Transformations of 3-aryl-2-chloro-2-imidoylaziridines: novel entries to 4-chloro-2,5-diaryl-1H-imidazoles and 2-chloro-2-acylaziridines

The reactivity of stereochemically defined 3-aryl-2-chloro-2-imidoylaziridines, an unexplored class of substituted aziridines, was investigated under various reaction conditions. 2-Chloro-2-imidoylaziridines underwent a novel thermal rearrangement by reflux in acetonitrile via C-C bond cleavage to 4-chloro-2,5-diarylimidazoles in high yield. Alternatively, a novel efficient entry toward 2-aroyl-2-chloroaziridines was based on the chemoselective hydrolysis of 2-chloro-2-imidoylaziridines with hydrochloric acid in aqueous tetrahydrofuran.

1,1-Dichloro- and 1,3-dichloro-2-alkanones react with 2-methylfuran in the presence of lithium perchlorate/triethylamine to form 2-chloro-1-methyl-8-oxabicyclo [3.2.1]oct-6-ene-3-ones predominantly.     

Synthesis of 1-aryl-5-chloro-3-(phenylsulfonyl)pyrazol-4-ols from 1,1-dichloro-3-phenylsulfonyl-2-propanone

1,1-Dichloro-3-phenylsulfonyl-2-propanone was treated with arenediazonium chlorides to give 1-arylhydrazono-3,3-dichloro-1-phenylsulfonyl-2-propanones, which were cyclized to 1-aryl-5-chloro-3-(phenylsulfonyl)pyrazol-4-ols on treatmemt with base.

Organozinc compounds prepared from dialkyl dibromomalonates and zinc react with 2-arylmethyl-eneindan-4,6-diones, 5-arylmethylene-2,2-dimethyl-1,3-dioxane-4,6-diones, as well as with 2-[4-(1,3-dioxoindan-2-ylidenemethyl)phenyl]methyleneindan-1,3-dione and 5-[4-(2,2-dimethyl-4,6-dioxo-1,3-dioxane-2-ylidenemethyl)phenyl]methylene-2,2-dimethyl-1,3-dioxane-4,6-diones to form dialkyl 3-aryl-1′3′-dioxaspiro(cyclopropane-2,2′-indan)-1,1-dicarboxylates, dimethyl 3-aryl-6,6-dimethyl-5,7-dioxa-4,8-dioxaspiro[2,5]octan-2,2-dicarboxylates, dialkyl 2-{4-[3,3-bis (alkoxycarbonyl)-1′,3′-dioxaspiro(cyclopropane-2,2′-indan)-1-yl]phenyl}-1′,3′-dioxaspiro[cyclopropane-2,2′-indan]-1,1-dicarboxylates, and dialkyl 2-{4-[2,2-bis(alkoxycarbonyl)-6,6′-dimethyl-4,8-dioxo-5,7-dioxaspiro[2,5]oct-1-yl]phenyl}-6,6-dimethyl-4,8-dioxo-5,7-dioxaspiro[2,5]octan-1,1-dicarboxylate respectively.

Synthese neuer Aryl-(3,-dichloro-4-pyridazinly)-ketone und ihre Reaktion mit N,N-Dinucleophilen

Synthesis of New Aryl (3,6-Dichloro-4-pyridazinyl) Ketones and their Reaction with N,N-Dinucleophiles The synthesis of the new aryl (3,6-dichloro-4-pyridazinyl) ketones 3a–e vïa Friedel-Crafts acylation of the aromatic compounds 2a–e with 3,6-dichloro-4-pyridazincarbonyl chloride (1) is described. The ketones 3a–e cyclized with N,N-dinucleophilic reagents to the 3-aryl-5-chloro-l H -pyrazolo[3,4-c]pyridazines 4a–d , and 3a–c are converted into the 3-aryl-5-chloro-l H -pyrazolo[3,4-c]pyridazin - l -ethanols 5a–c and to the hitherto unknown ring system of the 5-aryl-3-chloro-7,8-dihydro-9 H -pyridazino[3,4-e][1,4]diazepines ( 6a-c ).     

Reaction of polyfluorinated cyclohexa-2,4-dienones with aryl nitrile oxides

Reaction of 1,3-dipolar cycloaddition of 6-chloropentafluorocyclohexa-2,4-dienone, 6-chloro-3-(pentafluorophenoxy)tetrafluorocyclohexa-2,4-dienone, and perfluoro-6-phenoxycyclohexa-2,4-dienone with aryl nitrile oxides proceeds highly stereoselectively at the c C=O group of the dienone providing in a high yield mixtures of diastereomeric fluorine-containing 3-aryl-1,4-dioxa-2-azaspiro[4,5]deca-2,6,8-trienes. The reaction of the latter with sodium pentafluorophenolate proceeds along the type of allyl substitution affording polyfluorinated 3-aryl-8-phenoxy-1,4-dioxa-2-azaspiro[4,5]deca-2,6,9-trienes.     

