A colloidal suspension of nanostructured poly(N-butyl benzimidazole)-graphene sheets (PBBIns-Gs) was used to modify a gold electrode to form a three-dimensional PBBIns-Gs/Au electrode that was sensitive to hydrogen peroxide (H2O2) in the presence of acetic acid (AcOH). The positively charged nanostructured poly(N-butyl benzimidazole) (PBBIns) separated the graphene sheets (Gs) and kept them suspended in an aqueous solution. Additionally, graphene sheets (Gs) formed “diaphragms” that intercalated Gs, which separated PBBIns to prevent tight packing and enhanced the surface area. The PBBIns-Gs/Au electrode exhibited superior sensitivity toward H2O2 relative to the PBBIns-modified Au (PBBIns/Au) electrode. Furthermore, a high yield of glucose oxidase (GOD) on the PBBIns-Gs of 52.3 mg GOD per 1 mg PBBIns-Gs was obtained from the electrostatic attraction between the positively charged PBBIns-Gs and negatively charged GOD. The non-destructive immobilization of GOD on the surface of the PBBIns-Gs (GOD-PBBIns-Gs) retained 91.5% and 39.2% of bioactivity, respectively, relative to free GOD for the colloidal suspension of the GOD-PBBIns-Gs and its modified Au (GOD-PBBIns-Gs/Au) electrode. Based on advantages including a negative working potential, high sensitivity toward H2O2, and non-destructive immobilization, the proposed glucose biosensor based on an GOD-PBBIns-Gs/Au electrode exhibited a fast response time (5.6 s), broad detection range (10 μM to 10 mM), high sensitivity (143.5 μA mM−1 cm−2) and selectivity, and excellent stability. Finally, a choline biosensor was developed by dipping a PBBIns-Gs/Au electrode into a choline oxidase (ChOx) solution for enzyme loading. The choline biosensor had a linear range of 0.1 μM to 0.83 mM, sensitivity of 494.9 μA mM−1 cm−2, and detection limit of 0.02 μM. The results of glucose and choline measurement indicate that the PBBIns-Gs/Au electrode provides a useful platform for the development of oxidase-based biosensors.  相似文献

An amperometric choline biosensor was developed by immobilizing choline oxidase (ChOx) in a layer-by-layer (LBL) multilayer film on a platinum (Pt) electrode modified with Prussian blue (PB). 6-O-Ethoxytrimethylammoniochitosan chloride (EACC) was used to prepare the ChOx LBL films. The choline biosensor was used at 0.0 V versus Ag/AgCl to detect choline and exhibited good characteristics such as relative low detection limit (5 × 10−7 M), short response time (within 10 s), high sensitivity (88.6 μA mM−1 cm−2) and a good selectivity. The results were explained based on the ultrathin nature of the LBL films and the low operating potential that could be due to the efficient catalytic reduction of H2O2 by PB. In addition, the effects of pH, temperature and applied potential on the amperometric response of choline biosensor were evaluated. The apparent Michaelis-Menten constant was found to be (0.083 ± 0.001) ×10−3 M. The biosensor showed excellent long-term storage stability, which originates from a strong adsorption of ChOx in the EACC multilayer film. When the present choline biosensor was applied to the analysis of phosphatidylcholine in serum samples, the measurement values agreed satisfactorily with those by a hospital method.  相似文献

An ascorbic acid sensor was fabricated via the drop-casting of dodecylbenzene sulphonic acid (DBSA)-doped polyaniline nanoparticles onto a screen-printed carbon-paste electrode. The modified electrode was characterised with respect to the numbers of drop cast layers, optimum potential and operating pH. The sensor was found to be optimal at neutral pH and at 0 V vs. Ag/AgCl. Under these conditions, the sensor showed good selectivity and sensitivity in that it did not respond to a range of common interferents such as dopamine, acetaminophen, uric acid and citric acid, but was capable of the detection of ascorbic acid at a sensitivity of 0.76 μA mM−1 or 10.75 μA mM−1 cm−2 across a range from 0.5 to 8 mM (r2 = 0.996, n = 6), and a limit of detection of 8.3 μM (S/N = 3). The sensor was compared to a range of other conducting polymer-based ascorbate sensors and found to be comparable or superior in terms of analytical performance.  相似文献

