The purpose of this study was to clarify the effect of moisture on the impact toughness of sugar-coated tablets manufactured by the dusting method. We demonstrated that moisture plays an important role in the impact toughness of sugar-coated tablets. Moisturizing the sugar-coating layer resulted in enhancement of impact toughness of sugar-coated tablets, while reducing moisture in the sugar-coating layer resulted in weakening of the impact toughness. This was due to the characteristics of sucrose, the main ingredient of the sugar-coating layer, which is a soft and non-fragile material at high moisture levels, but hard and fragile at low moisture levels. We also demonstrated that friability as an indicator of impact toughness changed with time, and friability should be measured at 14 d after manufacture. This is due to moisture movement from outer sugar-coating layer into the inner sugar-coated tablets. Incorporating microcrystalline cellulose (MCC) in the subcoating layer resulted in sugar-coating layers with high resistance against impact even though moisture content of sugar-coated tablets was low. We confirmed the high impact toughness of the sugar-coated tablets with MCC whose moisture content was low from the results of both free fall and friability tests. We suggest that the dusting method using dusting powder containing MCC is a useful method for the production of sugar-coated tablets containing moisture sensitive drugs.  相似文献

Chemical imaging is a rapidly emerging analytical method in pharmaceutical technology. Due to the numerous chemometric solutions available, characterization of pharmaceutical samples with unknown components present has also become possible. This study compares the performance of current state-of-the-art curve resolution methods (multivariate curve resolution-alternating least squares, positive matrix factorization, simplex identification via split augmented Lagrangian and self-modelling mixture analysis) in the estimation of pure component spectra from Raman maps of differently manufactured pharmaceutical tablets. The batches of different technologies differ in the homogeneity level of the active ingredient, thus, the curve resolution methods are tested under different conditions. An empirical approach is shown to determine the number of components present in a sample. The chemometric algorithms are compared regarding the number of detected components, the quality of the resolved spectra and the accuracy of scores (spectral concentrations) compared to those calculated with classical least squares, using the true pure component (reference) spectra. It is demonstrated that using appropriate multivariate methods, Raman chemical imaging can be a useful tool in the non-invasive characterization of unknown (e.g. illegal or counterfeit) pharmaceutical products.  相似文献

The relation between the impact toughness and flow-induced crystalline orientation of high-density polyethylene (HDPE) was investigated. Flow-induced crystalline orientation was created in the samples via injection moulding and the amount of orientation was controlled through variation of processing conditions (injection temperature) and sample thickness. The impact toughness behaviour was found to be strongly dependent on the amount of crystalline orientation, whereas the loading direction also had a strong influence, e.g. giving highest impact properties in flow direction. Subsequently, injection moulded samples of HDPE modified with calcium carbonate filler particles were tested. In this case a similar relation between crystalline orientation and loading direction was found, whereas the total amount of flow-induced crystallization was observed to be strongly influenced by the presence of the filler particles.  相似文献

Polylactide (PLA) was successfully toughened by blending with bio-based poly(ester)urethane (TPU) elastomers which contained bio-based polyester soft segments synthesized from biomass diols and diacids. The miscibility, mechanical properties, phase morphology and toughening mechanism of the blend were investigated. Both DSC and DMTA results manifested that the addition of TPU elastomer not only accelerated the crystallization rate, but also increased the final degree of crystallinity, which proved that TPU has limited miscibility with PLA and has functioned as a plasticizer. All the blend samples showed distinct phase separation phenomenon with sea-island structure under SEM observation and the rubber particle size in the PLA matrix increased with the increased contents of TPU. The mechanical property variation of PLA/TPU blends could be quantitatively explained by Wu's model. With the variation of TPU, a brittle-ductile transition has been observed for the TPU/PLA blends. When these blends were under tensile stress conditions, the TPU particles could be debonded from the PLA matrix and the blends showed a high ability to induce large area plastic deformation before break, which was important for the dissipation of the breaking energy. Such mechanism was demonstrated by tensile tests and scanning electron microcopy (SEM) observations.  相似文献

In the present study three-point-bend impact experiments were conducted using an instrumented Charpy pendulum with a laser displacement measurement to better understand the correlation between impact velocity and the dynamic effects observed on the load-time curves. The experiments were performed at impact velocities ranging from 1 to 4 m/s.The aim of this work is to measure the dynamic fracture toughness at high impact velocities where the classical method is limited by the inertial effects. The direct measurements of the specimen deflection are successfully used for the toughness evaluation. The results obtained with this method, which are compared to other studies, indicate that this approach seems promising for brittle materials such as PMMA.  相似文献

