Condensation of thiosemicarbazones of furfural, 3-(2-furyl)acrolein, as well as their 5-nitro derivatives with chloroacetone by boiling in alcohol or acetic acid gives the corresponding 4-methylthiazolyl-(2)hydrazones. 4-Methylthiazolyl-(2)-hydrazones of 5-nitro-2-acetylfuran and 5-nitro-2-furfurilydeneacetone can be prepared by condensing the corresponding thiosemicarbazones with chloroacetone by heating with glacial acetic acid containing fused sodium acetate.For Part IV see [1].  相似文献

Reaction of N-methyl-2-amino-4-nitroaniline ( 1 ) with lactic acid afforded 2-(1-hydroxyethyl)-1-methyl-5-nitrobenzimidazole ( 2 ). Oxidation of compound 2 with chromic acid in acetic acid gave 2-acetyl-1-methyl-5-nitrobenzimidazole ( 3 ). Reaction of compound 3 with substituted 2-aminobenzaldehyde ( 4 ) under basic conditions yielded substituted 2-(1-methyl-5-nitro-2-benzimidazolyl)quinolines ( 5 ). Condensation and cyclization of o-aminoacetophenone (or substituted o-aminobenzophenones) with compound 3 under acetic condition afforded compound 7 . Condensation and cyclization of compound 1 with indole-3-carboxaldehyde ( 11 ) in ethanol in the presence of excess nitrobenzene gave 3-(1-methyl-5-nitro-2-benzimidazolyl)indole ( 12 ).  相似文献

The syntheses of 4- and 5-chlorosulfonylfuran-2-carboxylic acid (Ia,IIa), 4-chlorosulfonylfuran-2-carboxamide (Ib), 3,5-dimethylpyrazole and isoxazole-4-sulfonyl chlorides (IIIa,IVa) and 2,4-dimethylthiazole-5-sulfonyl chloride (Va) are described. The sulfonyl chlorides were converted into a range of amides, hydrazides and azides. Condensation of the sulfonohydrazides with β-dicarbonyl compounds, gave the corresponding β-ketohydrazones (VII), which, with the exception of the derivatives (VIIe,f,g,i), were converted to the sulfonylpyrazoles (VIII). The structures and spectral data of these compounds are briefly discussed. The reaction of the sodio derivative of acetylacetone with thiophene-2-sulfonyl chloride (VIc) gave 3-(thiophene-2-sulfonyl)pentane-2,4-dione (XII), which with hydrazine gave 4-(thiophene-2-sulfonyl)-3,5-dimethylpyrazole (XIII). However, the analogous reaction with thiophene-2-sulfonohydrazide (VIa) failed to give the expected 1,4-bisthiophenesulfonylpyrazole.  相似文献

T-reactions of anabasine derivatives were performed for the first time. Condensation of 5-nitro-2-[2-(3pyridyl)piperidino]benzaldehyde with 1,3-dimethylbarbituric acid formed an annelated tetrahydroquinoline system containing a spiropyrimidine moiety as the T-reaction product. Condensation of this same aldehyde with Meldrum’s acid was accompanied by recyclization that led to opening of the piperidine ring and closing of the tetrahydroquinolone system. A third version of the T-reaction that also led to opening of the piperidine ring but without recyclization was the condensation with dimedone.  相似文献

The condensation of 1,2-dimethylbenzimidazole with thiophene-2-aldehyde in the presence of fused zinc chloride results in 1-methyl-2-[β-(2-thienyl)vinylene]-1H-benzimidazole. Its electrophilic substitution reactions (nitration, bromination, sulfonation, formylation, acylation) were studied. All the reactions occur in the 5-position of the thiophene ring. Only in the case of bromination in dichloroethane 5′,5(6)-dibromo-derivative was obtained. Data of the quantum-chemical calculations of the total positive charge on the carbon atoms of the protonated molecules of thienylbenzimidazoles including the vinylene group and without it are reported.  相似文献

The reaction of 4-nitro-o-phenylenediamine (I) with acetoacetic ester at room temperature under acid catalysis gives ethyl 3-(2-amino-5-nitrophenylamino)crotonate (II), which is readily cyclized to 7-nitro-4-methyl-2,3-dihydro-1H-1,5-benzo-2-diazepinone (III) on heating with alkaline agents. The reaction of I with acetoacetic ester in refluxing xylene gives isomeric 8-nitro-4-methyl-2,5-dihydro-1H-1,5-benzo-2-diazepinone (IVa) or 8-nitro-4-methyl-2,3-dihydro-1H-1,5-benzo-2-diazepinone (IVb), which are readily interconverted. The synthesis of IV is complicated by the side formation of 5-nitro-2-methylbenzimidazole (V) and thermal rearrangement of IVa and IVb to 5-nitro-1-isopropenylbenzimidazolone (VI). 6-Nitro-1-isopropenylbenzimidazolone (VII) is similarly obtained on heating III.Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 5, pp. 696–699, May, 1972.  相似文献