The carbon ceramic electrode prepared with sol-gel technique is modified by a thin film of chlorogenic acid (CGA). By immersing the carbon ceramic electrode in aqueous solution of chlorogenic acid at less than 2 s a thin film of chlorogenic acid adsorbed strongly and irreversibly on the surface of electrode. The cyclic voltammetry of the resulting modified CCE prepared at optimum conditions shows a well-defined stable reversible redox couple due to hydroquinone/quinone system in both acidic and basic solutions. The modified electrode showed excellent electrocatalytic activity toward NADH oxidation and it also showed a high analytical performance for amperometric detection of NADH. The catalytic rate constant of the modified carbon ceramic electrode for the oxidation of NADH is determined by cyclic voltammetry measurement. Under the optimised conditions the calibration curve is linear in the concentration range 1-120 μm. The detection limit (S/N = 3) and sensitivity are 0.2 μM and 25 nA μM−1.The results of six successive measurement-regeneration cycles show relative standard deviations of 2.5% for electrolyte solution containing 1 mM NADH, indicating that the electrode renewal gives a good reproducible and antifouling surface. The advantages of this amperometric detector are: high sensitivity, excellent catalytic activity, short response time t < 2 s, remarkable long-term stability, simplicity of preparation at short time and good reproducibility.  相似文献

A new method was developed for simultaneous determination of trace arsenic and antimony in Chinese herbal medicines by hydride generation-double channel atomic fluorescence spectrometry with a Soxhlet extraction system and an n-octanol-water extraction system, respectively. The effects of analytical conditions on the fluorescence intensity were investigated and optimized. A water-dissolving and methanol-water-dissolving capability were compared. The contents of different species in five Chinese herbal medicines and their decoctions were analyzed. The concentration ratios of n-octanol-soluble As or Sb to water-soluble As or Sb were related to the kinds of medicine and the acidity of the decoction. Soxhlet extraction was found to be an effective method for plants pretreatment for determination of arsenic and antimony species in Chinese herbs; the interferences of coexisting ions were evaluated. The proposed method has the advantages of simple operation, high sensitivity and high speed, with 3σ detection limits of 0.094 μg g−1 for As(III), 0.056 μg g−1 for total As, 0.063 μg g−1 for Sb(III) and 0.019 μg g−1 for total Sb in a 1.0 g of the sample.  相似文献

A highly sensitive NOx sensor was designed and developed by electrochemical incorporation of copper nanoparticles (CuNP) on single-walled carbon nanotubes (SWCNT)-polypyrrole (PPy) nanocomposite modified Pt electrode. The modified electrodes were characterized by scanning electron microscopy and energy dispersive X-ray analysis. Further, the electrochemical behavior of the CuNP-SWCNT-PPy-Pt electrode was investigated by cyclic voltammetry. It exhibited the characteristic CuNP reversible redox peaks at −0.15 V and −0.3 V vs. Ag/AgCl respectively. The electrocatalytic activity of the CuNP-SWCNT-PPy-Pt electrode towards NOx is four-fold than the CuNP-PPy-Pt electrode. These results clearly revealed that the SWCNT-PPy nanocomposite facilitated the electron transfer from CuNP to Pt electrode and provided an electrochemical approach for the determination of NOx. A linear dependence (r2 = 0.9946) on the NOx concentrations ranging from 0.7 to 2000 μM, with a sensitivity of 0.22 ± 0.002 μA μM−1 cm−2 and detection limit of 0.7 μM was observed for the CuNP-SWCNT-PPy-Pt electrode. In addition, the sensor exhibited good reproducibility and retained stability over a period of one month.  相似文献

Metallic nanoparticles of rhodium were prepared by using the newly synthesized N,N-bis-succinamide-based dendrimer as stabilizers. The Rh nanoparticles were spherical shaped with a particle size of ∼2 nm. The dendrimer Rh-encapsulated nanoparticles (Rh-DENs) were immobilized on glassy carbon electrode (GCE) and their electrocatalytic activity towards hydrogen peroxide reduction was investigated using cyclic voltammetry and chronoamperometry. The Rh-DENs modified GCE showed excellent electrocatalytic activity for hydrogen peroxide reduction reactions. The steady-state cathodic current response of the modified electrode at −0.3 V (vs SCE) in phosphate buffer (pH 7.0) showed a linear response to hydrogen peroxide concentration ranging from 8 to 30 μM with a detection limit and sensitivity of 5 μM and 0.03103 × 10−6 A μM−1, respectively.  相似文献

