This work presents a modified method to analyze linear alkylbenzenesulfonates (LASs) in water samples. The method involves extraction of samples by a graphitized carbon black (GCB) cartridge, and direct derivatization in the GC injection port using a large-volume (10-20 microl) direct sample introduction (DSI) device with tetraalkylammonium (TAA) salts. The analytes were then identified and quantitated by ion-trap GC-MS. The large-volume DSI injection-port derivatization technique provides sensitivity, fast and reproducible results for LAS residues, to quantitation at 0.1 microg/l in 200 ml of water samples. The retention effect of TAA salts in the injection port was not detected. Enhanced selected mass chromatograms of [M-55]+ ions of butylated C10-C13 LASs by electron impact ionization MS allows one to determine LAS residues at trace levels in environmental samples. Recovery of total LASs in spiked variety water samples ranged from 89 to 112% while RSDs ranged from 2 to 13%.  相似文献

This work presents a modified method to analyze selected pharmaceutical residues (clofibric acid, ibuprofen, carbamazepine, naproxen, ketoprofen and diclofenac) in water samples. Various solid-phase extraction cartridges were investigated. The newly developed Oasis HLB (polystyrene-divinylbenzene-N-vinyl pyrrolidone terpolymer) solid-phase extraction (SPE) cartridge provides the optimal sample extraction results. The analytes were then identified and quantitatively determined by gas chromatography-mass spectrometry (GC-MS) via on-line derivatization in the injection-port using a large-volume (10 microl) sample injection with tetrabutylammonium (TBA) salts. This injection-port derivatization technique provides sensitivity, fast and reproducible results for pharmaceutical residues analysis. Mass spectra of butylated derivatives and tentative fragmentation profiles are proposed. Molecular ions and some characteristic ions were used as the quantitation ions to obtain maximum detection sensitivity and specificity. The quantitation limits of these compounds ranged from 1.0 to 8.0 ng/l in 500 ml tap water samples. Recovery of these residues in spiked various water samples ranged from 50 to 108% while RSD ranged from 1 to 10%. The selected analytes were detected in concentrations of 30 to 420 ng/l in wastewater treatment plant effluent and river water samples.  相似文献

Injection-port derivatization combined with solid-phase extraction (SPE) was developed and applied for the first time to determine five types of fecal sterols (coprostanol, cholestanol, epicholestanol, epicoprostanol and cholesterol) with gas chromatography–mass spectrometry (GC–MS). In this method, silylation of fecal sterols was performed with N,O-bis(trimethylsilyl)trifluoroacetamide (BSTFA) at GC injection-port. The factors influential to this technique such as injection-port temperature, purge-off time, derivatization reagent (BSTFA) volume, and the type of organic solvent were investigated. In addition, the conditions of SPE (including the type of SPE cartridge, the type of elution organic solvent) were also studied. After SPE followed by injection-port silylation by GC–MS, good linearity of analytes was achieved in the range of 0.02–10 ng/mL with coefficients of determination, R2 > 0.995. Good reproducibility was obtained with relative standard deviation less than 19.6%. The limits of detection ranged from 1.3 ng/mL to 15 ng/mL (S/N = 3) in environmental water samples. Compared with traditional off-line silylation of fecal sterols performed with water bath (60 °C, 30 min), this injection-port silylation method is much simpler and convenient. The developed method has been successfully applied for the analysis of fecal sterols from real environmental water samples.  相似文献

