In this study, a novel classical thermo‐ and salt‐sensitive semi‐interpenetrating polymer network (semi‐IPN) hydrogel composed of poly(N,N‐diethylacrylamide) (PDEAm) and κ‐carrageenan (KC) was synthesized by free radical polymerization. The structure of the hydrogels was studied by Fourier transform infrared spectroscopy (FTIR) and scanning electron microscopy (SEM). FTIR and SEM revealed that the semi‐IPN hydrogels possessed the structure of H‐bonds and larger number of pores in the network. Compared to the PDEAm hydrogel, the prepared semi‐IPN hydrogels exhibited a much faster response rate to temperature changes and had larger equilibrium swelling ratios at temperatures below the lower critical solution temperature (LCST). The salt‐sensitive behavior of the semi‐IPN hydrogels was dependent on the content of KC. In addition, during the reswelling process, semi‐IPN hydrogels showed a non‐sigmoidal swelling pattern. Summary: Specific temperature‐responsive biodegradable hydrogels were synthesized and characterized in terms of their regulation of enzymatic accessibility based on the physical properties of the temperature‐responsive polymers. The hydrogels consist of glycidyl methacrylate‐modified dextran grafted with the poly(N‐isopropylacrylamide) (PNIPAAm) homopolymer, and cross‐linked by co‐polymerization with NIPAAm and N,N‐dimethylacrylamide (DMAAm). The coil‐globule change in the grafted poly(NIPAAm) chains and only a slight dehydration of the poly(NIPAAm‐co‐DMAAm) cross‐linkers are effective in controlling the enzymatic degradation over a specific temperature range.

The thermo‐responses of the graft chains (steric hindrance) and the crosslinkers (slight deswelling of the hydrogel networks) control the enzymatic degradation of the hydrogel.  相似文献

Three series of water‐soluble semi‐rigid thermo‐responsive polymers with well‐defined molecular weights based on mesogen‐jacketed liquid crystal polymers, poly[bis(N‐(2‐hydroxypropyl) pyrrolidone) 2‐vinylterephthalate] [P(2‐HPPVTA)], poly[bis(N‐(1‐methyl‐2‐hydroxyethyl) pyrrolidone) 2‐vinylterephthalate] [P(1‐M‐2‐HEPVTA)] and poly[bis(N‐hydroxypropyl pyrrolidone) 2‐vinylterephthalate] (PHPPVTA) have been synthesized via reversible addition‐fragmentation chain transfer polymerization. The steric hindrance effects on liquid crystalline property and thermo‐responsive behaviors of semi‐rigid water‐soluble polymers (P(2‐HPPVTA), P(1‐M‐2‐HEPVTA), and PHPPVTA) were carefully investigated. From molecular structure, the steric hindrance of P(1‐M‐2‐HEPVTA) is stronger than that of P(2‐HPPVTA). Polarized light microscope and one‐dimensional wide‐angle X‐ray diffraction revealed that both the P(2‐HPPVTA) and P(1‐M‐2‐HEPVTA) display a columnar nematic phase, indicating that the steric hindrance effect do not affect liquid crystalline behavior of the polymers. The dynamic light scattering results demonstrated that P(1‐M‐2‐HEPVTA) exhibited lower cloud point compared with that of P(2‐HPPVTA) at the same mass concentration and the same molecular weight. The more significant molecular weight and concentration dependence on cloud point have been observed in P(2‐HPPVTA) solution than in P(1‐M‐2‐HEPVTA) solution. We also discovered that the cloud points of both P(2‐HPPVTA) and P(1‐M‐2‐HEPVTA) solution are lower in D2O than in H2O. It is noted that the cloud point of PM‐2 is 9.9 °C lower in D2O than in H2O, much less pronounced than the cloud point difference of PH‐2. The differences of thermo‐responsive behaviors between P(2‐HPPVTA) and P(1‐M‐2‐HEPVTA) were resulted from the steric hindrance effect existed in their side groups. © 2013 Wiley Periodicals, Inc. J. The semi‐IPN hydrogels consisting of poly(methacrylic acid) and guar gum (GG) are prepared at room temperature using water as solvent. 