A critical evaluation of the application of attenuated total reflectance–Fourier transform infrared spectroscopy (ATR–FTIR) and partial least squares (PLS) to the determination of several nutritional parameters, such as the energetic value and the carbohydrate, protein and calcium contents, in commercially available yogurt samples was carried out. To this end, a highly heterogeneous population of 48 samples covering a wide range of yogurts obtained from the Spanish market was used. After correcting the spectra, hierarchical cluster analysis was performed in order to select a representative calibration set and the corresponding validation sample set. Different PLS models and several spectral windows were tested in order to evaluate their prediction capabilities for the validation set. For all nutritional parameters, with the exception of fat content, the procedure developed here provided good precision; the values obtained complied with the statutory values declared by the US FDA.  相似文献

Near-infrared spectroscopy (NIRS) has been widely used in the pharmaceutical field because of its ability to provide quality information about drugs in near-real time. In practice, however, the NIRS technique requires construction of multivariate models in order to correct collinearity and the typically poor selectivity of NIR spectra. In this work, a new methodology for constructing simple NIR calibration models has been developed, based on the spectrum for the target analyte (usually the active principle ingredient, API), which is compared with that of the sample in order to calculate a correlation coefficient. To this end, calibration samples are prepared spanning an adequate concentration range for the API and their spectra are recorded. The model thus obtained by relating the correlation coefficient to the sample concentration is subjected to least-squares regression. The API concentration in validation samples is predicted by interpolating their correlation coefficients in the straight calibration line previously obtained. The proposed method affords quantitation of API in pharmaceuticals undergoing physical changes during their production process (e.g. granulates, and coated and non-coated tablets). The results obtained with the proposed methodology, based on correlation coefficients, were compared with the predictions of PLS1 calibration models, with which a different model is required for each type of sample. Error values lower than 1-2% were obtained in the analysis of three types of sample using the same model; these errors are similar to those obtained by applying three PLS models for granules, and non-coated and coated samples. Based on the outcome, our methodology is a straightforward choice for constructing calibration models affording expeditious prediction of new samples with varying physical properties. This makes it an effective alternative to multivariate calibration, which requires use of a different model for each type of sample, depending on its physical presentation.  相似文献

Real samples were used for PLS model calibration and validation steps, showing that this approach can be of value in preventing deviations in the results caused by the matrix effects for the simultaneous spectrophotometric determination of aluminum and iron in plant extracts. One hundred UV-vis spectra, obtained from samples of the 1997 to 2000 International Plant-Analytical Exchange (IPE) program (The Netherlands), were used for model development, with ICP-AES aluminum and iron determinations as reference values for model calculation. The plant extracts were analyzed both by ICP-AES and by the PLS models developed in this work, using calibrations with both aqueous standard solutions and with real sample extracts. In addition, since the use of smaller calibration sets could be of value in reducing both the cost and the time of analysis, sets with fewer calibration samples were also investigated, with the help of the Kennard and Stone algorithm for sample selection. Comparison of the predictability of the best model obtained with each calibration set was made using the ratio of their relative root mean square error (%RMSEV) for samples in the validation set, for aluminum or iron determinations, and were compared against F-test tabulated values. For all the models developed with real samples, the differences in the %RMSEV values for the aluminum or iron determinations were found not to be statistically significant, at a confidence level of 95%. Although it was observed that the aluminum, but not the iron, determinations with the PLS 2 model prepared with aqueous standards tend to be slightly lower than the ICP-AES determinations, this model has a good global prediction ability, as observed through the correlation curves presented, and can be used for screening determinations or for other agricultural purposes.  相似文献

The selection of an appropriate calibration set is a critical step in multivariate method development. In this work, the effect of using different calibration sets, based on a previous classification of unknown samples, on the partial least squares (PLS) regression model performance has been discussed. As an example, attenuated total reflection (ATR) mid-infrared spectra of deep-fried vegetable oil samples from three botanical origins (olive, sunflower, and corn oil), with increasing polymerized triacylglyceride (PTG) content induced by a deep-frying process were employed. The use of a one-class-classifier partial least squares-discriminant analysis (PLS-DA) and a rooted binary directed acyclic graph tree provided accurate oil classification. Oil samples fried without foodstuff could be classified correctly, independent of their PTG content. However, class separation of oil samples fried with foodstuff, was less evident. The combined use of double-cross model validation with permutation testing was used to validate the obtained PLS-DA classification models, confirming the results. To discuss the usefulness of the selection of an appropriate PLS calibration set, the PTG content was determined by calculating a PLS model based on the previously selected classes. In comparison to a PLS model calculated using a pooled calibration set containing samples from all classes, the root mean square error of prediction could be improved significantly using PLS models based on the selected calibration sets using PLS-DA, ranging between 1.06 and 2.91% (w/w).  相似文献

