Phosphorylation of 3',4',5,7-Tetramethyldihydroquercetin

3',4',5,7-Tetramethyldihydroquercetin was phosphorylated with phosphorous acid derivatives, and similar results were obtained with phosphorochloridites and phosphoramidites. The PIII derivatives obtained were subjected to oxidation. The structures of the products were determined by NMR spectroscopy.

Solid-state synthesis of poly(3',4'-dimethoxy-2,2':5',2"- terthiophene): comparison with poly(terthiophene) and poly(3',4'-ethylenedioxy-2,2':5',2"- terthiophene)

A new terthiophene monomer: 3',4'-dimethoxy-2,2':5',2"-terthiophene (TMT) was synthesized and characterized by 1H-NMR, 13C-NMR and FTIR. The solid-state oxidative polymerizations of TMT were performed in various ratios of oxidant (FeCl?) to monomer (TMT). The resulting polymers were characterized by 1H-NMR, FTIR, UV-vis-NIR, GPC, X-ray diffraction, CV, as well as TGA and conductivity measurements. The structure and properties of poly (TMT) were compared with those of polyterthiophene [poly(TT)] and poly (3',4'-ethylenedioxy-2,2':5',2"-terthiophene) [poly(TET)] prepared under the same polymerization conditions. After comparative analysis with poly(TT) and poly(TET), the effects of the dimethoxy substituent and FeCl? on the structural and physicochemical properties of the poly(TMT)s were discussed in depth. The comparison suggested that the dimethoxy-substituted polymer did not display higher crystallinity, thermal stability, conductivity and electrochemical activity than ethylenedioxy substituted one. The results also showed that the effect of FeCl? on poly(TMT) was similar that seen with the poly(TT), in which the oxidation degree, electrochemical activity and conductivity increased steadily with increasing [FeCl?/[TT] ratio. Furthermore, the poly(TMT) and poly(TT) are mostly made up of dimers with a small amount of higher molecular weight components.

以2-苯基-1,2,3-三唑-4-甲酰肼为原料合成了几种新的1-(2'-苯基-1',2',3'-三唑-4'-甲酰基)-4-芳基氨基硫脲,在不同条件下环化,制得一系列新的1,3,4-噻二唑、1,3,4-恶二唑和1,2,4-均三唑衍生物。化合物的结构经元素分析、IR、^1HNMR和MS确证,并对其波谱性质进行了讨论。     

Crystal and molecular structures of 2-(4-chlorophenyl)-5,7-dimethoxyquinolin-4-yl phenyl bis(2-chloroethyl) phosphoramidate

2-(4-Chlorophenyl)-5,7-dimethoxyquinolin-4-yl phenyl bis(2-chloroethyl)phosphoramidate is synthesized and characterized by NMR, IR and single crystal X-ray crystallography. The crystal is triclinic, P[`1]P\bar 1 space group, with a = 9.5188(19) ?, b = 12.856(3) ?, c = 13.250(3) ?, V = 1412.0(5) ?)³, and Z = 2 (at 291(2) K). The crystal packing arrangement indicates that the molecule is stacked through π...π aromatic stacking interactions.     

2',5,7-Trihydroxy-4',5'-(2,2-dimethylchromeno)-8-(3-hydroxy-3-methylbutyl) flavanone purified from Cudrania tricuspidata induces apoptotic cell death of human leukemia U937 cells

Cellular DNA topoisomerase I is an important target in cancer chemotherapy. A chloroform extract of the root barks of Cudrania tricuspidata showed an inhibitory effect on mammalian DNA topoisomerase I. The topoisomerase I inhibitory compound was purified and identified as 2',5,7-trihydroxy-4',5'-(2,2-dimethylchromeno)-8-(3-hydroxy-3-methylbutyl) flavanone. The compound, temporarily designated as PKH-3, was shown to inhibit the activity of topoisomerase I with IC50 about 1.0 mM. Concentration of 10 microM PKH-3 caused 50% growth inhibition of human cancer cell U937. PKH-3-induced cell death was characterized with the cleavage of poly(ADP-ribose) polymerase (PARP) and pro-caspase 3. Furthermore, PKH-3 induced the fragmentation of DNA into multiples of 180 b.p. (an apoptotic DNA ladder), indicating that the inhibitor triggered apoptosis. This induction of apoptosis by PKH-3 was also confirmed using flow cytometry analysis. Taken together, these results suggest that PKH-3 may function by inhibiting oncogenic disease, at least in part, through the inhibition of topoisomerase I activity.     

3-(2-二唑啉基)-色酮类化合物的合成研究

报道了取代的邻羟基苯乙酮(1a～1e)经Vilsmeier-Haack反应一步制得3-醛基色酮(2a～2e),2a～2e与取代的芳酰肼(3a～3c)缩合制得15个酰腙类化合物(4a～4o),4a～4o在醋酸酐作用下关环得15个3-(2-苝二唑啉基)-色酮(5a～5o),并通过元素分析,IR,~1H NMR和MS诺数据确证了上述化合物的结构。     

The synthesis and liquid crystal properties of certain 1-(4'-n-alkoxy-2,2',3,3',5,5',6,6'-octafluorobiphenyl-4-yl)- 2-(trans-4-n-pentylcyclohexyl)-ethanes and -ethenes

The synthesis and liquid crystal properties of eight members of the homologous series of 1-(4'-n-alkoxy-2,2',3,3',5,5',6,6'-octafluorobiphenyl-4-yl)-2-(trans-4-n-pentylcyclohexyl)-ethanes and eight members of the corresponding -ethanes, which have the E-configuration, are reported. These compounds are nematic, the ethenes having a considerably mesophase higher thermal stability than the analogous ethanes.
