The Friedel-Crafts reaction of alkyl glyoxylates with variously substituted furans was found to be efficiently catalyzed under simple, undemanding conditions by a 6,6'-dibromo-BINOL/Ti(IV) complex with high enantioselectivity. The reaction afforded chiral substituted 2-furanyl-hydroxyacetic acid esters, compounds of high synthetic interest, in good yield and enantiomeric excess, in most examples in the range of 90-97%.  相似文献

The synthesis of diarylmethanols via the reduction of a range of substituted benzophenone and benzoylpyridine derivatives with ketoreductase enzymes (KREDs) has afforded chiral products with high yield (>90%) and ee (up to >99%). Ortho, meta, and para substitutions with a variety of electron-donating, electron-withdrawing, and halogen groups were examined. Substitution at the ortho position and/or highly electronically dissymmetric molecules were not required for good selectivity, as is the case with conventional chemical catalyst reductions. Synthesis of a range of chiral non-racemic vinyl aziridines with varying substitutions on the alkene has been achieved by the reaction of (SS)-tert-butylphenylsulfinimine with a range of ylides derived by deprotonation of substituted allyltetrahydrothiophenium salts. Yields of the substituted vinyl aziridines range from 0% to 90%, with trans:cis ratios between 1:1 and 0:100 and diastereomeric excesses of up to 99%.  相似文献

The Friedel-Crafts reaction of (1R)-8-phenylmenthyl glyoxylate with variously substituted furans was found to be efficiently promoted by SnCl(4) or magnesium salts with high diastereoselectivities. MgBr(2) performs especially well under simple, undemanding conditions, giving both high yields and high diastereoselectivities (>90%). Enantioselective Friedel-Crafts alkylation reactions of a series of substituted indoles with methyl trifluoropyruvate, catalyzed by a chiral nonracemic C(2)-symmetric 2,2'-bipyridyl copper(II) triflate complex, are described. The corresponding 3,3,3-trifluoro-2-hydroxy-2-indole-3-yl-propionic acid methyl esters were formed in good yield and in high enantiomeric excess (up to 90%). This is the first report of the use of a chiral nonracemic 2,2'-bipyridyl ligand in catalytic and enantioselective Friedel-Crafts alkylation reactions. The structural characterization of a copper(II) chloride complex of the chiral 2,2'-bipyridyl ligand by X-ray crystallography is also presented. [reaction: see text]  相似文献

Abstract

The reaction of novel chiral selenonium and telluronium ylides was investigated with aldehydes and compared to the sulfur analogues. (2R,5R)-2,5-Dimethylselenolane was prepared and reacted as a catalyst for the benzylidenation of aldehydes. Disubstituted epoxides were readily prepared with a (surprising) absence of diastereoselectivity, and with enantiomeric excesses higher than 90%. The reaction of a tellurium analogue, (2S,5S)-2,5-diethyltellurolane, afforded the oxirane in very moderate yield and e.e.'s in the range of 62–82%. Though this was less productive, it is the first report of a chiral telluronium ylide leading to an asymmetric epoxidation of aldehydes.  相似文献

The asymmetric ring‐opening/cyclization of cyclopropyl ketones with primary amine nucleophiles was catalyzed by a chiral N,N′‐dioxide/scandium(III) complex through a kinetic resolution process. A broad range of cyclopropyl ketones and primary amines are suitable substrates of this reaction. The corresponding products were afforded in excellent enantioselectivities and yields (up to 97 % ee and 98 % yield) under mild reaction conditions. This method provides a promising access to chiral 2,3‐dihydropyrroles as well as an effective procedure for the kinetic resolution of 2‐substituted cyclopropyl ketones.  相似文献

A remarkable increase in catalytic activity is found for the asymmetric cyclopropanation of 2,5-dimethyl-2,4-hexadiene with diazoacetate by use of the chiral copper Schiff-base complexes, which are derived from substituted salicylaldehydes, chiral aminoalcohols, and copper acetate monohydrate. Furthermore, a combination of a chiral copper Schiff-base with a Lewis acid showed an increase in yield (up to 90%) and in enantioselectivity (up to 90% ee) for the asymmetric cyclopropanation of the diene with t-butyl diazoacetate at 20 °C.  相似文献

Chiral diarylmethylamines (DAMA) are important structural motifs widely present in pharmaceuticals, natural products, and chiral ligands. Herein, we reported a highly enantioselective synthesis of chiral DAMAs via cobalt-catalyzed enantioselective C−H alkoxylation strategy. The reaction features easy operation, the use of earth-abundant and cost-efficient cobalt(II) catalyst, and readily available ligand. A range of chiral DAMAs were efficiently synthesized in high yields with excellent enantioselectivities (up to 90 % yield and up to 99 % ee) through desymmetrization and parallel kinetic resolution. Moreover, this protocol was also compatible with the synthesis of chiral benzylamines via kinetic resolution.  相似文献

