A novel additive of multi‐walled carbon nanotubes (MWNTs) dispersed with cationic surfactants or mixed cationic/anionic surfactants was used for MEEKC separation of eight phenolic compounds, four glycosides, and one phenanthraquinone. In this context, several parameters affecting MEEKC separation were studied, including the dispersion agents of MWNTs, MWNTs content, oil type, SDS concentration, and the type and concentration of cosurfactant. Compared with conventional MEEKC, the addition of all types of MWNTs dispersions using single or mixed cationic surfactant solutions in running buffers was especially useful for improving the separation of solutes tested, as they influenced the partitioning between the oil droplets and aqueous phase due to the exceptional electrical properties and large surface areas of MWNTs. Use of cationic surfactant‐coated MWNTs (6.4 μg/mL) as the additive in a microemulsion buffer (0.5% octanol, 2.8% SDS, 5.8% isopropanol, and 5 mM borate buffer) yielded complete resolution of 13 analytes. The proposed method has been successfully applied for the detection and quantification of the studied compounds in a complex matrix sample (Compound Xueshuantong capsule).  相似文献

Microemulsion EKC (MEEKC) was developed for quantitative analysis of curcuminoids, such as curcumin (C), demethoxycurcumin (D), and bis-demethoxycurcumin (B). MEEKC separation of curcuminoids was optimized, and a change in resolution was explained using a modified equation for resolution in MEEKC without electroosmosis. The suitable MEEKC conditions for separation of curcuminoids were obtained to be the microemulsion buffer containing 50 mM phosphate buffer at pH 2.5, 1.1% v/v n-octane as oil droplets, 180 mM SDS as surfactant, 890 mM 1-butanol as cosurfactant, and 25% v/v 2-propanol as organic cosolvent; applied voltage of -15 kV; and separation temperature 25 degrees C. Achieved baseline resolution of C:D and D:B was obtained with R(s) -2.4 and analysis time within 18 min. In addition, high accuracy and precision of the method were obtained. This MEEKC method was used for quantitative determination of individual curcuminoids in medicinal turmeric capsules and powdered turmeric used as coloring additive in food, with simple sample preparation such as solvent extraction, dilution, and filtration, and without cleaning up by SPE.  相似文献

Microemulsion EKC (MEEKC) was used for the determination of ketorolac and its three impurities. The microemulsion system was optimized, for the first time in the literature, using a multivariate strategy involving a mixture design. A 13-run experimental plan covering an experimental domain defined by the components aqueous phase (10 mM borate buffer pH 9.2), oil phase (n-heptane) and surfactant/cosurfactant (SDS/n-butanol) was carried out. Good results were obtained with all microemulsions tested considering as responses analysis time and resolution, and according to the desirability function the best microemulsion system was constituted by 90.0% 10 mM borate buffer, 2.0% n-heptane, 8.0% of SDS/n-butanol in 1:2 ratio. Finally, with the aim of reducing analysis time, a response surface study was carried out in the experimental domain defined by the process variables temperature and voltage and the best values were 17 degrees C and -17 kV, respectively. Applying the optimised conditions, a complete resolution among the analytes was obtained in about 3 min using the short-end injection method. The method was validated for both drug substances and drug product and was applied to the quality control of ketorolac in coated tablets. A comparison of MEEKC, MEKC and CEC for assaying ketorolac and its related substances has been made.  相似文献

Microemulsion electrokinetic chromatography (MEEKC) was developed for quantitative analysis of avermectins, such as abamectin, doramectin and ivermectin, in commercial formulations, using the microemulsion buffer containing a 50 mM phosphate buffer at pH 2.5, 1.1% (v/v) n-octane as oil droplets, 180 mM sodium dodecylsulphate as surfactant, 890 mM 1-butanol as co-surfactant and 30% (v/v) ethanol as organic co-solvent. High accuracy and precision of the method were obtained. The contents of avermectins in commercial formulations determined by MEEKC were found to be insignificantly different with those determined by high performance liquid chromatography (HPLC). Therefore, MEEKC can be used an alternative method to HPLC for quantitative determination of avermectins.  相似文献

In this study, microemulsion electrokinetic chromatography (MEEKC) and micellar electrokinetic chromatography (MEKC) were compared for their abilities to separate and detect thirteen phenolic compounds (syringic acid, p-coumaric acid, vanillic acid, caffeic acid, gallic acid, 3,4-dihydroxybenzoic acid, 4-hydroxybenzoic acid, (+)-catechin, (-)-epigallocatechin, (-)-epicatechin gallate, (-)-epigallocatechin gallate, (-)-epicatechin, and (-)-gallocatechin), and two other ingredients (caffeine and theophylline) in teas and grapes. Separation of phenolic compounds was improved by changing the SDS concentration for MEEKC, but the SDS concentration rarely affected the resolution for MEKC. Organic modifier (acetonitrile or methanol) was found to markedly influence the resolution and selectivity for both MEEKC and MEKC systems. In addition, a higher voltage and a higher column temperature improved the separation efficiency without any noticeable reduction in resolution for MEEKC whereas they caused a poor resolution for the MEKC system. Although separations with baseline resolution were achieved by the optimized MEEKC and MEKC methods, the separation selectivity resulting from the proposed MEEKC method was completely different from that of MEKC.  相似文献