5‐aminosalicylic acid (5‐ASA) is entrapped in the hydrogel in the synthesis of hydrogel and all entrapment efficiencies are found above 85%. The hydrogel shows excellent pH‐sensitivity. It exhibited minimum swelling in an acidic pH medium through the formation of a complex hydrogen‐bonded structure and maximal swelling due to the electrostatic repulsion due to the ionization of the carboxylic groups in pH 7.4 medium. The degradation in vitro shows that the degree of degradation (R%) depended on the concentration of cross‐linking agent and content of GG. The hydrogel shows a minimum release of 5‐ASA due to the complex hydrogen bonded structure of the hydrogels in the medium of pH 2.2. The enzymatic degradation of hydrogels by cecal bacteria can accelerate the release of 5‐ASA entrapped in the hydrogel in pH 7.4 medium. Copyright © 2007 John Wiley & Sons, Ltd.  相似文献

A novel poly(N‐isopropylacrylamide) (PNIPA)/PNIPA interpenetrating polymer network (IPN) was synthesized and characterized. In comparison with conventional PNIPA hydrogels, the shrinking rate of the IPN hydrogel increased when gels, swollen at 20 °C, were immersed in 50 °C water. The phase‐transition temperature of the IPN gel remained unchangeable because of the same chemical constituent in the PNIPA gel. The reswelling kinetics were slower than those of the PNIPA hydrogel because of the higher crosslinking density of the IPN hydrogel. The IPN hydrogel had better mechanical strength because of its higher crosslinking density and polymer volume fraction. The release behavior of 5‐fluorouracil (5‐Fu) from the IPN hydrogel showed that, at a lower temperature, the release of 5‐Fu was controlled by the diffusion of water molecules in the gel network. At a higher temperature, 5‐Fu inside the gel could not diffuse into the medium after a burst release caused by the release of the drug on the surface of the gel. © 2004 Wiley Periodicals, Inc. Stimuli‐responsive bioconjugated hydrogels that can respond to a target antigen (antigen‐responsive hydrogels) were prepared by introducing antigen‐antibody bindings as reversible crosslinks into the gel networks. The preparation conditions of the antigen‐responsive hydrogels and the mechanism of the antigen‐responsive behavior were investigated, focusing on bioconjugated hydrogel structures. This article also focuses on the effect of semi‐interpenetrating polymer network (semi‐IPN) structures on the antigen‐responsive swelling/shrinking behavior of bioconjugated hydrogels with antigen‐antibody bindings. Poly(N‐vinyl‐pyrrolidone) (PVP) hydrogel has been considered as a very interesting and promising thermosensitive material. The most vital shortcoming of PVP hydrogel as thermosensitive material is that it does not exhibit thermosensitivity under usual conditions. In this work, semi‐interpenetrating polymer network (semi‐IPN) hydrogels based on PVP and carboxymethylcellulose (CMC) were prepared. The volume phase transition temperature (VPTT) of the hydrogels was determined by swelling behavior and differential scanning calorimetry (DSC). The results showed that the VPTT was significantly dependent on CMC content and the pH of the swelling medium. The amount of CMC in the semi‐IPN hydrogels was 0.050, 0.075, and 0.100 g, the VPTT in buffer solution of pH 1.2 was 29.9 °C, 27.5 °C and 24.5 °C, respectively. In addition, the VPTT occurred in buffer solution of pH 1.2, but did not appear in alkaline medium. Bovine serum albumin (BSA) as a model drug was loaded and the in vitro release studies were carried out in different buffer solutions and at different temperatures. The results of this study suggest that PVP/CMC semi‐IPN hydrogels could serve as potential candidates for protein drug delivery in the intestine. © 2010 Wiley Periodicals, Inc. An organosoluble polyimide based on bipyridyl moiety and an alkoxysilane dye have been developed for second‐order non‐linear optics (NLOs). This bipyridine‐containing polyimide exhibits a glass transition temperature of 254°C and a degradation temperature of 400°C. An NLO‐active