An InBr3-catalyzed direct and efficient alkynylation of nitrones with terminal alkynes was developed. The process enables practical synthesis of a wide range of synthetically useful N-hydroxy-propargyl amine derivatives in good yields under mild conditions. The application of this method to optically active propargylic N-hydroxyamines syntheses was also described. With chiral nitrones, good diastereoselectivities were obtained. Moreover, the first chiral indium(III) complex-catalyzed asymmetric alkynylation of nitrone was achieved with moderate enantioselectivity.  相似文献

Stereoselective intramolecular 1,3-dipolar cycloaddition of homochiral N-(alkenylglycosyl)nitrones, prepared by allylation of C-(glycosyl)nitrones and subsequent oxidation, is described. The previously described 2-aza-Cope rearrangement was not observed for these substrates, but evidences of E/Z isomerism during the cycloaddition were obtained. The obtained cycloadducts can serve as key precursors of imino disaccharide analogues. This is exemplified by a short route to a protected 2-furanosyl-4-hydroxy-6-phenyl piperidine.  相似文献

Cycloaddition of 3-O-allyl-1,2-isopropylidene N-Ph nitrones afforded appreciably increased yields of oxepanes compared to the corresponding N-Me or N-Bn nitrones. Higher yields permitted some useful further transformations of the oxepanes.  相似文献

Terminal alkynes readily form zinc acetylides in the presence of iPr2NEt and 20 mol % ZnBr2, then attack N-phenyl nitrones activated by trimethylsilyl trifluoromethanesulfonate. Deprotection with aqueous acid yields N-hydroxyl propargylamine. Yields are generally high for nitrones derived from aromatic aldehydes. Control experiments suggest that silyl triflate has a significant accelerating effect upon the reaction.  相似文献

A new synthetic method providing expedient access to a wide range of polyfunctionalized N-hydroxyindoles (IV) is reported. These unique constructs are assembled by nucleophilic additions to in situ generated α,β-unsaturated nitrones (III) through carbon-carbon and carbon-heteroatom bond formation. The new synthetic technology was applied to the synthesis of nocathiacin I (1) model systems (2 and 3a-c) containing the N-hydroxyindole structural motif.  相似文献

The generation of excess reactive oxygen species (ROS) in mitochondria is responsible for much of the oxidative stress associated with ageing (aging), and mitochondrial dysfunction is part of the pathology of neurodegeneration and type 2 diabetes. Lipophilic pyridinium ions are known to accumulate in mitochondria and this paper describes a general route for the preparation of nitrone-containing N-arylpyridinium salts having a range of lipophilicities, as potential therapeutic antioxidants. The compatibility of nitrones with the Zincke reaction is the key to their synthesis. Their trapping of carbon-centred radicals and the EPR spectra of the resulting nitroxides are reported.  相似文献

The N-hydroxyazetidines 2 and 3 are prepared startinfrom 2,3-dihydroazete 1-oxides (1a and 1b0 by reduction with sodium borohydride and by reaction with a nucleophile, respectively. The N-thydroxyazetidines 2 and 3 can be oxidized with mercury (II) oxide to the corresponding nitrones 1; oxidation of the N-hydroxyazetidine 2a (unsubstituted at C-4) with two equivalent of lead tetraacetate tyields the N-tacetoxy β-lactam 4.  相似文献

The reactions of unactivated bis(methylene)cyclopropanes with nitrones and nitrile oxides have been investigated. The 1,1-diaryl-2-isopropylidene-3-methylenecyclopropanes react with the C,N-diarylnitrones to give a mixture of 2,2-dimethyl-1,6-diaryl-3-(diarylmethylene)piperidin-4-ones and 5-methyl-1-aryl-1-(arylamino)-4-(diarylmethylene)hex-5-en-3-ones. 2,3-Dihydro-3-methylenepyridin-4(1H)-ones are obtained by reaction of 1,1-diaryl-2-isopropylidene-3-methylenecyclopropanes with nitrile oxides.  相似文献

A new addition reaction of a 3-alkoxy allenylzinc reagent to N-acyliminiums prepared in situ from imines and acid halides is reported. Unlike addition onto imines or nitrones that affords the trans adducts, acetylenic syn-1,2 amino ethers are yielded with good selectivities, thus providing a new entry to propargyl syn-1,2-aminoalcohol units.  相似文献

We succeeded in the synthesis of N,N-dimethyl-2-trifluoroacetyl-1-naphthylamine (10) by the regioselective deacylation of N,N-dimethyl-2,4-bis(trifluoroacetyl)-1-naphthylamine with trifluoroacetic acid and water. The aromatic nucleophilic substitutions of 10 with various amines, thiols and alcohols proceeded cleanly to give the corresponding N-N, N-S and N-O exchanged products in moderate to excellent yields.  相似文献

The synthesis of two N-aryl substituted 2-silaimidazolidenes 9a, b by metal-reduction of the appropriate silicon(IV) heterocycles is reported. Structural as well as spectroscopic data obtained for the N-aryl substituted N-heterocyclic silylenes (NHSi) are very close to those obtained previously for their N-alkyl substituted counterparts. NHSis 9a, b are used as starting materials for the synthesis of a series of dichalcogenadisiletanes 19-24 and for of a mono silylene tungsten complex 29. The reactivity studies revealed only marginally differences between the N-aryl substituted NHSis 9a, b and previously described N-alkyl substituted silylenes.  相似文献

Substituted five-membered cyclic nitrones (pyrroline N-oxides) have been obtained in good to high yields from tertiary γ-nitro ketones and nitriles employing aluminium amalgam as a reducing agent in moist diethyl ether or THF. Attempts to obtain cyclic amino nitrones from α- or β-nitro nitriles failed and only the corresponding hydroxylamines have been isolated. Both nitrones and hydroxylamines have been used for synthesis of tertiary C-nitroso nitriles or ketones.  相似文献

CuI/N,N-dimethylglycine catalyzed coupling of aryl bromides with substituted oxazolidinones took place at 120 °C in DMF, affording the corresponding N-arylation products with good to excellent yields. A number of functional groups, such as ketone, nitrile, nitro, methoxy, and hydroxyl were tolerated under these conditions, thereby allowing diversity synthesis of N-aryloxazolidinones.  相似文献

The synthesis of a set of new N-heteroarylpolyamines is reported. A multiple and regioselective alkylation on the exo nitrogen of pyridinium N-(heteroaryl)aminides with several polybromo compounds, followed by a clean N-N bond reduction of the corresponding pyridinium salts, provides an easy and general method to obtain the title compounds.  相似文献

Through systematical comparison of various N4-protected cytosine derivatives in the glycosylation step of gemcitabine synthesis, highly beta-stereoselective and high yielding TBAI catalyzed N-glycosylation was achieved with N4-Bz cytosine and anomeric mixture of 2,2‘-difluororibose mesylate donor. The subsequent global deprotection gave gemcitabine efficiently. Meanwhile, the anomeric chloride intermediate and fluoride-displaced side products of this N-glycosylation were identified, too. This new glycosylation method reveals the importance of N4-protection in the stereoselective preparation of pyrimidine nucleoside, also provides a potential alternative to current industrial process to gemcitabine.  相似文献
