In this study, the simultaneous determination of paracetamol, ibuprofen and caffeine in pharmaceuticals by chemometric approaches using UV spectrophotometry has been reported as a simple alternative to using separate models for each component. Spectra of paracetamol, ibuprofen and caffeine were recorded at several concentrations within their linear ranges and were used to compute the calibration mixture between wavelengths 200 and 400 nm at an interval of 1 nm in methanol:0.1 HCl (3:1). Partial least squares regression (PLS), genetic algorithm coupled with PLS (GA-PLS), and principal component-artificial neural network (PC-ANN) were used for chemometric analysis of data and the parameters of the chemometric procedures were optimized. The analytical performances of these chemometric methods were characterized by relative prediction errors and recoveries (%) and were compared with each other. The GA-PLS shows superiority over other applied multivariate methods due to the wavelength selection in PLS calibration using a genetic algorithm without loss of prediction capacity. Although the components show an important degree of spectral overlap, they have been determined simultaneously and rapidly requiring no separation step. These three methods were successfully applied to pharmaceutical formulation, capsule, with no interference from excipients as indicated by the recovery study results. The proposed methods are simple and rapid and can be easily used in the quality control of drugs as alternative analysis tools.  相似文献

The wavelength range and number of factors used in partial least-squares (PLS) calibration for the resolution of the dihydralazine (DHZ)-hydrochlorothiazide (HCT) binary mixture and the dihydralazine-hydrochlorothiazide-reserpine ternary mixture were optimized in terms of the relative standard error (R.S.E.) and relative mean standard error (R.M.S.E.). Under the optimum conditions thus established, synthetic mixtures of the analytes can be resolved with errors and relative standard deviations (R.S.D) less than 4.5 and 1.0%, respectively. The ensuing method, which was validated by comparison with high performance liquid chromatography (HPLC), also gives good results with real samples (pharmaceutical preparations).  相似文献

A ratio-spectra zero-crossing first-derivative spectrophotometric method and 2 chemometric methods have been used for the simultaneous determination of ternary mixtures of caffeine (A), 8-chlorotheophylline (B), and chlorphenoxamine hydrochloride (C) in bulk powder and dosage forms. In the ratio-spectra zero-crossing first-derivative spectrophotometric technique (1DD), calibration curves were linear in the range of 4-20 microg/mL for A, B, and C (r = 0.9992, 0.9994, and 0.9976, respectively). The measurements were carried out at 212, 209.2, and 231.4 nm for A, B, and C, respectively. The detection limits for A, B, and C were calculated to be 0.24, 0.34, and 0.13 microg/mL, and the percentage recoveries were 99.1 +/- 0.89, 100.1 +/- 0.95, and 100.1 +/- 1.0, respectively. Two chemometric methods, namely, the partial least-squares (PLS) model and the principal component regression (PCR) model, were also used for the simultaneous determination of the 3 drugs in the ternary mixture. A training set consisting of 15 mixtures containing different ratios of A, B, and C was used. The concentration used for the construction of the PLS and PCR models varied between 4 and 25 microg/mL for each drug. These models were used after their validation for the prediction of the concentrations of A, B, and C in mixtures. The detection limits for A, B, and C were calculated to be 0.13, 0.15, and 0.14 microg/mL, respectively, and the percent recoveries were found to be 99.8 micro 0.96, 99.9 micro 0.94, and 99.9 micro 1.18, respectively, for both methods. The 3 proposed procedures are rapid, simple, sensitive, and accurate. No preliminary separation steps or resolution equations are required; thus, they can be applied to the simultaneous determination of the 3 drugs in commercial tablets and suppositories or in quality-control laboratories.  相似文献