semi‐interpenetrating network (IPN) system was prepared by blending the polyimide with the alkoxysilane dye via in situ sol‐gel process of alkoxysilane. The selection of this bipyridine‐containing polyimide as the polymeric matrices provides improved solubility and thermal stability, and most importantly enhanced intermolecular interactions. No aggregation of the NLO chromophores in the polyimide matrices was observed through morphology and NLO studies. Under the limitation of chromophore degradation at elevated temperatures, the pristine poled/cured alkoxysilane dye exhibits poorer long‐term stability. By introducing the polyimide upon a silica network by the semi‐IPN system, randomization of the oriented chromophores can be effectively suppressed. Using in situ contact poling, the r33 coefficients of 2.2–17.0 pm/V were obtained for the optically clear semi‐IPN NLO materials. Preparation and characterization of novel semi‐interpenetrating polymer network (semi‐IPN) hydrogels based on partially hydrolyzed polyacrylamide (HPAM) and scleroglucan solution crosslinked with chromium triacetate are described. Effects of scleroglucan concentration on the gelation process and swelling behavior of synthesized hydrogels in different media were investigated using dynamic rheometery and swelling tests, respectively. Oscillatory shear rheology showed that the limiting storage modulus of the semi‐IPN gels increased with increase in scleroglucan concentration. It was also found that the viscous energy dissipating properties of the semi‐IPN gels decreased with increase in the crosslinker concentration of the gelation system. In addition, the loss factor slightly decreased by increasing the scleroglucan content, indicating that the viscous properties of this gelling system decreased more than its elastic properties. The swelling tests showed that the equilibrium swelling ratio (ESR) of the semi‐IPN networks decreased with increase in scleroglucan content, due to the decrease of ionic groups of polyelectrolyte hydrogel. However, the semi‐IPN gels showed lower salt sensitivity in synthetic oil reservoir water as compared with HPAM gels. To obtain a hydrogel‐like elastic membrane, we prepared semi‐interpenetrating polymer networks (IPNs) by the radical polymerization of methacrylates such as 2‐methacryloyloxyethyl phosphorylcholine (MPC), 2‐hydroxyethylmethacrylate, and triethyleneglycol dimethacrylate diffused into segmented polyurethane (SPU) membranes swollen with a monomer mixture. The values of Young's modulus for the hydrated semi‐IPN membranes were less than that for an SPU membrane because of higher hydration, but they were much higher than that for a hydrated MPC polymer gel (non‐SPU). According to a thermal analysis, the MPC polymer influenced the segment association of SPU. The diffusion coefficient of 8‐anilino‐1‐naphthalenesulfonic acid sodium salt from the semi‐IPN membrane could be controlled with different MPC unit concentrations in the membrane, and it was about 7 × 102 times higher than that of the SPU membrane. Fibroblast cell adhesion on the semi‐IPN membrane was effectively reduced by the MPC units. We concluded that semi‐IPNs composed of the MPC polymer and SPU may be novel polymer materials possessing attractive mechanical, diffusive‐release, and nonbiofouling properties. © 2002 Wiley Periodicals, Inc. Diblock copolymers of poly[2‐(dimethylamino)ethyl methacrylate]‐block‐poly[di(ethylene glycol) methyl ether methacrylate], PDMAEMA‐b‐PDEGMA, were synthesized by reversible addition–fragmentation chain transfer polymerization. The block ratio was varied to study the influence on the lower critical solution temperature and the corresponding phase transition in water. Therefore, turbidimetry, differential scanning calorimetry (DSC), dynamic light scattering (DLS), and laser Doppler velocimetry were applied. Additionally, asymmetric flow field‐flow fractionation (AF4) coupled