Fourier transform-near infrared (FT-NIR) and FT-Raman spectrometries have been used to design partial least squares (PLS) calibration models for the determination of the ethanol content of ethanol fuel and alcoholic beverages. In the FT-NIR measurements the spectra were obtained using air as reference, and the spectral region for PLS modeling were selected based on the spectral distribution of the relative standard deviation in concentration. In the FT-Raman measurements hexachloro-1,3-butadiene (HCBD) has been used as an external standard. In the PLS/FT-NIR modeling for ethanol fuel analysis 50 ethanol fuel standards (84.9-100% (w/w)) were used (25 in the calibration, 25 in the validation). In the PLS/FT-Raman modeling 25 standards were used (13 in the calibration, 12 in the validation). The PLS/FT-NIR and FT-Raman models for beverage analysis made use of 24 standards (0-100% (v/v)). Twelve of them contained sugars (1-5% (w/w)), one-half was used in the calibration and the other half in the validation. Different spectral pre-processing were used in the PLS modeling, depending on the type of sample investigated. In the ethanol fuel analysis the FT-NIR pre-processing was a 17 points smoothed first derivative and for beverages no spectral pre-processing was used. The FT-Raman spectra were pre-processed by vector normalization in the ethanol fuel analysis and by a second derivative (17 points smoothing) in the beverage analysis. The PLS models were used in the analysis of real ethanol fuel and beverage samples. A t-test has shown that the FT-NIR model has an accuracy equivalent to that of the reference method (ASTM D4052) in the analysis of ethanol fuel, while in the analysis of beverages, the FT-Raman model presents an accuracy equivalent to the reference method. The limits of detection for NIR and Raman calibration models were 0.05 and 0.2% (w/w), respectively. It has also been shown that both techniques, present better results than gas chromatography (GC) in evaluating the ethanol content of beverages.  相似文献

A solvent free, fast and environmentally friendly near infrared-based methodology (NIR) was developed for pesticide determination in commercially available formulations. This methodology was based on the direct measurement of the diffuse reflectance spectra of solid samples and a multivariate calibration model (partial least squares, PLS) to determine the active principle concentration in commercial formulations. The PLS calibration set was built on using the spiked samples by mixing different amounts of pesticide standards and powdered samples. Buprofezin, Diuron and Daminozide were used as test analytes. Concentration of Buprofezin in the samples was calculated employing a 4-factors PLS calibration using the spectral information in the range between 2231–2430 and 1657–1784 nm. For Diuron determination a 1-factor PLS calibration model using the spectral range 1110–2497 nm, after a linear removed correction. Daminozide determination was carried out employing a 4-factors PLS model using the spectral information in the ranges 1644–1772 and 2014–2607 nm without baseline correction. The root mean square errors of prediction (RMSEP) found were 1.1, 1.7 and 0.7% (w/w) for Buprofezin, Diuron and Daminozide determination, respectively. The developed PLS-NIR procedure allows the determination of 120 samples/h, does not require any sample pre-treatment and avoids waste generation.  相似文献

The use of chemometrics in order to improve the molecular selectivity of infrared (IR) spectra has been evaluated using classic least squares (CLS), partial least squares (PLS), science-based calibration (SBC), and multivariate curve resolution-alternate least squares (MCR-ALS) techniques for improving the discriminatory and quantitative performance of infrared hollow waveguide gas sensors. Spectra of mixtures of isobutylene, methane, carbon dioxide, butane, and cyclopropane were recorded, analyzed, and validated for optimizing the prediction of associated concentrations. PLS, CLS, and SBC provided equivalent results in the absence of interferences. After addition of the spectral characteristics of water by humidifying the sample mixtures, CLS and SBC results were similar to those obtained by PLS only if the water spectrum was included in the calibration model. In the presence of an unknown interferant, CLS revealed errors up to six times higher than those obtained by PLS. However, SBC provided similar results compared to PLS by adding a measured noise matrix to the model. Using MCR-ALS provided an excellent estimation of the spectra of the unknown interference. Furthermore, this method also provided a qualitative and quantitative estimation of the components of an unknown set of samples. In summary, using the most suitable chemometrics approach could improve the selectivity and quality of the calibration model derived for a sensor system, and may avoid the need to analyze expensive calibration data sets. The results obtained in the present study demonstrated that (1) if all sample components of the system are known, CLS provides a sufficiently accurate solution; (2) the selection between PLS and SBC methods depends on whether it is easier to measure a calibration data set or a noise matrix; and (3) MCR-ALS appears to be the most suitable method for detecting interferences within a sample. However, the latter approach requires the most extensive calculations and may thus result in limited temporal resolution, if the concentration of a component should be continuously monitored.  相似文献