The aim of this article was to determine the optimal ingredients for the rapidly disintegrating oral tablets prepared by the crystalline transition method (CT method). The effect of ingredients (diluent, active drug substance and amorphous sugar) on the characteristics of the tablets was investigated. The ingredients were compressed and the resultant tablets were stored under various conditions. The oral disintegration time of the tablet significantly depended on diluents, due to differences in the penetration of a small amount of water in the mouth and the viscous area formed inside the tablet. The oral disintegration time was 10-30 s for tablets with a tensile strength of approximately 1 MPa, when erythritol, mannitol or xylitol was used as the diluent. The increase in the tensile strength of tablets containing highly water-soluble active drug substances during storage was as large as that of tablets without active drug substances, while the increase in the tensile strength of tablets containing low water-soluble active drug substances was small. It was therefore found that highly water-soluble active drug substances were more suitable for the formulation prepared by the CT method than low water-soluble active drug substances. Irrespective of the type of amorphous sugar (amorphous sucrose, lactose or maltose) used, the porosity of tablets with 1 MPa of tensile strength was 30-40%, and their oral disintegration time was 10-20 s. The optimal ingredients for rapidly disintegrating oral tablets with reasonable tensile strength and disintegration time were therefore determined from these results.  相似文献

There has been significant interest in the binding of anthrax spores by molecular species, but with only limited success. Proteins and more recently peptides were used. However, despite the known presence of carbohydrates on the spore surface, carbohydrate-carbohydrate interactions have hardly been explored likely because of the lack of required specific platform for synthetic carbohydrates. We report the successful use of single-walled carbon nanotubes as a truly unique scaffold for displaying multivalent monosaccharide ligands that bind effectively to anthrax spores with divalent cation mediation to cause significant spore aggregation. A reversed-phase high-performance liquid chromatographic (HPLC) method is described for the analysis of cyproterone acetate in tablets. This steroid is extracted and quantitated using a C18 column. This procedure is shown to be rapid, simple, and valid in terms of recovery, linearity, and precision. Application of this method to analyze dilute samples obtained from dissolution testing is demonstrated.  相似文献

In order to modify the brittleness and flame retardant properties of poly(lactic acid) (PLA), a series of flame retardant toughened PLA composites were prepared using poly(ethylene glycol) 6000 (PEG6000) as a toughening and charring agent together with ammonium polyphosphate (APP) as an acid source and blowing agent. The fire and thermal behavior of PLA/PEG/APP composites was evaluated by limiting oxygen index (LOI), UL‐94, cone calorimeter, and thermogravimetric analysis (TGA). The results showed that the PLA/PEG/APP system had good charring ability and could improve the flame retardancy of PLA. When the content of APP in the composites was more than 5 wt%, all samples could reach UL‐94 V‐0 rating. The results of mechanical property tests demonstrated that the brittleness of PLA was also improved after blended with PEG6000. All the PLA/PEG/APP composites with an APP content of less than 10 wt% showed an obvious neck and fracture behavior, that is, the tensile behavior of PLA was changed from brittle to ductile. Summary Feed premixes usually contain highly effective ingredients. For different reasons it is suitable to determine a standard value for the quantity of the active ingredient(s) which is set free as dust.A test procedure was developed which enables such determinations in the laboratory. In this method dust is generated from the sample to be inspected under defined conditions in a modified, commercially available dust testing equipment. The generated dust is transported onto a filter membrane via an air stream. The content of active ingredient in the dust separated on the membrane is determined quantitatively by a suitable method. This dust testing procedure is described by the example of carbadox containing premixes. The results of the tests with various carbadox premixes and premixes with other ingredients are reported.

Zusammenfassung Futtermittelvormischungen enthalten in der Regel hochwirksame Substanzen. Aus verschiedenerlei Gründen ist es zweckmäßig, einen Richtwert für die in Form von Staub freiwerdende Menge des oder der Wirkstoffe quantitativ bestimmen zu können.Es wurde eine Bestimmungsmethode entwickelt, die derartige Untersuchungen im Labor ermöglicht. Dabei wird aus der zu untersuchenden Probe unter definierten Bedingungen in einer modifizierten, handelsüblichen Testapparatur Staub erzeugt und über einen Luftstrom auf eine Filtermembrane transportiert. Mittels geeigneter Analysenverfahren wird der Wirkstoffgehalt auf der Membrane quantitativ bestimmt. Am Beispiel Carbadox enthaltender Vormischungen wird die Durchführung der Staubmeßmethode beschrieben. Die Ergebnisse der Untersuchungen an verschiedenen Carbadoxvormischungen und Vormischungen mit anderen Wirkstoffen werden berichtet.