A rapid and environmental-friendly injection-port derivatization with gas chromatography-mass spectrometry (GC-MS) method was developed to determine selected low-molecular weight (LMW) dicarboxylic acids (from C2 to C10) in atmospheric aerosol samples. The parameters related to the derivatization process (i.e., type of ion-pair reagent, injection-port temperature and concentration of ion-pair reagent) were optimized. Tetrabutylammonium hydroxide (TBA-OH) 20 mM in methanol gave excellent yield for di-butyl ester dicarboxylate derivatives at injection-port temperature at 300 °C. Solid-phase extraction (SPE) method instead of rotary evaporation was used to concentrate analytes from filter extracts. The recovery from filter extracts ranged from 78 to 95% with relative standard deviation (RSD) less than 12%. Limits of quantitation (LOQs) ranged from 25 to 250 pg/m3. The concentrations of di-carboxylated C2-C5 and total C6-C10 in particles of atmospheric aerosols ranged from 91.9 to 240, 11.3 to 56.7, 9.2 to 49.2, 8.7 to 35.3 and n.d. to 37.8 ng/m3, respectively. Oxalic acid (C2) was the dominant LMW-dicarboxylic acids detected in aerosol samples. The quantitative results were comparable to the results obtained by the off-line derivatization.  相似文献

A new analytical method applying in situ derivatization was developed to enable the extraction of polar drug metabolites from water samples by solid-phase extraction (SPE). An additional derivatization by silylation was used to enhance the sensitivity of analyte detection by gas chromatography-mass spectrometry (GC-MS). Thus, the two metabolites 1,5-di-methyl-1,2-dehydro-3-pyrazolone (DP) and 4-(2-methylethyl)-1,5-dimethyl-1,2-dehydro-3-pyrazolone (PDP), postulated for the degradation of phenazone and propyphenazone, were identified and detected up to the microg/L level in raw and drinking water samples from public water supply.  相似文献

This work presents a modified method to analyze polar and water-soluble naphthalene monosulfonic acid (NS) isomers in industrial effluents and river water samples. The method involves extraction of samples by a styrene-divinylbenzene copolymer solid-phase extraction cartridge, and on-line derivatization in the GC injection port using a large-volume (10 microl) sample injection with tetrabutylammonium salts. The analytes were then identified and quantitatively determined by GC-MS. The large-volume injection-port derivatization technique provides sensitivity, fast and reproducible results for NS isomers, to quantitation at 0.05 microg/l in 200 ml of water sample. Enhanced extracted mass chromatograms of molecular ion and [M-56]+ ion of butylated NS isomers by electron impact ionization MS allows us to determine residues at trace levels in environmental samples. Recoveries of the NS isomers in spiked water samples ranged from 70 to 82% with RSDs around 10%. Naphthalene-2-sulfonic acid was found as a major pollutant and propagated in surface water and industrial effluents.  相似文献

Detection and identification of mycotoxin metabolites is a very challenging task. In order to achieve adequate sensitivity and specificity an analytical technique must overcome serious matrix interferences. Gas chromatography-mass spectrometry (GC-MS) which has the sensitivity and specificity to detect and identify mycotoxin metabolites requires hydrolysis of conjugated metabolites as well as derivatization. Thermospray high-performance liquid chromatography-mass spectrometry (HPLC-MS) offers the sensitivity, specificity, and structural information to detect and identify some mycotoxin metabolites in fecal and urine samples without derivatization. The mycotoxins evaluated in this study include deoxynivalenol (DON), T-2 toxin, and diacetoxyscirpenol. The de-epoxy and hydroxy metabolites of each toxin and the glucuronide conjugate of DON were isolated, extracted, and analyzed to detect their occurrence in animals. The thermospray mass spectra of the toxins showed an [M + H]+ ion and numerous structurally significant fragment ions in the positive ion detection mode. Negative ion detection exhibited primarily [M + acetate]- cluster ions with less fragmentation than observed by positive ion detection. The operation of the interface in the filament-on mode greatly increased the sensitivity in both positive and negative ion detection mode. Detection limits of 50-500 pg injected on column are obtained for these toxins and their metabolites using multiple ion detection. The urine and fecal extracts from rats, hens, and cows did not interfere with the HPLC-MS analysis for the specific metabolites or the glucuronide conjugate.  相似文献