It has been evaluated the potential of near-infrared (NIR) diffuse reflectance infrared Fourier transform spectroscopy (DRIFTS) as a way for non-destructive measurement of trace elements at μg kg−1 level in foods, with neither physical nor chemical pre-treatment. Predictive models were developed using partial least-square (PLS) multivariate approaches based on first-order derivative spectra. A critical comparison of two spectral pre-treatments, multiplicative signal correction (MSC) and standard normal variate (SNV) was also made. The PLS models built after using SNV provided the best prediction results for the determination of arsenic and lead in powdered red paprika samples. Relative root-mean-square error of prediction (RRMSEP) of 23% for both metals, arsenic and lead, were found in this study using 20 well characterized samples for calibration and 13 additional samples as validation set. Results derived from this study showed that NIR diffuse reflectance spectroscopy combined with the appropriate chemometric tools could be considered as an useful screening tool for a rapid determination of As and Pb at concentration level of the order of hundred μg kg−1.  相似文献

将中红外光谱筛选出的598个纯涤、纯棉及涤/棉混纺样本采用GB/T 2910.11-2009法测定其涤、棉准确含量,其中校正集样本252个,验证集样本346个。使用便携式近红外光谱仪获取样本的原始近红外光谱(NIRS)。校正集样本依据回归系数的分布趋势和范围选取最佳建模谱区,并采用差分一阶导、S-G平滑和均值中心化相结合的方法对原始光谱进行预处理,利用偏最小二乘法(PLS)建立涤/棉混纺织物中涤含量的近红外(NIR)定量分析模型。同时分析了样本颜色对NIRS的影响,探讨了斜线光谱样本、奇异样本和不同组织结构织物对模型预测效果的影响。结果表明:利用PLS法建立的涤/棉混纺织物定量分析模型最优组合包含1个光谱区间和9个主成分因子,校正集相关系数(RC)为0.998,标准偏差(SEC)为0.908。为验证所建模型的有效性和实用性,对346个未参与建模的涤棉样本进行了预测,并将预测结果与国标法测定值进行方差分析,两种方法结果无显著差异,预测正确率达97%以上。模型的建立为废旧涤/棉混纺织物快速、无损分拣提供了基础数据库。  相似文献

Diffuse reflectance near-infrared spectroscopy (NIR) combined with partial least squares (PLS) data treatment has been employed for the rapid and nondestructive determination of sedimentary humic substances. Forty one samples of surface estuarine sediments, taken during distinct seasonal periods from different locations across Ria de Arousa (northwest of Spain), were scanned at wavelengths from 833 to 2,976 nm (12,000 to 3,360 cm−1). Twenty four samples were randomly selected, from previous hierarchical cluster analysis of their NIR spectra, for the calibration set, and the 17 remaining samples were assigned to the validation set. NIR spectra of calibration samples were correlated to measured values of humic acids (HAs) and fulvic acids (FAs), which ranged from 1.53 to 28.17 mg/g and from 0.37 to 2.45 mg/g, respectively, using PLS regression and multiplicative scattering correction on the raw and first-derivative NIR spectra, respectively. Low root mean square error of prediction values of 4.3 mg HA/g sediment and 0.25 mg FA/g sediment were obtained. Good residual prediction deviation values of 1.16 and 1.2 were obtained for HA and FA, respectively, allowing the PLS models built to be considered as appropriate tools for screening purposes. The combination of infrared (MIR) and near-infrared (NIR) spectroscopy has been employed for the determination of important quality parameters of beers, such as original and real extract and alcohol content. A population of 43 samples obtained from the Spanish market and including different types of beer, was evaluated. For each technique, spectra were obtained in triplicate. In the case of NIR a 1 mm pathlength quartz flow cell was used, whereas attenuated total reflectance measurements were used in MIR. Cluster hierarchical analysis was employed to select calibration and validation data sets. The calibration set was composed of 15 samples, thus leaving 28 for validation. A critical evaluation of the prediction capability of multivariate methods established from the combination of NIR and MIR spectra was made. Partial least squares (PLS) and artificial neural networks (ANN) were evaluated for the treatment of data obtained in each individual technique and the combination of both. Different parameters of each methodology were optimized. A slightly better predictive performance was obtained for NIR-MIR combined spectra, and in all the cases ANN performs better than PLS, which may be interpreted from the existence of some non-linearity in the data. The root-mean-sqare-error of prediction (RMSEP) values obtained for the combined NIR-MIR spectra for the determination of real extract, original extract and ethanol were 0.076% w/w, 0.14% w/w and 0.091% v/v.  相似文献

