Zinc oxide nanoparticles (NanoZnO) uniformly dispersed in chitosan (CHIT) have been used to fabricate a hybrid nanocomposite film onto indium-tin-oxide (ITO) glass plate. Cholesterol oxidase (ChOx) has been immobilized onto this NanoZnO-CHIT composite film using physiosorption technique. Both NanoZnO-CHIT/ITO electrode and ChOx/NanoZnO-CHIT/ITO bioelectrode have been characterized using Fourier transform-infrared (FTIR), X-ray diffraction (XRD), cyclic voltammetry (CV), scanning electron microscopy (SEM) and electrochemical impedance spectroscopy (EIS) techniques, respectively. The ChOx/NanoZnO-CHIT/ITO bioelectrode exhibits linearity from 5 to 300 mg dl−1 of cholesterol with detection limit as 5 mg dl−1, sensitivity as 1.41 × 10−4 A mg dl−1 and the value of Michaelis-Menten constant (Km) as 8.63 mg dl−1. This cholesterol biosensor can be used to estimate cholesterol in serum samples.  相似文献

A highly sensitive disposable screen-printed butyrylcholine (BuCh) potentiometric sensor, based on heptakis (2,3,6-tri-o-methyl)-β-cyclodextrin (β-CD) as ionophore, was developed for butyrylcholinesterase (BuChE) activity monitoring. The proposed sensors have been characterized and optimized according to the constituents of homemade printing carbon ink including β-CD, anionic sites, and plasticizer. The fabricated sensor showed Nernstian responses from 10−6 to 10−2 mol L−1 with detection limit of 8 × 10−7 mol L−1, fast response time (1.6 s) and adequate shelf-life (6 months). Improved selectivity towards BuCh with minimal interference from choline (Ch) was achieved and the sensor was used for determination of 0.06-1.25 U mL−1 BuChE. The developed disposable sensors have been successfully applied for real-time intoxication monitoring through assaying cholinesterases (ChEs) activity in human serum. Determination of organophosphate pesticide was conducted by measuring their inhibition of BuChE with successful assaying of malathion in insecticide samples with high accuracy and precision.  相似文献

A new rapid, convenient and sensitive electrochemical method based on a gold nanoparticles modified ITO (Au/ITO) electrode is described for the detection of dopamine and serotonin in the presence of a high concentration of ascorbic acid. The electrocatalytic response was evaluated by differential pulse voltammetry (DPV) and the modified electrode exhibited good electrocatalytic properties towards dopamine and serotonin oxidation with a peak potential of 70 mV and 240 mV lower than that at the bare ITO electrode, respectively. The selective sensing of dopamine is further improved by applying square wave voltammetry (SWV) which leads to the lowering of its detection limit. A similar effect on the detection limit of serotonin was observed on using SWV. Linear calibration curves are obtained in the range 1.0 × 10−9-5.0 × 10−4 M and 1.0 × 10−8-2.5 × 10−4 M with a detection limit of 0.5 nM and 3.0 nM for dopamine and serotonin, respectively. The Au/ITO electrode efficiently determines both the biomolecules simultaneously, even in the presence of a large excess of ascorbic acid. The adequacy of the developed method was evaluated by applying it to the determination of the content of dopamine in dopamine hydrochloride injections. The proposed procedure was also successfully applied to simultaneously detect dopamine and serotonin in human serum and urine.  相似文献

Cholesterol oxidase (ChOx) has been covalently linked to Langmuir-Blodgett (LB) monolayers of polyaniline (PANI)-stearic acid (SA) prepared onto indium-tin-oxide (ITO) coated glass plates via glutaraldehyde (Glu) chemistry. These ChOx/Glu/PANI-SA LB film/ITO electrodes have been characterized by FT-IR, cyclic voltammetry, and scanning electron microscopy, respectively. The results of response measurements carried out on these bioelectrodes using linear sweep voltammetry (LSV) reveal linearity from 25 to 400 mg/dL of cholesterol concentration with sensitivity of 88.9 nA mg(-1) dL. The linear regression analysis of bioelectrode reveals standard deviation and correlation coefficient of 0.737 microA and 0.9988, respectively. The low value of the Michaelis-Menten constant of these bioelectrodes obtained as 1.21 mM for the immobilized enzyme indicates increased interaction between ChOx and cholesterol in the PANI-SA LB film.  相似文献

