The present study was performed to estimate the concentration of genistein and daidzein in ethanol extract of tubers of Pueraria tuberosa (Indian kudzu or Vidarikanda) and its various fractions (n-hexane, ethyl acetate, n-butanol, and aqueous) by high-performance thin-layer chromatography (HPTLC). The separation of bioactive compounds was performed using mobile phase, toluene:ethyl acetate:acetone:formic acid (20.0:4.0:2.0:1.0) and detected at wavelength 269?nm. The method was validated for linearity, accuracy, precision, limit of detection (LOD), limit of quantification (LOQ), etc. by International Conference on Harmonization guidelines. The calibration range was found to be 100–600?ng/band for both the bioactive compounds. Daidzein was separated with an Rf value of 0.39?±?0.02 and genistein with an Rf value of 0.54?±?0.02. Average recovery was 99.96 and 99.90% for genistein and daidzein, respectively. The LOD and LOQ were 14.786 and 44.805?ng, respectively, for genistein, and 9.607 and 29.114?ng, respectively, for daidzein. Both the phytoconstituents were found in ethanol extract and its ethyl acetate fraction only. The developed HPTLC method was simple, precise, robust, specific, rapid, and cost effective and could be used for quality control analysis and quantification of genistein and daidzein in different herbal formulations containing the plant species.  相似文献

Zusammenfassung Mittels Differentialkalorimetrie und Messung der Keimdichte an Mikrotomschnitten wird eine starke Abhängigkeit der keimbildenden Wirkung von p-tert-Butylbenzoesäure in isotaktischem Polypropylen von der Mischmethode festgestellt. Der Keimbildner wird in das Polypropylen einerseits in einer gemeinsamen Xylol-Lösung und andererseits durch eine mechanische Mischung der Pulver eingebracht. Bei der Beimischung über die Lösung zeigt der Keimbildner eine bedeutend geringere Wirkung als in mechanisch gemischten Proben. Zur Erklärung der Meßergebnisse ist es notwendig, eine Mindestkorngröße für aktive Keimbildner-Körner anzunehmen. Die Abhängigkeit der keimbildenden Wirkung von der Mischmethode, der Konzentration des Keimbildners und der Kristallisationstemperatur wird diskutiert.
Summary A strong dependence of the nucleating effectiveness of p-tert-butylbenzoic acid in isotactic polypropylene on the mixing method is found by differential scanning calorimetry and by measuring the nucleation density by means of polarization microscopy on microtome sections. The nucleating agent is incorporated into the polymer by dissolving polymer and nucleator in Xylene or by mechanical mixing of the powder. The nucleator is much more effective if incorporated by mechanical mixing than by means of a solution. To explain the results it is necessary to assume a minimum size for active grains of the nucleator. The dependence of the nucleating ability on the mixing method, the concentration of the nucleator and on the crystallization temperature is discussed.

Absolute Configuration of α-Doradexanthin and of Fritschiellaxanthin, a New Carotenoid from Fritschiella tuberosa IYENG . Fritschiellaxanthin, a new oxocarotenoid produced by the green alga Fritschiella tuberosa, in a nitrogen-deficient medium is now shown to be (3 S, 3′ R, 6′ R)-3, 3′-dihydroxy-β, ?-caroten-4-one ( 4b ). It is not identical with α-Doradexanthin ( 5b ) previously found in goldfish (Carassius auratus) and to which we assign the (3 S, 3′ S, 6′ R)-chirality. Consequently, fritschiellaxanthin and α-Doradexanthin are C(3′)-epimers of lutein-4-on. Furthermore, the so-called ′lutein′ from goldfish has now been found to be identical with 3′-epilutein (3) . Therefore, fritschiellaxanthin is probably biogenetically derived from lutein (2) , whereas α-Doradexanthin is formed from 3′-epilutein (3) with 3, O-didehydrolutein (=(3R, 6′R)-3-hydroxy-β, ?-caroten-3′-one 10) as a precursor. For comparison, optically active 10 and 3 have been prepared from lutein (2) and are fully characterised.  相似文献
