Nitrate determination in waters is generally carried out with cadmium filings and carcinogenic reagents or by reaction with phenolic compounds in highly concentrated sulfuric acid medium. In this work, it was developed a green analytical procedure for nitrate determination in natural waters based on direct spectrophotometric measurements in ultraviolet, using a flow-injection system with an anion-exchange column for separation of nitrate from interfering species. The proposed method employs only one reagent (HClO4) in a minimum amount (equivalent to 18 μL concentrated acid per determination), and allowed nitrate determination within 0.50-25.0 mg L−1, without interference of up to 200.0 mg L−1 humic acid; 1.0 mg L−1 NO2; 200.0 mg L−1 PO43−; 75.0 mg L−1 Cl; 50.0 mg L−1 SO42− and 15.0 mg L−1 Fe3+. The detection limit (99.7% confidence level) and the coefficient of variation (n = 20) were estimated as 0.1 mg L−1 and 0.7%, respectively. The results obtained for natural water samples were in agreement with those achieved by the reference method based on nitrate reduction with copperized cadmium at the 95% confidence level.  相似文献

A sequential injection method (SIA) for carbon speciation in inland bathing waters was developed comprising, in a single manifold, the determination of dissolved inorganic carbon (DIC), free dissolved carbon dioxide (CO2), total carbon (TC), dissolved organic carbon and alkalinity. The determination of DIC, CO2 and TC was based on colour change of bromothymol blue (660 nm) after CO2 diffusion through a hydrophobic membrane placed in a gas diffusion unit (GDU). For the DIC determination, an in-line acidification prior to the GDU was performed and, for the TC determination, an in-line UV photo-oxidation of the sample prior to GDU ensured the conversion of all carbon forms into CO2. Dissolved organic carbon (DOC) was determined by subtracting the obtained DIC value from the TC obtained value. The determination of alkalinity was based on the spectrophotometric measurement of bromocresol green colour change (611 nm) after reaction with acetic acid. The developed SIA method enabled the determination of DIC (0.24–3.5 mg C L−1), CO2 (1.0–10 mg C L−1), TC (0.50–4.0 mg C L−1) and alkalinity (1.2–4.7 mg C L−1 and 4.7–19 mg C L−1) with limits of detection of: 9.5 μg C L−1, 20 μg C L−1, 0.21 mg C L−1, 0.32 mg C L−1, respectively. The SIA system was effectively applied to inland bathing waters and the results showed good agreement with reference procedures.  相似文献

This work has evaluated the influence of humic acid and/or surfactants in the quantification of ammonium in waters with the indophenol-type reaction method. Thymol has been employed with the colorimetric method for sample ammonium concentrations between 0.25 and 1 mg L−1. In addition, SPE/diffuse reflectance method has been used for quantification of ammonium at low concentrations (between 0.025 and 0.25 mg L−1). Matrix effect owing to humic substances were observed with the colorimetric method when the concentration was equal or higher than 25 mg L−1. Lower concentrations of humic compounds produced matrix effects with the more sensitive SPE/diffuse reflectance method. Generalized H-point standard addition method (GHPSAM) was employed for evaluating the effect of humic acids and for eliminating the bias error produced by humic acids. Real water samples of different types were analyzed and accurate results for ammonium concentration were obtained with both procedures in presence or absence of humic acids. Cationic and anionic surfactants inhibited the derivatization reaction at percentages up to 0.001% and 0.5%, respectively, and non-ionic surfactants at percentages of 5.5%  相似文献

A rapid, simple miniaturised photometrical method was developed for the determination of nitrate and/or nitrite in freshwater samples. All procedures, including sample buffering, reduction by copperised cadmium granules, colour development and absorbance determination, were completed in a 96-well microplate. The factors governing the nitrate reduction and its recovery were investigated in detail, and the optimised analysing conditions were established. Nitrate was quantitatively reduced by copperised cadmium granules with a high reduction efficiency (96.59 ± 0.96%). The proposed method gave a linear calibration ranging from 0.01 to 1.50 mg L−1 for NO2-N and 0.02 to 1.50 mg L−1 for NO3-N. The detection limits for nitrite and nitrate were 2 and 4 μg L−1, respectively. The proposed method allowed at least 48 samples to be simultaneously analysed in duplicate, with good precision, within 90 min for nitrate and 30 min for nitrite, and was successfully applied to actual freshwater sample analysis with a recovery of 98.02 ± 1.04% for nitrite and 99.72 ± 1.39% for nitrate. This method produced accurate results comparable to standard methods, provided a much higher sample throughput than conventional methods and could be routinely used in actual freshwater sample monitoring.  相似文献