A feasible approach to construct multilayered enzyme film on the gold electrode surface for use as biosensing interface is described. The film was fabricated by alternate layer-by-layer deposition of periodate-oxidized glucose oxidase (GOx) and poly(allylamine) (PAA). The covalent attachment process was followed and confirmed by electrochemical impedance spectroscopy (EIS). X-ray diffraction (XRD) experiments revealed that the film was homogeneous and formed in an ordered manner with a thickness of 2.6 ± 0.1 nm per bilayer. The gold electrodes modified with the GOx/PAA multilayers showed excellent electrocatalytical response to the oxidation of glucose when ferrocenemethanol was used as an artificial redox mediator, which was studied by cyclic voltammetry (CV). From the analysis of voltammetric signals, the coverage of active enzyme on the electrode surface was estimated, which had a linear relationship with the number of GOx/PAA bilayers. This suggests that the analytical performance such as sensitivity, detection limit, and so on, is tunable by controlling the number of attached bilayers. The six GOx/PAA bilayer electrode exhibited a sensitivity of 15.1 μA mM−1 cm−2 with a detection limit of 3.8 × 10−6 M. In addition, the sensor exhibited good reproducibility and stability.  相似文献

The construction of amperometric enzymeless biosensors for phenolic compounds determination, using carbon paste electrode modified with copper phtalocyanine (CuPc) and histidine (His), based on the chemistry of the dopamine β-monooxygenase (DβM) enzyme that catalyzes the hydroxylation of the dopamine and its analogs is shown. The modified carbon paste was evaluated on electrodes constructed in two ways: putting the paste into a cavity of a rotating disk electrode and a platinum slide electrode fixed into a glass tube. The sensor in hydrodynamic conditions presented a linear response range between 30 and 250 μmol l−1, with a sensitivity of 4.6±0.1 nA l μmol−1 cm−2 for catechol, response time of 3 s and lifetime of about 50 days when stored at room temperature. The sensor in static conditions showed a linear response range from 40 to 250 μmol l−1, with a sensitivity of 0.30±0.01 nA l μmol−1 cm−2 for catechol. The sensors presented the following relative response order for dopamine and some analog species: catechol>dopamine>guaiacol>serotonin>phenol.  相似文献

In this work, a kojic acid electrochemical sensor, based on a non-covalent molecularly imprinted polymer (MIP) modified electrode, had been fabricated in the lab-on-valve system. The sensitive layer was synthesized by cyclic voltammetry using o-phenylenediamine as the functional monomer and kojic acid as the template. The template molecules were then removed from the modified electrode surface by washing with NaOH solution. Differential pulse voltammetry method using ferricyanide as probe was applied as the analytical technique, after extraction of kojic acid on the electrode. Chemical and flow parameters associated with the extraction process were investigated. The response recorded with the imprinted sensor exhibited a response in a range of 0.01-0.2 μmol L−1 with a detection limit of 3 nmol L−1. The interference studies showed that the MIP modified electrode had excellent selectivity. Furthermore, the proposed MIP electrode exhibited good sensitivity and low sample/reagent consumption, and the sensor could be applied to the determination kojic acid in cosmetics samples.  相似文献

A highly selective sequential injection (SI) method for the automated determination of weak-acid-dissociable cyanides is reported. The analytical procedure is based on the on-line reaction of the analyte with ninhydrin in carbonate medium to form a coloured product (λmax = 510 nm). Cyanides are removed from sample matrix by acidification through a gas-diffusion step incorporated in the SI manifold. The effect of instrumental and chemical variables was studied. By adopting an on-line standard addition protocol, the sensitivity of the proposed method was enhanced drastically, without affecting the determination range. The assay was validated in terms of linearity (up to 200 μg L−1), limit of detection (cL = 2.5 μg L−1), limit of quantitation (cQ = 7.5 μg L−1), precision (sr < 2.5% at 100 μg L−1) and selectivity. High tolerance against critical species such as sulfides and thiocyanates was achieved. The applicability of the method was demonstrated by analyzing tap and mineral water samples at levels below the limits established by international E.U. and U.S. organizations. The percent recoveries were satisfactory in all cases, ranging between 94.2 and 103.6%.  相似文献

The analytical potential of polystyrene (PS) spin-coated modified compact discs (CDs) surface as platforms for the development of microarray immunoassays is presented. The surface maintained the optical characteristics of compact discs, obtaining a transparent and smooth film polymer of 70 nm thickness, the track being read (λ 780 nm) without errors in a commercial CD reader/writer. The analytical capability of the methodology was demonstrated through an analysis of a neurotoxic compound (2560 spots per disc), reaching 0.08 μg L−1 as limit of detection. These figures demonstrate the enormous potential of using PS spin-coated compact discs in combination with CD players as an easy-to-operate and portable device to develop lab-on-a-disc analytical applications.  相似文献