In the present work, several MEEKC systems are studied to assess their suitability for lipophilicity determination of acidic, neutral, and basic compounds. Thus, several microemulsion compositions over a wide range of pH values (from 2.0 to 12.0), containing heptane, 1?butanol and different types and amounts of surfactant (SDS or sodium cholate: from 1.3 to 3.3%) are characterized using Abraham's solvation model. The addition of acetonitrile (up to 10%) is also studied, since it increases the resolution of the technique for the most lipophilic compounds. The system coefficients obtained are very similar to those of the 1?octanol/water, used as the reference lipophilicity index, allowing simple and linear correlations between the 1?octanol/water partition values (log Po/w) and MEEKC mass distribution ratios (log kMEEKC). Variations in the microemulsion composition (aqueous buffer, surfactant, concentration of ACN) did not significantly affect the similarity of the MEEKC systems to log Po/w partition.  相似文献

Microemulsion electrokinetic chromatography (MEEKC) is a CE separation technique, which utilizes buffered microemulsions as the separation media. In the past two decades, MEEKC has blossomed into a powerful separation technique for the analysis of a wide range of compounds. Pseudostationary phase composition is so critical to successful resolution in EKC, and several variables could be optimized including surfactant/co‐surfactant/oil type and concentration, buffer content, and pH value. Additionally, MEEKC coupled with online sample preconcentration approaches could significantly improve the detection sensitivity. This review comprehensively describes the development of MEEKC from the period 1991 to 2012. Areas covered include basic theory, microemulsion composition, improving resolution and enhancing sensitivity methods, detection techniques, and applications of MEEKC.  相似文献

A rapid screening assay for the determination of octanol-water partition coefficients (log P(OW)) of pharmaceuticals was developed by using pressure-assisted microemulsion electrokinetic chromatography (MEEKC). The microemulsion system contains 50 mM sodium dodecyl sulfate, 0.87 M l-butanol, 82 mM heptane, and 50 mM borate-phosphate (2:3) at pH 10. Ten standard compounds with known log P(OW) values from -0.26 to 4.88 were used for constructing the calibration curve of log P(OW) against the MEEKC retention factor, log k. The log P(OW) values of the compounds were calculated based on the log k values measured by MEEKC and the slope and intercept of the calibration curve. For 13 literature and 32 Roche compounds, about 90% of the log P(OW) values measured by MEEKC are within 0.5 log units of the values from the literature and potentiometric titration. The throughput is about 2 samples/h using +20 kV voltage plus 5 mbar air pressure for separation. This MEEKC method is applicable for log P(OW) screening of weakly basic, weakly acidic, and neutral pharmaceuticals with log P(OW) = 0-5 and pKa < or = 10.  相似文献

We describe herein the synthesis of a triptycene-based surfactant designed with the ability to solubilise single-walled carbon nanotubes (SWNTs) and C(60) in water through non-covalent interactions. Furthermore, an amphiphilic naphthalene-based surfactant with the same ability to solubilise SWNTs and C(60) has also been prepared. The compounds synthesised were designed with either two ionic or non-ionic tails to ensure a large number of supramolecular interactions with the solvent, thereby promoting strong solubilisation. The surfactants produced stable suspensions in which the SWNTs are dispersed and the surfactant/SWNT complexes formed are stable for more than one year. UV/Vis/NIR absorption spectroscopy, TEM and AFM were employed to probe the solubilisation properties of the dispersion of surfactants and SWNTs in water.  相似文献

A novel microemulsion based on a mixture of diethyl L-tartrate (DET) and SDS was developed for the microemulsion EKC (MEEKC) determination of structurally related steroids. The system consisted of 0.5% w/w DET, 1.7% w/w SDS, 1.2% w/w 1-butanol, 89.6% w/w phosphate buffer (40 mM, pH 7.0), and 7% w/w ACN. With an applied voltage of +10 kV, a baseline separation of aldosterone (A), cortisone acetate (CA), dexamethasone (D), hydrocortisone (H), hydrocortisone acetate (HA), prednisolone (P), prednisolone acetate (PA), prednisone (Ps), triamcinolone (T), and triamcinolone acetonide (TA) could be achieved. Under the optimized conditions, the reproducibility of the retention time (n = 4) for most of the compounds was less than +/-0.8% with the exception of A, Ps, and T. The average number of theoretical plates was 18 800 plates/m. The results were compared with those achieved by the modified micellar EKC (MEKC). MEEKC showed obvious advantages over MEKC for the separation of highly hydrophobic substances. To further evaluate the system, we tested the MEEKC method by analyzing corticosteroids in a spiked urine sample.  相似文献