A simple method for the rapid and simultaneous analysis of dichlorvos (DDVP), malathion, carbaryl, and 2,4-dichlorophenoxy acetic acid (2,4-D) in citrus fruit, which uses flow-injection ion spray ionization tandem mass spectrometry, has been developed for the first time. The method involves the combined use of stable isotopically labeled internal standards (DDVP-d6, malathion-d10, carbaryl-d7, and 2,4-D-d5) and a multiple reaction monitoring technique. The average recoveries for the pesticides at the same concentrations as their tolerance levels (DDVP: 0.1-0.2 μg g−1; malathion: 0.5-4.0 μg g−1; carbaryl: 1.0 μg g−1; 2,4-D: 1.0-2.0 μg g−1) ranged from 90 to 119% with the relative standard deviation (R.S.D.) ranging from 1.0 to 13.1% (n = 5). Analysis time, including sample preparation and determination, was only 15 min. The present method is effective for screening DDVP, malathion, carbaryl, and 2,4-D in citrus fruit.  相似文献

An electrochemical breast cancer biosensor based on a chitosan-co-polyaniline (CHIT-co-PANI) copolymer coated onto indium-tin-oxide (ITO) was fabricated by immobilizing the complementary DNA (cDNA) probe (42 bases long) associated with the breast cancer susceptible gene BRCA1. Both the CHIT-co-PANI/ITO and the cDNA/CHIT-co-PANI/ITO electrodes were characterized with Fourier transform infrared (FTIR) spectroscopy, atomic force microscopy (AFM), cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS). For the cDNA/CHIT-co-PANI/ITO electrode, the amperometric current decreased linearly with an increasing logarithm of molar concentration of the single-stranded target DNA (ssDNA) within the range of 0.05-25 fmol. The bioelectrode exhibited a sensitivity of 2.104 μA/fmol with a response time of 16 s. The cDNA/CHIT-co-PANI/ITO electrode had a shelf life of about six months, even when stored at room temperature.  相似文献

An electrochemical aptasensor based on chitosan-iron oxide nanocomposite (CHIT-IO) film deposited on fluorine tin Oxide (FTO) was developed for the detection of malathion. Iron oxide nanoparticles were prepared by co-precipitation method and characterized by Transmission electron microscopy and UV–Visible spectroscopy. The biotinylated DNA aptamer sequence specific to the malathion was immobilized onto the iron oxide doped-chitosan/FTO electrode by using streptavidin as linking molecule. Various characterization studies like Field Emission-Scanning Electron Microscopy (FE-SEM), Fourier Transform Infrared Spectroscopy (FT-IR), and Electrochemical studies were performed to attest the successful fabrication of bioelectrodes. Experimental parameters like aptamer concentration, response time, stability of electrode and reusability studies were optimized. Aptamer immobilized chitosan-iron oxide nanocomposite (APT/SA/CHIT-IO/FTO) bioelectrodes exhibited LOD of about 0.001 ng/mL within 15 min and spike-in studies revealed about 80–92% recovery of malathion from the lettuce leaves and soil sample.  相似文献

