探寻从微生物转化液中分离纯化5-氟尿苷(5-FUR)的工艺条件。先后尝试了大孔吸附树脂、大孔弱碱性阴离子交换树脂以及硅胶柱色谱的分离方法,首先利用大孔弱碱性阴离子交换树脂吸附,然后用1mol/L NaCl动态洗脱得到含量较高的两种含氟化合物,最终通过硅胶柱色谱,采用正己烷-乙酸乙酯(含10%醋酸)为流动相进行洗脱,得到了纯度为100%的纯品。在目标化合物和杂质理化性质极其相似,含量极其悬殊的条件下,成功去除含量极高的杂质,实现了5-FUR的分离纯化,圆满地达到了预期的目的。  相似文献

〕研究了离子交换纤维做固定相在离子色谱中的应用。结果表明,用VS-2型阴离子交换纤维和VS-型阳离子交换纤维分别填充分离柱和抑制柱,与树脂柱具有相同的效果,而纤维柱的阻力仅为树脂柱的十分之一左右。我们认为用离子交换纤维做固定相对离子色谱具有实际意义。  相似文献

D315, D311, D301和D335四种大孔弱碱型丙烯酸系阴离子交换树脂被用来吸附丙烯腈和二甲基亚砜中的Fe3+, 以纯化丙烯腈和二甲基亚砜. 通过改变温度、用量比和时间等实验条件以寻找最优的纯化条件, 实验结果证明D315大孔弱碱型丙烯酸系阴离子交换树脂的吸附效果是最佳的, 能够在室温下达到高效率低成本地纯化丙烯腈和二甲基亚砜中的Fe3+.  相似文献

阴阳离子交换色谱串联分离纯化苦瓜籽核糖体失活蛋白   总被引:11,自引:0,他引:11  
将阴阳离子交换色谱柱在灌注色谱工作站上串联,使苦瓜籽粗提液中未被阴离子色谱柱吸附的蛋白质直接在阳离子色谱柱上吸附,用盐线性梯度洗脱从阳离子色谱柱上分离出两个具有抗真菌活性的蛋白。两个抗真菌蛋白经鉴定都达到了电泳纯,相对分子质量均为30000左右,N-端氨基酸序列分别为DVSFRLSGADPRSYGMFI与DVNFDLSTATAK,表明所得到的两种蛋白为苦瓜籽中的2个Ⅰ型核糖体失活蛋白α-苦瓜素(α-MMC) 和β-苦瓜素(β-MMC)。抗菌实验表明,α-MMC对香蕉枯萎菌和果腐霉菌均具有较强的抑制作用,β  相似文献

采用阳离子交换树脂和大孔吸附树脂联用的方法,将倒提壶生物碱提取液依次经过阳离子交换树脂和两次大孔吸附树脂,对其中的天芥菜碱和毛果天芥菜碱进行分离纯化。结果表明,倒提壶生物碱溶液采用阳离子交换树脂经过水洗和NaCl洗脱与大孔吸附树脂两次经乙醚和50%乙醇洗脱后,毛果天芥菜碱与天芥菜碱的纯度分别从0.73%和2.2%提高至81.6%和79.2%,提取量分别由4.6 mg/g和10.7 mg/g提高至50.6mg/g和92.7 mg/g。该文所建立的分离纯化方法具有成本低、污染小、周期短、操作简便和分离纯化效率高等特点,适合工业化生产。  相似文献

化妆品中阴阳离子的快速测定及其用于化妆品鉴别的研究   总被引:3,自引:0,他引:3  
用离子色谱电导检测器对黑泥类化妆品中的阴、阳离子(Na+,NH+4,K+,Mg 2+ ,Ca 2+ ,Cl-,Br-,NO-2,NO-3和SO 2- 4)进行了测定。阳离子分离条件: 阳离子交换柱ICS C25,均苯四甲酸淋洗液浓度2.0 mmol/L ,流速0.6 mL/min ;阴离子分离条件: 阴离子交换柱Shim pack IC A1,淋洗液为2.5 mmol/L 邻苯二甲酸和2.4 mmol/L 三羟基氨基甲烷,流速1.0 mL/min 。结果表明,上述10种离子在较宽浓度范围内有良好  相似文献

采用烟叶和烟梗为原料,使用热裂解气相色谱法对烟草的燃烧时间、燃烧温度进行控制,精确模拟实际烟草致香物质释放环境,精确捕获、并半定量物质,从而在实验室模拟、分析、研究烟草燃烧物中致香物质的组成和特点。研究分别考察了两种生物质在500℃、550℃下释放的致香物质组成。结果表明,烟梗、烟叶中元素组成中硫、氮两种元素含量低,而C/H比值两种烟草生物质均较高。两个热解温度下烟叶的热解产物中致香物质的含量均明显高于烟梗。升高热解温度均有利于获得更高相对含量的热解成分。烟叶热解产物中烯烃类物质相对含量比烟梗中多,而烟梗热解产物中酚类物质含量比烟叶中多。提高热解温度有利于酮类致香物质含量产生。  相似文献

