Reduction of indolin-2-ones with lithium aluminium hydride (LAH) or diisobutylaluminum hydride (DIBAL) and desulfurization of indoline-2-thiones with Raney-Ni were investigated. Treatment of indoline-2-ones 1 with LAH or DIBAL yield indoles 4 and/or indolines 3 in moderate-to high yield depending on the substituents at N and C(3) of 1 . Indoline-2-thiones 2 were desulfurized with Raney-Ni to give indoles 4 and/or indolines 3 .  相似文献

Dearomative annulation of indoles has emerged as a powerful tool for the preparation of polycyclic indoline‐based alkaloids. Compared with well‐established methods towards five‐membered‐ring‐fused indolines, the six‐membered‐ring‐fused indolines are rarely accessed under thermal conditions. Herein, a dearomative [4+2] annulation between different indoles is developed through an electrochemical pathway. This transformation offers a remarkably regio‐ and stereoselective route to highly functionalized pyrimido[5,4‐b]indoles under oxidant‐ and metal‐free conditions. Notably, this electrochemical approach maintains excellent functional‐group tolerance and can be extended as a modification tactic for pharmaceutical research. Preliminary mechanism studies indicate that the electrooxidation annulation proceeds through radical–radical cross‐coupling between an indole radical cation and an N‐centered radical generated in situ.  相似文献

Unsaturated dialkylketenes 7a, 7b and 7c undergo intramolecular [2 + 2]cycloadditions to give 8a (45%), 9b (23%) and 9c (45%). Intramolecular cycloadditions of dialkylketenes give higher yields than intramolecular cycloadditions of monoalkylketenes, even though dialkylketenes are less reactive than monoalkylketenes. An intramolecular competition experiment with ketene 17 establishes that trans-alkenes are approximately 33 times more reactive than cis-alkenes in intramolecular cycloadditions. Ketene 36 furnishes 22% of the expected bicyclo[3.2.0]heptanone 37 and 28% of bicyclo[3.1.1]heptanone 38.  相似文献

Photochemical [2 2] cycloaddition of C60 with podophyllotoxin derivative containing a cyclohexadienone group in o-dichlorobenzene afforded an isomeric mixture of adducts and a pure adduct of C60-fused podophyllotoxin derivatives. Refluxing dimethyl acetylenedicarboxylate (DMAD) or 1,2-dibenzoylacetylene with isatin-3-thiosemicarbazone or isatin-3-thiocarbohydrazone in methanol produced 3-substituted oxindoles in good yields. Reaction of equimolecular amounts of 5,6-diamino-urasil-solfate, trans-(1R,2R)-diaminocyclohexane, and 3,4-diamino-1,2,4-triazol-5-methyl hydrochlorid with isatin in ethanol (85%) afforded tetracyclic ring systems and ring-opened products in mild reaction conditions.  相似文献

[2+2] Cycloaddition reactions of P2 with alkenes were predicted to have concerted paths, that is, pseudoexcitation, distorted 2πs+2πs, and 2πs+2πa processes without any interventions of intermediates. The pseudoexcitation and/or distorted 2πs+2πs paths with retention of configuration of alkenes are kinetically preferred to the 2πs+2πa path with inversion of configuration. The reactions were predicted from the appreciable difference in the calculated enthalpies of activation to be stereospecific.  相似文献

A chiral rhodium complex catalyzed an enantioselective [2+2+2] cycloaddition of unsymmetrical diynes with norbornene, and tetracyclic products were obtained in good to excellent ee. The photocycloaddition of benzothiazole-2-thiones to electron-rich and aryl-substituted alkenes are described. Irradiation of N-unsubstituted benzothiazole-2-thione ( 1 ) in the presence of alkenes 3 gave 2-(2′-mercaptoalkyl)benzothiazoles 4 , and 2-substituted benzothiazoles 5 and 6 (in the case of 3a and 3h , resp.) through the ring cleavage of an intermediate 2-aminothietane (Schemes 1 and 3 ). The latter was formed by [2+2] cycloaddition of the C?S bond of 1 and the C?C bond of 3 . Irradiation of N-methylbenzothiazole-2-thione ( 2 ) and 2-methylpropene ( 3a ) gave the spiro-1,3-dithiane 8 , 1,2,6-benzodithiazocin-5-one 9 , and disulfide 10 . Ruthenium-catalyzed [2+2] cycloadditions between bicyclic alkenes and ynamides were investigated. The ynamide moiety was found to be compatible with the ruthenium-catalyzed cycloaddition conditions giving the corresponding cyclobutene cycloadducts in moderate to good yields (up to 97%). Diastereoselective cycloaddition utilizing chiral cyclic ynamides were also examined and a low to moderate level of asymmetric induction was observed.  相似文献

The stereogenic centers at C3 and C12 of meloscine ( 3 ) can be established in the photochemical key step 1 → 2 . 1,2‐retro‐Benzilic acid rearrangement to a five‐membered ring, reductive amination, Claisen rearrangement, and ring‐closing metathesis are further key steps in the transformation of cyclobutane 2 into the target molecule 3 (14 steps, 9 % overall yield). Enantioselective access to (+)‐meloscine was possible when the [2+2]‐photocycloaddition was conducted in the presence of a chiral template.