A mixture of piracetam and vincamine was determined by 3 different methods. The first was the determination of piracetam and vincamine using the ratio-spectra first-derivative (DD1) spectrophotometric technique at 209 and 293 nm in concentration ranges of 10-45 and 2-14 microg/mL with mean recoveries of 99.22 +/- 0.72 and 99.67 +/- 0.79%, respectively. The second method was based on the resolution of the 2 components by bivariate calibration depending on a mathematic algorithm that provides simplicity and rapidity. The method depended on quantitative evaluation of the absorbencies at 210 and 225 nm in concentration ranges of 5-45 and 2-14 microg/mL, with mean recoveries of 100.33 +/- 0.54 and 100.44 +/- 0.98% for piracetam and vincamine, respectively. The third method was reversed-phase liquid chromatography using 0.05 M potassium dihydrogen phosphate-methanol (50 + 50, v/v) as the mobile phase, with the pH adjusted to 3.5 with phosphoric acid. The eluent was monitored at 215 nm in concentration ranges of 5-100 and 2-200 microg/mL, with mean recoveries of 99.62 +/- 0.67 and 99.32 +/- 0.85% for piracetam and vincamine, respectively. The suggested procedures were checked using laboratory-prepared mixtures and were successfully applied for the analysis of their pharmaceutical preparation. The methods retained their accuracy and precision when applying the standard addition technique. The results obtained by applying the proposed methods were statistically analyzed and compared with those obtained by the manufacturer's method.  相似文献

A spectrofluorimetric method for the simultaneous determination of dysprosium, europium, gadolinium and terbium in ternary and quaternary mixtures by the use of pyridine-2,6-dicarboxylic acid as a chelating agent was developed. The influence of chemical variables affecting the analytical reaction was evaluated. A partial least-squares procedure and PC Quant software were used to assess data obtained from a variable number of calibration solutions and wavelengths. The ensuing method was validated by applying it to the analysis of synthetic ternary (Eu-Dy-Tb) and quaternary mixtures (Eu-Dy-Gd-Tb) over the concentration ranges 60-550 mug Eu l(-1), 30-400 mug Dy l(-1) and 30-400 mug Tb l(-1) in the former, and 20-220 mug Eu l(-1), 20-235 mug Dy l(-1), 25-230 mug Gd l(-1) and 75-230 mug Tb l(-1) in the latter. The results obtained by using the two quantitation procedures are compared. The relative errors in the determinations were less than 8% in most cases.  相似文献

A binary mixture of hyoscine butylbromide and ketoprofen was determined by 4 different methods. The first involved determination of hyoscine butylbromide and ketoprofen using the ratio-spectra first-derivative spectrophotometric technique at 211 and 234 nm over the concentration ranges of 2-14 and 5-45 microg/mL with mean accuracies 99.84 +/-0.92 and 99.98+/- 0.64%, respectively. The second method utilized second-derivative spectrophotometry over the concentration ranges of 2-14 and 5-35 microg/mL with mean accuracies 99.32+/- 1.06 and 99.55+/-1.15%, respectively. The third method was based on the resolution of the 2 components by bivariate calibration depending on a simple and rapid mathematical algorithm and quantitative evaluation of the absorbances at 206 and 254 nm over concentration ranges of 2-16 and 5-35 microg/mL; mean accuracies of 100.21+/-1.30 and 100.19 +/-1.07% were obtained for hyoscine butylbromide and ketoprofen, respectively. The fourth method was reversed-phase liquid chromatography using 0.05 M ammonium dihydrogen phosphate-acetonitrile-methanol (20 + 30 + 6, v/v) as the mobile phase with ultraviolet detection at 220 nm over concentration ranges of 1-90 and 5-70 microg/mL; mean accuracies were 99.92+/-1.02 and 99.61+/- 0.98%, respectively. The suggested procedures were checked using laboratory-prepared mixtures and were successfully applied for the analysis of pharmaceutical preparations. The methods retained their accuracy and precision when the standard addition technique was applied. An automatic direct spectrophotometric method for the simultaneous determination of nitrite and nitrate by flow-injection analysis has been developed. Nitrite reacts with 3-nitroaniline in the presence of hydrochloric acid (0.96-1.8 M HCl or pH 0.5-0.7) to form a diazonium cation, which is subsequently coupled with N-(1-naphthyl)-ethylenediamine dihydrochloride to form a stable purple azo dye, the absorbance of which is measured at 535 nm. Nitrate is reduced on-line to nitrite in a copper-coated cadmium column which is then treated with azo dye reagent and the absorbance due to the sum of nitrite and nitrate is measured; nitrate is determined from the difference in absorbance values. A copper column incorporated into the reaction manifold before the copperised cadmium column not only improves the long-term accuracy, but also extends the life time of the copperised cadmium column. Various analytical parameters, such as effect of acidity (pH), flow rate, sample size, dispersion coefficient, time, temperature, reagent concentration and interfering species, were studied. The calibration graphs were rectilinear for 0.1-3.5 mug ml(-1) of NO(3) and 10 ng ml(-1)-2.2mug ml(-1) of NO(2). The method is successfully applied to some food samples (meat, flour and cheese), environmental waters (inland and surface), beer and soil samples. Up to 30 samples can be analysed per hour with a relative precision of approximately 0.1-2%.  相似文献

