The hydrolysis reaction mechanisms of a new generation Pd(II) anticancer drugs containing coumarin derivatives have been investigated combining density functional theory with the conductor-like dielectric continuum model approach. The first and the second aquation processes have been explored for the cis and trans counterpart of title PdL2 complex. Two possibilities for the second hydrolysis process have been analyzed for both compounds. From our data emerge that cis and trans PdL2 compounds have a different behavior in water. cis-isomer readily undergo first hydrolysis process generating a mono-aqua complex while both the pathways investigated for the second aquation reaction are more energetically demanding, suggesting that the mono-aqua complex could act as active species. On the contrary, for trans-compound, both the investigated reactions for the second aquation process occur by overcoming activation barriers comparable with that found for the first hydrolysis reaction. According to our data, trans-oriented PdL2 drug could undergo degradation process generating non-active compounds with the consequent lack of pharmacological activity.  相似文献

A new dichloroplatinum(II) complex with 5-methyl-1,10-phenanthroline (1) has been obtained and characterized by X-ray diffraction. MTT assay was used for in vitro cytotoxicity evaluation of this complex, along with a similar complex with 4,7-diphenyl-1,10-phenanthroline (2) against normal and cancerous cell lines. Interestingly, the IC50 values of the new complexes were higher for normal cells and in the case of complex 2, lower against all studied human cancer cells, in comparison with cisplatin.  相似文献

A series of Co(II), Ni(II), and Cu(II) complexes have been synthesized with Schiff bases (H2LI and H2LII) derived from 8-formyl-7-hydroxy-4-methylcoumarin or 5-formyl-6-hydroxycoumarin and o-aminophenol. Structures have been proposed from elemental analyses, spectral (IR, UV-Vis, FAB-mass, and Fluorescence), magnetic, and thermal studies. The measured low molar conductance values in DMF indicate that the complexes are non-electrolytes. Elemental analyses indicate ML · 3H2O [M = Co(II), Ni(II), and Cu(II)] stoichiometry. Spectroscopic studies suggest coordination through azomethine nitrogen, phenolic oxygen of o-aminophenol, and the coumarin via deprotonation. The Schiff bases and their complexes have been screened for antibacterial (Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa, and Salmonella typhi) and antifungal (Aspergillus niger, Aspergillus flavus, and Cladosporium) activities by minimum inhibitory concentration (MIC) method. A series of Co(II), Ni(II), and Cu(II) complexes ML?·?3H2O have been synthesized with Schiff bases derived from 3-substituted-4-amino-5-mercapto-1,2,4-triazole and 5-formyl-6-hydroxy coumarin. The complexes are insoluble in common organic solvents but soluble in DMF and DMSO. The measured molar conductance values in DMF indicate that the complexes are non-electrolytes. In view of analytical, spectral (infrared, UV-Vis, ESR, TG, and FAB-mass), and magnetic studies, it has been concluded that all the metal complexes possess octahedral geometry in which ligand is coordinated to metal through azomethine nitrogen, phenolic oxygen, and sulfur via deprotonation. The Schiff bases and their complexes have been screened for antibacterial (Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa, and Salmonella typhi) and antifungal activities (Aspergillus niger, Aspergillus flavus, and Cladosporium) by the minimum inhibitory concentration method. DNA cleavage is studied by agarose gel electrophoresis.  相似文献

Platinum(II) and palladium(II) complexes of the trithiacrown [9]aneS(3) containing a range of Group 15 donors are reviewed. These complexes have the general formula [M([9]aneS(3))(L(2))](n+) where L represents at least one Group 15 donor. Complexes involving pnictogens, with the exception of bismuth, are observed. The complexes generally have an elongated square pyramidal geometry with a long distance interaction to the third sulphur of the [9]aneS(3) which forms the apex of the square pyramid. This axial metal-sulphur distance is quite sensitive to the donor properties of L. Poorer donors such as Sb and As ligands show short axial distances whereas the better N donor ligands show longer distances. Pt(II) complexes of the formula [Pt([9]aneS(3))(EPh(3))(2)](2+) (E = P, As, Sb) show a considerable distortion towards a trigonal bipyramidal geometry due to intramolecular π-π interactions. Over seventy of these types of complexes have been crystallographically characterized and are discussed in this article. Other unique features of the complexes, including NMR spectroscopy, redox chemistry, and electronic spectroscopy, are also discussed.  相似文献

