Chalcones are known to act on various physiological targets. As a result, structural modifications of chalcones have been studied extensively. Benzochalcones, in which the A‐ring of chalcone is substituted with a naphthalene unit, inhibits breast cancer resistance protein. Chalcones in which the α,β‐unsaturated carbonyl group is switched with a pyrazoline moiety are potent cytotoxic agents against various cancer cell lines, and chalcones with a pyrazoline‐1‐carbothioamide group instead of an α,β‐unsaturated carbonyl group exhibit antimicrobial activities. The present report describes hybrid molecules designed from benzochalcone and pyrazoline–carbothioamide. Methoxylation of plant‐derived polyphenols alters their hydrophobicity, resulting in changes in biological function and intracellular compartmentation. In the current study, 22 novel methoxylated 3‐(naphthalen‐2‐yl)‐N,5‐diphenyl‐pyrazoline‐1‐carbothioamide derivatives were prepared. This report provides complete assignments of their 1H and 13C NMR data, which can be used to subsequently identify chalcones bearing pyrazoline–carbothioamide groups. In the present investigation, the first incorporation of both ferrocene scaffold and 1,2‐dihydro‐2‐oxopyridine‐3‐carbonitrile pharmacophore leading to a series of structurally novel ferrocene‐based hybrids has been achieved, involving the condensation reaction of ferrocenyl substituted chalcones with 2‐cyanoacetamide in a freshly prepared EtONa solution at 70°. The molecular structures of these newly synthesized products were confirmed by IR, and 1H‐ and 13C‐NMR analyses.  相似文献

Several types of chalcones containing 2H‐chromen group were synthesized. Claisen–Schmidt condensation of 2H‐chromen‐3‐carbaldehydes (I) with methoxy substituted acetophenones afforded (E)‐3‐(2H‐chromen‐3‐yl)‐1‐(methoxyphenyl)prop‐2‐en‐1‐ones (chromenylchalcones, 1–7). Other types of chromenylchalcone, (E)‐1‐(6‐methoxy‐2H‐chromen‐3‐yl)‐3‐(methoxyphenyl)prop‐2‐en‐1‐ones (8–13) were also obtained between reaction of methoxy substituted benzaldehydes and 1‐(6‐methoxy‐2H‐chromen‐3‐yl)ethanone (II). Dichromenylchalcones (14–16) were also synthesized through the same reaction between aldehydes (I) and ketone (II). Their complete 1H‐NMR and 13C‐NMR assignments are reported here and more polysubstituted chromenylchalcones synthesized or isolated from the natural sources in the future can be identified on the basis of the NMR data reported here. To find potent new chemotherapy drugs, we designed and synthesized a series of naphthochalcones bearing naphthalenyl‐phenyl‐pyrazoline moieties. The complete 1H and 13C NMR data for these compounds are reported here and can be used to identify further new naphthochalcones bearing the desired pyrazoline moieties. An unsymmetrical heterocyclic diamine, 1,2‐dihydro‐2‐(4‐aminophenyl)‐4‐[4‐(4‐aminophenoxy)‐4‐phenyl]‐(2H)phthalazin‐1‐one, was synthesized. Its 1H and 13C NMR spectra were completely assigned by utilizing the two‐dimensional heteronuclear 13C–1H multiple‐bond coherence (HMBC) spectroscopy, and heteronuclear 13C–1H one‐bond correlation spectroscopy, homonuclear shift correlation spectroscopy (H,H‐COSY) and rotating frame Overhauser enhancement spectroscopy (ROESY). The structure of the compound was shown to be the phthalazinone rather than the phthalazine ether from cross peaks and chemical shifts of the protons. 2-Acetyl naphthalene reacts with various methoxy substituted benzaldehyde in the presence of 10 % sodium hydroxide solution giving functionalized chalcones. The synthesized chalcones when further reacted with hydrazine hydrate in the presence of acetic acid afforded N-acetyl pyrazolines. All the synthesized products were confirmed by various spectral data such as FTIR, 1H NMR, 13C NMR, and HRMS studies. All the synthesized compounds were screened for cytotoxicity against various cell lines.  相似文献