This paper reports the fabrication of highly-sensitive cholesterol biosensor based on cholesterol oxidase (ChOx) immobilization on well-crystallized flower-shaped ZnO structures composed of perfectly hexagonal-shaped ZnO nanorods grown by low-temperature simple solution process. The fabricated cholesterol biosensors reported a very high and reproducible sensitivity of 61.7 μA μM−1 cm−2 with a response time less than 5 s and detection limit (based on S/N ratio) of 0.012 μM. The biosensor exhibited a linear dynamic range from 1.0-15.0 μM and correlation coefficient of R = 0.9979. A lower value of apparent Michaelis-Menten constant (Kmapp), of 2.57 mM, exhibited a high affinity between the cholesterol and ChOx immobilized on flower-shaped ZnO structures. Moreover, the effect of pH on ChOx activity on the ZnO modified electrode has also been studied in the range of 5.0-9.0 which exhibited a best enzymatic activity at the pH range of 6.8-7.6. To the best of our knowledge, this is the first report in which such a very high-sensitivity and low detection limit has been achieved for the cholesterol biosensor by using ZnO nanostructures modified electrodes.  相似文献

An analytical procedure with improved sensitivity was developed for cyanide determination in natural waters, exploiting the reaction with the complex of Cu(I) with 2,2′-biquinoline 4,4′-dicarboxylic acid (BCA). The flow system was based on the multi-pumping approach and long pathlength spectrophotometry with a flow cell based on a Teflon AF 2400® liquid core waveguide was exploited to increase sensitivity. A linear response was achieved from 5 to 200 μg L−1, with coefficient of variation of 1.5% (n = 10). The detection limit and the sampling rate were 2 μg L−1 (99.7% confidence level), and 22 h−1, respectively. Per determination, 48 ng of Cu(II), 5 μg of ascorbic acid and 0.9 μg of BCA were consumed. As high as 100 mg L−1 thiocyanate, nitrite or sulfite did not affect cyanide determination. Sulfide did not interfere at concentrations lower than 40 and 200 μg L−1 before or after sample pretreatment with hydrogen peroxide. The results for natural waters samples agreed with those obtained by a fluorimetric flow-based procedure at the 95% confidence level. The proposed procedure is then a reliable, fast and environmentally friendly alternative for cyanide determination in natural waters.  相似文献

A multi-reversed flow system software-assisted was developed for improvement of sensitivity in flow analysis. The performance of the flow system proposed was evaluated by using as a model the conventional Griess’ colorimetric reaction for determination of nitrite in waters. The manifold incorporated three 3-way solenoid valves, a relay box solenoid actuated, a peristaltic pump, and a photometric detector. A tailored software was designed and written in Visual Basic 6.0 which allows full control of all flow system components and simultaneous acquisition and processing of the data. The sensitivity measured as the slope of the calibration curve was improved 2.5- and 1.4-fold regarding those obtained by continuous- and stopped-flow systems, respectively. Other valuable features such as analytical throughput of 55 determinations per hour, limit of detection of 5 μg L−1 (3σblank/slope), relative standard deviation < 2% (n = 8), and a linear dynamic range up to 1800 μg L−1 were also achieved.  相似文献

Point-of-care platforms can provide fast responses, decrease the overall cost of the treatment, allow for in-home determinations with or without a trained specialist, and improve the success of the treatment. This is especially true for microfluidic paper-based analytical devices (μPAD), which can enable the development of highly efficient and versatile analytical tools with applications in a variety of biomedical fields. The objective of this work was the development of μPADs to identify and quantify levels of nitrite in saliva, which has been proposed as a potential marker of periodontitis. The devices were fabricated by wax printing and allowed the detection of nitrite by a colorimetric reaction based on a modified version of the Griess reaction. The presented modifications, along with the implementation of a paper-based platform, address many of the common drawbacks (color development, stability, etc.) associated with the Griess reaction and are supported by results related to the design, characterization, and application of the proposed devices. Under the optimized conditions, the proposed devices enable the determination of nitrite in the 10–1000 μmol L−1 range with a limit of detection of 10 μmol L−1 and a sensitivity of 47.5 AU [log (μmol L−1)]−1. In order to demonstrate the potential impact of this technology in the healthcare industry, the devices were applied to the analysis of a series of real samples, covering the relevant clinical range.  相似文献