Polymeric light-emitting diodes (PLEDs) using high-performance hole-transport polyurethanes (PUs) have been fabricated. The PUs were prepared from the condensation polymerization of (EE)-1,4-bis(2-hydroxystyryl)benzene, an oligo para-phenylene-(E)-vinylene (OPV) unit, with toluene diisocyanate (TDI), isophorone diisocyanate (IPDI) or dicyclohexylmethane 4,4′-diisocyanate (H12MDI), respectively. The condensation polymerization was end-capped with 4-tert-butylphenol as the terminal group. The PLED having the PU layer inserted between PEDOT:PSS (HIL) and MEH-PPV (EML) demonstrated superior current efficiency and low turn-on voltage when comparing to the reference devices of ITO/MEH-PPV(50 nm)/Ca(10 nm)/Ag(100 nm) as well as ITO/PEDOT:PSS(30 nm)/MEH-PPV(50 nm)/Ca(10 nm)/Ag(100 nm). In particularly, the best device performance was realized with the PU of OPV-IPDI as the hole-transport layer, resulting 53 times and 2.72 times of current efficiency enhancement as well as 1.5 V and 1 V voltage reduction of the turn-on voltage, respectively, when compared against the reference devices. Besides, our experiments also showed that the PU polymer could also be applied for flexible PLED with similar performance enhancement. Based on the promising results, we concluded that OPV-IPDI was a good hole-transport material for light-emitting diode application.  相似文献

A composite film (MWCNTs-PNF) which contains multi-walled carbon nanotubes (MWCNTs) along with the incorporation of poly(new fuchsin) (PNF) has been synthesized on glassy carbon electrode (GCE), gold (Au) and indium tin oxide (ITO) by potentiostatic methods. The presence of MWCNTs in the composite film enhances surface coverage concentration (Γ) of PNF to ≈176.5%, and increases the electron transfer rate constant (ks) to ≈346%. The composite film also exhibits promising enhanced electrocatalytic activity towards the mixture of biochemical compounds such as adenine (AD), guanine (GU) and thymine (THY). The surface morphology of the composite film deposited on ITO has been studied using scanning electron microscopy and atomic force microscopy. These two techniques reveal that the PNF incorporated on MWCNTs. Electrochemical quartz crystal microbalance study reveals the enhancement in the functional properties of MWCNTs and PNF. The electrocatalytic responses of analytes at MWCNTs and MWCNTs-PNF films were measured using both cyclic voltammetry (CV) and differential pulse voltammetry (DPV). From electrocatalysis studies, well separated voltammetric peaks have been obtained at the composite film for AD, GU and THY, with the peak separation of 320.3 and 132.7 mV between GU-AD and AD-THY respectively. The sensitivity of the composite film towards AD, GU and THY in DPV technique is 218.18, 12.62 and 78.22 mA M−1 cm−2 respectively, which are higher than MWCNTs film. Further, electroanalytical studies of AD, GU and THY present in single-strand deoxyribonucleic acid (ssDNA) have been carried out using semi-derivative CV and DPV.  相似文献

We report results of the studies relating to the fabrication and characterization of a conducting polymer based molecularly imprinted para-nitrophenol (PNP) sensor. A water pollutant, para-nitrophenol is electrochemically imprinted with polyvinyl sulphonic acid (PVSA) doped polyaniline onto indium tin oxide (ITO) glass substrate. This PNP imprinted electrode (PNPI-PANI-PVSA/ITO) prepared via chronopotentiometric polymerization and over-oxidation is characterized by Fourier transform infra-red spectroscopy (FT-IR), UV–visible (UV–vis) spectroscopy, contact angle (CA), scanning electron microscopy (SEM), cyclic voltammetry (CV) and differential pulse voltammetry (DPV) studies. The response studies of PNPI-PANI-PVSA/ITO electrode carried out using DPV reveal a lower detection limit of 1 × 10−3 mM, improved sensitivity as 1.5 × 10−3 A mM−1 and stability of 45 days. The PNPI-PANI-PVSA/ITO electrode shows good precision with relative standard deviation of 2.1% and good reproducibility with standard deviation of 3.78%.  相似文献