A new route for the synthesis of 3,5‐diarylcyclohex‐2‐enones is reported. The 4‐acetyl‐1,3‐diarylhexane‐1,5‐diones were obtained by the addition of pentane‐2,4‐dione to chalcones. The reaction of 4‐acetyl‐1,3‐diarylhexane‐1,5‐diones with NH4Cl/HCl in EtOH under reflux conditions gave the 3,5‐diarylcyclohex‐2‐enones in good yields. All synthesized compounds were characterized by spectroscopic methods (1H‐, 13C‐NMR, and IR), and elemental analyses.  相似文献

15N NMR chemical shifts of 2‐aryl‐1,3,4‐oxadiazoles were assigned on the basis of the 1H–15N HMBC experiment. Chemical shifts of the nitrogen and carbon atoms in the oxadiazole ring correlate with the Hammett σ‐constants of substituents in the aryl ring (r2 ≥ 0.966 for N atoms). 15N NMR data are a suitable and sensitive means for characterizing long‐range electronic substituent effects. 3,4,5‐Triamino‐1,2,4‐triazolium iodide ( 1 ) was obtained in good yield and purity and characterized using vibrational (IR, Raman) and multinuclear NMR spectroscopy (1H, 13C, 15N), EA, MS, DSC, and X‐ray crystallography. The compound was synthesized by two different methods rendering two different polymorphs (α and β) as proved by X‐ray measurements, vibrational spectroscopy and DSC. 1‐Methyl‐3,4,5‐triamino‐1,2,4‐triazolium iodide ( 2 ) was synthesized by reaction of guanazine with methyliodide and fully characterized by the same techniques mentioned above. Both compounds showed to be suitable starting materials for the synthesis of guanazinium salts of energetic interest.  相似文献

A novel series of bis(3‐thienyl‐4,5‐dihydropyrazoles) has been synthesized by the cyclization reactions of bischalcones with phenyl hydrazine in basic medium. The O‐alkylation reactions of chalcones with suitable 1,ω‐dibromoalkanes in the presence of anhydrous K2CO3, dry acetone, and Bu4N+I as PTC lead to the formation of bischalcones in good yields. The chalcone required was obtained from the Claisen–Schmidt condensation reaction of 2‐acetylthiophene with 3‐hydroxybenzaldehyde. Structures of prepared compounds were elucidated from their IR, 1H‐NMR, 13C‐NMR, and ESI‐MS spectral data. Newly synthesized compounds were screened for their antimicrobial potencies against Gram‐positive, Gram‐negative bacterial strains, and fungal strains using serial tube dilution method. Docking simulations have also been carried out to visualize the possible interaction of synthesized scaffold 2(a – g) and 3(a – g) at the active sites of Escherichia coli .  相似文献