An allylic zinc reagent bearing a chiral bisoxazoline ligand adds to a substituted cyclopropenone acetal to produce an optically active cyclopropanone acetal possessing a quaternary chiral center in high yield with 97.8-99.8% ee. The steric effects of the bulky bisoxazoline ligand overwhelm the regioselectivity inherent to the electronic property of the olefinic acceptor. High pressure exerts favorable effects on the reaction rate without affecting the high enantio- or regioselectivity.  相似文献

Two kinds of novel chiral ionic liquids based on imidazolium have been synthesized from chiral pool by a simple and straightforward procedure in good overall yields (44-60%). Methyl Grignard undergoes highly regio (>90/10) and enantioselective (ee 91-96%) copper catalyzed allylic substitution on cinnamyl-type chlorides. CuBr (3%) and 3.3% of a chiral phosphoramidite ligand are sufficient for a complete reaction. The synthesis of a precursor of (+)-Naproxen is described. The reaction can be extended to alkyl substituted allylic chlorides (ee 72%).  相似文献

A broad range of substituted anilines were tolerated, and the corresponding products were obtained in high yield (up to 99 %) with good enantioselectivity (up to 94 % ee) under mild reaction conditions. Benzylzinc reagent reacted with α-imino ester (2) at the α-carbon exclusively, though other organometallic reagents such as Mg. Al, Cu, Tl, and B derivatives reacted at the nitrogen atom. Use of the (S)-amine as a chiral auxiliary of 2 created the R chirality at the imino carbon. Very high chiral Induction was realized In the reaction of prenylzinc reagent wlih α-imino 8-(-)phenylmenthyl ester (10). The reaction of 2 with heteroatom substituted allylic organometalic compounds (15) gave the corresponding α-heterostom substituted amino acid derivatives (16). Here again, the allylic zinc reagent gave the adduct in higher yield than the corresponding Tl, Al, and B reagents.  相似文献

The conjugate addition of thioacetic acid to methacrylamides with chiral C2-symmetric trans-2,5-disubstituted pyrrolidines afforded the addition products in excellent stereoselectivities (>99% de) and good yields (80–90%). The high selectivity was attributed mainly to the steric effect of the chiral auxiliaries. The cyclic nature of the chiral auxiliaries seemed also important for both the stereoselectivity and the reaction rate. Acidic hydrolysis of the adduct containing (2R,5R)-bis(methoxymethyl)pyrrolidine gave (S)-3-mercapto-2-methylpropanoic acid, a key intermediate for captopril, in 98% ee and 96% yield. The chiral auxiliary was recovered in the demethylated form of N-Boc-(2R,3R)-bis(hydroxymethyl)pyrrolidine in 90% yield.  相似文献

The iridium phosphoramidite complex-promoted regio- and enantioselective reaction of allylic carbonates with sodium triisopropylsilanethiolate produced allylic sulfides in 40-77% yields with up to 97 : 3 (branched?:?linear) and 89% ee, which were readily transformed into chiral thiol in 68% yield with 87% ee or disulfides with two chiral C-S bond centers in 40-73% yields with up to 90 : 10 dr and 99% ee.  相似文献

Highly efficient catalytic asymmetric Claisen rearrangements of O‐propargyl β‐ketoesters and O‐allyl β‐ketoesters have been accomplished under mild reaction conditions. In the presence of the chiral N,N′‐dioxide/NiII complex, a wide range of allenyl/allyl‐substituted all‐carbon quaternary β‐ketoesters was obtained in generally good yield (up to 99 %) and high diastereoselectivity (up to 99:1 d.r.) with excellent enantioselectivity (up to 99 % ee).  相似文献

The s-BuLi-sparteine base combination deprotonated the C-2' position of 1,2,3,4,5-pentamethylazaferrocene and subsequent reaction with a range of electrophiles gave C-2 substituted products in 76-93% yield and approximately 80% ee. The products could be recrystallised to enrich ee's to >90%. Resubjection of the initial addition products ( approximately 80% ee) to the deprotonation conditions led to a kinetic resolution to give products with >90% ee and superior overall yields compared to recrystallisation for the cases where the electrophiles were Ph2CO, MeI and Ph2S2. Transmetallation of the 2-lithiopentamethylazaferrocene ( approximately 80% ee) with ZnCl2 allowed palladium catalysed cross coupling with a variety of C-2 haloaryl, heteroaryl and vinyl groups to give some novel C-2' substituted pentamethylazaferrocene derivatives in 61-77% yield in 80% ee. Potential N,N-chelate ligands were recrystallised to >95% ee. A novel C2-symmetric bis-pentamethylazaferrocene could be synthesised by an iron catalysed oxidative coupling of the enatioenriched C-2 lithio derivative and in the presence of a PhMe-Et2O solvent mixture proceeded in 97% ee.  相似文献