The effect of surface modification of indium tin oxide (ITO) by multi wall carbon nanotube (MWNT) and gold nanoparticles attached multi wall carbon nanotube (AuNP-MWNT) has been studied to determine tryptophan, an important and essential amino acid for humans and herbivores. A detailed comparison has been made among the voltammetric response of bare ITO, MWNT/ITO and AuNP-MWNT/ITO in respects of several essential analytical parameters viz. sensitivity, detection limit, peak current and peak potential of tryptophan. The AuNP-MWNT/ITO exhibited a well defined anodic peak at pH 7.2 at a potential of ∼669 mV for the oxidation of tryptophan as compared to 760 mV at MWNT/ITO electrode. Under optimum conditions linear calibration curve was obtained over tryptophan concentration range 0.5-90.0 μM in phosphate buffer solution of pH 7.2 with detection limit and sensitivity of 0.025 μM and 0.12 μA μM−1, respectively. The oxidation of tryptophan occurred in a pH dependent, 2e and 2H+ process and the electrode reaction followed adsorption controlled pathway. The method has been found selective and successfully implemented for the determination of tryptophan in human urine and plasma samples using standard addition method. The electrode exhibited an efficient catalytic response with good reproducibility and stability.  相似文献

A series of alternating fluorene and p-phenylenevinylene copolymers containing non-conjugated spacer have been synthesized through the Wittig polycondensation reaction. These amorphous copolymers are highly soluble in common organic solvents and can be spin-cast to obtain transparent films. The effects of non-conjugated spacers in the main chain and the methoxyl groups on the side chain on the thermal behavior, photoluminescence (PL) and electroluminescence (EL) properties of these copolymers have been investigated in detail. Single-layered light-emitting diodes (LEDs) have been fabricated in the configuration of ITO/PEDOT/copolymer/Ca/Al and emitted blue light in the range of 456-492 nm. The measurements of current vs voltage show turn-on voltages at 6.2-12.4 V. Among the LEDs based on the six copolymers, the maximum EL brightness and efficiency of the LED based on P1 containing 4CH2 aliphatic segment length in the main chain and without methoxyl groups on side chain are reached 3936 cd/m2 and 0.70 cd/A, respectively.  相似文献

Amperometric acetylcholinesterase (AChE) biosensors have been developed to resolve mixtures of chlorpyrifos oxon (CPO) and chlorfenvinfos (CFV) pesticides. Three different biosensors were built using the wild type from electric eel (EE), the genetically modified Drosophila melanogaster AChE B394 and B394 co-immobilized with a phosphotriesterase (PTE). Artificial Neural Networks (ANNs) were used to model the combined response of the two pesticides. Specifically two different ANNs were constructed. The first one was used to model the combined response of B394 + PTE and EE biosensors and was applied when the concentration of CPO was high and the other, modelling the combined response of B394 + PTE and B394 biosensors, was applied with low concentrations of CPO. In both cases, good prediction ability was obtained with correlation coefficients better than 0.986 when the obtained values were compared with those expected for a set of six external test samples not used for training.  相似文献

As continuous outbreak of avian influenza (AI) has become a threat to human health, economic development and social stability, it is urgently necessary to detect the highly pathogenic avian influenza H5N1 virus quickly. In this study, we fabricated indium-tin-oxide thin-film transistors (ITO TFTs) on a glass substrate for the detecting of AI H5N1. The ITO TFT is fabricated by a one-shadow-mask process in which a channel layer can be simultaneously self-assembled between ITO source/drain electrodes during magnetron sputtering deposition. Monoclonal anti-H5N1 antibodies specific for AI H5N1 virus were covalently immobilized on the ITO channel by (3-glycidoxypropyl)trimethoxysilane. The introduction of target AI H5N1 virus affected the electronic properties of the ITO TFT, which caused a change in the resultant threshold voltage (VT) and field-effect mobility. The changes of IDVG curves were consistent with an n-type field effect transistor behavior affected by nearby negatively charged AI H5N1 viruses. The transistor based sensor demonstrated high selectivity and stability for AI H5N1 virus sensing. The sensor showed linear response to AI H5N1 in the concentrations range from 5 × 10−9 g mL−1 to 5 × 10−6 g mL−1 with a detection limit of 0.8 × 10−10 g mL−1. Moreover, the ITO TFT biosensors can be repeatedly used through the washing processes. With its excellent electric properties and the potential for mass commercial production, ITO TFTs can be promising candidates for the development of label-free biosensors.  相似文献