Nickel(II) complexes with 6-benzylaminopurine (BAP) derivatives, namely 6-(3-chlorobenzylamino)purine (HL1), 6-(4-chlorobenzylamino)purine (HL2) and 6-(4-fluorobenzylamino)purine (HL3), have been prepared and characterized by elemental analyses, i.r., u.v.–v.i.s., ES+ and FAB+ mass spectroscopy, magnetic susceptibility and conductivity measurements, and by thermal analysis. The complexes are: [Ni(L1(H2O)2Cl] · H2O, [Ni(L1)(H2O)-(NO3)] · H2O, [Ni(L2)(H2O)2Cl], [Ni(L2)(H2O)2(NO3)] · H2O, [Ni(HL2)(H2O)Cl2] · EtOH and [Ni(L3)(H2O)2Cl]. They have been tested in vitro for their possible cytotoxic activity against G-361 (human malignant melanoma), HOS (human osteogenic sarcoma), K-562 (human chronic myelogenous leukemia) and MCF-7 (human breast adenocarcinoma) cell lines.  相似文献

New thioether-phosphanes 2-RSC6H4CH2PPh2(RS-PPh2: R = Me, tBu, Ph) and the corresponding complexes [PdCl2(MeS-PPh2)], [PdCl2(tBuS-PPh2)], [PdCl2(PhS-PPh2)], [PdClMe(MeS-PPh2)] and [PtMe2(MeS-PPh2)] have been prepared, characterized and the X-ray crystal structures of all complexes determined. Whilst Pd(II) complexes of RS-PPh2 show low activity for CO/ethene copolymerisation, the complexes [PdCl2(RS-PPh2)] have been found to be very efficient for the Heck arylation of n-butylacrylate with bromobenzene under aerobic conditions.  相似文献

Complexes of composition L2MCl2 [M=Pt, R=H (I), Me (II), Ph (III)], and LMC12 [M=Pd, R=H (IV)] are prepared by reaction of 4,6-R2-2,5-diphenyl-1,3,2,5-dioxaboraphosphorinanes (L) with MCl2. Far-IR and31P NMR spectroscopy are used to demonstrate that I is cis whereas II and III are trans complexes in the solid. The conformational behavior of I is studied by31P and1H NMR. The synthesis and characterization (elemental analysis, 1H and 13C NMR spectroscopy and single crystal X-ray diffraction) of the first ortho-metallated Pd(II) complexes containing N-benzoyl thiourea ligands are described. Four of these compounds showed liquid crystal properties which were investigated by mean of DSC and polarised light microscopy and their mesogenic properties are compared to those of ortho-metallated imine Pd(II) complexes with other types of co-ligands.  相似文献

CoII, NiII and CuII chloride complexes with acetamidrazone derivatives (L 1 )–(L 8 ) have been prepared, by reacting the corresponding metal chloride with the ligand in a 1:2 ratio, and characterised by chemical analysis and physical measurements. The ligand behaviour and the geometry have been assigned on the basis of i.r. spectroscopy, electronic reflectance spectra and molar conductivity values and only, where possible, the geometry has been confirmed as octahedral by X-ray structure determination. Complexes cytotoxicity on a human cellular line of adenocarcinoma was also tested. Three of the complexes present a weak cytotoxicity in vitro.  相似文献

The mononuclear palladium(II) (1) and platinum(II) (2) complexes containing phenylglycine have been synthesized and characterized by elemental analysis, IR spectra, and 1H NMR spectra. The structure of 1 was determined by X-ray diffractometry. The interaction between the complexes and fish sperm DNA (FS-DNA), adenosine-5′-triphosphate (ATP), and adenine (Ade) were investigated by UV absorption spectra, the interaction mode of the complex binding to DNA was studied by fluorescence spectra and viscometry. The results indicate that the two complexes have different binding affinities to DNA, complex 2 > complex 1. Gel electrophoresis assay demonstrates that the two complexes have the ability to cleave pBR322 plasmid DNA. Cytotoxicity experiments were carried out toward four different cancer cell lines, and 1 shows lower inhibitory efficiency than 2, consistent with the binding affinities towards DNA.  相似文献