to DLS and multiangle laser light scattering (MALLS) was established as an alternative route to characterize these systems in terms of molar mass of the polymer chain and size of the colloids after the phase transition. It was found that AF4–MALLS allowed accurate determination of molar masses in the studied range. Nevertheless, some limitations were observed, which were critically discussed. The cloud point and phase transition of all materials, as revealed by turbidimetry, could be confirmed by DSC. For block copolymers with block ratios in the range of 50:50, a thermo‐induced self‐assembly into micellar and vesicular structures with hydrodynamic radii (Rh) of around 25 nm was observed upon heating. At higher temperatures, a reordering of the self‐assembled structures could be detected. The thermo‐responsive behavior was further investigated in dependence of pH value and ionic strength. Variation of the pH value mainly influences the solubility of the PDMAEMA segment, where a decrease of the pH value increases the transition temperature. A calix‐conjugated thermo‐responsive hydrogel containing 15% tetra(5‐hexenyloxy)‐p‐tert‐butylcalix[4]arene (HBCalix), P(NIPAM‐co‐HBCalix), was used to remove nickel(II) ions from water. Both thermo‐sensitive properties and the Ni2+‐adsorption capabilities of the prepared P(NIPAM‐co‐HBCalix) hydrogels are investigated. Introduction of the monomer HBCalix considerably enhanced the adsorption of Ni2+ onto the hydrogel by chelation between hexenyloxy groups and metal ion. When HBCalix units capture Ni2+ and forms HBCalix/Ni2+ host–guest complexes, the lower critical solution temperature of the hydrogel shifts to a higher temperature due to both the repulsion between charged HBCali/Ni2+ groups and the osmotic pressure within the hydrogel. Adsorption studies were carried out by varying contact time, counter ion and initial concentration of Ni2+. The evaluation of adsorption properties showed that the hydrogel exhibited better correlation with Langmuir isotherm model. P(NIPAM‐co‐HBCalix) could be used repeatedly with little loss in adsorption capacity by simply changing the environmental temperature. A new type of glucose‐responsive hydrogel with rapid response to blood glucose concentration change at physiological temperature has been successfully developed. The polymeric hydrogel contains phenylboronic acid (PBA) groups as glucose sensors and thermo‐responsive poly (N‐isopropylacrylamide) (PNIPAM) groups as actuators. The response rate of the hydrogel to environmental glucose concentration change was significantly enhanced by introducing grafted poly(N‐isopropylacrylamide‐co‐3‐acrylamidophenylboronic acid) [poly(NIPAM‐co‐AAPBA)] side chains onto crosslinked poly(NIPAM‐co‐AAPBA) networks for the first time. The synthesized comb‐type grafted poly(NIPAM‐co‐AAPBA) hydrogels showed satisfactory equilibrium glucose‐responsive properties, and exhibited much faster response rate to glucose concentration change than normal type crosslinked poly(NIPAM‐co‐AAPBA) hydrogels at physiological temperature. Such glucose‐responsive hydrogels with rapid response rate are highly attractive in the fields of developing glucose‐responsive sensors and self‐regulated drug delivery systems. A series of granulated semi‐interpenetrating polymer network (semi‐IPN) superabsorbent hydrogels composed of chitosan‐g‐poly(acrylic acid) (CTS‐g‐PAA) and poly(vinyl alcohol) (PVA) were prepared by solution polymerization using ammonium persulfate (APS) as an initiator and N,N′‐methylenebisacrylamide (MBA) as a crosslinker. The effects of reaction conditions such as the concentration of MBA, the weight ratio of AA to CTS, and the content of PVA on water absorbency were investigated. Infrared (IR) spectra and differential scanning calorimetry (DSC) analyses confirmed that AA had been grafted onto CTS backbone, and PVA semi‐interpenetrating into CTS‐g‐PAA networks. SEM analyses