Derivatized β‐cyclodextrin (β‐CD) functionalized monolithic columns were prepared by a “one‐step” strategy using click chemistry. First, the intended derivatized β‐CD monomers were synthesized by a click reaction between propargyl methacrylate and mono‐6‐azido‐β‐CD and then sulfonation or methylation was carried out. Finally, monolithic columns were prepared through a one‐step in situ copolymerization of the derivatized β‐CD monomer and ethylene glycol dimethacrylate. The sulfated β‐CD‐based monolith was successfully applied to the hydrophilic interaction liquid chromatography separation of nucleosides and small peptides, while the methylated β‐CD‐functionalized monolith was useful for the separation of nonpolar compounds and drug enantiomers in capillary reversed‐phase liquid chromatography. The structures of the monomers were characterized by Fourier transform infrared spectroscopy and mass spectrometry. The physicochemical properties and column performance of monoliths were evaluated by scanning electron microscopy and micro high performance liquid chromatography. This strategy has considerable prospects for the preparation of other derivatized CD‐functionalized methacrylate monoliths.  相似文献

To further evaluate the feasibility and applicability of the one‐pot strategy in monolithic column preparation, two novel β‐cyclodextrin‐functionalized organic polymeric monoliths were prepared using two β‐cyclodextrin derivatives, i.e. mono(6‐amino‐6‐deoxy)‐β‐cyclodextrin and heptakis(6‐amino‐6‐deoxy)‐β‐cyclodextrin. In this improved method, mono(6‐amino‐6‐deoxy)‐β‐cyclodextrin or heptakis(6‐amino‐6‐deoxy)‐β‐cyclodextrin reacted with glycidyl methacrylate to generate the corresponding functional monomers and were subsequently copolymerized with ethylene dimethacrylate. The polymerization conditions for both monoliths were carefully optimized to obtain satisfactory column performance with respect to column efficiency, reproducibility, permeability, and stability. The obtained poly(glycidyl methacrylate‐mono(6‐amino‐6‐deoxy)‐β‐cyclodextrin‐co‐ethylene dimethacrylate) and poly(glycidyl methacrylate‐heptakis(6‐amino‐6‐deoxy)‐β‐cyclodextrin‐co‐ethylene dimethacrylate) monoliths exhibited a uniform structure, good permeability, and mechanical stability as indicated by scanning electron microscopy and micro‐high‐performance liquid chromatography experimental results. Because of the probable existence of multi‐glycidyl methacrylate linking spacers on the poly(glycidyl methacrylate‐heptakis(6‐amino‐6‐deoxy)‐β‐cyclodextrin‐co‐ethylene dimethacrylate) monolith, the effect of the ratio of glycidyl methacrylate/heptakis(6‐amino‐6‐deoxy)‐β‐cyclodextrin was especially studied, and satisfactory reproducibility could still be achieved by strictly controlling the composition of the polymerization mixture. To investigate the effect of the degree of amino substitution of β‐cyclodextrin on column performance, a detailed comparison of the two monoliths was also carried out using series of analytes including small peptides and chiral acids. It was found that the β‐cyclodextrin‐functionalized monolith with mono‐glycidyl methacrylate linking spacers demonstrated better chiral separation performance than that with multi‐glycidyl methacrylate linking spacers.  相似文献

To evaluate the effect of the preparation strategy on the enantioseparation performance of β‐cyclodextrin‐functionalized monoliths, a series of β‐cyclodextrin‐functionalized organic polymeric monolithic columns were prepared through two‐step, single‐step, and one‐pot approaches, using the same cyclodextrin, linker–spacer, and crosslinker. Physicochemical characterization of the columns was carried out by determining the morphology, β‐cyclodextrin density, permeability, and chromatographic efficiency. For each type of monolithic column, the enantioresolution of 22 chiral compounds, including mandelic acid derivatives, nonsteroidal anti‐inflammatory drugs, N‐derivatized amino acids, and herbicides, was comparatively studied under optimum chromatographic conditions. The β‐cyclodextrin‐functionalized monolithic columns prepared through the one‐pot approach exhibited higher enantioresolution for most chiral compounds, and they have the advantage of good controllability and simple preparation. On the other hand, the enantioresolution obtained on columns prepared through the single‐step approach was quite unsatisfactory, and therefore the effect of using different linking spacers and crosslinkers was studied. A significant improvement of enantioresolution for 2‐chloro‐mandelic acid was obtained by using N ,N‐methylenebisacrylamide instead of ethylene dimethacrylate as the crosslinker in the single‐step preparation.  相似文献

