A series of novel muti‐substituted pyrido[4,3‐d]pyrimidin‐4‐one derivatives 5a , 5b , 5c , 5d , 5e , 5f , 5g , 5h , 5i , 5j , 5k , 5l were designed and synthesized by the muti‐step reaction. N,S‐acetal 1 reacted with acetyl acetamide in the presence of zinc nitrate to obtain muti‐substituted pyridine 2 , which reacted with triethyl orthoformate to give 8‐cyano‐5‐methyl‐7‐methylthio‐pyrido[4,3‐d]pyrimidin‐4‐one 3 ; the target compounds 5 were obtained in good yields by the oxidation of 3 with H2O2 in a catalytic amount of sodium tungstate then by the substitution with various substituted phenols. Their structures were confirmed by IR, 1H NMR, EI‐MS, and elemental analyses. The preliminary bioassay indicated that some of them displayed moderate herbicidal activity against dicotyledonous weed Brassica campestris L. at the concentration of 100 mg/L. For example, compounds 5a , 5f , and 5g possessed 76.0%, 62.7%, and 60.2% inhibition against B. campestris at the concentration of 100 mg/L. Moreover, 5a exhibited 58.2% inhibition against B. campestris at the concentration of 10 mg/L.  相似文献

This work reports the synthesis of thiazolidin‐4‐ones and thiazinan‐4‐ones analogous to rosiglitazone, a potent antidiabetic drug. The desired compounds were synthesized with moderate to good yields by one‐pot reactions between different primary amines, mercaptoacetic or mercaptopropionic acids, and the 4‐(2‐(methyl(pyridin‐2‐yl)amino)ethoxy)benzaldehyde. The cyclocondensation reactions were carried out for 20 h, and all the products were characterized by 1H and 13C nuclear magnetic resonance spectroscopy, mass spectrometry, and one example by X‐ray diffraction.  相似文献

The 5,6,7,8,9,10‐hexahydro‐2‐methylthiopyrimido[4,5‐b]quinolines 4a , 4b , 4c , 4d , 5a , 5b , 5c , 5d and their oxidized forms 6a , 6b , 6c , 6d , 7a , 7b , 7c , 7d were obtained from the reaction of 6‐amino‐2‐(methylthio)pyrimidin‐4(3H)‐one 2 or 6‐amino‐3‐methyl‐2‐(methylthio)pyrimidin‐4(3H)‐one 3 and α,β‐unsaturated ketones 1a , 1b , 1c , 1d using BF3.OEt2 as catalyst and p‐chloranil as oxidizing agent. Some of the new compounds were evaluated in the US National Cancer Institute (NCI), where compound 5a presented remarkable activity against 46 cancer cell lines, with the most important GI50 values ranging from 0.72 to 18.4 μM from in vitro assays.  相似文献

We report herein one‐pot synthesis and the antibacterial and antitubercular activities of 2,5‐disubstituted‐1,3,4‐oxadiazole compounds obtained by hybridization of a well‐known antitubercular agent isoniazid (INH ) with four broad‐spectrum antibiotics belonging to fluoroquinolone (FQ ) class. The work is aimed at designing and developing potential antimicrobial agents having synergistic action due to the coupling of INH and FQ through the biologically active 1,3,4‐oxadiazole nucleus. The synthesized compounds are expected to have low toxicity as compared to INH due to the absence of free hydrazide group in the chemical structure of the prepared derivatives. The antibacterial activities of the 1,3,4 oxadiazole derivatives were also tested against several Gram‐positive and Gram‐negative pathogenic bacterial strains. The antitubercular activity was evaluated against M. tuberculosis H37Rv strain, and the results were compared with that of the positive control INH . The title compounds showed excellent antimicrobial and promising antitubercular activity in comparison to the parent fluoroquinolones and INH , respectively.  相似文献

Their structures were clearly verified by IR spectroscopy, 1H‐NMR spectroscopy, EI‐MS, and elemental analysis, and in the case of compound 6i , further analyzed by single‐crystal X‐ray diffraction. Sulfonylurea herbicides have been widely used because of their low application rates,good crop selectivities and low mammalian toxicities.However,some sulfonylureas might persist unfavourably in the environment with residual problems.In order to look for ecologically safer and environmentally benign sulfonylureas,and on keeping the pyrimidine ring being monosubstituted,15 novel C5-monosubstituted benzenesulfonylurea compounds were synthesized.The structures of all the compounds synthesized were confirmed by elemental analysis and 1H NMR.Preliminary herbicidal activities of these new sulfonylurea compounds were determined by ALS screening(in vitro)and pot bioassay experiments(in vivo).The herbicidal results show that some novel sulfonylureas are comparable to commercial Foramsulfuron and Monosulfuron.  相似文献

An efficient, simple, and green procedure for the synthesis of isoxazol‐5(4H)‐one derivatives are described here through a convenient one‐pot, three‐component reaction at room temperature. The title compounds are isolated in high to excellent yields and after short reaction times, and are characterized by various spectroscopic methods such as IR, 1H NMR, and 13C NMR. The synthesized compounds 4a–c and 4e–i were tested for their in vitro activity against a panel of Gram‐positive and Gram‐negative bacteria, demonstrating their ability to inhibit microorganisms with a zone of inhibition ranging from 15 to 30 mm, minimum inhibitory concentration between 250 and 900 μg/mL, and minimum bacterial concentration between 700 and 1000 μg/mL.  相似文献

