Summary A simple and sensitive isocratic LC method is described for the determination of erythromycins in fermentation broths. A simple technique utilizing acetone-methyl ethyl ketone, 1∶1, as extraction solvent was coupled with suitable chromatographic conditions—compounds were separated on a 250 mm×4.6 mm i.d., 5 μm, reversed-phase column at 65°C with acetonitrile-0.2m K2HPO4 pH7.0-water, 35:5:60 (v/v), as mobile phase at a flow rate of 1.0 mL min−1. UV detection was performed at 215 nm. Separation of erythromycin F from polar components of the fermentation liquid was sufficient. Erythromycins A, B, C, D, and E, andN-desmethylerythromycin A were also separated, as were known decomposition products of erythromycin A and several unknown components. The method is suitable for monitoring the progress of erythromycin fermentation.  相似文献

建立了采用高效液相色谱(HPLC)同时测定发酵液中底物赤藓糖醇和产物L-赤藓酮糖含量的方法。采用Lichrospher 5-NH2色谱柱(250 mm×4.6 mm),柱温30 ℃,以乙腈-水(体积比为9:1)为流动相,流速1.0 mL/min。用示差折光检测器检测赤藓糖醇,检测器温度为35 ℃。用紫外检测器在室温下检测L-赤藓酮糖,检测波长为277 nm。所得赤藓糖醇的线性范围为1.00~100.00 g/L,相关系数为0.9985,检出限为0.10 g/L,定量限为0.45 g/L;所得L-赤藓酮糖的线性范围为1.00~100.00 g/L,相关系数为0.9958,检出限为0.50 g/L,定量限为0.87 g/L;赤藓糖醇的日内和日间相对标准偏差(RSD)分别小于3.28%和5.30%, L-赤藓酮糖的日内和日间RSD分别小于2.16%和2.25%;回收率均大于99%。取不同时间的发酵液样品分别用上述方法测定,结果表明所建立的HPLC法不受发酵液中其他组分的影响,可同时测定底物赤藓糖醇和产物L-赤藓酮糖的含量。  相似文献

Domperidone is a dopamine D2 receptor antogonist, which has been used as antiemetic agent in human beings. It has been found in wastewater released by some pharmaceutical industries leading to the contamination of surface and ground water. Therefore, a sensitive, inexpensive and reproducible HPLC-SPE method was developed for the analysis of domperidone in the wastewater. The column used was Waters symmetry C18 (15 cm × 0.46 mm, 5 μm). The mobile phase used was phosphate buffer (50 mM, pH 3.5) acetonitrile (80:20, v/v) at the flow rate 2.0 mL/min. The detection was achieved by using UV mode at 230 nm. The retention, separation and resolution factors were 2.63, 3.00 and 3.20, respectively. The percentage recovery of domperidone from wastewater was 95.0%. Celiprolol was used as the internal standard to access the percentage extraction of domperidone from wastewater.  相似文献

研究了固相萃取富集和预分离,高效液相色谱测定枸杞中的类胡萝卜素的方法;枸杞中的类胡萝卜素用WatersXterraTMRP18固相萃取小柱预分离,以WatersXterraTMRP18(3.9×150mm,5μm)液相色谱柱为固定相,甲醇 四氢呋喃(4 1)为流动相分离,用二极管矩阵检测器检测,检测波长为450nm。方法标准回收率为95%~103%。用该方法测定了几种枸杞样品中的类胡萝卜素。  相似文献

以可代替使用的水溶性合成色素-亮黑BN、固绿FCF、酸性红、酸性黄36为研究对象,建立了高效液相色谱同时测定食品中4种合成色素的方法。样品通过WAX固相萃取柱富集净化,10%氨化甲醇洗脱,采用C18色谱柱分离,以甲醇和10 mmol/L乙酸铵水溶液作为流动相梯度洗脱,外标法定量。通过固相萃取柱净化后,杂质对待测物的干扰明显降低,优化色谱条件下,4种色素在0.10~10.0 mg/L质量浓度范围内,线性良好,相关系数(r)≥0.9995。方法回收率为92.6%~101.8%,相对标准偏差(RSD)≤5.4%,方法检出限为0.01~0.04 mg/kg。该方法灵敏度高,准确可靠,适合食品中亮黑BN、固绿FCF、酸性红、酸性黄36的同时测定。  相似文献