The separation of neutral hydrophobic corticosteroids (cortisone, cortisone acetate, hydrocortisone, hydrocortisone acetate, prednisolone and prednisolone acetate) by microemulsion electrokinetic chromatography (MEEKC) was studied. In the preparation of microemulsion, heptane was the solvent, n-butanol the co-surfactant and, as anionic surfactants, sodium dodecyl sulfate (SDS) or taurodeoxycholic acid sodium salt (STDC) were employed. Using an acidic running buffer, (phosphate pH 2.5) a strong suppression of the electroosmotic flow (EOF) was observed; this resulted in a fast anodic migration of the analytes partitioned into the negatively charged microemulsion droplets. Under these conditions, STDC showed better separation of corticosteroids than the conventional SDS; however, the use of a single anionic surfactant did not provide the required selectivity. The addition of the neutral surfactant polyoxyethylene glycol octadecyl ether (Brij 76) significantly altered the migration of each analytes allowing a better tuning of separation; however, in order to obtain adequate resolution between couples of adjacent critical peaks, the addition of neutral cyclodextrins (CDs) was found to be essential. This apparently complex system (CD-MEEKC), was optimized by studying the effect of the most important parameters affecting separation: STDC concentration, Brij 76 concentration, nature and concentration of cyclodextrins. Following a rational step-by-step approach, the optimised conditions providing the complete separation of the analytes were found to be: 4.0% STDC, 2.5% Brij 76, 6.6% n-butanol, 1.36% heptane and 85.54% of a solution 5 mM beta-CD in 50 mM phosphate buffer (pH 2.5). The optimized system was preliminary applied to the detection of corticosteroids related substances at impurity level and it could be considered a useful orthogonal alternative to HPLC methods.  相似文献

A novel microemulsion based on sodium bis(2-ethylhexyl) sulfosuccinate (AOT) was developed for the simultaneous determination of natural and synthetic estrogens by microemulsion EKC (MEEKC). The microemulsion system consisted of 1.4% w/w AOT, 1.0% w/w octane, 7.0% w/w 1-butanol and 90.6% w/w 20 mM sodium salt of 3-(cyclohexylamino)-2-hydroxy-1-propanesulfonic acid (CAPSO) and 10 mM phosphate buffer at pH 12.5. A baseline resolution in the separation of estrone, 17beta-estradiol, estriol, estradiol 17-hemisuccinate, etinilestradiol, estradiol 3-benzoate, and estradiol 17-valerate was achieved in comparison to the traditional MEEKC system with SDS in less than 15 min. The optimized electrophoretic conditions included the use of an uncoated-silica capillary of 60 cm of total length and 75 microm id, an applied voltage of 25 kV, a temperature of 25 degrees C and 214 UV-detection. Parameters of validation such as specificity, linearity, accuracy, LOD, LOQ and robustness were evaluated according to international guidelines. Due to its simplicity, accuracy, and reliability, the proposed method can be an advantageous alternative to the traditional methodologies for the analysis of natural and synthetic estrogens in different pharmaceutical forms.  相似文献

Microemulsion electrokinetic chromatography (MEEKC) has been applied to the separation of some UV filters (Eusolex 4360, Eusolex 6300, Eusolex OCR, Eusolex 2292, Eusolex 6007, Eusolex 9020, Eusolex HMS, Eusolex OS, and Eusolex 232) commonly found in suntan lotions. The composition of the microemulsion employed was optimized with respect to the best possible separation of the selected analytes using artificial neural networks (ANNs). Two parameters namely the composition of the mixed surfactant system comprising the anionic sodium dodecyl sulfate (SDS) and neutral Brij 35 and the amount of organic modifier (2-propanol) present in the aqueous phase of the microemulsion were modeled. Using an optimized MEEKC buffer consisting of 2.25 g SDS, 0.75 g Brij 35, 6.6 g 1-butanol, 0.8 g n-octane, 17.5 g 2-propanol, and 72.1 g of 10 mM borate buffer (pH 9.2), eight target analytes could be separated in under 25 min employing a diode-array detector to segregate the overlapping signals obtained for Eusolex 9020 and Eusolex HMS. Detection limits from 0.8 to 6.0 nug/mL were obtained and the calibration plots were linear over at least one order of magnitude. The optimized method could be applied to the determination of Eusolex 6300 and Eusolex 9020 in a commercial suntan lotion.  相似文献