Tetraalkyl ammonium (TAA) salts are of great importance in many areas of chemistry (organic, physical etc.). Tetraalkylammonium (TAA) chlorides, bromides or iodides are usually prepared by direct quaternization of trial-kyl amines with the corresponding alkyl halides. However other tetraalkylammonium (TAA) salts - cyanides, rhoda-nides, azides etc. cannot be prepared in this direct way. A typical method of preparation of these salts consists in ion exchange between available TAA halides and various salts containing the required anions. In some instances simply treating the TAA halides with aqueous solution of silver salts can be applied. This procedure, however, besides being costly is limited to the cases when the required silver salts are soluble in water1.  相似文献

A reliable, sensitive and eco-friendly injection-port trimethylsilylated (TMS) derivatization and gas chromatography–tandem mass spectrometry (GC–MS/MS) with furan chemical ionization (furan-CI) method was developed to determine melamine and cyanuric acid in powdered milk samples. The effects of several parameters related to the TMS-derivatization process (i.e., injection-port temperature, residence time and volume of silylating agent) and of various CI agents were investigated. Addition of a solution (3 μL) of bis(trimethyl)silyltrifluoroacetamide (BSTFA) containing 1% trimethylchlorosilane (TMCS) reagent to a 20-μL extract from the powdered milk sample gave an excellent yield of the tris-TMS-derivatives of melamine and cyanuric acid at an injection-port temperature of 90 °C. Furthermore, using furan as the CI agent in conjunction with tandem mass spectrometry provided the greatest sensitivity and selectivity of detection. The limits of quantitation (LOQs) for melamine and cyanuric acid were 0.5 and 1.0 ng/g in 0.5-g of powdered milk samples, respectively. The recoveries from spiked samples – after simple ultra-sonication with 5% dimethyl sulfoxide in acetonitrile coupled with n-hexane liquid–liquid extraction – ranged from 72% to 93% with relative standard deviations of lower than or equal to 18%. In three of four real powdered milk samples, melamine was detected at concentrations ranging from 36 to 1460 ng/g; and cyanuric acid was detected in two of these samples at concentrations of 17 and 180 ng/g.  相似文献

A novel derivatization method was developed for the simultaneous determination of six acidic metabolites of catecholamine and serotonin by gas chromatography–mass spectrometry (GC‐MS). The metabolites were converted to O‐ethoxycarbonyl/tert‐butyldimethylsilyl (EOC/TBDMS) derivatives for the direct GC‐MS analysis in selected ion monitoring mode. Their mass spectral pattern as EOC/TBDMS derivatives showed characteristic fragment ions of [M – 15]+ and [M – 57]+, which permitted rapid and accurate structural confirmation of acidic metabolites. The present method was linear (r ≥ 0.998), reproducible (percentage relative standard deviation = 1.0–10.0) and accurate (% relative error = ?9.7–9.8) with detection limits of 0.001–4.7 ng/mL. A liquid chromatography/tandem mass spectrometry (LC/MS/MS) method has been developed for the determination of 14 parent triazole fungicides and 8 of their metabolites found in apples, peaches, flour, raw water, and tap water. The triazole fungicides chosen for this multiresidue method development project included propiconazole, fenbuconazole and its RH-9129 and RH-9130 metabolites, cyproconazole, difenoconazole, tebuconazole and its HWG 2061 metabolite, hexaconazole, bromuconazole (both stereoisomers), epoxiconazole, tetraconazole, triticonazole and its RPA-404886 and RPA-406341 metabolites, triadimefon, triadimenol, and myclobutanil. Of special concern to the U.S. Environmental Protection Agency were the metabolites common to all triazole fungicides: free triazole, 1,2,4-triazole (T), and its 2 conjugates: triazolylalanine (TA) and triazolylacetic acid (TAA). These metabolites were the primary focus of this project. All samples we cleaned up by a combination of C18 solid-phase extraction (SPE), mixed-mode cationic SPE, and mixed-mode anionic SPE columns. A triple-stage quadrupole mass spectrometer, equipped with electrospray ionization in the positive-ion mode, was used to determine the compounds of interest. T, TA, and TAA were quantitated using isotopically labeled internal standards (IS), in which the 1,2,4-triazole ring had been synthesized by using 13C and 15N (IS_T, IS_TA, and IS_TAA). These isotopically labeled internal standards were necessary to correct for matrix effects. The T, TA, and TAA metabolites were quantitated at the 25-50 parts-per-billion (ppb) level in food commodities and at 0.50 ppb in water. Recoveries were 70-101% from apples, 60-121% from peaches, 57-118% from flour, 75-99% from raw water, and 79-99% from tap water.  相似文献