indicated that CTS‐g‐PAA/PVA has improved porous surface and PVA was uniformly dispersed in CTS‐g‐PAA network. The semi‐IPN hydrogel containing 10 wt% PVA shows the highest water absorbency of 353 and 53 g g?1 in distilled water and 0.9 wt% NaCl solution, respectively. Swelling behaviors revealed that the introduction of PVA could improve the swelling rate and enhance the pH stability of the superabsorbent hydrogel. Poly(vinyl alcohol) (PVA) was blended with sodium alginate (Alg) in various ratios and crosslinked with calcium chloride and made into hydrogel membranes. The dependence of the swelling behavior of these Alg‐Ca/PVA hydrogels on pH was investigated. The temperature‐dependent swelling behavior of the semi‐interpenetrating network (semi‐IPN) hydrogels was examined at temperatures from 2 to 45°C and the enthalpy of mixing (ΔHmix) was determined at various temperatures. The molecular structure of the hydrogels was studied by infrared spectroscopy and their water structure in the semi‐IPN hydrogels was measured by differential scanning calorimetry (DSC). The influence of Ca2+ content on the network structure of Alg‐Ca/PVA hydrogels was investigated in terms of the compressive elastic modulus, effective crosslinking density, and the polymer–solvent interaction parameter based on the Flory theory. The loading of alizarin red S (ARS) followed the Langmuir isotherm mechanism and the release kinetics of ARS from the Alg‐Ca/PVA hydrogels followed the Fickian diffusion mechanism. Novel stimuli‐responsive hydrophilic microspheres were prepared by free radical polymerization of hydroxyethyl methacrylate (HEMA) and methacrylic acid (MA), as hydrophilic monomers, and N‐isopropylacrylamide (NIPAAm) and N,N′‐ethylenebisacrylamide (EBA), as thermo‐sensitive monomer and crosslinker, respectively. Hydrophilic comonomers were introduced in the macromolecular network to synthesize materials with tunable thermal behavior. In addition, by introducing in the polymerization feed both a hydrophilic and a pH‐sensitive monomer, such as MA, dual stimuli‐responsive (pH and temperature) hydrogels were synthesized. The incorporation of monomers in the network was confirmed by infrared spectroscopy, while the network density and the shape of hydrogels was found to strictly depend on the concentration of monomers in the polymerization feed. Thermal analyses showed negative thermo‐responsive behavior with pronounced water affinity of microspheres at a temperature lower than lower critical solution temperature (LCST). In our experiment, the LCST values of the hydrogels were in the range 34.6–37.5°C, close to the body temperature, and the amount of hydrophilic moieties in the polymeric network allows to collect shrinking/swelling transition temperatures higher than the LCST of NIPAAm homopolymers. In order to test the preformed materials as drug carriers, diclofenac diethylammonium salt (DDA) was chosen and drug entrapment percent was determined. Drug release profiles, in media at different temperature and pH, depend on hydrogels crosslinking degree and drug–bead interactions. In this thesis, we studied the convenient synthesis and characterizations of thermo‐responsive materials with double response. To achieve these, AB‐type diblock copolymers comprising of poly(N‐isopropylacrylamide) (NIPAAm) segment and poly(NIPAAm‐co‐(N‐(hydroxymethyl)acrylamide) (HMAAm)) one were designed. That was synthesized in one‐pot using an atom transfer radical polymerization (ATRP) technique. Poly(NIPAAm‐co‐HMAAm)s synthesized separately showed sensitive thermo‐response and the cloud point was completely tunable by the composition of HMAAm. As expected, the block copolymers exhibited double thermo‐responsive profiles in aqueous solution. The responsive behavior was discussed by precise trace by 1H NMR and turbidity measurements. Novel itaconates having a pyrrolidinonyl moiety, i. e. 4‐methyl 1‐(2‐pyrrolidonylethyl) itaconate (PyEMI) and 1‐methyl 4‐(2‐pyrrolidonylethyl) itaconate (MPyEI) were synthesized by the reaction of 2‐N‐hydroxyethyl‐pyrrolidinone (HEPy) with 4‐methyl or 1‐methyl itaconyl chloride in the presence of triethylamine. The polymerization of PyEMI and MPyEI were conducted in two ways. By photopolymerization using 2,2‐dimethoxy‐2‐phenylacetophenone as a photosensitizer, P(PyEMI)‐A and P(MPyEI) were obtained. By redox polymerization using N,N,N,N‐tetra‐methylethylenediamine – potassium persulfate as an initiating system, P(PyEMI)‐B was obtained. It was found that both polymers exhibit thermo‐responsive behavior in water. The thermo‐responsive behavior of P(MPyEI) is similar to that of P(PyEMI)‐A, however, P(PyEMI)‐B is more sensitive to a change in temperature than P(PyEMI)‐A. The fluid dynamic diameter of P(PyEMI)‐A and P(PyEMI)‐B in water were detected by a laser scattering method, and it was shown that the fluid dynamic diameter of the latter is about 5 times that of the former. Due to the difference in polymer chain size and shape, the thermo‐responsive behavior of P(PyEMI)‐B is more sensitive in the same mass concentration. The swelling ratios of PyEMI hydrogels with various amounts of cross‐linker at different temperature were determined. The thermo‐responsive behavior of the PyEMI hydrogel was compared with that of the P(PyEMI) aqueous solution.  相似文献

Statistical copolymerizations of 2‐(dimethylamino)ethyl methacrylate (DMAEMA) with 2‐vinylpyridine (2VP) with 80 to 99 mol % DMAEMA in the feed utilizing a succinimidyl ester‐terminated alkoxyamine unimolecular initiator (NHS‐BlocBuilder) at 80 °C in bulk were performed. The effectiveness of 2VP as a controlling comonomer is demonstrated by linear increases in number‐average molecular weight versus conversion, relatively low PDI (1.5–1.6 with up to 98% DMAEMA) and successful chain extensions with 2VP. Additional free nitroxide does not significantly improve control for the DMAEMA/2VP copolymerizations. The succinimidyl ester on the initiator permits coupling to amine‐terminated poly(propylene glycol) (PPG), yielding an effective macroinitiator for synthesizing a doubly thermo‐responsive block copolymer of PPG‐block‐P(DMAEMA/2VP). A detailed study of the thermo‐ and pH‐sensitivities of the statistical and block copolymers is also presented. The cloud point temperature of the statistical copolymers is fine tuned from 14 to 75 °C by varying polymer composition and pH. © 2012 Wiley Periodicals, Inc. Poly(ethylene glycol)(PEG)‐based interpenetrating polymeric network (IPN) hydrogels were prepared for the application of enzyme immobilization. Poly(acrylamide)(PAAm) was chosen as the other network of IPN hydrogel and different concentration of PAAm networks were incorporated inside the PEG hydrogel to improve the mechanical strength and provide functional groups that covalently bind the enzyme. Formation of IPN hydrogels was confirmed by observing the weight per cent gain of hydrogel after incorporation of PAAm network and by attenuated total reflectance/Fourier transform infrared (ATR/FTIR) analysis. Synthesis of IPN hydrogels with higher PAAm content produced more crosslinked hydrogels with lower water content (WC), smaller Mc and mesh size, which resulted in enhanced mechanical properties compared to the PEG hydrogel. The IPN hydrogels exhibited tensile strength between 0.2 and 1.2 MPa while retaining high levels of hydration (70–81% water). For enzyme immobilization, glucose oxidase (GOX) was immobilized to PEG and IPN hydrogel beads. Enzyme activity studies revealed that although all the hydrogels initially had similar enzymatic activity, enzyme‐immobilizing PEG hydrogels lost most of the enzymatic activity within 2 days due to enzyme leaching while IPN hydrogels maintained a maximum 80% of the initial enzymatic activity over a week due to the covalent linkage between the enzyme and amine groups of PAAm.