Copper(0)‐catalyzed one‐pot reaction combining living radical polymerization and “click chemistry” was investigated. By precisely tuning reaction time, three novel well‐defined polymers with different degree of carboxyl substitution, poly(propargyl methacrylate) (PPgMA), poly(1‐(4‐carboxyphenyl)‐[1,2,3]triazol‐4‐methyl methacrylate) (PCTMMA), and poly(1‐(4‐carboxyphenyl)‐[1,2,3]triazol‐4‐methyl methacrylate‐co‐propargyl methacrylate) (PCTMMA‐co‐PPgMA) were selectively obtained via Cu(0) powder/N,N,N′,N″,N″‐pentamethyldiethylenetriamine (PMDETA) cocatalyzed LRP and click chemistry. In addition, gel permeation chromatography and 1H NMR analysis in conjunction with FTIR spectroscopy elucidate that one‐pot process undergoes three steps due to a pronounced rate enhancement of click reaction: (1) generating new monomer, 1‐(4‐carboxyphenyl)‐[1,2,3]triazol‐4‐methyl methacrylate (CTMMA); (2) copolymerization of two monomers (CTMMA and PgMA); (3) building homopolymer PCTMMA. Surprisingly, in contrast to typical Cu(I)‐catalyzed atom transfer radical polymerization (ATRP), copper(0)‐catalyzed one‐pot reaction showed high carboxylic acid group tolerance. © 2012 Wiley Periodicals, Inc. Neutral diol methacrylate‐based monoliths were developed for normal phase chromatography (NPC) and NP‐CEC of polar compounds including N‐glycans. Four different diol methacrylate‐based monoliths were synthesized via the copolymerization of a functional monomer using either glyceryl monomethacrylate (GMM) or glycidyl methacrylate (GMA) and a crosslinker either ethylene dimethacrylate (EDMA) or trimethylolpropane trimethacrylate (TRIM). While the GMM‐based monoliths yield in one reaction step polar diol methacrylate monoliths that are ready for use in NPC or NP‐CEC, the GMA‐based monoliths required a postmodification with hot sulfuric acid to convert the epoxy functions into diols before use in NPC or NP‐CEC. All the four monoliths are neutral and void of fixed charges on their surfaces but yet exhibited relatively strong EOF in NP‐CEC. The EOF is attributed to the adsorption of ions from the mobile phase thus forming the electric double layer necessary for producing a bulk mobile phase flow. Under the same in situ copolymerization conditions of GMM or GMA with either EDMA or TRIM, the GMM–EDMA monolith was the best choice in terms of retention, separation efficiency, EOF velocity in CEC and linear flow velocity in Nano‐LC.  相似文献

Synthesis of some new oxadiazole derivatives starting from 1,2,3-benzo[d]triazole-1-acetic hydrazide (1) is described. The target compounds 2-(N-substituted-aminocarbonylmethylthio)-5-(1,2,3-benzo[d]triazol-1-ylmethyl)- 1,3,4-oxadiazole (4a—4i) and 2-[2-(N-substituted-aminocarbonyl)ethylthio]-5-(1,2,3-benzo[d]triazol-1-ylmethyl)- 1,3,4-oxadiazole (5a—5i) were obtained in good yields via cyclisation of 1 and subjected to antibacterial activity test against pathogenic bacteria. The halogen containing mono- and di-substituted derivatives showed excellent antibacterial activity compared to other analogues.  相似文献