A comparison between chiral cyclodextrin‐modified microemulsion electrokinetic chromatography (CD‐MEEKC) and cyclodextrin‐modified micellar electrokinetic chromatography (CD‐MEKC) for the enantiomeric separation of esbiothrin was carried out. For both methods, the separation conditions were optimized by varying CD types and concentration, running buffer pH and compositions, organic modifiers, and temperature. The optimal CD‐MEEKC conditions were 0.8% n‐heptane, 2.3% SDS, 6.6% n‐butanol, 90.3% 10 mM sodium tetraborate containing 3% (w/v, the ratio of CD mass to microemulsion volume) methyl‐β‐cyclodextrin, pH 10, 25°C. The optimized CD‐MEKC conditions were 3.3% SDS, 96.7% 10 mM sodium tetraborate containing 5% (w/v) β‐CD, pH 10, 25°C. The difference in physicochemical properties of the buffer and CDs resulted in different optimal CD type. The competitive distribution between the microemulsion (or micelle) and chiral CD contributed to the chiral separation. Both methods provided excellent separation (Rs ~? 3) with similar migration time (ca. 15 min). CD‐MEEKC provided higher separation efficiencies (>300000) than CD‐MEKC (>200000). The LODs for CD‐MEEKC and CD‐MEKC were 4.7 μg/mL and 3.2 μg/mL, respectively. The RSDs of migration time and peak area for CD‐MEEKC were slightly higher than for CD‐MEKC. Both the demonstrated CD‐MEEKC and CD‐MEKC methods provided high efficiencies, low LODs, and reproducible enantioseparations of esbiothrin.  相似文献

The enantiomeric resolution of 15 different pharmaceutical compounds was explored using chiral microemulsion electrokinetic chromatography (MEEKC). The microemulsion employed was comprised of the chiral surfactant dodecoxycarbonylvaline (DDCV), 1-butanol, and ethyl acetate, at an initial composition of 1% w/v:1.2% v/v:0.5% v/v, respectively. The effect of varying the background buffer composition, voltage, and ultimately the surfactant concentration and/or aggregate phase ratio were examined. Changing from a zwitterionic buffer ((2-[2-amino-2-oxoethyl)amino]ethanesulfonic acid, ACES) to the same concentration of phosphate buffer improved the efficiency and decreased overall analysis time, but also resulted in a decrease in chiral resolution. Furthermore, using phosphate buffer while simultaneously increasing the percent DDCV from 1 to 4% increased the efficiencies from a range of 34,000 to 59,000 N/m to a range of 160,000 to 400,000 N/m. While the enantioselectivities did not change significantly, the improvement in efficiencies, elution range, and retention factors provided an increase in both resolution and the number of enantiomers that were separated. Using an optimized microemulsion comprised of phosphate buffer and 4% DDCV, chiral separation was achieved for all 11 pairs of enantiomers, with a resolution ranging from 0.90 to 4.71. Moreover, the average resolution doubled in going from nonoptimized to optimized conditions for five of the eleven compounds. Finally, a comparison was made of the effect of increasing only the surfactant concentration by a factor of 4 versus increasing the overall composition (or phase ratio) by a factor of 4. Ultimately, the microemulsion containing 4% DDCV provided a larger elution range, greater resolution, and more optimal retention than that provided by the 4x phase increase.  相似文献

As-synthesized single-walled carbon nanotubes (SWNTs) are bundled mixtures of different species. The current challenge in the field of carbon nanotube research lies in the processing and separation of SWNTs, which first require efficient dispersion of individual SWNTs in solvents. We report DNA-mimicking polysoap surfactants that disperse SWNTs in aqueous solutions more effectively than DNA. The polysoaps are synthesized by functionalizing the side chain of poly(styrene-alt-maleic acid) with aminopyrene. The synthetic nature of the polysoap opens a new approach to further optimization of not only SWNT dispersion efficiency but also multi-functional SWNT dispersing surfactant.  相似文献

In this study, the separation of 13 homologous stick-like hydrophobic solutes, i.e., biphenyl nitrile derivatives, by organic-solvent-based micellar electrokinetic chromatography (MEKC) was investigated in terms of separation medium composition, species and concentration of surfactant, other additives, separation voltage and temperature. The results showed that the 13 strong hydrophobic compounds were baseline separated in 25 min with a repeatability of less than 1.3% (RSD) for migration time. The separation medium was a mixture of methanol, 2-propanol and water (58.5:10:31.5), containing 150 mM cetyltrimethylammonium bromide (CTAB) and 20 mM sodium borate. Variety of solvent composition, temperature and applied voltage all showed remarkable effect on the separation. The organic-solvent-based MEKC method proved to be superior to the aqueous MEKC and microemulsion electrokinetic chromatography (MEEKC) methods for the separation of strongly hydrophobic compounds.  相似文献