A cloud point extraction (CPE) process using the nonionic surfactant Triton X-114 to simultaneous extraction and spectrophotometric determination of uranium and zirconium from aqueous solution using partial least squares (PLS) regression is investigated. The method is based on the complexation reaction of these cations with Alizarin Red S (ARS) and subsequent micelle-mediated extraction of products. The chemical parameters affecting the separation phase and detection process were studied and optimized. Under the optimum experimental conditions (i.e. pH 5.2, Triton X-114?=?0.20%, equilibrium time 10?min and cloud point 45?°C), calibration graphs were linear in the range of 0.01?C3?mg?L?1 with detection limits of 2.0 and 0.80???g L?1 for U and Zr, respectively. The experimental calibration set was composed of 16 sample solutions using an orthogonal design for two component mixtures. The root mean square error of predictions (RMSEPs) for U and Zr were 0.0907 and 0.1117, respectively. The interference effect of some anions and cations was also tested. The method was applied to the simultaneous determination of U and Zr in water samples.  相似文献

A kinetic spectrophotometric method for the simultaneous determination of iodate and periodate in mixtures was proposed. The method is established on the different kinetic behaviours of the analytes which react with starch–iodide in the presence of sodium chloride in sulfuric acid medium. The kinetic data were collected from 260 to 900 nm every 10 nm, within a time range of 0–180 s at 1 s interval, and the absorbance collected at 291, 354 and 585 nm, respectively, increased linearly with the concentration between 0.1–1.2 mg L− 1 for both iodate and periodate. The mechanism investigation revealed that the iodate/periodate–iodide–starch system is a consecutive reaction. Subsequently, the mathematical model for the quantitative kinetic determination based on the consecutive reactions by utilizing chemometric methods was deduced, and the simultaneous determination of synthetic mixtures of iodate and periodate was then applied. Kinetic data collected at 291, 354 and 585 nm, were processed by chemometric methods, such as classical least square (CLS), principal component regression (PCR), partial least square (PLS), back-propagation artificial neural network (BP-ANN), radial basis function–artificial neural network (RBF-ANN) and principle component–radial basis function–artificial neural network (PC-RBF-ANN). The results showed that calibration model with the data collected at 354 nm had some advantages for the prediction of the analytes as compared with the ones of other two wavelengths, and the PLS and PC-RBF-ANN gave the lower prediction errors than other chemometric methods. The proposed method was applied to the simultaneous determination of iodate and periodate in several real samples; and the standard addition method yielded satisfactory recoveries in all instances.  相似文献

A rapid and simple sample preparation method was developed for simultaneous determination of three triazine herbicides in honey samples. The selected herbicides were extracted from honey samples by ionic liquid dispersive liquid–liquid microextraction, separated on a C18 column (250 mm × 4.6 mm id, 5 μm) using acetonitrile and H2O as the mobile phase with gradient elution, and then detected by high‐performance liquid chromatography. The parameters, such as the type and volume of the extraction and disperser solvent, ion strength, pH, extraction time, and centrifuge time were optimized in order to provide the excellent extraction performance. Good linearity was showed for all the target herbicides over the tested concentration range with correlation coefficient higher than 0.994. Three spiked levels (0.005, 0.05, 0.10 mg/kg) were applied for determination of the recoveries of the targets in honey samples in the range of 80–103% with relative standard deviations not larger than 10.6%. The limits of quantification for the analytes ranged between 1.5 and 4.0 μg/kg. The developed method was applied for determination of the target compounds residues in real samples.  相似文献