The reactions of PhSe, PhS and Se2− with N-{2-(chloroethyl)}pyrrolidine result in N-{2-(phenylseleno)ethyl}pyrrolidine (L1), N-{2-(phenylthio)ethyl}pyrrolidine (L2), and bis{2-pyrrolidene-N-yl)ethyl selenide (L3), respectively, which have been explored as ligands. The complexes [PdCl2(L1/L2)] (1/7), [PtCl2(L1/L2)] (2/8), [RuCl(η6-C6H6)(L1/L2)][PF6] (3/9), [RuCl(η6-p-cymene)(L1/L2)][PF6] (4/10), [RuCl(η6-p-cymene)(NH3)2][PF6] (5) and [Ru(η6-p-cymene)(L1)(CH3CN)][PF6]2·CH3CN (6) have been synthesized. The L1-L3 and complexes were found to give characteristic NMR (Proton, Carbon-13 and Se-77). The crystal structures of complexes 1, 3-6, 9 and 10 have been solved. The Pd-Se and Ru-Se bond lengths have been found to be 2.353(2) and 2.480(11)/2.4918(9)/2.4770(5) Å, respectively. The complexes 1 and 7 have been explored for catalytic Heck and Suzuki-Miyaura coupling reactions. The value of TON has been found up to 85 000 with the advantage of catalyst’s stability under ambient conditions. The efficiency of 1 is marginally better than 7. The Ru-complexes 3 and 9 are good for catalytic oxidation of primary and secondary alcohols in CH2Cl2 in the presence of N-methylmorpholine-N-oxide (NMO). The TON value varies between 8.0 × 104 and 9.7 × 104 for this oxidation. The 3 is somewhat more efficient catalyst than 9.  相似文献

A series of novel Pd(II) and Pt(II) complexes, [PdL(2)Cl(2)]·DMF (1), [Pd(2)(L-H)(2)(bpy)Cl(2)]·(H(2)O)(2)·DMF (2), [Pd(2)(L-H)(2)(phen)Cl(2)]·2H(2)O (3), [PtL(2)Cl(2)]·H(2)O (4), [Pt(2)(L-H)(2)(bpy)Cl(2)]·2H(2)O (5), and [Pt(2)(L-H)(2)(phen)Cl(2)]·H(2)O (6), where bpy = 2,2'-bipyridine, phen = 1,10-phenanthroline, and L = 1,3-benzothiazol-2-amine, have been synthesized and characterized. The competitive binding of the complexes to DNA has been investigated by fluorescence spectroscopy. The values of the apparent DNA binding constant, calculated from fluorescence spectral studies, were 3.8 × 10(6) (K(app)(4)), 2.9 × 10(6) (K(app)(1)), 2.4 × 10(6) (K(app)(6)), 2.0 × 10(6) (K(app)(5)), 1.2 × 10(6) (K(app)(3)), and 6.9 × 10(5) (K(app)(2)). The binding parameters for the fluorescence Scatchard plot were also determined. On the basis of the data obtained, it indicates that the six complexes bind to DNA with different binding affinities in the relative order 4 > 1 > 6 > 5 > 3 > 2. Viscosity studies carried out on the interaction of complexes with Fish Sperm DNA (FS-DNA) suggested that all complexes bind by intercalation. Gel electrophoresis assay demonstrates that all the complexes can cleave the pBR 322 plasmid DNA and bind to DNA in a similar mode. The cytotoxic activity of the complexes has been also tested against four different cancer cell lines. The results show that all complexes have activity against KB, AGZY-83a, Hep-G2, and HeLa cells. In general, the Pt(II) complexes were found to be more effective than the isostructural Pd(II) complexes. The mononuclear complexes exhibited excellent activity in comparison with the dinuclear complexes in these four cell lines. Moreover, on the KB cell line (the human oral epithelial carcinoma), the observed result seems quite encouraging for the six complexes with IC(50) values ranging from 1.5 to 8.6 μM. Furthermore, apoptosis assay with hematoxylin-eosin staining shows treatment with the six complexes results in morphological changes of KB cells. The results induce apoptosis in KB cells.  相似文献