Condensation of 3,4-dichloro-6-[(trimethylsilyl)oxy] pyridazine ( 3 ) with 1-O-acetyl-2,3,5-tri-O-benzoyl-β- D -ribofuranose ( 4 ), by the stannic chloride catalyzed procedure, has furnished 3,4-dichloro-1-(2,3,5-tri-O-benzoyl-β- D -ribofuranosyl) pyridazin-6-one ( 5 ). Nucleophilic displacement of the chloro groups and removal of the benzoyl blocking groups from 5 has furnished 3-chloro-4-methoxy-, 3,4-dimethoxy-, 4-amino-3-chloro-, 3-chloro-4-methylamino-, 3-chloro-4-hydroxy-, and 4-hydroxy-3-methoxy-1-β- D -ribofuranosylpyridazin-6-one. An unusual reaction of 5 with dimethylamine is reported. Condensation of 4,5-dichloro-3-nitro-6-[(trimethylsilyl)oxy]pyridazine with 4 yielded 4,5-dichloro-3-nitro-1-(2,3,5-tri-O-benzoyl-β- D -ribofuranosyl)pyridazin-6-one ( 24 ). Nucleophilic displacement of the aromatic nitro groups from 24 is discussed. Condensation of 3 with 3,5-di-O-p-toluoyl 2-deoxy- D -erythro-pentofuranosyl chloride ( 28 ) afforded an α, β mixture of 2-deoxy nucleosides. The synthesis of certain 3-substituted pyridazine 2′-deoxy necleosides are reported.  相似文献

2-(2-Cyano-acetylamino)-4,5,6,7-tetrahydro-benzo[b]thiophene-3-carboxamide(3) was used as starting material for synthesis of 4-thiazolidinone, thiazolidine, and thiophene derivatives 6, 7a, b, and 8a, b, respectively. Thiocarbomyl derivative 5, 4-thizolidinone 9, and thioxothiazolidine 10 were obtained from reaction of 3 with thioglycolic acid and phenyl isothiocyanate/sulfur, respectively. Condensation of 3 with selected cyclic ketones and aromatic aldehydes yielded the arylidine derivatives 11a, b and 13, respectively. Refluxing of 11a, b with sulfur and morpholine yielded the thiophene derivatives 12a, b, respectively. Diazocoupling of compound 3 withp-tolyl diazonium chloride yielded the hydrazone derivative 14. The newly synthesized compounds were characterized by infrared, 1H NMR, and mass spectral studies. Representative compounds of the synthesized product were tested and evaluated as antimicrobial agents. Compound 12b gives very high antimicrobial activity against Ampicillin.  相似文献

A study is made of the nitration of thionaphthene-2-aldehyde, its diacetate, and of 2-(-nitrovinyl)thionaphthene. UV and IR spectroscopy show that nitration of thionaphthene-2-aldehyde in acetic anhydride gives a mixture of 3-nitrothionaphthen-2-aldehyde diacetate, 4-nitrothionaphthene-2-aldehyde diacetate, and 6-nitrothionaphthene-2-aldehyde diacetate in the ratios 106624, while nitration of thionaphthene-2-aldehyde diacetate gives a mixture of 3-nitrothionaphthene-2-aldehyde diacetate, 4-nitrothionaphthene-2-aldehyde diacetate, and 6-nitrothionaphthene-2-aldehyde diacetate in the ratios 363826. Nitration of 2-(-nitrovinyl)thionaphthene gives 4-nitro-2-(-nitrovinyl)thionaphthene.  相似文献

1-Bis(methoxy)-4-bis(methylthio)-3-buten-2-one has been shown to be a useful three carbon synthon for efficient regiospecific synthesis of a variety of five (pyrazole and isoxazole) and six membered (pyrimidines, pyridone and pyridines) heterocycles with masked (or unmasked) aldehyde functionality by cyclocondensation with bifunctional heteronucleophiles such as hydrazine, hydroxylamine, guanidine, thiourea, cyanoacetamide and substituted β-lithioaminoacrylonitriles. Reaction of 1-bis(methoxy)-4-bis(methylthio)-3-buten-2-one with methylene iodide and Zn-Cu couple under Simmons-Smith reaction conditions afforded 2-(methylthio)thiophene-4-aldehyde in good yield, whereas cycloaromatization with thiophene-2- and 3-acetonitriles gave the corresponding substituted benzothiophene-4- and 7-aldehydes in good yields.  相似文献

Condensation of 2-formyl-or 2-cyano-3-chloro-1,4-dimethoxyanthraquinone with methyl thioglycolate in the presence of base gave methyl 4,11-dimethoxyanthra[2,3-b]thiophene-5,10-dione-2-carboxylate and its 3-amino derivative respectively. Hydrolysis of the ester group in methyl 4,11-dimethoxyanthra[2,3-b]thiophene-5,10-dione-2-carboxylate and subsequent decarboxylation of the carboxylic acid formed gave 4,11-dimethoxyanthra[2,3-b]thiophene-5,10-dione. The electronic absorption spectra of ten chalcone analogs derived from 4- and 5-nitropyrroles have been measured in solution in ethanol, dioxane, concentrated sulfuric acid, and 20% ethanolic caustic potash and interpreted. The IR spectra of these compounds and also of 4- and 5-nitro-2-acetylpyrroles and 5-nitropyrrole-2-aldehyde have been measured in the 3700-500 cm–1 region, an assignment of frequencies has been carried out, and considerations have been put forward on the connection between the characteristic features of the absorption and the chemical structure.  相似文献