A novel strategy for building and maintaining calibration models has been developed for use when the future boundaries of the sample set are unknown or likely to change. Such a strategy could have an impact on the economics and time required to obtain and maintain a calibration model for routine analysis. The strategy is based on both principal component analysis (PCA) and partial least squares (PLS) multivariate techniques. The principal action of the strategy is to define how “similar” a new sample is to the samples currently defining the calibration dataset. This step is performed by residuals analysis, following PCA. If the new sample is considered to have a spectrum “similar” to previously available spectra, then the model is assumed able to predict the analyte concentration. Conversely, if the new sample is considered “dissimilar”, then there is new information in this sample, which is unknown to the calibration model and the new sample is added automatically to the calibration set in order to improve the model. The strategy has been applied to a real industrial dataset provided by BP Amoco Chemicals. The data consists of spectra of 102 sequential samples of a raw material. The strategy produced an accurate calibration model for both target components starting with only the first four samples, and required a further 17 reference measurements to maintain the model for the whole sampling sequence, which was over a 1-year period.  相似文献

Two-dimensional correlation spectroscopy (2DCOS) and near-infrared spectroscopy (NIRS) were used to determine the polyphenol content in oat grain. A partial least squares (PLS) algorithm was used to perform the calibration. A total of 116 representative oat samples from four locations in China were prepared and the corresponding near-infrared spectra were measured. Two-dimensional correlation spectroscopy was employed to select wavelength bands for the PLS regression model for the polyphenol determination. The number of PLS components and intervals was optimized according to the coefficients of determination (R2) and root mean square error of cross validation (RMSECV) in the calibration set. The performance of the final model was evaluated using the correlation coefficient (R) and the root mean square error of validation (RMSEV) in the prediction set. The results showed the band corresponding to the optimal calibration model was between 1350 and 1848?nm and the optimal spectral preprocessing combination was second derivative with second smoothing. The optimal regression model was obtained with an R2 of 0.8954 and an RMSECV of 0.06651 in the calibration set and R of 0.9614 and RMSEV of 0.04573 in the prediction set. These measurements reveal the calibration model had qualified predictive accuracy. The results demonstrated that the 2DCOS with PLS was a simple and rapid method for the quantitative determination of polyphenols in oats.  相似文献

Photoacoustic spectroscopy (PAS) is based on the absorption of electromagnetic radiation by analyte molecules, and this technique has emerged as a valuable tool for the study of materials like biological, chemical and geological samples. In this paper, Fourier transform mid-infrared photoacoustic spectroscopy (FTIR-PAS) was used in the prediction of soil properties. Air-dried soil samples (n = 56) from Fengqiu Ecology Experimental Station Chinese Academy of Sciences were involved in this experiment, and FTIR-PAS spectra of these soil samples were recorded. These FTIR-PAS spectra indicated abundant soil information, but overlapping of absorption made it difficult to make direct measurement of soil properties. Partial least squares (PLS) models based on soil FTIR-PAS spectra was developed to predict available nitrogen (N), phosphorus (P), potassium (K) and organic matter content of soil. 42 soil samples were firstly used in leave-one-out cross-validation, and calibration error, calibration coefficient, validation error and ratio of standard deviation to prediction error (RPD) were obtained to optimize the PLS factor number; then based on the optimized PLS models the soil properties of the other 14 soil samples were predicted. The calibration statistics showed that the PLS model was suitable to use in the prediction of available N, P, K and organic matter content of soil. This prediction technique was non-destructive, and no sample pre-treatment was needed, which made FTIR-PAS a very promising method for fast evaluation of soil properties as well as soil quality.  相似文献

The aim of this study was to assess the feasibility of near infrared spectroscopy (NIRS) for analysis of acyclovir in plasma. This methodology was based on the direct measurement of the transmission spectra of liquid samples and a multivariate calibration model (partial least squares, PLS) to determine the acyclovir concentration in plasma sample. The PLS calibration set was built on using the spiked samples by mixing different amounts of acyclovir. Concentration of acyclovir in the plasma samples was calculated employing a 6-factors PLS calibration using the spectral information in the range of 6102-5450 cm− 1. The root mean square errors of prediction (RMSEP) found was 1.21 for acyclovir. The developed PLS-NIRS procedure allows the determination of 120 samples/h does not require any sample pretreatment and avoids waste generation.  相似文献

