The 1-trimethylsilylmethylvinyl group, as a 1-hydroxymethylvinyl equivalent, was readily introduced to epoxides with the corresponding Grignard reagent derived from 2-bromoallyltrimethylsilane. Obtained 2-(2-hydroxy-ethyl)-allylsilanes were converted to α-methylene-γ-lactones via diols.  相似文献

It was shown that ethyl 2-nitroacetoacetate is a synthetic precursor of ethoxycarbonylnitrile oxide as well as of isoxazole- and isoxazoline-3-carboxylic acids and their esters. The elimination of acetic acid from ethyl 2-nitroacetoacetate occurs in a mixture of acetic acid and acetic anhydride in the presence of strong mineral acids,e.g., H2SO4, at room temperature and gives isoxazolines in yields of up to 85–91 %.Translated fromIzvestiya Akademii Nauk. The work described deals with the isolation and characterization of epoxides from 6-deoxyhex-5-enopyranosides and preliminary exploration of their synthetic potential. Prolonged epoxidation reaction times led to their hydrolysis in situ and gave novel protected D-hexos-5-ulose derivatives (sugar 1,5-dicarbonyls). Some reactions of the hexos-5-uloses were studied, and in some cases septanoside (seven-membered-ring saccharide) derivatives were isolated. Novel routes to D-xylo-hexos-5-ulose and D-lyxo-hexos-5-ulose, of interest as intermediates in the synthesis and biosynthesis of inositols and aza sugars, are also described. The structures of the epoxides and novel hexos-5-uloses were established by NMR and X-ray crystallographic methods.  相似文献

An investigation of Hermecz and Hartenstein’s cyclization methods was conducted. Overcoming the limited substrate scope allowed the expansion of the synthetic application of 1,3,5-triazine as an umpolung hydrogen cyanide equivalent. The reaction proceeded under mild conditions to provide various isoquinolinone and 2-pyridone derivatives with excellent yields.  相似文献

An MT-sulfone group was converted to a trifluoromethyl group by treatment with IF(5) after an alkylation reaction. Therefore, an MT-sulfone anion can be used as a trifluoromethyl anion equivalent. The formal asymmetric Michael-addition of a trifluoromethyl anion to crotonaldehyde was also performed.  相似文献

The synthesis of the novel amino acid 6-(hydroxymethyl)-2-piperidinecarboxylic acid ( 1a ) and its ethyl ester 1b is reported. In the hydrogenation of 6-(hydroxymethyl)pyridine-2-carboxylates, hydrogenolysis of the alcohol group appeared as an unusual side reaction. Optimization of the reaction conditions allowed us to minimize hydrogenolysis and afforded pure 1 .  相似文献

alpha-Phenyl-N-tert-butylnitrone (PBN) derivatives bound to beta-cyclodextrin derivatives have been synthesized. Inclusion of the PBN group into the beta-cyclodextrin moiety is host- and temperature-dependent. In the case of the nitrone linked to permethylated cyclodextrin (Me3CD-PBN), the thermokinetic parameters are in favour of a slow chemical exchange between a tight and a loose complex. In contrast, 2,6-di-O-Me-beta-cyclodextrin-grafted PBN (Me2CD-PBN) exists either in a fast exchange or as a strongly self-associated complex. The covalent cyclodextrin-PBN compounds have been used to trap carbon and oxygen-centred free radicals. The self-associated forms of the beta-CD-spin-traps are compatible with effective spin-trapping, affording spin-adducts with enhanced EPR signal intensities relative to noncovalent CD-nitrone systems or the nitrone alone. This kind of cyclodextrin-bound nitrone is the first type of covalent supramolecular spin-trap and should open new possibilities for the study of biological free radicals in vivo.  相似文献

Seven isomers of cyclomaltooctaose (γ-cyclodextrin) whose C6s were unsymmetrically disubstituted with both an azido group and an arenesulfonyloxy group were prepared and each of them was isolated by reversed phase chromatography. The assignment of the modified positions in each regioisomers was unambiguously performed by chemical correlation with the authentic compounds and chemical conversion to the 3,6-anhydro derivatives followed by high-resolution 2D-1H NMR (COSY, TOCSY, and HOHAHA) analyses. The compounds are versatile synthetic and authentic intermediates to prepare sophisticated derivatives with two different functionalities at desired positions on the molecule for supramolecular chemistry.  相似文献

The synthesis of D-lyxo-hexos-5-ulose (5-ketomannose, 1,5-dicarbonyl sugar), a synthetic precursor to the glycoprocessing inhibitor deoxymannojirimycin, was carried out by an in situ epoxidation and hydrolysis of a trimethylsilyl-protected 6-deoxyhex-5-enopyranoside followed by facile removal of the protecting groups. A novel nine-step synthesis of deoxymannojirimycin has also been achieved from methyl alpha-D-mannopyranoside; this involved methanolysis of epoxides derived from an acetylated 1-azido-6-deoxyhex-5-enopyranoside followed by deprotection and catalytic hydrogenation.  相似文献

Sulfur substituted 3-vinylpyrrole 10 was prepared from 3-thio-acetylpyrrole 9 by alkylation with alkyl halide in the presence of propylene oxide. Functionalized 4-alkylthioindoles were made by Diels-Alder reaction of the 3-vinylpyrrole 10 with dienophiles. Reaction of 1,3-dihalo-3-methy]-2-butanone with t-butyl acetoacetate (NaH) gave t-butyl 3,4-dihydro-2,2,6-trimethyl-4-oxo-2H-pyran-5-carboxylate via the Favorskii-type rearrangement. An efficient protocol for the modular synthesis of sulfones and sulfonyl derivatives has been developed utilizing sodium tert-butyldimethylsilyloxymethanesulfinate (TBSOMS-Na) as a sulfoxylate (SO22−) equivalent. TBSOMS-Na, easily prepared from the commercial reagents Rongalite™ and TBSCl, serves as a potent nucleophile in S-alkylation and Cu-catalyzed S-arylation reactions with alkyl and aryl electrophiles. The sulfone products thus obtained can undergo the second bond formation at the sulfur center with various electrophiles without a separate unmasking step to afford sulfones and sulfonyl derivatives such as sulfonamides and sulfonyl fluorides.

Key intermediates are azabicycloalkenes 9 and 17, which are prepared via Diels-Alder reactions and subsequent mild deprotection. These unsaturated bicyclic heterocycles are versatile intermediates for different dipeptide mimetics of the aza- and diazabicycloalkane type, which is demonstrated by the synthesis of diazabicycloalkanes 11 and 19 in only 3-6 steps and good overall yield.  相似文献

Synthesis of oligothiophenes of well-defined structures that possess 2-8 thiophene units is performed with a new synthetic strategy involving C-H homocoupling of bromothiophenes and cross-coupling with organostannanes. 2-Oxo-1,2-ethylenedioxy (2-OED) functionality has been prepared on a soluble MPEG polymer and solid (HM resin and Lanterns) supports as platforms for discovery of drug-like small molecules. The functionality is cleaved either by Yb(OTf)3 or TMSCHN2 in MeOH, or by various amines to release small molecules synthesized on the platform in good yield.