Herein, we describe the synthesis and complete assignment of the 1H and 13C NMR chemical shifts of a series of antimicrobial 4‐arylamino‐3‐nitrocoumarin derivatives based on a combination of 1H and 13C NMR, 1H‐1H‐COSY, NOESY, HSQC and HMBC experiments. Conformational effects upon the chemical shifts of the coumarin moiety arising from the anisotropy of the aryl side group are briefly discussed. This study provides the first complete and fully assigned NMR data for this important group of antimicrobial compounds and bridges the gap existing in the literature with regard to NMR structural data for 4‐arylamino‐3‐nitrocoumarins. 1H, 13C and two‐dimensional NMR analyses were applied to determine the NMR parameters of 6‐(2′,3′‐dihydro‐1′H‐inden‐1′‐yl)‐1H‐indene. The measurements were accomplished with 0.5 mg of the substance, this quantity being sufficient to determine the chemical shifts of all the H and C atoms, and also the appropriate coupling constants and to give the complete NMR resonance assignments of the molecule. The predicted patterns of the four different H atoms of the methylene groups of the indane structural element coincided completely with the complex patterns in the NMR spectra. A total of 13 of the 16 possible methylol derivatives of 2,4′‐ and 4,4′‐dihydroxydiphenylmethane have been synthesized, isolated, and identified. These compounds are found as intermediates in the cure process of resol phenol–formaldehyde (PF) resins. Analysis of the 13C NMR spectra (in acetone‐d6) of these compounds provided a way to evaluate the seven methylolphenol ring types (methylol derivatives of 2‐hydroxyphenyl and 4‐hydroxyphenyl rings) found in typical resol PF resins using the ipso carbon region from 150 to 160 ppm. A simple diagnostic test was developed using the chemical shift values of the methylol methylene carbon atoms to identify the presence of intermediates containing either a 2‐hydroxyphenyl or a 4‐hydroxyphenyl ring. The assignments of 1H and 13C NMR chemical shifts together with x‐ray diffraction data for synthesized diosgenyl 3,4,6‐tri‐O‐acetyl‐2‐deoxy‐2‐tetrachlorophthalimido‐β‐D ‐glucopyranoside are described. The structure of this glycoside was established by using homo‐ and heteronuclear two‐dimensional NMR techniques. X‐ray diffraction data for this compound are also reported. 1‐Acetyl‐ and 1‐propionyl‐2‐pyrazolines 11‐27 have been synthesized by the reaction of (3‐coumarinyl)chalcones 1‐10 with hydrazine in hot acetic acid or propionic acid. While 5‐aryl‐3‐(3‐coumarinyl)‐1‐phenyl‐2‐pyrazolines 28‐35 have been prepared by the reaction of (3‐coumarinyl)chalcones 1,3,5‐10 with phenylhydrazine in hot pyridine. Structures of all new compounds have been elucidated by microanalyses, 1H and 13C nmr spectroscopies.  相似文献

Paramagnetic effects from lanthanide ions present powerful tools for protein studies by nuclear magnetic resonance (NMR) spectroscopy provided that the lanthanide can be site‐specifically and rigidly attached to the protein. A new, particularly small and rigid lanthanide‐binding tag, 3‐mercapto‐2,6‐pyridinedicarboxylic acid (3MDPA), was synthesized and attached to two different proteins via a disulfide bond. The complexes of the N‐terminal domain of the E. coli arginine repressor (ArgN) with seven different paramagnetic lanthanide ions and Co2+ were analyzed in detail by NMR spectroscopy. The magnetic susceptibility anisotropy (Δχ) tensors and metal position were determined from pseudocontact shifts. The 3MDPA tag generated very different Δχ tensor orientations compared to the previously studied 4‐mercaptomethyl‐DPA tag, making it a highly complementary and useful tool for protein NMR studies.  相似文献

N‐acetyl‐4‐nitrotryptophan methyl ester (2), N‐acetyl‐5‐nitrotryptophan methyl ester (3), N‐acetyl‐6‐nitrotryptophan methyl ester (4) and N‐acetyl‐7‐nitrotryptophan methyl ester (5) were synthesized through a modified malonic ester reaction of the appropriate nitrogramine analogs followed by methylation with BF3‐methanol. The configuration at C‐3 of the 3α‐ and 3β‐hydroxy metabolites of tibolone was studied by extensive application of one‐ and two‐dimensional 1H and 13C NMR spectroscopy combined with molecular modeling performed at the B3LYP/6–31G(d) level. Using HF and DFT GIAO methods, shielding tensors of the two molecules were computed; comparison of the calculated NMR chemical shifts with the experimental values revealed that the density functional methods produced the best results for assigning proton and carbon resonances. Although steroids are relatively large molecules, the present approach appears accurate enough to allow the determination of relative configurations by using calculated 13C resonances; the chemical shift of pairs of geminal α/β hydrogen atoms can also be established by using calculated 1H resonances. A small library of thirty‐two 2′‐triazolyl uridine and 2′‐triazolyl‐5‐methyluridine has been synthesized by Cu(I)‐catalyzed condensation of 2′‐azido‐2′‐deoxyuridine and 2′‐azido‐2′‐deoxy‐5‐methyluridine with different alkynes and aryl propargyl ethers in almost quantitative yields. Triazolo‐nucleoside conjugates, which can be evaluated for different biological activity for suitable drug development, were unambiguously identified on the basis of 1H NMR, 13C NMR, IR, and HRMS data analysis. These compounds have been synthesized for the first time and have not been reported in the literature earlier.  相似文献