Summary High performance liquid chromatography on an octadecyl silica column has been used to determine both the monensin A: monensin B ratio and by the method of standard addition, the concentration of both in the fermentation broth ofStreptomyces cinnamonensis. Refractive index detection was preferred to ultraviolet owing to the presence of UV-absorbing components which could not be completely separated from the substances of interest. A linear relationship was obtained from the calibration data. The coefficients of variation for the estimation both of the ratio and the concentrations of the compounds were better then 5%. The estimated limit of detection for both substances was about 1 g/ml. The results obtained from the determination of the ratios of monensins were compared with those obtained by chemical ionization mass spectrometry. Chromatographic separation of monensins on the silica gel column is also described.  相似文献

A new method was developed for the analysis of illicit drugs in human urine by coupling carrier-mediated liquid phase microextraction (LPME) to high performance liquid chromatography (HPLC). By adding an appropriate carrier in organic phase, simultaneous extraction and enrichment of hydrophilic (morphine and ephedrine) and hydrophobic (pethidine) drugs were achieved. Effects of the types of organic solvents and carriers, the carrier concentration in the organic phase, the HCl concentration in the acceptor solution, the stirring rate, and the extraction time on the enrichment factor of analytes were investigated. Under the optimal experimental conditions, high enrichment factors (202-515) were obtained. The linear detection ranges were 0.1-10 mg L−1 for the studied drugs. The limits of detection (LOD) at signal-to-noise ratio of 3 were 0.05 mg L−1 for both morphine and ephedrine, and 0.02 mg L−1 for pethidine. This method was successfully applied to analysis of ephedrine in real urine specimens, revealing that the determination of illicit drugs in urine was feasible.  相似文献

报道了固相萃取-高效液相色谱法测定浓缩苹果汁中多菌灵残留量的方法.样品经适量水稀释后,C18固相萃取柱提取净化,用V(甲醇)∶V(二氯甲烷)=1∶1淋洗,HPLC法测定.在添加水平为0.10,0.50,2.0 mg/kg时,多菌灵的回收率在92.6%~108.3%之间;RSD<3% (n=6),检出限为0.02 mg/kg,该方法的测定结果满足农药残留量的检测要求.  相似文献

A novel core-shell composite (SiO(2)-nLPD), consisting of micrometer-sized silica spheres as a core and nanometer titania particles as a surface coating, was prepared by liquid phase deposition (LPD). Here, we show the resulting core-shell composite to have better efficient and selective enrichment for mono- and multi-phosphopeptides than commercially available TiO(2) spheres without any enhancer. The material exhibited favorable characteristics for HPLC, which include narrow pore size distribution, high surface area and pore volume. We also show that the core-shell composite can efficiently separate adenosine phosphate compounds due to the Lewis acid-base interaction between titania and phosphate group when used as HPLC packings. After coating the silica sphere with titania by LPD, the silanol of silica spheres will be shielded and that the stationary phase, C(18) bonded SiO(2)-3LPD, could be used under extreme pH condition.  相似文献

采用高效液相色谱法(HPLC)同时检测丁二酸发酵液中的有机酸和水溶性维生素。以RP-HPLC为分离模式,选用Hedera ODS-2色谱柱(5μm,4.6×250 mm),20 mmol/L的磷酸盐缓冲溶液(pH2.9)和乙腈为流动相,梯度洗脱,检测波长为210 nm,建立了同时测定分析8种水溶性维生素和3种有机酸的方法,15 min内所有组分都洗脱完全。8种水溶性维生素和3种有机酸在线性范围内峰面积与浓度呈良好的线性关系,相关系数为0.9989~0.9997;加标回收率在92.5%~100.3%之间;RSD(n=5):0.9%~1.9%。本方法采用同一种流动相既可以测定发酵液中的有机酸又可以测定水溶性维生素的含量。  相似文献