The presence of trace neutral organonitrogen compounds as carbazole and indole in derivative petroleum fuels plays an important role in the car's engine maintenance. In addition, these substances contribute to the environmental contamination and their control is necessary because most of them are potentially carcinogenic and mutagenic. For those reasons, a reliable and sensitive method was proposed for the determination of neutral nitrogen compounds in fuel samples, such as gasoline and diesel using preconcentration with modified silica gel (Merck 70-230 mesh ASTM) followed by differential pulse voltammetry (DPV) technique on a glassy carbon electrode. The electrochemical behavior of carbazole and indole studied by cyclic voltammetry (CV) suggests that their reduction occurs via a reversible electron transfer followed by an irreversible chemical reaction. Very well resolved diffusion controlled voltammetric peaks were obtained in dimethylformamide (DMF) with tetrabutylammonium tetrafluoroborate (TBAF4 0.1 mol L−1) for indole (−2.27 V) and carbazole (−2.67 V) versus Ag|AgCl|KClsat reference electrode. The proposed DPV method showed a good linear response range from 0.10 to 300 mg L−1 and a limit of detection (L.O.D) of 7.48 and 2.66 μg L−1 for indole and carbazole, respectively. The results showed that simultaneous determination of indole and carbazole presents in spiked gasoline samples were 15.8 ± 0.3 and 64.6 ± 0.9 mg L−1 and in spiked diesel samples were 9.29 ± 1 and 142 ± 1 mg L−1, respectively. The recovery was evaluated and the results shown the values of 88.9 ± 0.4 and 90.2 ± 0.8% for carbazole and indole in fuel determinations. The proposed method was also compared with UV-vis spectrophotometric measures and the results obtained for the two methods were in good agreement according to the F and t Student's tests.  相似文献

This paper describes selenium determination based on Se0 preconcentration in the imprinted polymer (synthesized with 2.25 mmol SeO2, 4-vinylpyridine and 1-vinylimidazole) with subsequent detection on-line in HG-FAAS. During the synthesis, SeO2 is reduced to Se (0). Therefore, there are no MIP neither IIP in the present work, thus we denominated: AIP, i.e., atomically imprinted polymers. For the optimization of analytical parameters Doehlert design was used. The method presented limit of detection and limit of quantification of 53 and 177 ng L−1, respectively, and linear range from 0.17 up to 6 μg L−1 (r = 0.9936). The preconcentration factor (PF), consumptive index (CI) and concentration efficiency (CE) were 232; 0.06 mL and 58 min−1 respectively. The proposed method was successfully applied to determine Se in Brazil nuts (0.33 ± 0.03 mg kg−1), apricot (0.46 ± 0.02 mg kg−1), white bean (0.47 ± 0.03 mg kg−1), rice flour (0.47 ± 0.02 mg kg−1) and milk powder (0.22 ± 0.01 mg kg−1) samples. It was possible to do 12 analyzes per hour. Accuracy was checked and confirmed by analyzing certified reference material (DORM-2, dogfish muscle), and samples precision was satisfactory with RSD lower than 10%.  相似文献

In the present study, an analytical procedure was developed for the determination of short-chain fatty acids (SCFAs) in landfill leachate and municipal wastewater employing injection of aqueous samples to gas chromatograph with flame ionization detector (GC-FID). Chromatographic conditions such as a separation system, injection volume, oven temperature program were investigated and selected. With two columns, one with a polar (polyethylene glycol) and one with a non-polar (dimethylpolisiloxane) stationary phase, good separation of SCFAs, containing from 2 to 8 carbon atoms, was achieved. The sample volume was 2 μL and the temperature program 80 °C (30 s) then 7 °C min−1 to 220 °C (2 min). LOQs values were below 0.25 mg L−1. The concentrations of the acids in the landfill leachate studied ranged from 0.45 ± 0,059 (average ± extended uncertainty) mg L−1 for pentanoic acid to 15.2 ± 0.73 mg L−1 for ethanoic acid. Concentrations of SCFAs in the municipal wastewater were lower than LOQs.  相似文献