The fatty acid composition of tobacco seed oil revealed that the oil is rich in unsaturated fatty acids, having linoleic acid (71.63%), oleic acid (13.46%) and palmitic acid (8.72%) as the most abundant unsaturated and saturated fatty acids respectively. So the tobacco oil was characterized as semi-drying type on the basis of fatty acid composition. The synthesis of alkyd resin was carried out by alcoholysis or monoglyceride process using an alkali refined tobacco seed oil, pentaerythritol, cis-1,2,3,6-tetrahydrophthalic anhydride along with lithium hydroxide as catalyst. The alkyd resin so prepared was found to be bright and of low color with high gloss. The drying and hardness properties and adhesion of the tobacco seed oil derived alkyd resin were also found a bit superior to those of other alkyd resins of the same oil length. In addition, the water and acid resistance of the said alkyd was also found comparable to the other alkyds.  相似文献

A method for removimg glucoraphenin from the extract of Radish pigment by anion exchange resin 201×7 was studied.The adsorption capacity of 201×7 resin for glucoraphenin was 72.8mg/ml resin,the equilibrium time 55 minutes,and the optinum pH5.5.All glucoraphenin that had been adsorbed on 201×7 resin was eluted by 1.5BV.hr^-1, eluent in whinc concentration of NaOH was 0.05mol·L^-1 at the flow rate of 1.5BV/h.Extracting solution of deglucoraphenin was enriched by vacuum and spray drying.A powder product of Radish pigment was obtained and E1cm^1%=4.30.  相似文献

采用带MS Vent装置的气质联用仪,同时实现MS定性和FID定量,对烤烟烟梗中性致香成分进行定性定量分析,共分离鉴定出40种成分.方法回收率介于85.69%~100.39%之间,精密度达到0.28%.结果表明,该方法简便、快捷、可靠,为全面评价烟草质量提供一种有效的分析手段.  相似文献

Indirect photometric chromatography, which couples analyte separation by ion exchange with indirect photometric detection, is applied to the analysis of complex solutions encountered in the pharmaceutical industry. Applications of primary interest include simultaneous acetate, lactate, chloride, or phosphate determinations, a bisulfite/sulfate assay, and quantitation of trace levels of chloride in deionized water. Three commercially available ion chromatography columns are studied for their suitability in these applications. Due primarily to (A) the acid-base chemistry of the phosphate and bisulfite ions, (B) the need to stabilize the readily oxidized bisulfite, and (C) resin stability at high pH, the column packed with a polymer-based resin is capable of producing analytically superior separations of Cl-, HPO4-2, and matrix components and of SO3-2/SO4-2 in the samples of interest. Under rigid relative concentration conditions, the columns packed with a silica backbone resin are capable of separating acetate, lactate, Cl-, and H2PO4- with baseline resolution and are effective for determining Cl- levels at concentrations of less than one part-per-million.  相似文献

The study of the behavior of hydrogen in ion exchange columns is of theoretical and practical interest. This problem can be solved by using tritium as radioactive hydrogen tracer. The present paper describes the first results obtained in an experimental study of the statics and dynamics of the distribution and movement of tritium in an ion exchanger-water system.The materials studied were Ku-2 cation exchange resin in the H- and Sr-forms, and AN-2F and AV-17 anion exchange resins in the Cl-form. The isotopic effects in the distribution of tritium have been estimated. It has been shown that tritium is concentrated to a greater extent in the water than in the resin. Study of the filtration of water labeled with tritium in ion exchange columns has established that, as a result of isotopic exchange sorption, the rate of movement of the water front labeled with tritium is less than the rate of flow of water. The effluent curves have been interpreted theoretically on the basis of the general theory of the dynamics of sorption, and values for the constants of the dynamics of sorption have been estimated.  相似文献

New non-destructive method for characterization of ion exchange chromatographic columns based on transient pH formed by a step change in ionic strength of buffer solutions was examined. The method was used to distinguish between cation and anion or weak and strong ion exchange chromatographic supports and to determine the capacity of the chromatographic resins. The general scheme to distinguish between most commonly used types of ion exchange chromatographic columns was proposed. The duration of pH transient was shown to be linearly proportional to the total ionic capacity and was used to estimate protein dynamic binding capacity of the resin. The effects of pH, concentration and temperature on transient pH duration were examined.  相似文献

为了分析烤烟中游离氨基酸含量与烤烟香型、产地的关系,以国内14个烟草种植省份的138个调制后的烤烟中部叶片为材料,采用柱前衍生-超高效液相色谱-单重四极杆质谱法对烤烟中的20种游离氨基酸进行分析。方法表征结果显示该方法满足分析要求,适合烤烟中游离氨基酸的检测。对调制后烤烟中部叶片的游离氨基酸分析结果显示,烤烟的游离氨基酸含量在产地间差异很大(28.50%~94.20%),其中天冬酰胺、谷氨酰胺含量的相对标准偏差(RSD)超过80%;在3种典型香型(浓香型、清香型和中间香型)烤烟中,浓香型烤烟游离氨基酸含量的RSD大于其他两种香型。对同一香型的不同种植省份和同一省份不同香型的烤烟游离氨基酸分析结果显示,其含量呈现一定的规律。  相似文献
