Two series of combinatorial library of 3,6‐disubstituted‐7H‐1,2,4‐triazolo‐[3,4‐b]‐1,3,4‐thiadiazines bearing an amide linkage were synthesized. All the newly synthesized compounds were characterized by spectral analyses. The newly synthesized compounds were screened for their cytotoxicity, anti‐inflammatory, and analgesic activities. Among the tested compounds, the compound 9g (Ar = 4‐(methoxybenzyl)piperazine) is the most promising molecule with half maximal inhibitory concentration (IC50) value of 14.24 μM in (MCF‐7) cells. Compounds 9f (Ar = 4‐(chlorobenzyl)piperazine), 9g , and 9k (Ar = 2‐(fluorophenyl)piperazine) exhibited excellent anti‐inflammatory activity at a dose level of 50 mg/kg, almost comparable with the standard drug. In case of analgesic activity among the tested compounds, the compounds 9f , 9g , and 9k showed more potent and consistent activity in both 100 and 200 mg/kg/po doses with less ulcerogenic risk.  相似文献

A series of 3‐(4‐phenylisothiazol‐5‐yl)‐2H‐chromen‐2‐one ( 6a – l ) derivatives has been efficiently synthesized by straightforward sequential reactions. Tandem Vilsmeier Hack reaction/cyclization/bromination/Suzuki cross‐coupling reactions were successfully applied to the preparation of title compounds in good‐to‐high yields. In the synthetic sequences, 3‐chloro‐3‐(2‐oxo‐2H‐chromen‐3‐yl)acrylaldehydes ( 2 ) were found to react with ammonium thiocyanate to yield the corresponding 3‐(isothiazol‐5‐yl)‐2H‐chromen‐2‐ones ( 3 ). These derivatives were brominated with N‐bromo succinamide to yield the corresponding regioselective 3‐(4‐bromoisothiazol‐5‐yl)‐2H‐chromen‐2‐one ( 4 ). Finally, compound 4 was treated with various phenyl/pyrazole/7H –pyrrolo[2,3‐d]pyrimidinyl boronic acids 5a – l in the presence of K2CO3 and Pd catalyst in dimethylformamide to yield the corresponding title derivatives 6a – l . All the synthesized compounds were characterized by analytical and spectral studies. All the final compounds were screened against different cancer cell lines (A549, PC3, SKOV3, and B16F10), and among these compounds, 6b , 6g , 6h , and 6l displayed moderate cytotoxic activity against the tested cell lines.  相似文献

In the present study, a series of novel pyrido[1,2‐a]pyrimidin‐4‐one derivatives ( 1 – 45 ) were synthesized, characterized, and evaluated for their anti‐inflammatory activity. The structures of all newly synthesized compounds were confirmed by 1H NMR, 13C NMR, mass spectroscopy, and C, H, and N analyses. Preliminary these newly synthesized compounds were evaluated for their in vitro cyclooxygenase (COX)‐2/COX‐1 inhibitory activity. The celecoxib, a COX‐2 inhibitor, was used as a reference standard drug. In this inhibitory study, compounds 42 , 43 , 44 , and 45 were found to have significant in vitro inhibitory profile as compared with the reference drug. These compounds were then subjected to their in vivo anti‐inflammatory assay by using carrageenan‐induced rat paw edema method in next level of screening. Later, these same compounds were tested for their ulcerogenic property. Based on these activity data, the compound 43 (in vitro COX‐2 activity—IC50 = 0.4 μM, SI = 400, in vivo anti‐inflammatory activity—72% inhibition after 3 h, and 0.38%—Ulcer index) was emerged as most promising anti‐inflammatory agent with very low ulcerogenic action.  相似文献

A series of 2‐substituted‐6‐(morpholinyl/piperidinyl)pyridazin‐3(2H )‐ones was synthesized and the structures were established using various spectroscopic techniques. The target compounds were screened for anti‐inflammatory and analgesic activities at 20 and 40 mg/kg. The safety of the synthesized derivatives was evaluated by assessing anti‐platelet activity and ulcer index. The obtained pharmacological data revealed that 6‐morpholinyl derivatives 4a–12a were found to be somewhat more potent than 6‐piperidinyl derivatives 4b–6b. The 6‐morpholinyl substituted pyridazinone 12a exhibited maximum anti‐inflammatory and analgesic activities. Homoveratrylamine substituted compounds 6a and 6b emerged as promising leads in both the series with good anti‐inflammatory and analgesic activities without any ulcerogenicity. Anti‐platelet activity results of the compounds of both the series showed significantly low bleeding time in comparison with standard drug aspirin indicating the cardiovascular safety of new pyridazinones.  相似文献