Anilines are important pollutants occurring in the environment as industrial discharges as well as the transformation products (i.e., metabolites) of a wide variety of commonly used pesticides. In this report, we describe the precolumn derivatization of anilines with fluorescein isothiocyanate (FITC) and their subsequent separation and detection by capillary electrophoresis-laser induced fluorescence (CE-LIF) detection. The FITC-aniline derivatives were readily detected at the 10(-10) M level. This limit of detection (LOD) was achieved in the presence of glycosidic surfactants complexed with borate at alkaline pH yielding the so-called in situ charged micelles. The glycosidic surfactants evaluated were n-octyl- and n-nonylglucopyranoside. Furthermore, and under optimum conditions, the FITC precolumn derivatization of the anilines was performed in real water (e.g., tap and lake water) spiked with anilines at the LOD level. The water matrices showed marginal effects on the extent of derivatization at the LOD level, and the possible interferents in the water samples did not affect the FITC-solute signal due to the selectivity of the derivatization and detection schemes. Besides filtration from microparticles, the real water samples did not necessitate extensive sample cleanup prior to derivatization.  相似文献

Sulfonamides are widely used in veterinary medicine for prophylactic purposes and for the treatment of various infections of food-producing animals. This means that residues of these drugs and their possible metabolites may occur in food of animal origin. In Belgium, a zero tolerance level for sulfonamides in edible animal tissues has been set. In order to check this zero level on a routine basis, a rapid and sensitive method has to be available. For this purpose, a quantitative high-performance thin-layer chromatographic (HPTLC) method for the detection of sulfonamide residues in animal tissue and milk samples has been developed. The sample preparation consists of a liquid extraction followed by a solid phase extraction (SPE) on disposable columns for the meat samples and a matrix solid phase dispersion (MSPD) for the milk samples. A three-multiple development chromatographic system is used for the separation and a derivatization with fluorescamine decreases the minimal detectable quantity per spot from 1.42 to 0.32 ng. The limit of quantification is 4 micrograms/kg for milk and meat samples.  相似文献

The commercial hair conditioners and fabric softeners were analyzed for the content of alkyltrimethylammonium compounds (ATMACs) by gas chromatography-mass spectrometry (GC-MS) with electron impact (EI) and low-pressure positive-ion chemical ionization (PICI) modes. The method involves mixed diluted samples (adjust pH to 10.0) with potassium iodide to enhance the extraction of iodide-ATMA+ ion pairs by direct liquid-liquid extraction. The iodide-ATMA+ pairs were then demethylated to their corresponding nonionic alkyldimethylamines (ADMAs) by thermal decomposition in a GC injection-port. A high abundance of ADMAs was detected at the temperature above 300 degrees C in the GC injection-port. The enhanced selectivity of quasi-molecular ion chromatograms of C12-C18-ADMA, obtained using methanol PICI-MS enables ADMAs to be identified. The accuracy and precision of the method was validated and was successfully applied to determine contents of ATMAC in commercial hair conditioners and fabric softeners. The contents of total measured ATMAC ranged from 0.4 to 6.9% for hair conditioners, and from 3.3 to 4.6% for fabric softeners.  相似文献

