromatography are reviewed. The applicability and inherent limitations of headspace microextraction are also discussed. The future direction of research in this field and general trends toward commercial applications are considered.  相似文献

The packings most widely used for solid-phase extraction are hydrophobic and make poor surface contact with aqueous samples unless the resins are first treated with an activating organic solvent such as methanol. Insertion of an acetyl- or hydroxymethyl group into a porous polystyrene-divinylbenzene resin provides a more hydrophilic surface that is easily wetted by water alone. Small columns of the chemically modified resins were found to be very efficient for the solid-phase extraction of many types of organic solutes from aqueous samples. Comparative recovery studies showed that the modified resins are superior to both silica packings and unmodified organic resins for the solid-phase extraction of organic compounds, and especially for polar organics such as phenols.  相似文献

Solid-phase extraction is an attractive alternative in sample preparation because it overcomes many drawbacks of liquid-liquid extraction and makes on-line determination possible by hyphenation with chromatographic techniques. Driven by the need for more effective and more selective sorbents, advances in solid-phase extraction include the development of new materials. This paper describes different types of chemically modified sorbents for the solid-phase extraction of compounds from aqueous samples. Chemical introduction of different functional groups into a polymeric resin improves the efficiency of solid-phase extraction by providing better surface contact with the aqueous samples; also, these sorbents have a greater capacity than the typical solid-phase materials for polar compounds have. The most important new sorbents are the chemically modified resins based on styrene-divinylbenzene copolymers. Preparation of these new sorbents is described, and advantages and drawbacks of off-line procedures and on-line procedures are also discussed. Applications for off-line and on-line chromatographic determinations of polar compounds are presented.  相似文献

In this review, recent methods developed for the determination of degradation intermediates of personal care products in environmental matrixes focusing on the extraction and determination steps are discussed. The five classes of personal care products evaluated are stimulants, fragrances, sunscreens, antimicrobials, and insect repellents. Methods are critically reviewed in terms of the analytical steps involved in the analysis, sample pretreatment, separation, and detection as well as the different confirmation strategies employed. Preconcentration from aqueous matrixes was performed by solid-phase extraction, liquid–liquid extraction, or solid-phase microextraction, allowing the simultaneous extraction of parent compounds and their degradation intermediates. Following the extraction and cleanup steps, the identification and quantification of degradation intermediates of personal care products at environmental levels (i.e., parts per trillion to parts per billion range) is usually performed by using mass spectrometry techniques such as single quadrupole mass spectrometry and more recently by time-of-flight mass spectrometry or tandem mass spectrometry. The main scope of this review is to critically evaluate the current state of the art of the analytical techniques used and to identify the research needs in the determination of degradation intermediates of personal care products in environmental matrixes.  相似文献

A new strategy has been developed for conjugation of peptides to oligonucleotides. The method is based on the "native ligation" of an N-terminal thioester-functionalized peptide to a 5'-cysteinyl oligonucleotide. Two new reagents were synthesized for use in solid-phase peptide and oligonucleotide synthesis, respectively. Pentafluorophenyl S-benzylthiosuccinate was used in the final coupling step in standard Fmoc-based solid-phase peptide assembly. Deprotection with trifluoracetic acid generated in solution peptides substituted with an N-terminal S-benzylthiosuccinyl moiety. O-trans-4-(N-alpha-Fmoc-S-tert-butylsulfenyl-L-cysteinyl)aminoc yclohe xyl O-2-cyanoethyl-N,N-diisopropylphosphoramidite was used in the final coupling step in standard phosphoramidite solid-phase oligonucleotide assembly. Deprotection with aqueous ammonia solution generated in solution 5'-S-tert-butylsulfenyl-L-cysteinyl functionalized oligonucleotides. Functionalized peptides and oligonucleotides were used without purification in native ligation conjugation reactions in aqueous/organic solution using tris-(2-carboxyethyl)phosphine to remove the tert-butylsulfenyl group in situ and thiophenol as a conjugation enhancer. A range of peptide-oligonucleotide conjugates were prepared by this route and purified by reversed-phase HPLC.  相似文献

Capillary liquid chromatography (cLC) hyphenated with tandem mass spectrometry (MS-MS) was used to separate and quantitate trace concentrations of five estrogens in aqueous samples. New C(18)-based sorption materials bound to the silica support by monomeric and polymeric mechanisms were compared and tested for solid-phase extraction (SPE) of selected analytes with respect to optimization of their preconcentration yield. Application of an endcapped, monomer-bound preconcentration Discovery DSC-18Lt column under the optimized conditions provides yields in the range from 95 to 100% with a high repeatability (n=3, RSD≤7.2%). Using the electrospray ionization in the positive mode (ESI+), the cLC-MS-MS system (the Zorbax SB C18 capillary column and a binary mobile phase of acetonitrile and water containing 0.1% formic acid in both the components) was optimized to attain a sufficient retention of the early eluting estriol, a satisfactory resolution of the analytes and the maximum sensitivity of the determination. Both the isocratic and gradient elution were used and the optimized gradient method permitted analyses of aqueous environmental samples in 14 min within a linearity range from 6.1 to 25.0 (LOQ of analytes) to 500 ng/L and with a very good linearity (r>0.9981) for all the estrogens studied. The detection limits are in the range from 3.0 to 6.8 ng/L (1 μL injection volume). Six environmental water samples were analyzed and the studied estrogens were found in the Vltava river sample collected in Prague (13.2 ng/L for 17β-estradiol) and in the inlet to the wastewater treatment plant in Prague, at an overall concentration of 371.4 ng/L.  相似文献

