We report on a facile immunoassay for porcine circovirus type 2 (PCV2) based on surface enhanced Raman scattering (SERS) using multi-branched gold nanoparticles (mb-AuNPs) as substrates. The mb-AuNPs in the immunosensor act as Raman reporters and were prepared via Tris base-induced reduction and subsequent reaction with p-mercaptobenzoic acid (pMBA). They possess good stability and high SERS activity. Subsequently, the modified mb-AuNPs were covalently conjugated to the monoclonal antibody (McAb) against the PCV2 cap protein to form SERS immuno nanoprobes. These were captured in a microtiterplate via a immunoreaction in the presence of target antigens. The effects of antibody concentration, reaction time and temperature on the sensitivity of the immunoassay were investigated. Under optimized assay conditions, the Raman signal intensity at 1,076 cm−1 increases logarithmically with the concentrations of PCV2 in the concentration ranging from 8 × 102 to 8 × 106 copies per mL. The limit of detection is 8 × 102 copies per mL. Compared to conventional detecting methods such as those based on PCR, the method presented here is rapid, facile and very sensitive.

We describe a novel surface-enhanced Raman scattering (SERS) tag that is based on Au/Ag core-shell nanostructures embedded with p-aminothiophenol. The Au/Ag core-shell sandwich nanostructures demonstrate bright and dark stripe structure and possess very strong SERS activity. Under optimum conditions, the maximum SERS signal was obtained with a 10?nm thick Ag nanoshell, and the enhancement factor is 3.4?×?104 at 1077?cm?1. After conjugation to the antibody of muramidase releasing protein (MRP), the Au/Ag core-shell nanostructures were successfully applied to an SERS-based detection scheme for MRP based on a sandwich type of immunoassay.

Silver nanoparticles (Ag NPs) modified with sodium 2-mercaptoethanesulfonate (mesna) exhibit strong surface-enhanced Raman scattering (SERS). Their specific and strong interaction with heavy metal ions led to a label-free assay for Hg(II). The covalent bond formed between mercury and sulfur is stronger than the one between silver and sulfur and thus prevents the adsorption of mesna on the surface of Ag NPs. This results in a decrease of the intensity of SERS in the presence of Hg(II) ions. The Raman peak at 795?cm?1 can be used for quantification. The effect of the concentration of mesna, the concentration of sodium chloride, incubation time and pH value on SERS were optimized. Under the optimal conditions, the intensity of SERS decreases with increasing concentration of Hg(II). The decrease is linear in the 0.01 and 2?μmol L?1 concentration range, with a correlation coefficient (R2) of 0.996 and detection limit (S/N?=?3) is 0.0024?μmol L?1. The method was successfully applied to the determination of the Hg(II) in spiked water samples.

A glassy carbon electrode (GCE) was modified with pyrocatechol violet (PCV) that was electrodeposited on single walled carbon nanotubes (SWCNTs) via continuous cycling between 0 and 0.9 V (vs. SCE). The resulting electrode exhibits excellent electrocatalytic activity towards the oxidation of hydrazine at 0.3 V. The apparent surface coverage of the electrode is at least 24 times higher (2.7?×?10?10 mol cm?2) than that obtained with a bare GCE (1.1?×?10?11 mol cm?2). This is attributed to a remarkably strong synergistic effect between the acid-pretreated SWCNTs and the electrodeposited PCV coating. Response is fast (2 s) and sensitive (281 mA M?1 cm?2). Other features include a wide linear range (150 nM to 0.4 mM) and a low detection limit (150 nM at an SNR of 3). The sensor has been successfully applied to the determination of hydrazine in water and cigarette samples with good accuracy and precision. In addition, the morphology and the wetting properties of the coating were studied by scanning electromicroscopy and contact angle measurements.

We have synthesized silver nanoparticles (AgNPs) decorated with α-cyclodextrin (CD) by using the traditional silver mirror reaction in the presence of CD. The CD-AgNPs were used as substrate in surface-enhanced Raman spectroscopy (SERS) for determining melamine. The intensity of the Raman band of melamine at 704 cm?1 was used to determine melamine in milk and milk powder. The use of CD-AgNPs as the SERS substrate rather than classical silver nanoparticles makes the method more sensitive in giving an enhancement by a factor of up to?~?106 in scattering efficiency. The effects of the volume of solutions (of CD-AgNPs, NaCl, NaOH, melamine) and of mixing time were optimized. The standard addition method was employed for quantitative analysis. The correlation coefficient of the calibration plot is 0.9995, and the limit of detection is 3.0 μg L?1. The method was successfully applied to the determination of melamine in milk and milk powder, with relative standard deviations of <10 % and recoveries between 89 and 104 %.