建立了一种简便、直接进样的超高效液相色谱-三重四极杆质谱法(UPLC-MS/MS)快速测定环境水样中草甘膦、氨甲基膦酸、草铵膦及乙烯利的残留。环境水样经0.22 μm滤膜过滤或冷冻离心去除杂质后,滤液无需衍生化直接进行定量分析。4种农药通过Metrosep A Supp 5柱(150 mm×4.0 mm,5 μm)分离,以碳酸氢铵-氨水溶液为流动相进行梯度洗脱,在负离子模式下以MRM方式进行检测。结果表明,4种农药在0.50~50.0 μg/L范围内相关系数(r)均大于0.999,线性关系良好,方法检出限为0.05~0.09 μg/L。实际水样在低、中、高3种加标浓度水平下,回收率分别为76.3%~108%、83.0%~107%和87.0%~105%,相对标准偏差分别为2.0%~12.3%、2.4%~5.6%和2.7%~6.8%。使用该方法对海南省34个水样进行测定,其中30个饮用水源地水样中均未检出4种农药,槟榔园附近3个水样均检出草甘膦和氨甲基膦酸,香蕉园附近的1个水样检出草铵膦和氨甲基膦酸。与传统的衍生化方法比较,该方法操作简便,重现性好,准确性高,不受基体干扰,适用于环境水样中草甘膦、氨甲基膦酸、草铵膦及乙烯利的残留检测。  相似文献

3,4-Methylenedioxymethamphetamine (MDMA, ecstasy) is consumed as the racemate but some metabolic steps are enantioselective. In addition, chiral properties are preserved during MDMA biotransformation. A quantitative analytical methodology using gas chromatography/mass spectrometry (GC/MS) to determine enantioselective disposition in the body of MDMA and its main metabolites including 3,4-methylenedioxyamphetamine (MDA), 4-hydroxy-3-methoxymethamphetamine (HMMA), and 4-hydroxy-3-methoxyamphetamine (HMA) was developed. Plasma and urine samples were collected from a male volunteer. The analysis of MDMA, MDA, and 4-hydroxy-3-methoxy metabolites by GC/MS required a two-step derivatization procedure. The first step consisted of derivatization of the amine with enantiomerically pure Mosher's reagent ((R)-MTPCl). Triethylamine was used as a base to neutralize hydrochloric acid formed during the reaction allowing quantitative derivatization, which resulted in a substantial improvement in the sensitivity of the method compared with other previously described techniques. Further treatment with ammonium hydroxide was required since both amine and hydroxyl groups underwent derivatization in the reaction. Ammonium hydroxide breaks bonds formed with hydroxyl groups without affecting amine derivatives. The second derivatization step using hexamethyldisilazane was needed for metabolites containing phenol residues. This derivatization method permitted the stereochemically specific study of MDMA and its main monohydroxylated metabolites by GC/MS. A detailed study of the chemical reactions involved in the derivatization steps was indispensable to develop a straightforward, sensitive, and reproducible method for the analysis of the parent drug compound and its metabolites.  相似文献

An efficient method is described for the simultaneous determination of phenol and 49 substituted phenols present in aqueous samples. The method is based on the extractive two-phase isobutoxycarbonyl (isoBOC) derivatization with subsequent solid-phase extraction (SPE) for the direct analysis by gas chromatography (GC) and gas chromatography-mass spectrometry (GC-MS). Phenolic hydroxyl groups in acidic aqueous solutions were allowed to react with isobutyl chloroformate present in the dichloromethane phase containing triethylamine. The resulting isoBOC derivatives were then recovered by SPE using Chromosorb P in normal-phase partition mode, followed by direct GC and GC-MS analysis. Using this combined procedure, linear detector responses were obtained in the concentration range of 0.5-8 microg ml(-1), with correlation coefficients varying from 0.925 to 0.999 for most of the phenols studied except for 2,4-dinitorphenol (0.789). The temperature-programmed retention index (I) sets as measured on DB-5 and DB-17 dual-capillary columns of different polarity were characteristic of each isoBOC phenol derivative and thus, useful in the screening for isomeric phenols by I matching only. The mass spectral patterns, exhibiting characteristic [M-100]+, [M-200]+ and [M-300]+ ions for the mono-, di- and trihydroxybezenes, respectively with common ions at m/z 57, facilitated their rapid structural confirmation. The present method allowed rapid screening for phenols when applied to water samples spiked with phenols.  相似文献