2‐Bromo‐4‐(pyrrolidin‐1‐yl)pyridine‐3‐carbonitrile obtained from 2‐(1,3‐bis(pyrrolidin‐1‐yl)allylidene)malononitrile has been used as a substrate for the synthesis of new cyanopyridine derivatives: 2‐methoxy, 2‐phenoxy, 2‐aminoethylthio, and 2‐thioxo. 4‐(Pyrrolidin‐1‐yl)‐2‐thioxo‐1,2‐dihydropyridine‐3‐carbonitrile 7 in reaction with suitable alkyl and aminoalkyl halides gave respective sulfides. All synthesized compounds were evaluated for their antimicrobial activity against 26 aerobic and anaerobic bacteria. Determined minimal inhibitory concentration values ranged from 6.2 to 100 µg/mL. Derivatives 1 , 3 , 4 , 6 , and 12 were the most active compounds.  相似文献

Independent of the substrate structure and reaction conditions, 3‐amino‐2‐cyanothioacrylamides, which contain two active electrophilic centers, were shown to interact with various active halo methylene compounds under mild conditions to afford 5‐acyl‐2‐amino‐3‐cyanothiophenes as the only products. A series of new polyfunctional thiophene derivatives with a rare combination of functionalities were synthesized, and their photophysical properties were experimentally and computationally investigated. The calculated electronic characteristics of the ground and excited states were compared to the experimental results, which provided a good understanding of the relationship between the optoelectronic properties and the molecular structures. After absorption of light quanta, the systems populate an intramolecular charge‐transfer (ICT) Franck–Condon state, and emission occurs from a twisted ICT minimum.  相似文献

A simple and efficient approach to synthesize novel polysubstituted quinazolin‐4(3H)‐ones has been developed, and the key step is a sequential procedure involved iron‐mediated reduction and acid‐catalytic cyclization. The present method provides a convenient and practical strategy for the synthesis of quinazolin‐4(3H)‐one derivatives.  相似文献

The acid hydrazide derivatives 2 were converted into pyrazolone, triazinone, and schiff bases. However, the reaction of Schiff base 12 with malononitrile or thioglycolic acid gives the pyrazolotriazinone or thiazolidinone derivative, respectively. The structures of the newly synthesized compounds were established on the basis of IR, 1H‐NMR, mass spectral data, and elemental analyses. The antimicrobial activities of the synthesized compounds were examined against two types of bacteria and two types of fungi. Some of the tested compounds showed promising activities.  相似文献

A series of novel N ,N'‐disubstituted‐1,2,3,4‐tetrahydroquinoxalines were designed and synthesized by cyclization and acylation. The structures of all the novel compounds were confirmed by IR, 1H NMR, 13C NMR, and high‐resolution mass spectrometry. The configuration of 4d was determined by X‐ray diffraction. The preliminary biological tests showed that all the products could protect maize against the injury caused by acetochlor to some extent.  相似文献

A series of nitrogen‐tethered allenynes (‘5‐aza‐1,2‐dien‐7‐ynes’) 1 were transformed to the corresponding 3‐acyl‐4‐alkenylpyrrolidines 3 when treated with a catalytic amount of PtCl2 in MeOH at 70°. Initial Pt‐promoted cyclization forms a nonclassical carbocationic intermediate. In contrast to the cycloisomerization in toluene, which produced the bicyclic cyclobutenes 2 , the intermediate is intercepted by addition of an oxygen nucleophile to achieve the formal hydrative cyclization.  相似文献

Seven new substituted 5‐phenoxy‐1,2,4‐dithiazole‐3‐ones were prepared in modest yield (53–76%) from corresponding O‐phenyl thiocarbamates and chlorocarbonylsulfenyl chloride in dry ether at ?10 °C. All of the compounds were characterized by NMR and elemental analysis and some of them by X‐ray diffraction. Preliminary kinetic measurements showed that the parent 5‐phenoxy‐1,2,4‐dithiazole‐3‐one is a very efficient sulfurizing agent toward triphenyl phosphite. J. Heterocyclic Chem., (2011)  相似文献

A novel series of 2,4‐disubstituted‐1,2,4‐triazolo[1,5‐a]quinazolin‐5(4H)‐ones were prepared by Dimroth rearrangement of their respective isomers namely 1,4‐disubstituted‐[1,2,4]triazolo[4,3‐a]‐quinazolin‐5(4H)‐ones. The latter were prepared via new synthetic strategy based on 1,5‐elecrocyclization of the respective N‐(4‐oxo‐3‐phenylquinazolin‐2‐yl)nitrilimines.  相似文献

Various 3‐fluoropyrrolidines and 4‐fluoropyrrolidin‐2‐ones were prepared by 5‐exo‐trig iodocyclisation from allylic fluorides bearing a pending nitrogen nucleophile. These bench‐stable precursors were made accessible upon electrophilic fluorination of the corresponding allylsilanes. The presence of the allylic fluorine substituent induces syn‐stereocontrol upon iodocyclisation with diastereomeric ratios ranging from 10:1 to > 20:1 for all N‐tosyl‐3‐fluoropent‐4‐en‐1‐amines and amides. The sense and level of stereocontrol is strikingly similar to the corresponding iodocyclisation of structurally related allylic fluorides bearing pending oxygen nucleophiles. These results suggest that the syn selectivity observed upon ring closure involves I2–π complexes with the fluorine positioned inside.  相似文献