A biosensor for hydrogen peroxide was constructed by immobilizing horseradish peroxidase on chitosan-wrapped NiFe2O4 nanoparticles on a glassy carbon electrode (GCE). The electron mediator carboxyferrocene was also immobilized on the surface of the GCE. UV?Cvis spectra, Fourier transform IR spectra, scanning electron microscopy, and electrochemical impedance spectra were acquired to characterize the biosensor. The experimental conditions were studied and optimized. The biosensor responds linearly to H2O2 in the range from 1.0?×?10?5 to 2.0?×?10?3?M and with a detection limit of 2.0?×?10?6?M (at S/N?=?3).

A carbon paste electrode (CPE) was modified with multi-wall carbon nanotubes and successfully applied to the determination of silver ion by differential pulse anodic stripping voltammetry. Compared to a conventional CPE, a remarkably improved peak current response and sensitivity is observed. The analytical procedure consisted of an open circuit accumulation step for 2?min in ?0.4?V, this followed by an anodic potential scan between +0.2 and?+?0.6?V to obtain the voltammetric peak. The oxidation peak current is proportional to the concentration of silver ion in the range from 1.0?×?10?8 to 1.0?×?10?5?mol?L?1, with a detection limit of 1.8?×?10?9?mol?L?1 after an accumulation time of 120?s. The relative standard deviation for 7 successive determinations of Ag(I) at 0.1???M concentration is 1.99%. The procedure was validated by determining Ag(I) in natural waters.

Microextraction by packed sorbents (MEPS) combined with Surface-enhanced Raman spectroscopy (SERS) was investigated, and applied to the determination of musk ketone (MK) in river water samples. The full MEPS–SERS method includes analyte enrichment by MEPS preconcentration with C18 sorbent followed by SERS detection supported by silver nanoparticles. An eluent drop containing the analyte is deposited directly from the MEPS syringe on a CaF2 glass plate. When the drop has dried, a specific volume of silver nanoparticles solution is added on it before each SERS measurement. Several experimental variables were studied in depth; under the optimum experimental conditions MK can be extracted from a 500 μL sample with recoveries in the range 47–63 %. The limit of detection was 0.02 mg L?1 and the relative standard deviation 15.2 % (n?=?4). Although not investigated in this work, the proposed method might be suitable for in-situ monitoring, because of the portability of the Raman spectrometer used.

We have developed a surface-enhanced Raman scattering (SERS) probe for the determination of mercury(II) using methimazole-functionalized and cyclodextrin-coated silver nanoparticles (AgNPs). These AgNPs in pH 10 solution containing sodium chloride exhibit strong SERS at 502 cm?1. Its intensity strongly decreases in the presence of Hg(II). This effect serves as the basis for a new method for the rapid, fast and selective determination of trace Hg(II). The analytical range is from 0.50 μg L?1 to 150 μg L?1, and the limit of detection is 0.10 μg L?1. The influence of 11 metal ions commonly encountered in environmental water samples was found to be quite small. The method was applied to the determination of Hg(II) in spiked water samples and gave recoveries ranging from 98.5 to 105.2 % and with relative standard deviations of <3.5 % (n?=?5). The total analysis time is <10 min for a single sample.

A novel kind of carbon paste electrode (CPE) was prepared by mixing graphite powder, liquid paraffin and the ionic liquid 1-ethyl-3-methylimidazolium ethylsulfate. The resulting electrode was used for the simultaneous determination of guanosine and adenosine by differential pulse voltammetry. Compared to a conventional CPE, the oxidation peak currents are largely increased, and the oxidation peak potentials are negatively shifted. The electrochemical responses to guanosine and adenosine were investigated. Under optimized conditions, the calibration curves are linear in the concentration range from 1.0?×?10-6?mol?L-1 to 1.6?×?10-4?mol?L-1 for guanosine, and from 1.0?×?10-6?mol?L-1 to 2.7?×?10-4?mol?L-1 for adenosine at pH 3.5. Substances potentially interfering in the biological matrix do no interfere. The method was successfully applied to detect adenosine and guanosine in human urine without sample treatments.

A novel enzyme-free electrochemical sensor for H2O2 was fabricated by modifying an indium tin oxide (ITO) support with (3-aminopropyl) trimethoxysilane to yield an interface for the assembly of colloidal gold. Gold nanoparticles (AuNPs) were then immobilized on the substrate via self-assembly. Atomic force microscopy showed the presence of a monolayer of well-dispersed AuNPs with an average size of ~4 nm. The electrochemical behavior of the resultant AuNP/ITO-modified electrode and its response to hydrogen peroxide were studied by cyclic voltammetry. This non-enzymatic and mediator-free electrode exhibits a linear response in the range from 3.0?×?10?5 M to 1.0?×?10?3 M (M?=?mol?·?L?1) with a correlation coefficient of 0.999. The limit of detection is as low as 10 nM (for S/N?=?3). The sensor is stable, gives well reproducible results, and is deemed to represent a promising tool for electrochemical sensing.