Synthesis of new imidazo[2,1-<Emphasis Type="Italic">b</Emphasis>][1,3]thiazole derivatives from 2-amino-4-(2,2-dichlorovinyl)-1,3-thiazole and <Emphasis Type="Italic">N</Emphasis>-(2,2-dichloro-2-phenylethylidene)arenesulfonamides

2-Amino-4-(2,2-dichlorovinyl)-1,3-thiazole reacted with highly electrophilic N-(2,2-dichloro-2-phenylethylidene)- and N-(2,2,2-trichloroethylidene)arenesulfonamides through the exocyclic amino group to give products of nucleophilic addition to the azomethine bond, N-[2,2-di(or 2,2,2-tri)chloro-1-(1,3-thiazol-2-ylamino)ethyl]arenesulfonamides in good yields. Intramolecular heterocyclization of the latter afforded N-[3-(2,2-dichloroethyl)-6-phenylimidazo[2,1-b][1,3]thiazol-5-yl]arenesulfonamides.     

Synthesis of 3,4-dihydro-2H-pyrans by hetero-Diels-Alder reactions of functionalized alpha,beta-unsaturated carbonyl compounds with N-vinyl-2-oxazolidinone

Cycloadditions of 3-aryl-2-benzoyl-2-propenenitriles and 3-phenylsulfonyl-3-buten-2-one to N-vinyl-2-oxazolidinone proceed regio- and diastereoselectively yielding cis and trans diastereoisomers of 4-aryl-3,4-dihydro-2-(2-oxo-3-oxazolidinyl)-2H-pyrans in 37-65% yield. Cycloadducts cis- were the major products. Reaction of 5-arylidene-1,3-dimethylbarbituric acids with dienophile afforded mixtures of 2H-pyrano[2,3-d]pyrimidine-2,4(3H)-diones trans and products resulted from an elimination of 2-oxazolidinone, in 50-52% yield. To confirm the experimental results, semiempirical AM1 and PM3 calculations of frontier orbital energies have been performed.     

Asymmetric hydrogen-transfer hydrogenation on rhodium(I) complexes with new optically active salen ligands derived from (4<Emphasis Type="Italic">S</Emphasis>,5<Emphasis Type="Italic">S</Emphasis>)-4,5-bis(aminomethyl)-2,2-dimethyl-1,3-dioxolane

New optically active C ₂-symmetric salen-type ligands were synthesized on the basis of (4S,5S)-4,5-bis(aminomethyl)-2,2-dimethyl-1,3-dioxolane. These ligands were used to obtain cationic (trifluoromethanesulfonate) and neutral (chloride) rhodium(I) complexes with [(4S,5S)-2,2-dimethyl-5-{[(E)-pyridin-2-ylmethylidene]aminomethyl}-1,3-dioxolan-4-yl]-N-[(E)-pyridin-2-ylmethylidene]methanamine and [2,2-dimethyl-5-{[(E)-quinolin-2-ylmethylidene]aminomethyl}-1,3-dioxolan-4-yl]-N-[(E)-quinolin-2-ylmethylidene] methanamine. The latter complex ensured preparation of (S)-2-phenylethanol with an optical yield of 34.8% by transfer hydrogenation of acetophenone.     

Reactions of bromine-containing organozinc compounds derived from α,α-dibromo ketones with 2-arylmethylideneindan-1,3-diones and 5-arylmethylidene-2,2-dimethyl-1,3-dioxane-4,6-diones

Bromine-containing organozinc compounds generated from 1,1-dibromo-3,3-dimethylbutan-2-one reacted with 2-arylmethylideneindan-1,3-diones and 5-arylmethylidene-2,2-dimethyl-1,3-dioxane-4,6-diones to give 3-aryl-2-(2,2-dimethylpropanoyl)spiro[cyclopropane-1,2′-indan]-1′,3′-diones and 1-aryl-6,6-dimethyl-2-(2,2-dimethylpropanoyl)-5,7-dioxaspiro[2.5]octan-4,8-diones, respectively. Reactions of 2-arylmethylideneindan-1,3-diones with bromine-containing zinc enolates derived from 1-aryl-2,2-dibromopropan-1-ones and 2,2-dibromo-1,2,3,4-tetrahydronaphthalen-1-one resulted in the formation of 2-aroyl-3-aryl-2-methylspiro-[cyclopropane-1,2′-indan]-1′,3′-diones and 2,3: 8,9-dibenzo-12-phenyldispiro[4.0.5.1]dodecane-1,4,7-trione, respectively.     

Synthesis of 5-Aryl-2,2-dimethyl-4-oxaspiro[5,5]undecane-1,3-diones by Reformatsky Reaction

Alkyl 3-(1-bromocyclohexyl)-2,2-dimethyl-3-oxopropanoates react with zinc and aromatic aldehydes to yield 5-aryl-2,2-dimethyl-4-oxaspiro[5,5]undecane-1,3-diones.     