建立了高效液相色谱(HPLC)同时测定生物质乳酸发酵液中有机酸及糖类的分析方法。使用Bio-Rad Aminex HPX-87H色谱柱,以5 mmol/L的H2SO4为流动相,在柱温55 ℃,流速0.6 mL/min条件下,采用示差折光检测器进行检测。结果表明,该方法可在17 min内实现发酵液中各种有机酸和糖类化合物等的完全分离与定量,6种有机酸和3种糖类化合物在0.15~5.19 g/L范围内的线性关系良好,回归方程的线性相关系数在0.9998以上。将该法用于米根霉发酵液的检测,两个水平的加标回收率为96.91%~103.11%,相对标准偏差(n=6)为0.81%~4.61%。该法适用于微生物发酵液中多种有机酸和糖类的快速、高效分离和定量测定。  相似文献

This study aimed to establish a rapid analytical method to determine antioxidants in essence. A simple,efficient and practical, vortex-assisted, cloud-point extraction(VACPE) procedure is proposed for extracting and pre-concentrating four different of synthetic phenolic antioxidants(SPAs), propyl gallate(PG), tert-butylhydroquinone(TBHQ), butylated hydroxyanisole(BHA), butylated hydroxytoluene(BHT) in essence prior to high performance liquid chromatography(HPLC) analysis. The non-ionic surfactant, fatty alcohol polyoxyethylene ether-9(AEO-9), was used as extractant and vortex-mixing was utilized to reduce extraction time and improve extraction efficiency. The effective parameters of the extraction process, such as volume of extraction solvent, pH, vortex-mixing time, equilibration temperature and time, were optimized. Under the optimum conditions, the linear range of PG, TBHQ,BHA and BHT was 8.0–800 ng/mL. All correlation coefficients of the calibration curves were higher than0.996 and relative standard deviations(RSD, n = 5) were 2.36%–5.46%. The proposed method was successfully applied to the extraction and determination of antioxidants in essence samples with satisfactory relative recoveries of 89.4%–103.5%. The results confirmed the SPAs of essence could be effectively monitored by this method and also established good reference criteria for essence.  相似文献

An emulsification liquid phase microextraction followed by on-line phase separation coupled to high performance liquid chromatography (HPLC) is introduced based on a novel idea for the separation of dispersed organic phase from aqueous phase. In this method, the dispersed organic extraction phase was filtered using an in-line filter and it was separated from the water sample. The new approach is simple and, in addition to improving some limitations of the conventional emulsification liquid phase microextraction, eliminates the need for centrifugation in the phase separation step.  相似文献

研究了用快速分离柱高效液相色谱法测定烟草样品中的苯酚、苯二酚和甲基苯酚. 烟草样品中的酚经水蒸汽蒸馏分离后用Waters Sep-Pak-C18固相萃取小柱富集, 以ZORBAX Stable Bound (4.6 mm i.d.×20 mm, 1.8 μm) 快速分离柱为固定相, 0.05 mol/L KH2PO4缓冲溶液-甲醇梯度为流动相, 几种主要酚在2.0 min 内可达到基线分离;该方法的相对标准偏差为2.1%~3.6%, 标准加入的回收率为88%~97%, 已用于几种烟草样品测定.  相似文献

采用加速溶剂萃取-凝胶色谱净化建立了动物源性食品中胆固醇检测的高效液相色谱方法。结果表明,胆固醇浓度在1.0~40.0mg/L范围内线性关系良好,加标回收率为89.1%~99.8%,相对标准偏差为3.1%~7.8%。该方法准确、灵敏度高,适用于富含脂类的动物源性食品中胆固醇的检测。  相似文献