In the presented work, a disposable immunosensor for the detection of testosterone, an endogenous steroid hormone, in bovine urine has been developed using screen-printed electrodes (SPEs). Due to concerns over the use of steroid hormones as growth promoters, the EU prohibits their use in food producing animals. Consequently, rigorous screening procedures have been implemented in all member states to detect the illegal administration of such compounds. Competitive immunoassays were developed, initially by enzyme linked immunosorbent assay (ELISA), and subsequently transferred to an electrochemical immunosensor format using disposable screen-printed carbon electrodes. Horseradish peroxidase (HRP) was the enzyme label of choice and chronoamperometric detection was carried out using a tetramethylbenzidine/hydrogen peroxide (TMB/H2O2) substrate system, at +100 mV. The EC50 values obtained for the assay in buffer and urine gave relatively comparable results, 710 pg mL−1 and 960 pg mL−1, respectively. The linear range obtained for the assay in buffer extended from 0.03 ng mL−1 to 40 ng mL−1; while that in urine ranged from 0.03 ng mL−1 to 1.6 ng mL−1. The corresponding limits of detection (LOD) in buffer and urine were 26 pg mL−1 and 1.8 pg mL−1. Cross reactivity profiles of the antibody have been examined, with notable cross reactivities with 19-nortestosterone (11.6%) and boldenone (9.86%). Precision studies for the sensor demonstrated adequate reproducibility (CV < 13%, n = 3) and repeatability (CV < 9%, n = 3). Recovery data obtained showed good agreement between spiking studies and known concentrations of analyte. Sensors showed stability for 4 days at +4 °C. A sensitive, highly specific, inexpensive, disposable immunosensor, showing excellent overall performance for the detection of testosterone in bovine urine, has been developed.  相似文献

This work reports monoamine oxidase (MAO)/horseradish peroxidase (HRP) and diamine oxidase (DAO)/horseradish peroxidase (HRP) based biosensors using screen-printed carbon electrodes for the determination of biogenic amines (BA). The enzymes have been covalently immobilized onto the carbon working electrode, previously modified by an aryl diazonium salt, using hydroxysuccinimide and carbodiimide. The detection has been performed by measuring the cathodic current due to the reduction of the mediator hydroxymethylferrocene at a low potential, 250 mV vs screen-printed Ag/AgCl reference electrode. The experimental conditions for the enzymes immobilization, as well as for the main variables that can influence the chronoamperometric current have been optimized by the experimental design methodology. Under these optimum conditions, the disposable biosensors have been characterized. A linear response range from 0.2 up to 1.6 μM and from 0.4 to 2.4 μM of histamine was obtained for DAO/HRP and MAO/HRP based biosensors, respectively. The biosensor construction was highly reproducible, yielding relative standard deviations of 10% and 11% in terms of sensitivity for DAO/HRP and MAO/HRP based biosensors, respectively. The capability of detection, 0.18 ± 0.01 μM in the case of DAO/HRP and 0.40 ± 0.04 μM (α = 0.05 and β = 0.005) for MAO/HRP based biosensors, and the biosensor sensitivity towards different BA has also been analyzed. Finally, the developed biosensors have been applied to the determination of the total amine content in fish samples.  相似文献

Bismuth-coated glassy carbon electrodes have been successfully applied for catalytic adsorptive stripping voltammetric measurements of low levels of vanadium(V) in the presence of chloranilic acid (CAA) and bromate ion. The new protocol is based on the accumulation of the vanadium-chloranilic acid complex from an acetate buffer (pH 5.5) solution at a preplated bismuth film electrode held at −0.35 V (versus Ag/AgCl), followed by a square-wave voltammetric scan. Factors influencing the adsorptive stripping performance, including the CAA and bromate concentrations, solution pH, and accumulation potential or time have been optimized. The response compares favorably with that observed at mercury film electrodes. A linear response is observed over the 5-25 μg/L concentration range (2 min accumulation), along with a detection limit of 0.20 μg/L vanadium (10 min accumulation). High stability is indicated from the reproducible response of a 50 μg/L vanadium solution (n = 25; R.S.D. = 3.1%). Applicability to a groundwater sample is illustrated.  相似文献