A rapid and sensitive LC-electrospray tandem mass spectrometry method has been developed for the quantitation of 4-chloro-2-methylphenoxyacetic acid (MCPA) and 4-chloro-2-methylphenol in both water and soil samples. Soil samples were extracted in alkaline media and cleaned-up by solid-phase extraction with C18 cartridges before LC-MS analysis. The selectivity and sensitivity offered by the triple quadrupole allowed the direct injection of the water samples rendering a sample throughput of around 100 samples per day, without any sample pretreatment, rendering for MCPA a limit of detection of 40 ng/l. In order to increase the method sensitivity, mainly for metabolite, a previous solid-phase extraction step was also performed. The method was validated by means of recovery experiments using fortified water and soil samples, obtaining satisfactory recoveries for both compounds in water and for MCPA in soil. The validated procedures can be used for the specific monitoring of residues of MCPA and its main metabolite in environmental samples, as ground and surface waters and soils, providing more selectivity and sensitivity than the current UV-based methodology. Besides, sample manipulation is greatly reduced in comparison to other GC-MS based methods which require a previous derivatization.  相似文献

A procedure is presented which allows the ultratrace level determination of phenylurea herbicides (PUHs) in natural waters. Samples were enriched by solid-phase extraction (SPE) on Carbopack B and alkylated with iodoethane and sodium hydride to yield thermostable products. After derivatization, the aqueous samples were extracted and injected by SPME. The use of iodoethane instead of iodomethane allowed the differentiation between parent compounds and the N-demethylated metabolites. Limits of detection were between 0.3 and 1.0 ng/l for the parent compounds. Standard deviations below 10% were achieved for samples containing more than 4 ng/l in very different matrices including Nanopure water, lake water, and waste water treatment plant (WWTP) effluent. Moreover, the para-hydroxylated metabolite of diuron could be quantified with the same procedure. The presence of further metabolites was assessed qualitatively. Chromatography was stable over a large number of measurements even with dirty samples from WWTP effluent. The precision and sensitivity of the developed analytical method allowed the investigation of the fate of PUHs in lakes, their degradation during drinking water treatment and their transport within the North Sea.  相似文献

Synthetic surfactants are economically important chemicals, as they are widely used in household cleaning detergents, textiles, paints, polymers and personal care products. In this work we have developed a method capable of the isolation and analysis of the most widely used surfactants (linear alkylbenzene sulfonates, LAS, nonylphenol ethoxylates, NPEO, and alcohol ethoxylates, AEO) and their main degradation products (sulfophenyl carboxylic acids, SPC, nonylphenol ethoxycarboxylates, NPEC, and polyethylene glycols, PEG) in aqueous and solid environmental matrices. First, analytes were extracted by ultrasonic extraction from sediments and suspended solids using methanol at 50°C as solvent and 3 cycles (30 min per cycle). Clean-up and pre-concentration of the extracts and water samples were carried out by solid-phase extraction (SPE), using Oasis HLB cartridges. Recoveries were generally about 80% for most compounds. Identification and quantification of target compounds were performed by liquid chromatography-time-of-flight-mass spectrometry (LC-ToF-MS), which has been much less used in the field of environmental analysis than other MS techniques. Examples which illustrate the possible advantages of this technique for multi-analyte analysis of target and non-target contaminants in environmental samples are provided. Finally, the methodology developed here was validated by measuring the concentration of surfactants and their metabolites in selected marine sediment and seawater samples collected in Long Island Sound (NY), and in influent and effluent wastewater from Stony Brook treatment plant (NY). This paper presents some of the first data relative to the occurrence of PEG in the environment, especially in sediments where concentrations were generally higher (up to 1490 μg/kg) than those for other classes of targeted surfactants and their metabolites.  相似文献