In this research, a mixed immunoassay design for multiple chemical residues detection based on combined reverse competitive enzyme-linked immunosorbent assay (ELISA) procedure was developed. This method integrated two reverse ELISA reactions in one assay by labeling horseradish peroxidase to deoxynivalenol (DON) and orbifloxacin. Within this method, IC50 of the two mAbs for each analyte we produced ranged from 23?~?68 ng?mL?1 for DONs and 4.1?~?49 ng?mL?1 for quinolones (QNs). The limit of detection measured by IC10 was achieved at 0.45–1.3 ng?mL?1 for DONs and 0.59–6.9 ng?mL?1 for QNs, which was lower than the maximum residue levels. Recoveries in negative samples spiked at concentrations of 100, 200, and 500 ng?mL?1 ranged from 91.3 to 102.2 % for DONs and 88.7–98.05 % for QNs with relative standard deviation less than 9.88 and 12.67 %. The results demonstrated that this developed immunoassay was suitable for screening of low molecular weight contaminants.

SiO2 nanosheets (SNS) have been prepared by a chemical method using montmorillonite as raw material and were characterized by scanning electron microscopy and X-ray diffraction. SiO2 nanosheet–Nafion nanocomposites with excellent conductivity, catalytic activity, and biocompatibility provided an extremely hydrophilic surface for biomolecule adhesion. Chitosan was used as a cross-linker to immobilize acetylcholinesterase (AChE), and Nafion was used as a protective membrane to efficiently improve the stability of the AChE biosensor. The AChE biosensor showed favorable affinity for acetylthiocholine chloride and catalyzed the hydrolysis of acetylthiocholine chloride with an apparent Michaelis–Menten constant of 134 μM to form thiocholine, which was then oxidized to produce a detectable and fast response. Based on the inhibition by pesticides of the enzymatic activity of AChE, detection of the amperometric response from thiocholine on the biosensor is a simple and effective way to biomonitor exposure to pesticides. Under optimum conditions, the biosensor detected methyl parathion, chlorpyrifos, and carbofuran at concentrations ranging from 1.0?×?10?12 to 1?×?10?10?M and from 1.0?×?10?10 to 1?×?10?8?M. The detection limits for methyl parathion, chlorpyrifos, and carbofuran were 5?×?10?13?M. The biosensor developed exhibited good sensitivity, stability, reproducibility, and low cost, thus providing a new promising tool for analysis of enzyme inhibitors.

A simple and sensitive method, based on surface-enhanced Raman scattering (SERS), for immunoassay and label-free protein detection is reported. A series of bowl-shaped silver cavity arrays were fabricated by electrodeposition using a self-assembled polystyrene spheres template. The reflection spectra of these cavity arrays were recorded as a function of film thickness, and then correlated with SERS enhancement using sodium thiophenolate as the probe molecule. The results reveal that SERS enhancement can be maximized when the frequency of both the incident laser and the Raman scattering approach the frequency of the localized surface plasmon resonance. The optimized array was then used as the bottom layer of a silver nanoparticle–protein–bowl-shaped silver cavity array sandwich. The second layer of silver was introduced by the interactions between the proteins in the middle layer of the sandwich architecture and silver nanoparticles. Human IgG bound to the surface of this microcavity array can retain its recognition function. With the Raman reporter molecules labeled on the antibody, a detection limit down to 0.1 ng mL?1 for human IgG is easily achieved. Furthermore, the SERS spectra of label-free proteins (catalase, cytochrome C, avidin and lysozyme) from the assembled sandwich have excellent reproducibility and high quality. The results reveal that the proposed approach has potential for use in qualitative and quantitative detection of biomolecules.

We describe the synthesis of ß-cyclodextrin modified magnetic nanoparticles (CD-mNPs) as a material for solid-phase extraction of the cancer biomarker 5-hydroxy-indole-3-acetic acid (5-HIAA) from urine. The CD-mNPs were characterized by TEM, FTIR, and XRD, and the kinetics and adsorption isotherms were studied. The strong interaction between the CD-mNPs and 5-HIAA is the main driving force for recognition and extraction, while the magnetic core of the NPs allows their separation from the sample matrix. Recovery of 5-HIAA from the adsorbent using an adequate solvent regenerated the adsorbent for further use. 5-HIAA was then quantified by fluorometry of its complex with ß-CD. The method works in the 1?×?10?7 to 1?×?10?5 mol L?1 (R2 0.9982–0.9996) concentration range, and the limits of detection (3σ) and quantification (10 σ) of the method are 1.2?×?10?8 mol L?1 and 4.01?×?10?8 mol L?1 5-HIAA, respectively. The recovery of 5-HIAA from urine samples spiked with 5-HIAA in three concentrations (1.4?×?10?6, 4.50?×?10?6 and 1.0?×?10?5 mol L?1) are within 63?±?3 %.