In view of developing novel bioactive compounds, a series of 2‐(5‐[2‐methyl‐6‐arylpyridin‐3‐yl]‐1,3,4‐oxadiazol‐2‐ylthio)‐1‐arylethanones (6a–n) were designed and synthesized in good yield. Novel compounds were evaluated for their antibacterial and anti‐inflammatory activities. All synthesized compounds were screened for their antibacterial activity against Staphylococcus aureus, Bascillus subtilis, Eschericia coli, and Pseudomonas aeruginosa strains. Compounds 6a , 6b , 6c , 6h , and 6i displayed the highest antibacterial activity with minimal inhibitory concentration (MIC) values ranging from 6.25–12.5 μg/mL in comparison with the standard Ciprofloxacin. The results of anti‐inflammatory activity of carrageenan‐induced footpad edema assay indicated that tested compounds exhibited remarkable anti‐inflammatory activity with percentage of inhibition of 63.9–70.1% (potency 96.8–106.20% of indomethacin activity) after 3 hr. Particularly, 6c – e and 6j – l were found to be excellent inhibitors of inflammation, with potential higher than that of the standard, Indomethacin.  相似文献

A novel class of 3‐(4‐chlorophenyl)‐2‐(substituted)quinazolin‐4(3H)‐one derivatives were synthesized, and the structure of synthesized compounds was characterized by IR, 1H NMR, and mass spectroscopy. The newly synthesized compounds ( 4a–g and 6a–g ) were tested for their in vitro cyclooxygenase (COX) inhibition activity. The compounds have inhibitory profile against both COX‐1 and COX‐2, and some of the compounds are found to be selective against COX‐2. The compound 6g showed distinct inhibitory activity on COXs. The synthesized compounds were evaluated for their potential anti‐inflammatory activity as inhibitors of the proinflammatory cytokines IL‐6. Compounds 4d – g showed the highest level of inhibition among all the tested compounds. Thus, our data suggested that these compounds may represent a new class of potent anti‐inflammatory agents.  相似文献

A new quinazolinone derivative, 3‐[1‐(2‐hydroxyphenyl)ethylideamino]‐2‐phenyl‐3,4‐dihydroquinazolin‐4(3H)‐one ( LH ) was synthesized by the condensation of 2‐hydroxyacetophenone‐2‐aminobenzoylhydrazone and benzaldehyde. The cyclization to form 1,2‐dihydroquinazolinone was confirmed by IR, 1D and 2D HETCOR studies. Coordination compounds of Co(II), Ni(II), Cu(II) and Zn(II) of LH were synthesized and characterized using various physico‐chemical studies like stoichiometric, conductivity, magnetic moment measurements and spectral techniques such as IR, NMR, UV‐vis and EPR spectroscopy. The elemental analysis and thermal studies suggested a general stoichiometry [M(HEPDQ)Cl] for all the complexes. A four‐coordinate geometry was assigned to all the complexes. The complexes along with the parent ligand were screened for their anti‐inflammatory activity, using carrageenan‐induced rat paw edema, and for their analgesic activity by Eddy's hot plate method. The activity of the ligand was enhanced on complexation with metal ions. This enhanced activity was attributed to the increased lipophilic nature of the complexes. Notable anti‐inflammatory activity was observed for Ni(II), Cu(II) and Zn(II) complexes. The analgesic activity of the ligand was greater than the standard at 60 min. and at a 10 mg kg−1 dose, whereas the activity of Ni(II) and Cu(II) complexes at 10 mg kg−1 dose was comparable with the standard used. Some new derivatives of 3‐chloro‐1‐(4a,10b‐diazaphenanthrene‐2‐yl)‐4‐phenyl azetidin‐2‐one were synthesized through the reaction of N‐{4‐[phenyldiazenyl] phenyl}‐N‐[phenyl methylene] amine with 4‐[phenyldiazenyl] aniline. The resulting 3‐chloro‐4‐phenyl‐1‐{4‐[phenyldiazenyl] phenyl} azetidin‐2‐one intermediate in benzene was irradiated in a Pyrex vessel with 350 nm UV light in a photochemical reactor to give the desired derivatives (4a–j) . Structures of the new compounds were verified on the basis of spectral and elemental methods of analyses. Nine of the prepared compounds were tested for their anti‐inflammatory effects; most of these compounds showed potent and significant results compared with indomethacin.  相似文献