The development of a highly sensitive amperometric sensor for nitrite using a glassy carbon electrode modified with alternated layers of iron(III) tetra-(N-methyl-4-pyridyl)-porphyrin (FeT4MPyP) and cobalt(II) tetrasulfonated phthalocyanine (CoTSPc) is described. The modified electrode showed an excellent catalytic activity and stability for the nitrite oxidation decreasing the peak potentials about 200 mV toward less positive values and presenting much higher peak currents than those obtained on the bare GC electrode. A linear response range of 0.2-8.6 μmol l−1, with a sensitivity of 0.37 μA l μmol−1 and detection limit of 0.04 μmol l−1 were obtained with this sensor. The repeatability of the proposed sensor, evaluated in term of relative standard deviation, was verified to be 1.4% for 10 measurements of 0.2 μmol l−1 nitrite solution. Interference caused by common ions has been investigated in simulated mixtures containing high concentration level of interfering ions and the sensor was found to be tolerant against these ions. The developed sensor was applied for the nitrite determination in water samples and the results were in agreement with those obtained by a comparative method described in the literature. The average recovery for these samples was 100.1 (±0.7)%.  相似文献

H2O2 is a widely occurring molecule which is also a byproduct of a number of enzymatic reactions. It can therefore be used to quantify the corresponding enzymatic substrates. In this study, the time-resolved fluorescence emission of a previously described complex consisting of phthalic acid and terbium (III) ions (PATb) is used for H2O2 detection. In detail, glucose oxidase and choline oxidase convert glucose and choline, respectively, to generate H2O2 which acts as a quencher for the PATb complex. The response time of the PATb complex toward H2O2 is immediate and the assay time only depends on the conversion rate of the enzymes involved. The PATb assay quantifies glucose in a linear range of 0.02–10 mmol L−1, and choline from 1.56 to 100 μmol L−1 with a detection limit of 20 μmol L−1 for glucose and 1.56 μmol L−1 for choline. Both biomolecules glucose and choline could be detected without pretreatment with good precision and reproducibility in human serum samples and infant formula, respectively. Furthermore, it is shown that the detected glucose concentrations by the PATb system agree with the results of a commercially available assay. In principle, the PATb system is a universal and versatile tool for the quantification of any substrate and enzyme reaction where H2O2 is involved.  相似文献

The determination of tetracycline by fluorescence spectrophotometry in complex matrices has some difficulties, because the presence of other compounds in the matrix affects the analytical signal. In this work, the effect of some inorganic species that are present in whey milk on the fluorescence signal of tetracycline is studied using a D-optimal experimental design. Next, an experimental strategy is proposed in conjunction with Parallel Factor Analysis, PARAFAC, modeling that leads to suitably modeling the severe matrix effect in the determination of tetracycline in whey milk. A specific design is performed in such a way that the lack of trilinearity due to the effect of the presence of interferents on the signal is obviated. Then, ten test samples from three brands of milk, spiked with different quantities of tetracycline and measured in 2 days were analysed using this methodology (mean of the absolute value of the relative errors: 5.1%). The developed analytical method fulfils the property of trueness, the relative errors being, both in calibration and prediction, inside the interval set by Commission Decision 2002/657/EC at these concentration levels. Decision limits (CCα) at x0 = 0 μg L−1 and at x0 = 100 μg L−1 were 13.2 and 112.4 μg L−1 respectively, for α = 0.05; whereas detection capabilities (CCβ) were 25.9 μg L−1 and 124.4 μg L−1 respectively for α = β = 0.05.  相似文献

This work presents a new methodology to quantify ethyl carbamate (EC) in fortified wines. The presented approach combines the microextraction by packed sorbent (MEPS), using a hand-held automated analytical syringe, with one-dimensional gas chromatography coupled with mass spectrometry detection (GC–MS). The performance of different MEPS sorbent materials was tested, namely SIL, C2, C8, C18, and M1. Also, several extraction solvents and the matrix effect were evaluated. Experimental data showed that C8 and dichloromethane were the best sorbent/solvent pair to extract EC. Concerning solvent and sample volumes optimization used in MEPS extraction an experimental design (DoE) was carried out. The best extraction yield was achieved passing 300 μL of sample and 100 μL of dichloromethane. The method validation was performed using a matrix-matched calibration using both sweet and dry fortified wines, to minimize the matrix effect. The proposed methodology presented good linearity (R2 = 0.9999) and high sensitivity, with quite low limits of detection (LOD) and quantification (LOQ), 1.5 μg L−1 and 4.5 μg L−1, respectively. The recoveries varied between 97% and 106%, while the method precision (repeatability and reproducibility) was lower than 7%. The applicability of the methodology was confirmed through the analysis of 16 fortified wines, with values ranging between 7.3 and 206 μg L−1. All chromatograms showed good peak resolution, confirming its selectivity. The developed MEPS/GC–MS methodology arises as an important tool to quantify EC in fortified wines, combining efficiency and effectiveness, with simpler, faster and affordable analytical procedures that provide great sensitivity without using sophisticated and expensive equipment.  相似文献