Synthesis of some ω- (2,5-dichloro-3-thienyl)alkanoic acids and their intramolecular ring closure

The synthesis of 1-chloro-4,5,6,7-tetrahydrobenzo[c]-4-thiophenone and 1-chloro-5,6,7,8-tetrahydro-4H-cyclohepta[c]-4-thiophenone was accomplished by the cyclization of the acid chlorides of -(2,5-dichloro-3-thienyl)butyric acid and -(2,5-dichloro-3-thienyl)valeric acid, respectively, to 1,3-dichloro-4,5,6,7-tetrahydrobenzo[c]-4-thiophenone and 1,3-dichloro-5,6,7,8-tetrahydro-4H-cyclohepta[c]-4-thiophenone and by partial dehalogenation of the latter by heating with copper metal in propionic acid.Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 10, pp. 1358–1360, October, 1971.     

The reaction of 2-hetarylacetonitriles with heterocyclic haloaldehydes

The reaction of 4-oxo-3,4-dihydroquinazolyl-and benzimidazolylacetonitriles with 2-chloro-2-quinolinecarbaldehydes and 1-aryl-5-chloro-3-methyl-1H-pyrazole-4-carbaldehydes gave the corresponding 3-(2-chloro-3-quinolyl)-2-(4-oxo-3,4-dihydro-2-quinazolyl)-2-propenenitriles and 3-(1-aryl-5-chloro-3-methyl-1H-4-pyrazolyl)-2-hetaryl-2-propenenitriles. Intramolecular cyclization of these compounds gives 15-oxo-15H-benzo[6,7][1,8]naphthyridino[2,1-b]quinazoline-6-carbonitriles, 1-aryl-3-methyl-11-oxo-1,11-dihydropyrazolo[4′,3′:5,6]pyrido[2,1-b]quinazoline-5-carbonitriles, and 1-aryl-3-methyl-1H-benzo[4,5]imidazo[1,2-a]pyrazolo[4,3-e]pyridine-5-carbonitriles.     

Reaction of dihalocarbenes with 2-(benzylideneamino)pyridines. Cyclization of 2-(benzylideneamino)pyridinium dihalomethylids to give 2-aryl-3-haloimidazo[1,2-α]pyridines

Pyridinium dichloromethylids, formed in the reaction of dichlorocarbene with 2-(benzylideneamino)pyridines, undergo intramolecular 1,5-cyclization to give 2-aryl-3-chloroimidazo[1,2-a]pyridines, which undergo partial conversion to 2-aryl-3-chloro-4H-pyrido[1,2-a]pyrimidin-4-ones under the reaction conditions. 2-Aryl-3-bromoimidazo[1,2-a]pyridines and 2-[N-(1-aryl-2,2,2-tribromoethyl)amino]pyridines, respectively, are obtained in the reaction of dibromocarbene and the tribromomethyl onion generated in the reaction of bromoform with potassium hydroxide.Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 6, pp. 810–816, June, 1991.     

1,1,3-Trisubstituted cyclobutanes containing thiazole and thiourea fragments

The synthesis of 1-aryl-3-(2-chloro-1-hydroxyethyl)-1-methylcyclobutanes, 1-aryl-3-(2-chloro-1-oxoethyl)-1-methylcyclobutanes, 2-amino-4-(3-aryl-3-methylcyclobutyl)thiazoles, and N-[4-(3-aryl-3-methylcyclobutyl)thiazole-2-yl]-N′-arylthioureas is reported. Synthesis and Properties of 7-Acetyl-8-aryl-9-cyano-3-hydroxy-6-methyl-3,4-dihydro-2H,8H-pyrido[2,1-<Emphasis Type="Italic">b</Emphasis>][1,3]thiazines

7-Acetyl-8-aryl-2-(1-chloro-2-hydroxy-3-propyl)thio-9-cyano-6-methyl-1,4-dihydropyridines were obtained by treatment of 1,4-dihydropyridine-2(3H)-thiones with epichlorohydrin in the presence of sodium bicarbonate. When treated with NaOMe, these compounds are readily intramolecularly alkylated with formation of 7-acetyl-8-aryl-3-hydroxy-9-cyano-6-methyl-3,4-dihydro-2H,8H-pyrido[2,1-b]-[1,3]thiazines. Synthesis and mass spectral studies of 1-[5-chloro-1-substituted-2(1H)-pyrazin-2-on-3-yl]-5-aryl-3-methylpyrazoles

Sixteen new 1-[5-chloro-1-substituted-2(1H)-pyrazin-2-on-3-yl]-5-aryl-3-methylpyrazoles V have been synthesized by condensation of 5-chloro-1-substituted-3-hydrazino-2(1H)-pyrazin-2-ones III and 1-aryl-1,3-butanediones IV in dry 1,4-dioxane. The general mass spectral fragmentation mode of these compounds has been studied.