A series of 3,4‐bis‐chalcone‐N‐arylpyrazoles 3a‐h was prepared conveniently from diacetyl pyrazoles 2a,b . All reactions were carried out under conventional thermal heating and/or microwave irradiation. The structure of the latter functionally pyrazoles was confirmed under the bases of their IR, mass, 1H NMR and 13C NMR. The X‐ray diffraction of compound 3e not only confirmed the chemical structure of 3a‐h , but also showed the E configuration of their chalcone moieties. Treatment of compound 3e with phenyl hydrazine in presence of acetic acid afforded the tri‐pyrazle 4 . The anti‐inflammatory activity of the newly synthesized compounds was investigated. Some of these compounds showed a moderate activity when compared with indomethacin as a reference drug. The combination between chalcone and pyrazole moieties revealed a variable effect in anti‐inflammatory activity.  相似文献

A series of novel N‐{5‐[2‐(4,6‐dimethoxy‐pyrimidin‐2‐yloxy)‐phenyl]‐[1,3,4]thiadiazol‐2‐yl}2‐aroxy‐propanamides were designed and synthesized by the multistep reactions. 2‐(4,6‐Dimethoxy‐pyrimidin‐2‐yloxy)‐benzaldehyde ( 1 ) reacted with aminothiourea to yield 2 , which undergoes ring closure to give 5‐[2‐(4,6‐dimethoxy‐pyrimidin‐2‐yloxy)‐phenyl]‐[1,3,4]thiadiazol‐2‐amine ( 3 ) in the presence of ferric chloride in refluxing ethanol. 3 reacted with 2‐aroxy‐propionyl chlorides to give the target compounds 4a , 4b , 4c , 4d , 4e , 4f , 4g , 4h , 4i in moderate to good yields. Their structures were confirmed by IR, 1H‐NMR, EIMS, and elemental analyses. The preliminary bioassay indicated that some of them displayed moderate to good selective herbicidal activity against Brassica campestris L. at the concentration of 100 mg/L. However, these compounds did not possess inhibitory activity against Echinochloa crus‐galli at the tested concentrations.  相似文献

Synthesis, spectral analysis, and antibacterial activity of new coumarin derivatives are described in this paper. Twelve new coumarin derivatives were synthesized in moderate to good yields by the react with 4‐methyl‐6‐(prop‐2‐ynyloxy)‐2H‐chromen‐2‐one ( 3a – c ) and ethyl azide ( 4a – l ) and done by the click reaction to obtained 6‐[(l‐ethyl‐lH‐l,2,3‐triazol‐4‐yl)methoxy]‐4‐methyl‐2H‐chromen‐2‐ones ( 5a – l ). The structures of all the newly synthesized molecules were assigned by elemental analysis and spectral data. In continuation to our search for new heterocyclic system‐based anti‐inflammatory activity, a series of novel 1,2,3‐benzotriazine derivatives were synthesized. The pharmacological screening showed that many of these compounds have good anti‐inflammatory activity. A novel series of 2,4‐disubstituted oxazole derivatives were synthesized, screened for their anti‐tumor activity against three cell lines MCF‐ 7 , TK‐10, and UACC‐62. Molecular docking study was carried out against epidermal growth factor receptor. A new series of 2‐phenyl‐4‐substituted oxazole derivatives were synthesized. A series of chiral α‐amino acid derivatives 6 , 7 , 8 , 9 , 10 , 11 , 12 , 13 , 14 , 15 were synthesized by coupling various l ‐acylated amino acid azide 3. The synthesized compounds were tested for their in vitro antitumor activity against MCF‐7, TK‐10, and UACC‐62 cell lines. Compound 6 exhibited the strongest inhibitory activity against TK cell lines, while compound 12 showed the highest activity against MCF‐7 cell lines. Compound 14 was the most active against UACC‐62 cell lines. Furthermore, a molecular docking study of the most active compounds was carried out using epidermal growth factor receptor X‐ray 3D structure (protein data bank ID 1 M17). Docking results revealed that compound 6 showed the highest binding energy of ΔG = ?78.17 Kcal/mol.  相似文献