The multivariate calibration methods, partial least squares (PLS) and principle component regression (PCR) have been used to determine phenanthridine, phenanthridinone and phenanthridine N-oxide in spiked human plasma samples. Resolution of binary and ternary mixtures of analytes with minimum sample pre-treatment and without analyte separation has been successfully achieved analyzing the UV spectral data. The net analyte signal (NAS) concept was also used to calculate multivariate analytical figures of merit such as limit of detection, selectivity and sensitivity. The simultaneous determination of three analytes was possible by PLS and PCR processing of sample absorbance in the 210–355 nm region. Good recoveries were obtained for both synthetic mixtures and spiked human plasma samples.  相似文献

The flow injection principle is used in the photometric determination of nitrite and nitrate with sulfanilamide and N-(1-naphthyl)ethylenediamine as reagents. An on-line copper-coated cadmium reductor reduces nitrate to nitrite. The detection limit is 0.05 μM for nitrite and 0.1 mM for nitrate at a total sample volume of 200 μM. Two rapid, accurate and reproducible methods are reported for the simultaneous spectrophotometric determination of tetravalent and hexavalent selenium in the presence of each other. Both procedures involve the sequential complexation and solvent extraction of the selenium(IV)-diethyldithiocarbamate complex by chloroform in the presence of perchlorate ions followed by reduction of hexavalent selenium in the remaining aqueous phase to tetravalent selenium by 25% hydrobromic acid and its subsequent determination after complexation with 3,3′-diaminobenzidine or sodium diethyldithiocarbamate.  相似文献

Two methods are described for the simultaneous determination of mebeverine hydrochloride (MB) and sulpiride (SU) in their combinations. The first method depends on the first derivative of the ratio spectra by measurement of the amplitudes at 263.7 and 234.9 nm for MB and SU, respectively. The linear ranges and detection limits are 4.0-40.0 and 0.72 microg/ml for MB and 1.0-10.0 and 0.34 microg/ml for SU. In the second case, a chemometric (classical least squares) method was developed. The concentration data matrices were obtained by using different concentrations of pure drugs in 0.1 M HCl. The absorbance data matrix corresponding to each concentration data matrix was obtained by the measurements of absorbances in the range 200-300 nm in their zero order spectra; then calibration was obtained by using the absorbance data matrix and the concentration data matrix for the prediction of the unknown concentrations of MB and SU in their mixture. The numerical values were calculated by using Matlab R12 version 6.0 and Origin 5.0 software. The procedures do not require any separation steps. These two methods were successfully applied for assaying the pharmaceutical formulation, of Colona tablets.  相似文献

A new catalytic spectrophotometric method is reported for the simultaneous determination of nitrite and nitrate by flow injection analysis, based on the catalytic effect of nitrite on the redox reaction between pyrogallolsulfonephthalein and potassium bromate in acidic media. Nitrate can also be on-line reduced to nitrite with a modified copper-coated cadmium reduction column. The reaction was monitored spectrophotometrically by measuring the decrease in the absorbance of pyrogallolsulfonephthalein at 465 nm. Various analytical parameters such as effects of acidity, reagent concentrations, flow rates, sample sizes, lengths of the reaction coil and temperatures were studied and were optimized. Under the optimized conditions, the calibration graph was linear for 2.4 to 160 ng ml(-1) of nitrite and 4.0 to 100 ng ml(-1) of nitrate. The influences of potential interfering cations and anions for nitrite and nitrate determination were studied. The method is successfully applied for food and water samples. Up to ten samples can be analyzed per hour.  相似文献

A method for the simultaneous spectrophotometric determination of cadmium, copper and zinc based on the formation of their complexes with 1,5-bis(di-2-pyridylmethylene)thiocarbonohydrazide is proposed. The absorption curves of these complexes overlap severely in the scanning range 380–480 nm. The analyte concentrations are calculated by a least squares fit of the pure spectra to the mixture spectra. A linear determination range of 0.1–1.7 μg/ml for cadmium, 0.1–1.3 μg/ml for copper and 0.2–1.2 μg/ml for zinc were obtained. The effect of interference was studied. The method has been applied to the determination of these metal ions in various type of materials.  相似文献