An analytical procedure with improved sensitivity was developed for cyanide determination in natural waters, exploiting the reaction with the complex of Cu(I) with 2,2′-biquinoline 4,4′-dicarboxylic acid (BCA). The flow system was based on the multi-pumping approach and long pathlength spectrophotometry with a flow cell based on a Teflon AF 2400® liquid core waveguide was exploited to increase sensitivity. A linear response was achieved from 5 to 200 μg L−1, with coefficient of variation of 1.5% (n = 10). The detection limit and the sampling rate were 2 μg L−1 (99.7% confidence level), and 22 h−1, respectively. Per determination, 48 ng of Cu(II), 5 μg of ascorbic acid and 0.9 μg of BCA were consumed. As high as 100 mg L−1 thiocyanate, nitrite or sulfite did not affect cyanide determination. Sulfide did not interfere at concentrations lower than 40 and 200 μg L−1 before or after sample pretreatment with hydrogen peroxide. The results for natural waters samples agreed with those obtained by a fluorimetric flow-based procedure at the 95% confidence level. The proposed procedure is then a reliable, fast and environmentally friendly alternative for cyanide determination in natural waters.  相似文献

A new and simple flow injection system procedure has been developed for the indirect determination of cyanide. The method is based on insertion of aqueous cyanide solutions into an on-line cadmium carbonate packed column (25% m/m suspended on silica gel beads) and a sodium hydroxide with pH 10 is used as the carrier stream. The eluent containing the analyte as cadmiumcyanide complexes, produced from reaction between cadmium carbonate and cyanide, measured by flame atomic absorption spectrometry. The absorbance is proportional to the concentration of cyanide in the sample. The linear range of the system is up to 15 mg L−1 with a detection limit 0.2 mg L−1 and sampling rate 72 h−1. The method is suitable for determination of cyanide in industrial waste waters with a relative standard deviation better than 1.22%.  相似文献

A sequential injection analysis (SIA) using lab-on-valve with air segmentation and spectrophotometric detection was designed for copper(II) determination. It is based on the reaction of copper(II) and 2-carboxy-2′-hydroxy-5′-sulfoformazyl benzene (Zincon) in a weak alkaline solution between the air zones. Beer's Law was obeyed over the range of 0.1-2.0 mg L−1 copper(II) with a correlation coefficient 0.9985 and a slope of 0.2893 absorbance unit/mg L−1. The relative standard deviation was 2.0% for a series of 10 measurements of 0.5 mg L−1 copper(II) solution. The detection limit (3 S/N) and the limit of quantification (LOQ) were 0.05 and 0.17 mg L−1 respectively. This method has been successfully applied to determination of copper(II) in wastewater with a sample throughput of 120 h−1. The method is superior to the batchwise method in that it provides fully automation, rapidity, less reagents and sample consumption with little waste generation.  相似文献

A simple, specific and sensitive sequential injection analysis (SIA) system based on non-immunoassay fluorescent detection has been developed for the determination of urinary albumin. The specific binding of the dye Albumin Blue 580 (AB 580) to albumin in urine generated high emission fluorescent signals. The excitation and emission wavelengths were set at 590 and 610 nm, respectively. The analytical range was obtained from 1 to 100 mg L−1, with a detection limit of 0.3 mg L−1 (S/N = 3). The SIA system gave high precision with relative standard deviations (R.S.D.s) of 0.9% and 1.4% when evaluated with 15 and 100 mg L−1 albumin (n = 15), respectively. The method exhibited good reproducibility, as assessed by performing four analytical curves on different days, and intra-run CVs (2.3-3.3%) and inter-run CVs (3.8%) were obtained. Rapid operation was achieved with a sample throughput of 37 h−1. This method was successfully applied to the determination of urinary albumin, and the method was highly correlated with the immunoturbidimetric method (r2 = 0.965; n = 72).  相似文献

An imidazole derivative, 2-(2′-pyridyl)imidazole (PIMH), was developed as a colorimetric probe for the qualitative analysis of Fe2+ in aqueous solution. PIMH was then used to post-functionalize poly(vinylbenzyl chloride) (PVBC) nanofibers after electrospinning so as to afford a solid state colorimetric probe. Upon treatment with Fe2+ the probe displayed a distinctive color change both in liquid and solid platforms. The linear dynamic range for the colorimetric determination of Fe2+ was 0.0988–3.5 μg mL−1. The ligand showed a high chromogenic selectivity for Fe2+ over other cations with a detection limit of 0.102 μg mL−1 in solution (lower than the WHO drinking water guideline limit of 2 mg L−1), and 2 μg mL−1 in the solid state. The concentration of Fe2+ in a certified reference material (Iron, Ferrous, 1072) was found to be 2.39 ± 0.01 mg L−1, which was comparable with the certified value of 2.44 ± 0.12 mg L−1. Application of the probe to real samples spiked with Fe2+ achieved recoveries of over 97% confirming accuracy of the method and its potential for on-site monitoring.  相似文献