A multi-determinant artificial antigen was prepared by haptens of four pesticides (chlorpyrifos, triazophos, carbofuran and parathion methyl) conjugating to the carrier protein BSA in turn. Male New Zealand white rabbits were immunized with this multi-determinant immunogen to produce the polyclonal antibodies (PAbs), which can recognize the four pesticides. The PAbs displayed high level for each relative hapten-OVA conjugate, with the favorable titers of 4.49 × 104, 8.98 × 104, 2.24 × 104 and 1.86 × 104, for CHBu-OVA, THHe-OVA, BFNB-OVA and MP5-OVA, respectively. Characterization studies of the PcAbs showed that it has high affinity and specificity to the four relative pesticides. An indirect competitive ELISA was developed for multi-residue determination. The I50 value for the four pesticides was 0.290, 0.065, 0.582 and 2.824 μg mL−1, with the detection limit (I10) of 0.022, 0.005, 0.015 and 0.115 μg mL−1 for carbofuran, triazophos, chlorpyrifos and parathion methyl, respectively. The linear rang was 0.016-2.000, 0.005-0.500, 0.010-2.000 and 0.063-5.000 μg mL−1, respectively, for carbofuran, triazophos, chlorpyrifos and parathion methyl. Results indicated that, this study provided a new strategy to develop immunoassays through artificial antigen design for pesticides multi-residue determination.  相似文献

A new high temperature, flexible and colorless indium-tin-oxide (ITO) coated plastic substrate has been prepared from a thermally stable, high glass transition temperature (Tg) and colorless polyimide film. The polyimide was synthesized from 3,3′-diaminodiphenylsulfone (3,3′-DDS) and 4,4′-(hexafluoroisopropylidene) diphthalic anhydride (6FDA) monomers. Its heat distortion temperature was 278 °C. The ITO was deposited on the polyimide film using the magnetron-sputtering process. After annealing at 250 °C under nitrogen for 1 h, the resistivity of the ITO film was 4.0 × 10−4 Ω cm, and its transmittance was 83.5%. Scanning electron microscope (SEM) and X-ray diffraction (XRD) were used to observe the surface and morphology of the ITO film. UV-visible spectroscopy and the four-probe method were used to study their optical and electrical properties. The high performance ITO-plastic substrate can be used in the next generation flat panel displays.  相似文献

A compact hand-held heated fluorometric instrument for performing real-time isothermal nucleic acid amplification and detection is described. The optoelectronic instrument combines a Printed Circuit Board/Micro Electro Mechanical Systems (PCB/MEMS) reaction detection/chamber containing an integrated resistive heater with attached miniature LED light source and photo-detector and a disposable glass waveguide capillary to enable a mini-fluorometer. The fluorometer is fabricated and assembled in planar geometry, rolled into a tubular format and packaged with custom control electronics to form the hand-held reactor. Positive or negative results for each reaction are displayed to the user using an LED interface. Reaction data is stored in FLASH memory for retrieval via an in-built USB connection. Operating on one disposable 3 V lithium battery >12, 60 min reactions can be performed. Maximum dimensions of the system are 150 mm (h) × 48 mm (d) × 40 mm (w), the total instrument weight (with battery) is 140 g. The system produces comparable results to laboratory instrumentation when performing a real-time nucleic acid sequence-based amplification (NASBA) reaction, and also displayed comparable precision, accuracy and resolution to laboratory-based real-time nucleic acid amplification instrumentation. A good linear response (R2 = 0.948) to fluorescein gradients ranging from 0.5 to 10 μM was also obtained from the instrument indicating that it may be utilized for other fluorometric assays. This instrument enables an inexpensive, compact approach to in-field genetic screening, providing results comparable to laboratory equipment with rapid user feedback as to the status of the reaction.  相似文献