We report on a simple and reliable method for the determination of trace cadmium ion using a glassy carbon electrode (GCE) modified with cupferron, ß-naphthol and MWCNTs. The operational mechanism consists of several steps: first, the ligand cupferron on the modified electrode reacts with Cd2+ ion to form a chelate compound. Next, this chelate is adsorbed by the carrier ß-naphthol following the principle of organic co-precipitation. Finally, the coprecipitated complex is detected by the GCE. This scheme is interesting because it combines preconcentration and electrochemical detection. Two linear responses are obtained, one in the concentration range of 5.0?×?10?11 to 1.6?×?10?8 M, the other in the range of 1.6?×?10?8 to 1.42?×?10?6 M, with a lower detection limit of 1.6?×?10?11 M. This modified GCE does not suffer from significant interferences by Cu(II), Hg(II), Ag(I), Fe(III), Pb(II), Cr(III), Zn(II), NO3?, Cl?, SO 4 2? ions and EDTA. The response of the electrode remained constant for at least 3 weeks of successive operation. The method presented here provides a new way for the simultaneous separation, enrichment, and electrochemical detection of trace cadmium ion.

We report on the detection of trace quantities of melamine (MA) by a colorimetric method that exploits the conformational change of hemin G-quadruplex-DNAzyme. The addition of MA to hemin G-quadruplex-DNAzyme structure containing thymine bases causes the thymine in the DNAzyme to interact with MA via a stable triple H-bond and leads to a conformational change. This, in turn, affects the peroxidase-like activity of hemin which is determined colorimetrically at 450 nm by adding 3,3’,5,5’-tetramethylbenzidine and hydrogen peroxide. The method was applied to the colorimetric determination of MA over a wide range of concentrations (0.2 to 24 μM) with a detection limit of 80 nM. The effect also can be detected with bare eyes. The method was successfully applied to the determination of MA in spiked milk powder.

Deoxyribonucleic acid (DNA) was electrochemically deposited on a carbon ionic liquid electrode to give a biosensor with excellent redox activity towards paraquat as shown by cyclic voltammetry and differential pulse voltammetry. Experimental conditions were optimized with respect to sensing paraquat by varying the electrochemical parameters, solution pH, and accumulation time of DNA. Under the optimized conditions, a linear relation exists between the reduction peak current and the concentration of paraquat in the range from 5?×?10?8 mol L?1 to 7?×?10?5 mol L?1, with a detection limit of 3.6?×?10?9 mol L?1. The utility of the method is illustrated by successful analysis of paraquat in spiked real water samples.

A novel glucose biosensor is presented as that based on a glassy carbon electrode modified with hollow gold nanoparticles (HGNs) and glucose oxidase. The sensor exhibits a better differential pulse voltammetric response towards glucose than the one based on conventional gold nanoparticles of the same size. This is attributed to the good biological conductivity and biocompatibility of HGNs. Under the optimal conditions, the sensor displays a linear range from 2.0?×?10?6 to 4.6?×?10?5?M of glucose, with a detection limit of 1.6?×?10?6?M (S/N?=?3). Good reproducibility, stability and no interference make this biosensor applicable to the determination of glucose in samples such as sports drinks.

We report on a fluorometric method for the determination of the fluoroquinolones levofloxacin (LEV) and moxifloxacin (MOXI). It is based on the Tb(III)-sensitized luminescence that is plasmonically enhanced by silver nanoparticles (Ag NPs). The emission of the Tb(III) complexes has maximum at 545?nm after excitation at 284?nm and is strongly enhanced in the presence of the colloidal Ag NPs. Under optimum experimental conditions, luminescence intensity increases linearly with the concentration in the range from 4.16?×?10-17-3.59?×?10-15?M of LEV, and from 4.98?×?10-17-2.49?×?10-15?M for MOXI with correlation coefficients of 0.9996 and 0.9996, respectively. The limits of detection are 7.19?×?10-18?M and 8.47?×?10-18?M, respectively, and the relative standard deviations are 1.3 and 1.5% for 5 replicate measurements at 6.08?×?10-14?M of LEV and 5.48?×?10-14?M of MOXI. The method was successfully applied to the determination of LEV and MOXI in pharmaceutical samples, in urine and in serum.