A novel construction strategy of monolithic capillary column for selectively enriching perfluorinated persistent organic pollutants was proposed. The organic–inorganic hybrid fluorous monolithic capillary column was synthesized by a “one‐pot” approach via the polycondensation of γ‐methacryloxypropyltrimethoxy‐silane, then in situ copolymerization of 1H,1H,7H‐dodecafluoroheptyl methacrylate and vinyl group on the precondensed siloxanes. The obtained monolithic columns were systematically characterized. The results demonstrated that the optimal column possessed good mechanical stability and high permeability. The adsorption capacities of the optimized monolithic column for perfluorooctanoic acid and perfluorooctane sulfonate were 0.257 and 0.513 μg/mg, respectively. Adsorption capacities of the monoliths were proved to increasing with increasing the amounts of fluorinated monomers in the fluorous monoliths. Sodium 1‐octanesulfonate, as a comparison compound, was hardly adsorbed on the fluorous monolith. In addition, the trace amounts of perfluorooctanoic acid and perfluorooctane sulfonate in water samples can be successfully concentrated about 160 times to their original concentrations by this monolithic column. These results demonstrated that the capacity and selectivity of the affinity fluorous column is high and can be applied to the selective enrichment for the perfluorinated persistent organic pollutants from environmental samples.  相似文献

Herein, we report the selective mono‐derivatization of heptakis[6‐deoxy‐6‐(2‐aminoethylsulfanyl)]‐β‐CD ( 1 ) through a guest‐mediated covalent capture strategy. The use of guests functionalized with cleavable linkers enables the installation of an amine‐orthogonal thiol group on the primary rim of 1 as a handle for further transformations to the β‐CD scaffold. Applying this methodology, two novel monoderivatized β‐CDs were obtained in good yield and high purity. Both of these monoacylated CDs were amenable to facile linker cleavage and further modification at the resulting thiol group. This methodology can be applied towards the synthesis heterofunctionalized β‐CD constructs for analyte sensing, drug delivery, and other applications.  相似文献

In this paper, a novel highly cross‐linked porous monolithic stationary phase having a long alkyl chain ligand (C16) was introduced and evaluated in CEC. The monolithic stationary phase was prepared by in situ copolymerization of 1‐hexadecene, trimethylolpropane trimethacrylate, and 2‐acrylamido‐2‐methyl‐1‐propanesulfonic acid (AMPS) in the presence of ternary porogenic solvent (cyclohexanol/1,4‐butanediol/water). In preparing monoliths, the ternary cross‐linker trimethylolpropane trimethacrylate was usually applied to preparing molecularly imprinted polymers or molecularly imprinted solid‐phase extraction, instead of binary cross‐linker ethylene dimethacrylate. 1‐Hexadecene was introduced to provide the non‐polar sites (C16) for chromatographic retention, while AMPS was used to generate the EOF for transporting the mobile phase through the monolithic capillary. Monolithic columns were prepared by optimizing proportion of porogenic solvent and AMPS content in the polymerization solution as well as the cross‐linkers. The monolithic stationary phases could generate a strong and stable EOF in various pH values and exhibit an RP‐chromatographic behavior for neutral compounds. For charged compounds, the separation was mainly based on the association of hydrophobic, electrostatic and electrophoretic interaction.  相似文献

Orthometalation at IrIII centers is usually facile, and such orthometalated complexes often display intriguing electronic and catalytic properties. By using a central phenyl ring as C?H activation sites, we present here mono‐ and dinuclear IrIII complexes with “click”‐derived 1,2,3‐triazole and 1,2,3‐triazol‐5‐ylidene ligands, in which the wingtip phenyl groups in the aforementioned ligands are additionally orthometalated and bind as carbanionic donors to the IrIII centers. Structural characterization of the complexes reveal a piano stool‐type of coordination around the metal centers with the “click”‐derived ligands bound either with C^N or C^C donor sets to the IrIII centers. Furthermore, whereas bond localization is observed within the 1,2,3‐triazole ligands, a more delocalized situation is found in their 1,2,3‐triazol‐5‐ylidene counterparts. All complexes were subjected to catalytic tests for the transfer hydrogenation of benzaldehyde and acetophenone. The dinuclear complexes turned out to be more active than their mononuclear counterparts. We present here the first examples of stable, isomer‐pure, dinuclear cyclometalated IrIII complexes with poly‐mesoionic‐carbene ligands.  相似文献