A centrifugal microanalysis system using spectrophotometric detection has been developed for the determination of nitrite and chromium VI. The system was designed to illustrate the potential of centrifugal microfluidic systems for on-site (field) analysis of waters. The performance was significantly better than manual/visual field test systems and approaches that of bench top systems with detection limits of 0.008 mg L−1 NO2-N and 0.03 mg L−1 Cr6+. The system used pre-loaded solid reagents on the disc, providing ease of use, and could process up to 24 samples simultaneously. Precision was enhanced by the use of a multiwavelength ratiometric blank estimation technique.  相似文献

An ion mobility spectrometer equipped with an ultraviolet lamp was used for the qualitative and quantitative determination of acetone in urine samples. This analyte can be used as a biomarker for some fat metabolism-related diseases in humans and cows. Samples require no pretreatment other than warming at 80 °C for 5 min, after which an N2 stream is used to drive volatile analytes to the ion mobility spectrometer. The precision of the ensuing method, expressed as relative standard deviation (%RSD), is better in all cases than 6.7% for peak height and calculated at three levels of concentration. The analyte concentration range studied was from 5 to 80 mg L−1, its limit of detection in the aqueous matrix 3 mg L−1 and recoveries from spiked urine samples 109 ± 3%. The calculated reduced mobility for acetone in the urine samples, 1.75 ± 0.04 cm2 V−1 s−1, was similar to previously reported values. Also, the results were consistent with those provided by test strips used for reference. The proposed method provides a new vanguard screening system for determining acetone in urine samples.  相似文献

The usefulness of the secondary line at 252.744 nm and the approach of side pixel registration were evaluated for the development of a method for sequential multi-element determination of Cu, Fe, Mn and Zn in soil extracts by high-resolution continuum source flame atomic absorption spectrometry (HR-CS FAAS). The influence of side pixel registration on the sensitivity and linearity was investigated by measuring at wings (248.325, 248.323, 248.321, 248.329, and 248.332 nm) of the main line for Fe at 248.327 nm. For the secondary line at 252.744 nm or side pixel registration at 248.325 nm, main lines for Cu (324.754 nm), Mn (279.482 nm) and Zn (213.875 nm), sample flow-rate of 5.0 mL min−1 and calibration by matrix matching, analytical curves in the 0.2-1.0 mg L−1 Cu, 1.0-20.0 mg L−1 Fe, 0.2-2.0 mg L−1 Mn, 0.1-1.0 mg L−1 Zn ranges were obtained with linear correlations better than 0.998. The proposed method was applied to seven soil samples and two soil reference materials (IAC 277; IAC 280). Results were in agreement at a 95% confidence level (paired t-test) with reference values. Recoveries of analytes added to soil extracts containing 0.15 and 0.30 mg L−1 Cu, 7.0 and 14 mg L−1 Fe, 0.60 and 1.20 mg L−1 Mn, 0.07 and 0.15 mg L−1 Zn, varied within the 94-99, 92-98, 93-101, and 93-103% intervals, respectively. The relative standard deviations (n = 12) were 2.7% (Cu), 1.4% (Fe - 252.744 nm), 5.7% (Fe - 248.325 nm), 3.2% (Mn) and 2.8% (Zn) for an extract containing 0.35 mg L−1 Cu, 14 mg L−1 Fe, 1.1 mg L−1 Mn and 0.12 mg L−1 Zn. Detection limits were 5.4 μg L−1 Cu, 55 μg L−1 Fe (252.744 nm), 147 μg L−1 Fe (248.325 nm), 3.0 μg L−1 Mn and 4.2 μg L−1 Zn.  相似文献

A multi-pumping flow system (MPFS) for the spectrophotometric determination of dissolved orthophosphate and dissolved organic phosphorus in wastewater samples is proposed. The determination of orthophosphate is based on the vanadomolybdate method. In-line ultraviolet photo-oxidation is employed to mineralise organic phosphorus to orthophosphate prior to detection. A solenoid valve allows the deviation of the flow towards the UV-lamp to carry out the determination of organic phosphorus.Calibration was found to be linear up to 20 mg P L−1, with a detection limit (3sb/S) of 0.08 mg P L−1, an injection throughput of 75 injections h−1 and a repeatability (R.S.D.) of 0.6% for the direct determination of orthophosphate. On the other hand, calibration graphs were linear up to 40 mg P L−1, with a detection limit (3sb/S) of 0.5 mg P L−1, an injection throughput of 11 injections h−1 and a repeatability (R.S.D.) inferior to 2.3% for the procedures involving UV photo-oxidation.  相似文献