A series of novel isoxazolyl‐4‐(2‐oxo‐2,3‐dihydro‐1H‐3‐indolyl)pyrrole‐3‐carboxylates ( 17a – i) were synthesized by a three‐component reaction of 4‐amino‐3‐methyl‐5‐styrylisoxazole 14 , β‐keto ester 15 , and 3‐phenacylideneoxindole 16 , in the presence of CAN catalyst in ethanol. The structures of the synthesized compounds have been established on the basis of spectral and analytical data. The title compounds 17a – i were evaluated for their anti‐inflammatory activity. In the present investigation, a novel series of 3‐(4‐(2‐substituted thiazol‐4‐yl)phenyl)‐2‐(4‐methyl‐2‐substituted thiazol‐5‐yl)thiazolidin‐4‐one derivatives were synthesized by condensation of 2‐substituted‐4‐methylthiazole‐5‐carbaldehyde with 4‐(2‐substituted thiazol‐4‐yl)benzenamine followed by cyclo‐condensation with thioglycolic acid in toluene. All the newly synthesized compounds were characterized by spectral (IR, 1H NMR, 13C NMR, and Mass) methods. The title compounds were screened for quantitative antibacterial activity (minimal inhibitory concentration). All compounds 7a , 7b , 7c , 7d , 7e , 7f , 7g , 7h and 8a , 8b , 8c , 8d , 8e , 8f , 8g , 8h show moderate to good antimicrobial activity, whereas compounds ( 7a , 7b , 7c , 7d , 7e , 7f , 7g , 7h ) also show moderate antifungal activity.  相似文献

A series of novel 4‐(3,3‐dimethylspiro{bicyclo[2.2.1]heptan‐2,5′‐isoxazoline‐2}‐3′‐yl)‐2‐phenyl‐2,3‐dihydro‐1H‐1,5‐benzodiazepines were synthesized. An efficient and rapid synthesis of coumarin derivatives was accomplished via reactions of 3‐(3‐(4‐methoxyphenyl)acryloyl)‐2H ‐chromen‐2‐one ( 3 ) with different carbon nucleophiles such as ethyl acetoacetate, ethyl cyanoacetate, malononitrile, and ethyl benzoylacetate via conventional heating and microwave irradiation conditions and were used as source of pyran and pyridine derivatives bearing coumarin moiety 4 – 11 . Compound 9a was condensed with different carbon electrophiles triethylorthoformate, phenylisothiocyanate, carbon disulfide, benzoylchloride, and acetylchloride that afforded the corresponding chromen derivatives 12 – 16 . All the newly synthesized compounds were characterized by elemental and spectroscopic evidences. All of synthesized compounds were tested for in vitro cytotoxicity. The preliminary screening results showed that most of the compounds had moderate cytotoxic activity against HCT‐116 and MCF‐7 cell lines. Nevertheless, compound 10 exhibited potent activity against the two cell lines, which was comparable with the standard drug 5‐fluorouracil.  相似文献

Condensation of coumarin‐4‐acetic acids ( 1 ) with ortho‐phenylenediamine ( 2 ) in anhydrous phosphoric acid afforded 4‐((1H‐benzo[d]imidazol‐2‐yl)methyl)‐2H‐chromen‐2‐ones ( 3 ). Attempted Mannich reaction of 3 with formalin and primary amines resulted in 4‐(2‐phenyl‐1,2,3,4‐tetrahydrobenzo[4,5]imidazo[1,2‐c]pyrimidin‐4‐yl)‐2H‐chromen‐2‐ones ( 6 ). The structures of synthesized compounds were elucidated by analyses including 2D HETCOR and DEPT experiments. Synthesized compounds have been subjected for anti‐inflammatory activity. Compound 6j exhibited promising anti‐inflammatory activity.  相似文献

A series of novel 3‐(coumarin‐4‐yl)tetrahydroisoxazoles 5a,b, 7, 9 and 3‐(coumarin‐4‐yl)dihydropyra‐zoles 13a‐d, 14,15a,b were synthesized from coumarin‐4‐carboxaldehyde 1 via the intermediate N‐methyl nitrone 3 and N‐phenyl or N‐methyl hydrazones 11a,b . These coumarin derivatives were isolated, characterized and evaluated in vitro for their ability to inhibit trypsin, β‐glucuronidase, soybean lipoxygenase and to interact with the stable radical 1,1‐diphenyl‐2‐picrylhydrazyl. The compounds were tested in vivo as anti‐inflammatory agents in the rat carrageenin paw edema assay. Compound 15a seems to be a lead molecule to be modified in order to improve the lipoxygenase inhibition. The results are discussed in terms of structural characteristics.  相似文献