To improve resolution power of chiral selector and enantiomeric peak efficiency in CE, single isomer negatively charged β‐CD derivatives, mono(6‐deoxy‐6‐sulfoethylthio)‐β‐CD (SET‐β‐CD) bearing one negative charge and mono[6‐deoxy‐6‐(6‐sulfooxy‐5,5‐bis‐sulfooxymethyl)hexylthio]‐β‐CD (SMHT‐β‐CD) carrying three negative charges, were synthesized. The structure of these two β‐CD derivatives was confirmed by 1H NMR and MS. SET‐β‐CD and SMHT‐β‐CD successfully resolved the enantiomers of several basic model compounds. SMHT‐β‐CD provided for a significantly greater enantioseparation than SET‐β‐CD at lower concentrations. This appears to be due to the higher binding affinity of SMHT‐β‐CD to the model compounds and the wider separation window resulting from an increased countercurrent mobility of the selector. Overall, the new chiral selectors provided enantioseparations with high peak efficiency while avoiding peak distortion due to polydispersive and electrodispersive effects. The information obtained from an apparent binding constant study suggested that the enantioseparation of the model compounds followed the predictions of charged resolving agent migration model and that the observed degree of enantioseparation difference were due to the magnitude of differences in both enantiomer‐chiral selector binding affinities (ΔK) and the mobilities of the complexed enantiomers (Δμc).  相似文献

Ultrashort monolithic columns (disks) were thoroughly studied as efficient stationary phases for precipitation–dissolution chromatography of synthetic polymers. Gradient elution mode was applied in all chromatographic runs. The mixtures of different flexible chain homopolymers, such as polystyrenes, poly(methyl methacrylates), and poly(tert‐butylmethacrylates) were separated according to their molecular weights on both commercial poly(styrene‐co‐divinylbenzene) disks (12 id × 3 mm and 5 × 5 mm) and lab‐made monolithic columns (4.6 id × 50 mm) filled with supports of different hydrophobicity. The experimental conditions were optimized to reach fast and highly efficient separation. It was observed that, similar to the separation of monoliths of other classes of (macro)molecules (proteins, DNA, oligonucleotides), the length of column did not affect the peak resolution. A comparison of the retention properties of the poly(styrene‐co‐divinylbenzene) disk‐shaped monoliths with those based on poly(lauryl methacrylate‐co‐ethylene dimethacrylate), poly(butyl methacrylate‐co‐ethylene dimethacrylate), and poly(glycidyl methacrylate‐co‐ethylene dimethacrylate) supports demonstrated the obvious effect of surface chemistry on the resolution factor. Additionally, the results of the discussed chromatographic mode on the fast determination of the molecular weights of homopolymers used in this study were compared to those established by SEC on columns packed with sorbent beads of a similar nature to the monoliths.  相似文献