This paper proposes a new method to divide a pool of samples into calibration and validation subsets for multivariate modelling. The proposed method is of value for analytical applications involving complex matrices, in which the composition variability of real samples cannot be easily reproduced by optimized experimental designs. A stepwise procedure is employed to select samples according to their differences in both x (instrumental responses) and y (predicted parameter) spaces. The proposed technique is illustrated in a case study involving the prediction of three quality parameters (specific mass and distillation temperatures at which 10 and 90% of the sample has evaporated) of diesel by NIR spectrometry and PLS modelling. For comparison, PLS models are also constructed by full cross-validation, as well as by using the Kennard-Stone and random sampling methods for calibration and validation subset partitioning. The obtained models are compared in terms of prediction performance by employing an independent set of samples not used for calibration or validation. The results of F-tests at 95% confidence level reveal that the proposed technique may be an advantageous alternative to the other three strategies.  相似文献

In ordinary multivariate calibration methods, when the calibration set is determined to build the model describing the relationship between the dependent variables and the predictor variables, each sample in the calibration set makes the same contribution to the model, where the difference of representativeness between the samples is ignored. In this paper, by introducing the concept of weighted sampling into partial least squares (PLS), a new multivariate regression method, optimized sample-weighted PLS (OSWPLS) is proposed. OSWPLS differs from PLS in that it builds a new calibration set, where each sample in the original calibration set is weighted differently to account for its representativeness to improve the prediction ability of the algorithm. A recently suggested global optimization algorithm, particle swarm optimization (PSO) algorithm is used to search for the best sample weights to optimize the calibration of the original training set and the prediction of an independent validation set. The proposed method is applied to two real data sets and compared with the results of PLS, the most significant improvement is obtained for the meat data, where the root mean squared error of prediction (RMSEP) is reduced from 3.03 to 2.35. For the fuel data, OSWPLS can also perform slightly better or no worse than PLS for the prediction of the four analytes. The stability and efficiency of OSWPLS is also studied, the results demonstrate that the proposed method can obtain desirable results within moderate PSO cycles.  相似文献

This investigation describes a reliable and sensitive method for simultaneously determining bisphenol A (BPA) and two major phytoestrogens, daidzein and genistein, in powdered milks and infant formulas by gas chromatography-mass spectrometric analysis after trimethylsilylation. To reduce the matrix interference associated with the constituents of the formulas, the dissolved formula solutions were firstly ultra-centrifuged and the analytes in the supernatant were then extracted using a C18 solid-phase extraction column. The accuracy and precision of the method were determined and the technique was successfully employed to measure trace concentrations of BPA, daidzein and genistein in powdered formulas. The results show that BPA, daidzein and genistein were detected in all the testing samples (n = 6) at concentrations from 45 to 113 ng/g (except one infant formula), 20 to 2050 ng/g and 21 to 6510 ng/g, respectively. The highest concentrations of daidzein and genistein (i.e., 2050 and 6510 ng/g) were detected in a soy-based powdered infant formula. The quantitation limits were 1.0 ng/g for BPA, and 10 ng/g for daidzein and genistein using 0.5 g powdered milk samples.  相似文献

Near infrared spectroscopy (NIR) has been used to determine important indicators of the quality of beers, for example original and real extract and alcohol content, using a partial least squares (PLS) calibration approach. A population of 43 samples, obtained commercially in Spain and including different types of beer, was used. Cluster hierarchical analysis was used to select calibration and validation data sets. Absorbance sample spectra, in transmission mode, were obtained in triplicate by using a 1-mm pathlength quartz flow cell and glass chromatography vials of 6.5 mm internal diameter. The two methods of sample introduction were compared critically, on the basis of spectral reproducibility for triplicate measurements and after careful selection of the best spectral pre-processing and the spectral range for building the PLS model, to obtain the best predictive capability. For each mode of sample introduction two calibration sets were assayed, one based on the use of 15 samples and a second extended based on use of 30 samples, thus leaving 28 and 13 samples, respectively, for validation. The best results were obtained for 1 mm flow cell measurements. For this method original zero-order spectra data in the ranges 2220–2221 and 2250–2350 nm were chosen. For the real extract, original extract, and alcohol dx-y and sx-y values of –0.04 and 0.07% w/w, –0.01 and 0.13% w/w, and –0.01 and 0.1% v/v, respectively, were obtained. The maximum errors in the prediction of any of these three indicators for a new sample were 2.2, 1.2, and 1.9%, respectively. This method compares favorably with the automatic reference method in terms of speed, reagent consumed, and waste generated.  相似文献