The paper describes methods for the synthesis and isolation of solid phases of the individual stereoisomeric Pd(II) bis(amino acid) complexes with serine (SerH = NH2C*H(CH2OH)COOH, α-amino-β-hydroxypropionic acid), threonine (ThrH CH3C*H(OH)C*H(NH2)COOH, threo-α-amino-β-hydroxybutyric acid), and allothreonine (alloThrH is erythro-α-amino-β-hydroxybutyric acid): cis-[Pd(S-Ser)2], trans-[Pd(R-Ser)(S-Ser)], cis-[Pd(S-Thr)2], trans-[Pd(S-Thr)2], trans-[Pd(R-Thr)(S-Thr)], and cis-[Pd(R-alloThr)(S-alloThr)] (R, S are the absolute configurations of the asymmetric C* atom connected to the NH2 group). In the search for antitumor active metal complexes several ruthenium complexes have been reported to be promising. A series of mononuclear Ru(II) complexes, [Ru(T)2(S)]2+, where T?=?2,2′-bipyridine/1,10-phenanthroline and S?=?CH3-bitsz, Cl-bitsz, Br-bitsz, tmtsz, dmtsz, have been prepared and characterized by UV-Vis, IR, 1H-NMR, FAB-mass spectroscopy, and elemental analysis. The complexes were subjected to in vivo anticancer activity against a transplantable murine tumor cell line Ehrlich's ascitic carcinoma (EAC) and in vitro cytotoxic activity against human cancer cell line Molt 4/C8, CEM, and murine tumor cell line L1210. Ruthenium complexes showed promising biological activity especially in decreasing tumor volume and viable ascitic cell counts. Treatment with these complexes prolonged the life span of EAC-tumor-bearing mice by 10–48%. In vitro evaluation of these ruthenium complexes revealed cytotoxic activity from 0.21 to 24?µmol?L?1 against Molt 4/C8, 0.16–19?µmol?L?1 against CEM, and 0.75–32?µmol?L?1 against L1210 cell proliferation, depending on the nature of the compound.  相似文献

A series of Co(II), Ni(II) and Cu(II) complexes have been synthesized with Schiff bases derived from 5-amino-1,3,4-thiadiazole-2-thiol and 8-formyl-7-hydroxy-4-methylcoumarin/8-acetyl-7-hydroxy-4-methylcoumarin. The chelation of the complexes has been proposed in the light of analytical, spectral (IR, UV–Vis), ESI-mass, magnetic, ESR and thermal studies. The measured molar conductance values indicate that the complexes are non-electrolytes. TG and DTA provide the useful information about the coordination of water molecules to the metal ion and the stability of the complexes. TG and DTA curves show that the Co(II) complexes decomposition takes place in two stages corresponds to loss due to water molecules and Schiff base moiety. Whereas, Ni(II) and Cu(II) complexes decomposition took place in three steps corresponding to the loss of coordinated water molecules, 1,3,4-thiadiazole moiety and coumarin moiety, respectively. The Schiff bases and their complexes have been screened for their antibacterial and antifungal activities. The results of these studies show the metal complexes to be more antibacterial and antifungal as compared to the uncomplexed coumarins.  相似文献

Novel mixed complexes of copper (II) and cobalt (II) with 3-amino-5,5-dimethylhydantoin were synthesized and their in vitro anticancer activity was investigated. The structures of the compounds were confirmed by IR, UV–Vis spectrometry, voltammetry and elemental analysis. The cytotoxic effects of novel complexes of copper (II) and cobalt (II) with 3-amino-5,5-dimethylhydantoin were tested against a panel of human tumor cell lines. All of the compounds investigated exhibited different concentration-dependent antiproliferative effects against the HT29, MDA-MB-231, HepG2 and HeLa cell lines after 24 h of treatment. The most sensitive cells were the HepG2 cells at various concentrations of both tested compounds followed by HT29.  相似文献

Treatment of the functionalized Schiff base ligands with boronic esters 1a, 1b, 1c and 1d with palladium (II) acetate in toluene gave the polynuclear cyclometallated complexes 2a, 2b, 2c and 2d, respectively, as air-stable solids, with the ligand as a terdentate [C,N,O] moiety after deprotonation of the -OH group. Reaction of 1j with palladium (II) acetate in toluene gave the dinuclear cyclometallated complex 5j. Reaction of the cyclometallated complexes with triphenylphosphine gave the mononuclear species 3a, 3b, 3c, 3d and 6j with cleavage of the polynuclear structure. Treatment of 2c with the diphosphine Ph2PC5H4FeC5H4PPh2 (dppf) in 1:2 molar ratio gave the dinuclear cyclometallated complex 4c as an air-stable solid.Deprotection of the boronic ester can be easily achieved; thus, by stirring the cyclometallated complex 3a in a mixture of acetone/water, 3e is obtained in good yield. Reaction of the tetrameric complex 2a with cis-1,2-cyclopentanediol in chloroform gave complex 2c after a transesterification reaction. Under similar conditions complexes 3a and 3d behaved similarly: with cis-1,2-cyclopentanediol, pinacol or diethanolamine complexes 3c, 3b, 3g and 3f, were obtained. The pinacol derivatives 3b and 3g experiment the Petasis reaction with glyoxylic acid and morpholine in dichloromethane to give complexes 3h, and 3i, respectively.  相似文献