Recently, multi-wall carbon nanotubes (MWCNTs) as adsorbents of solid-phase extraction are attractive because they can be used for enrichment of organic compounds and metal ions at trace levels. In this study, we use the carboxyl modified multi-wall carbon nanotubes (CMMWCNTs) as adsorbents of solid-phase extraction for extraction of linear alkylbenzene sulfonates (LAS), which are widely used anion surfactant with different homologues, and detected by HPLC-UV. The effect of eluent and its volume, sample pH and flow rate, sample volume and the ultrasonic time of sample, the content of the electrolyte (NaCl) were investigated and optimized. The detection limit for LAS homologues was 0.02-0.03 μg L−1 with R.S.D. (n = 6) ranging from 2.04 to 10.03%. The recoveries of LAS homologues in the spiked environmental water samples ranged from 84.8 to 106.1%. The proposed method has been applied successfully to the analysis of LAS in aqueous environmental samples, which demonstrates that CMMWCNTs-based solid-phase extraction is a precision and convenient enrichment method and can be used for analysis of LAS homologues in water samples.  相似文献

Speciation and separation of chromium (VI) and chromium (III) from aqueous solutions were investigated using amino-propyl functionalised mesoporous silica (AP-MCM-41) as an adsorbent. The as-synthesised adsorbent was produced following a simple synthesis method at room temperature prior to template removal using microwave digestion. The maximum adsorption capacity at 111.1mg/g was calculated according to the Langmuir isotherm model, suggesting a 1:1 monolayer adsorption mechanism. Moreover, AP is a simple chelate, yet it can extract Cr (VI) exclusively from solutions containing other mixed metal ions simply by tuning the solution pH. Recovery of Cr (VI) from loaded sorbents is equally easy to perform with 100% extraction efficiencies allowing reuse of the sorbent and recovery of Cr (VI) from aqueous solutions containing a complex mixture of ions. The material would find use in environmental remediation applications, as a selective adsorbent of Cr (VI) or even as a solid-phase extraction stationary phase to remove and pre-concentrate Cr (VI) from aqueous solutions; this study demonstrates enrichment factors of 100 although higher levels are also possible.  相似文献

Superporous monolithic hydrogels (cryogel monoliths) are elastic, sponge-like materials that can be prepared in an aqueous medium through a cryotropic gelation technique. These monoliths show interesting properties for the development of high-throughput solid-phase extraction supports to treat large volumes of aqueous samples. In this work, a cryogel-supported molecularly imprinted solid-phase extraction approach for the endocrine disruptor bisphenol A (BPA) from river water and wine samples is presented. An imprinted polymer with molecular recognition properties for BPA was prepared in acetonitrile by thermal polymerization of a mixture of 4,4′-dihydroxy-2,2-diphenyl-1,1,1,3,3,3-trifluoropropane as a mimic template of BPA, 4-vinylpyridine and trimethylolpropane trimethacrylate in a molar ratio of 1 + 6 + 6. Fine imprinted particles (<10 μm) were embedded in a poly-acrylamide-co-N,N′-methylenbisacrylamide cryogel obtained by ammonium persulfate-induced cryopolymerization at −18 °C. The resulting monolithic gel was evaluated for its use as a sorbent support in an off-line solid-phase extraction approach to recover BPA from dilute aqueous samples with minimum pre-loading work-up. The optimized extraction protocol resulted in a reliable MISPE method suitable to selectively extract and preconcentrate BPA from river water and red wine samples, demonstrating the practical feasibility of cryogel-trapped imprinted polymers as solid-phase extraction materials  相似文献

We have developed a general, robust, and cost-effective method for the (E)- or (Z)-stereocomplementary enol tosylation of beta-ketoesters using TsCl- N-methylimidazole (NMI)-Et3N or LiOH. TsCl coupled with NMI formed a highly reactive N-sulfonylammonium intermediate. Stereocongested secondary alcohols were smoothly sulfonylated using Ts(Ms)Cl-NMI-Et3N. beta-Ketoesters underwent (E)-selective tosylation using TsCl-NMI-Et3N and (Z)-selective tosylation using TsCl-NMI-LiOH (total of 23 examples; 60%-99% yield). Stereoretentive Negishi and Sonogashira couplings using enol tosylates proceeded successfully to give trisubstituted alpha,beta-unsaturated esters.  相似文献

An analytical procedure for the determination of steroid estrogens and their conjugates was developed and applied to aqueous environmental samples. The analytes of 15 compounds were solid-phase extracted and fractionated into two fractions: one containing unconjugated (free) steroids and the other containing conjugates (sulfates and glucuronides). Identification and quantification were carried out using liquid chromatography coupled with tandem mass spectrometry. The recoveries for each compound ranged from 57 to 116% and reproducibilities represented as RSD ranged from 2.9 to 17%. Some of the sulfates and free compounds were detected in environmental samples, whereas most of the conjugates were below the detection limits.  相似文献