Composites based on MF-4SC perfluorinated cation-exchange membranes were manufactured by casting from a polymer solution with a precursor for subsequent use in the synthesis of hydrous silica. Their proton conductivity was studied as a function of composition, temperature, and relative humidity. Modification of MF-4SC with small dopant amounts increases the water content and ion conductivity, in a reduced humidity, too. Materials containing 3% SiO2 by weight have a maximal conductivity.  相似文献

Controlled Nucleation Thermochemical Deposition (CNTD) has emerged from classical chemical deposition (CVD) technology. This paper describes the techniques of thermochemical grain refinement. The effects of such refinement on mechanical properties of materials at room temperature and at elevated temperatures are outlined. Emphasis is given to high temperature structural ceramic materials such as SiC, Si3N4, AiN, and TiB2 and ZrB2. An example of grain refinement accompanied by improvements in mechanical properties is SiC. Grain sizes of 500 to 1000 Å have been observed in CNTD SiC with room temperature MOR of 1380 to 2070 MPa (4 pt bending) and MOR of 3450 to 4140 MPa (4 pt bending) at 1350°C. Various applications of these materials to the solution of high temperature structural problems are described.  相似文献

A simple, rapid, accurate, precise and sensitive colorimetric method for the determination of finasteride in tablets is described. The proposed methods are based on the formation of ion-pair complexes between the examined drug with bromophenol blue (BPB), bromocresol green (BCG) and bromothymol blue (BTB), which can be measured at the optimum lambda(max). Beer's law is obeyed in the concentration ranges 3.0-15.0, 3.0-15.0 and 5.0-20 microg/mL with BPB, BCG and BTB, respectively. The detection limits of FIN was found to be 1.16 microg/mL for BPB, 1.17 for BCG, 1.76 microg/mL for BTB. All the methods gave similar results and were validated for selectivity, linearity, precision and sensitivity. The proposed methods were directly and easily applied to the pharmaceutical preparation with accuracy, resulting from recovery experiments between 100.11 and 100.33% for BPB, 100.17 and 100.67% for BCG and 100.33 and 100.60% for BTB methods. The low relative standard deviation values indicate good precision and high recovery values indicate accuracy of the proposed methods. The proposed methods have been applied to the determination of drug in commercial tablets. Results obtained from the analysis of commercial preparations with the proposed methods are in good agreement with those obtained with the official HPLC method.  相似文献

A simple, rapid, and sensitive visible spectrophotometric method was developed, for the first time, for analysis of desloratadine (DE) in tablets. The method is based on the deep-blue colored TCNQ*- radical anion formed by interaction of the drug (n-donor) with 7,7,8,8-tetracyanoquinodimethane (TCNQ, pi-acceptor) in acetonitrile at ambient temperature. Optimum conditions for the reaction were investigated, absorbances were read at 843 nm, and the linearity range for concentrations of DE was found to be 1.5-13 microg/mL. The reaction product remains stable up to 8 h when kept at room temperature in the dark. The developed method was validated and successfully applied to the determination of DE in tablets. The tablets were also analyzed with a column liquid chromatography method reported in literature. The results from both methods were statistically compared by t- and F-tests. No significant difference was found for the means and standard deviations at 95% confidence level. Accuracy was examined through recovery studies. Being very simple and reliable, the method can be recommended for routine quality control analysis of DE in tablets.  相似文献

The aim of this study was to prepare, using taste-masked granules, tablets which can rapidly disintegrate in saliva (rapidly disintegrating tablet), of drugs with bitter taste (pirenzepine HCl or oxybutynin HCl). The taste-masked granules were prepared using aminoalkyl methacrylate copolymers (Eudragit E-100) by the extrusion method. None of the drugs dissolved from the granules (% of dissolved, < 5%) even at 480 min at pH 6.8 in the dissolution test. However, the drugs dissolved rapidly in the medium at pH 1.2 in the dissolution test. Rapidly disintegrating tablets were prepared using the prepared taste-masked granules, and a mixture of excipients consisting of crystalline cellulose (Avicel PH-102) and low-substituted hydroxypropylcellulose (L-HPC, LH-11). The granules and excipients were mixed well (mixing ratio by weight, crystalline cellulose: L-HPC = 8:2) with 1% magnesium stearate, and subsequently compressed at 500-1500 kgf in a single-punch tableting machine. The prepared tablets (compressed at 500 kgf) containing the taste-masked granules have sufficient strength (the crushing strength: oxybutynin tablet, 3.5 kg; pirenzepine tablet, 2.2 kg), and a rapid disintegration time (within 20 s) was observed in the saliva of healthy volunteers. None of the volunteers felt any bitter taste after the disintegration of the tablet which contained the taste-masked granules. We confirmed that the rapidly disintegrating tablets can be prepared using these taste-masked granules and excipients which are commonly used in tablet preparation.  相似文献