The reaction of anthranilic acid and methoxy- and nitro-substituted anthranilic acid with p-toluenesulfonyl chloride in pyridine was investigated. Anthranilic acid and its methoxy-substituted derivatives react to give the corresponding 2-(2-tosylaminophenyl)-4H-3,1-benzoxazin-4-ones and N,N'-ditosyldianthranilides. At 20°C, 4-nitroanthranilic acid gives primarily 2-(2-amino-4-nitrophenyl)-7-nitro-4H-3,1-benzoxazin-4-one, while its tosyl derivative is obtained at 114°. The products of the reaction of 5-nitroanthranilic acid with p-toluenesulfonyl chloride are 2-nitro-11H-pyrido[2,1-b]quinazolin-11-one and 2-(2-amino-5-nitrophenyl)-6-nitro-4H-3, 1-benzoxazin-4-one. Syntheses of Sulfonated Derivatives of 4-Amino-1, 3-dimethylbenzene and 2-Amino-1, 3-dimethylbenzene Direct sulfonation of 4-amino-1, 3-dimethylbenzene (1) and sulfonation of 4-nitro-1,3-dimethylbenzene ( 4 ) to 4-nitro-1,3-dimethylbenzene-6-sulfonic acid ( 3 ) followed by reduction yield 4-amino-1,3-dimethylbenzene-6-sulfonic acid ( 2 ). The isomeric 5-sulfonic acid ( 5 ) however is prepared solely by baking the acid sulfate salt of 1 . Reaction of sulfur dioxide with the diazonium chloride derived from 2-amino-4-nitro-1,3-dimethylbenzene ( 7 ) leads to 4-nitro-1,3-dimethylbenzene-2-sulfonyl chloride ( 8 ), which is successively hydrolyzed to 4-nitro-1,3-dimethylbenzene-2-sulfonic acid ( 9 ) and reduced to 4-amino-1, 3-dimethylbenzene-2-sulfonic acid ( 6 ). Treatment of 4-amino-6-bromo-1,3-dimethylbenzene ( 12 ) and 4-amino-6-chloro-1, 3-dimethylbenzene ( 13 ), the former obtained by reduction of 4-chloro-6-nitro-1,3-dimethyl-benzene ( 10 ) and the latter from 4-chloro-6-nitro-1, 3-dimethylbenzene ( 11 ), with oleum yield 4-amino-6-bromo-1,3-dimethylbenzene-2-sulfonic acid ( 14 ) and 4-amino-6-chloro-1,3-dimethylbenzene-2-sulfonic acid ( 15 ) respectively; subsequent carbon-halogen hydrogenolyses of 14 and 15 lead also to 6 (Scheme 1). Baking the acid sulfate salt of 2-amino-1, 3-dimethylbenzene ( 17 ) gives 2-amino-1, 3-dimethylbenzene-5-sulfonic acid ( 16 ), whereas the isomeric 4-sulfonic acid ( 18 ) can be prepared by either of the following three possible pathways: Sulfonation of 2-nitro-1,3-dimethylbenzene ( 20 ) to 2-nitro-1,3-dimethylbenzene-4-sulfonic acid ( 21 ) followed by reduction or sulfonation of 2-acetylamino-1,3-dimethylbenzene ( 19 ) to 2-acetylamino-1,3-dimethylbenzene-4-sulfonic acid ( 22 ) with subsequent hydrolysis or direct sulfonation of 17 . 5-(2-Thienylsulfanyl)thiophene-2-carbaldehyde reacted with propane-1-thiol and propane-1,3-dithiol in the presence of chloro(trimethyl)silane to give previously unknown 5-[bis(propylsulfanyl)methyl]-2-(2-thienylsulfanyl)thiophene and 2-[5-(2-thienylsulfanyl)thiophen-2-yl]-1,3-dithiane. Chloromethylation of 5-(2-thienylsulfanyl)thiophene-2-carbaldehyde with formaldehyde in a stream of hydrogen chloride in the presence of zinc chloride resulted in the formation of an oligomeric product consisting of thiophene rings connected alternately by sulfur and methylene bridges. The oligomer is formed via fast polycondensation of the primary chloromethylation product with the initial aldehyde. 5-(2-Thienylsulfanyl)thiophene-2-carbaldehyde was oxidized at the sulfide and aldehyde groups with 30% hydrogen peroxide in glacial acetic to produce 5-(2-thienylsulfonyl)thiophene-2-carboxylic acid.  相似文献