An efficient novel method for the synthesis of a covalent molecularly imprinted polymer (MIP) highly specific to β‐estradiol have been developed. MIP prepared by both covalent and non covalent techniques, demonstrated high selectivity toward β‐estradiol. MIPs were synthesized by radical polymerization of 17‐β‐estradiol 4‐vinyl‐benzene carboxyl or sulfonyl esters used as covalent functional monomers, methacrylic acid as noncovalent functional monomer, ethylene glycol dimethacrylate as crosslinking agent, and acetonitrile as swelling and porogenic component. Almost 35% (w/w) of 17‐β‐estradiol was successfully removed from the polymer network by basic hydrolysis. The binding ability of MIP was 10.73 μg/mg MIP following removal of 17‐β‐estradiol in the 2 mg/mL β‐estradiol solution. Selective rebinding of β‐estradiol toward MIP was tested in the presence of competitive binders including estrone, 19‐nortestosterone, epiandrosterone, and cholesterol. Estrone having closest similar chemical structure to β‐estradiol exhibited only 0.6 μg/mg MIP competitive binding, being exposed to equivalent concentrations. Moreover, other competitive steroids demonstrated negligible affinity toward MIP indicating high selectivity of novel MIP system toward β‐estradiol. © 2009 Wiley Periodicals, Inc. The radical polymerization of Ntert‐butyl‐N‐allylacrylamide (t‐BAA) was carried out in a dimethyl sulfoxide/H2O mixture in the presence of β‐cyclodextrin (β‐CD). The polymerization proceeded with the complete cyclization of the t‐BAA unit and yielded optically active poly(t‐BAA). The IR spectrum of the obtained polymer showed that the cyclic structure in the polymer was a five‐membered ring. The optical activity of poly(t‐BAA) increased with an increasing molar ratio of β‐CD to the t‐BAA monomer. The interaction of β‐CD with t‐BAA was confirmed by 1H NMR and 13C NMR analyses of the polymerization system. It is suggested that interaction of the t‐BAA monomer with the hydrophobic cavity of β‐CD plays an important role in the asymmetric cyclopolymerization of t‐BAA. The radical copolymerization of t‐BAA with styrene (St), methyl methacrylate, ethyl methacrylate, or benzyl methacrylate (BMA) also produced optically active copolymers with a cyclic structure from the t‐BAA unit. St and BMA carrying a phenyl group were predicted to compete with t‐BAA for interaction with β‐CD in the copolymerization system. © 2000 John Wiley & Sons, Inc. A well‐defined structure liquid crystal heptakis [6‐deoxy‐6‐(1‐H‐1,2,3‐triazol‐4‐yl)(methyl)6‐(4‐methoxybiphenyl‐4′‐yloxy) hexanoyl]‐β‐cyclodextrin (H6B‐β‐CD) was synthesized from propargyl 6‐(4‐methoxybiphenyl‐4′‐yloxy) hexanoate (P6B) and heptakis (6‐deoxy‐6‐azido)‐β‐cyclodextrin ((N3)7‐β‐CD) by click reaction. The chemical structure of H6B‐β‐CD was confirmed by 1H NMR, FTIR, and MALDI‐TOF MS. The thermal stability of the compound was investigated by thermogravimetric analysis (TGA). The liquid crystalline behavior was studied by differential scanning calorimetry (DSC), polarizing optical microcopy (POM), and wide‐angle X‐ray diffraction (WAXD) measurement. These investigations have shown that the supramolecular structure of H6B‐β‐CD are consisted of a large scale ordered lamellar structure and a small scale ordered structure (SmE) at low temperature region. As the temperature increases, the small scale structure becomes disordered relatively in the first instance, from smectic E to smectic A. The synthesis of a variety of 2‐(1H‐1,2,3‐triazol‐4‐yl)‐pyridines by click chemistry is demonstrated to provide straightforward access to mono‐functionalized ligands. The ring‐opening polymerization of ε‐caprolactone initiated by such a mono‐functionalized ligand highlights the synthetic potential of this class of bidentate ligands with respect to polymer chemistry or the attachment onto surfaces and nanoparticles. The coordination to RuII ions results in homoleptic and heteroleptic complexes with the resultant photophysical and electrochemical properties strongly dependent on the number of these ligands attached to the RuII core.  相似文献

New single‐isomer, cationic β‐cyclodextrins, including mono‐6‐deoxy‐6‐pyrrolidine‐β‐cyclodextrin chloride (pyCDCl), mono‐6‐deoxy‐6‐(N‐methyl‐pyrrolidine)‐β‐cyclodextrin chloride (N‐CH3‐pyCDCl), mono‐6‐deoxy‐6‐(N‐(2‐hydroxyethyl)‐pyrrolidine)‐β‐cyclodextrin chloride (N‐EtOH‐pyCDCl), mono‐6‐deoxy‐6‐(2‐hydroxymethyl‐pyrrolidine)‐β‐cyclodextrin chloride (2‐MeOH‐pyCDCl) were synthesized and used as chiral selectors in capillary electrophoresis for the enantioseparation of carboxylic and hydroxycarboxylic acids and dansyl amino acids. The unsubstituted pyCDCl exhibited the greatest resolving ability. Most analytes were resolved over a wide range of pH from 6.0 to 9.0 with this chiral selector. In general, increasing pH led to a decrease in resolution. The effective mobilities of all the analytes were found to decrease with increasing CD concentration. The optimal concentration for most carboxylic acids and dansyl amino acid was in the range 5–7.5 mM and >15 mM for hydroxycarboxylic acids. 1H NMR experiments provided direct evidence of inclusion in the CD cavity.  相似文献