An interfacing has been developed to connect a spectrophotometer with a personal computer and used as a readout system for development of a simple, rapid and sensitive reversed flow injection (rFI) procedure for chlorate determination. The method is based on the oxidation of indigo carmine by chlorate ions in an acidic solution (dil. HCl) leading to the decrease in absorbance at 610 nm. The decrease in absorbance is directly related to the chlorate concentration present in the sample solutions. Optimum conditions for chlorate were examined. A linear calibration graph over the range of 0.1-0.5 mg L−1 chlorate was established with the regression equation of Y = 104.5X + 1.0, r2 = 0.9961 (n = 6). The detection limit (3σ) of 0.03 mg L−1, the limit of quantitation (10σ) of 0.10 mg L−1 and the RSD of 3.2% for 0.3 mg L−1 chlorate (n = 11) together with a sample throughput of 92 h−1 were obtained. The recovery of the added chlorate in spiked water samples was 98.5 ± 3.1%. Major interferences for chlorate determination were found to be BrO3, ClO2, ClO and IO3 which were overcome by using SO32− (as Na2SO3) as masking agent. The method has been successfully applied for the determination of chlorate in spiked water samples with the minimum reagent consumption of 14.0 mL h−1. Good agreement between the proposed rFIA and the reference methods was found verified by Student's t-test at 95% confidence level.  相似文献

The complexation behavior of Aldrich humic acid (AHA) and a modified humic acid (AHA-PB) with blocked phenolic hydroxyl groups for trivalent lanthanides (Ln) is compared, and their influence on the mobility of Ln(III) in an aquifer is analyzed. As speciation technique, capillary electrophoresis (CE) was hyphenated with inductively coupled plasma mass spectrometry (ICP-MS). For metal loading experiments 25 mg L−1 of AHA and different concentrations (cLn(Eu+Gd) = 100–6000 μg L−1) of Eu(III) and Gd(III) in 10 mM NaClO4 at pH 5 were applied. By CE-ICP-MS, three Ln-fractions, assumed to be uncomplexed, weakly and strongly AHA-complexed metal can be detected. For the used Ln/AHA-ratios conservative complex stability constants log βLnAHA decrease from 6.33 (100 μg L−1 Ln3+) to 4.31 (6000 μg L−1 Ln3+) with growing Ln-content. In order to verify the postulated weaker and stronger humic acid binding sites for trivalent Eu and Gd, a modified AHA with blocked functional groups was used. For these experiments 500 μg L−1 Eu and 25 mg L−1 AHA and AHA-PB in 10 mM NaClO4 at pH-values ranging from 3 to 10 have been applied. With AHA-PB, where 84% of the phenolic OH-groups and 40% of the COOH-groups were blocked, Eu complexation was significantly lower, especially at the strong binding sites. The log β-values decrease from 6.11 (pH 10) to 5.61 at pH 3 (AHA) and for AHA-PB from 6.01 (pH 7) to 3.94 at pH 3. As a potential consequence, particularly humic acids with a high amount of strong binding sites (e.g. phenolic OH- and COOH-groups) can be responsible for a higher metal mobility in the aquifer due to the formation of dissolved negatively charged metal-humate species.  相似文献

A simple, automatic and practical system for successive determination of albumin and creatinine has been developed by combining sequential injection analysis (SIA) and highly sensitive dye-binding assays. Albumin detection was based on the increase in the absorbance due to complex formation between albumin and eosin Y in acidic media. The absorbance of the complex was monitored at 547 nm. For the creatinine assay, the concentration of creatinine was measured by reaction with alkaline picrate to form a colored product which absorbs at 500 nm. The influences of experimental variables such as effects of pH, reagent concentration, standard/sample volume and interferences were investigated. Under optimal conditions, the automated method showed linearity up to 20 mg L−1 for albumin and 100 mg L−1 for creatinine. The 3σ detection limits were 0.6 and 3.5 mg L−1 for albumin and creatinine, respectively, and the relative standard deviations (n = 10) were 2.49% for 20 mg L−1 albumin, and 3.14% for 20 mg L−1 creatinine. Application of the proposed method to the direct analysis of urinary samples yielded results which agreed with those obtained from the Bradford protein assay and a creatinine enzymatic assay according to a paired t-test. The results obtained should be a step towards developing a fully automated and reliable analytical system for clinical research, which requires direct determination of albumin and creatinine and/or its ratios.  相似文献