In this study, a novel method for the highly selective enrichment of phosphopeptides using erbium phosphate doped poly(glycidyl methacrylate/ethylene dimethacrylate) spin columns is presented. Erbium phosphate was synthesized by precipitation from boiling phosphoric acid and incubated overnight in erbium chloride solutions. The resulting powder was embedded in a monolithic poly(glycidyl methacrylate/ethylene dimethacrylate) polymer. The monolith was synthesized in a spin column by radical polymerization. Erbium phosphate demonstrated a high affinity and selectivity for phosphopeptides due to the strong interaction of trivalent erbium ions with the phosphate groups of phosphopeptides. The high selectivity and performance of the designed spin columns were demonstrated by successfully enriching phosphopeptides from tryptically digested protein mixtures containing the model phosphoproteins α‐ and β‐casein, bovine milk, and human saliva. By the implementation of several washing steps, unspecific components were removed and the enriched phosphopeptides were effectively eluted from the spin columns under alkaline conditions. The selective performance of the presented method was further demonstrated by the enrichment of two synthetic phosphopeptides, which were spiked in tryptically digested and dephosphorylated HeLa cell lysates at low ratios. Finally, the presented approach was compared to conventional phosphopeptide enrichment by titanium oxide and revealed higher recoveries for the erbium phosphate doped monoliths.  相似文献

Two novel polymeric monoliths for anion-exchange capillary liquid chromatography of proteins were prepared in a single step by a simple photoinitiated copolymerization of 2-(diethylamino)ethyl methacrylate and polyethylene glycol diacrylate (PEGDA), or copolymerization of 2-(acryloyloxy)ethyl trimethylammonium chloride and PEGDA, in the presence of selected porogens. The resulting monoliths contained functionalities of diethylaminoethyl (DEAE) as a weak anion-exchanger and quaternary amine as a strong anion-exchanger, respectively. An alternative weak anion-exchange monolith with DEAE functionalities was also synthesized by chemical modification after photoinitiated copolymerization of glycidyl methacrylate (GMA) and PEGDA. Important physical and chromatographic properties of the synthesized monoliths were characterized. The dynamic binding capacities of the three monoliths (24 mg/mL, 56 mg/mL and 32 mg/mL of column volume, respectively) were comparable or superior to values that have been reported for various other monoliths. Chromatographic performance was also similar to that provided by a modified poly(GMA-ethylene glycol dimethacrylate) monolith. Separation of standard proteins was achieved under gradient elution conditions using these monolithic columns. Peak capacities of 34, 58 and 36 proteins were obtained with analysis times of 20–30 min. This work represents a successful attempt to prepare functionalized monoliths via direct copolymerization of monomers with desired functionalities. Compared to earlier publications, additional surface modifications were avoided and the PEGDA crosslinker helped to improve the biocompatibility of the monolithic backbone.  相似文献

A polymer monolith microextraction method coupled with high‐performance liquid chromatography was developed for the determination of adenosine triphosphate, adenosine diphosphate, and adenosine monophosphate. The monolithic column was synthesized inside fused‐silica capillaries using thermal initiation free‐radical polymerization with glycidyl methacrylate as the monomer, ethylene dimethacrylate as the cross‐linker, cyclohexanol, and 1‐dodecanol as the porogen. N‐Methylolacrylamide, an important hydrophilic monomer, was incorporated into the polymerization mixture to enhance the hydrophilicity of the poly(glycidyl methacrylate‐co‐ethylene dimethacrylate) column. The obtained poly(glycidyl methacrylate‐coN‐methylolacrylamide‐co‐ethylene dimethacrylate) monolith was characterized by scanning electron microscopy, Fourier‐transform infrared spectra, and X‐ray photoelectron spectroscopy. Optimum conditions for the preconcentration and separation of the target adenosines were also investigated. Under the optimum conditions, we obtained acceptable linearities, low limits of detection, and good relative standard deviations. The developed polymer monolith microextraction with high‐performance liquid chromatography method exhibited a good performance with recovery values in the range of 76.9?104.7% when applied to the determination of the adenosines in five royal jelly samples.  相似文献
