The effectiveness of monochromatic light on chlorophyll formation over a range of 420–80 nm was examined in the regreening cells of Chlorella protothecoides in the presence of chloro-phenyl dimethylurea (CMU). An action spectrum showed two maxima with a minimum near 450 nm. At the most effective wavelength examined (444 nm), 0.2 W. m -2 was sufficient for half saturation. The activity of 5-aminolevulinic acid (ALA) production was examined in the greening cells under irradiation with white, blue and red light. The activity was always limited by availability of substrate, especially in the case where the greening cells were incubated with cycloheximide or transferred to darkness. Decay of the activity in these cases was delayed by provision of organic compounds such as glycine, pyruvate or glucose. The effectiveness of blue light on ALA* production observed in the presence of CMU was inferred to be brought about by the increased availability of endogenous substrates.  相似文献

Abstract— We have investigated the possibility of the involvement of a blue light fluence-rate sensing photoreceptor in the light acclimation of chloroplast components in light-grown pea seedlings. Low lightgrown seedlings were acclimated for 2 days to either 20 or 200 μmolm-2s-2 of white, blue-enriched, or broad-band red light. An increase in blue-enriched light fluence rate was more effective than that of red light in bringing about both inhibition of internode growth and the enhancement of the chlorophyll a/b ratio. Ribulose 1,5-bisphosphate carboxylase/oxygenase and cytochrome f protein levels, per unit cell, also increased more markedly (around two-fold) in response to an increase in blue light. The 23 kDa polypeptide of the oxygen-evolving complex and the light-harvesting chlorophyll d b protein of photosystem II apoprotein levels vaned under all wavelengths to a lesser extent, correlating with total protein levels or greening. These data are consistent with the hypothesis of a role for a blue photoreceptor in detecting low versus high fluence rate of light, and subsequently controlling the light acclimation responses. Nevertheless photosynthesis or other mechanisms of fluence-rate photoperception must also be involved.  相似文献

Abstract— The in vivo photostationary state, φfr= ([ P fr]fr/[ P ]), of phytochrome in far red light has been determined in mustard seedling cotyledons by three different methods. The φfr is a function of the length of time of etiolation ( t = 36 hr, φfr= 0·14; t = 72 – 120 hr, φfr= 0·075). The calculated φr= 0·8. The amount of P tot is strongly dependent on the time of onset of far red light. These data imply that it would be almost impossible to maintain a constant level of P fr in mustard cotyledons over a considerable period of time.  相似文献

Abstract— The hypocotyl of the tomato ( Lycopersicon esculentum ) seedling synthesizes large amounts of anthocyanin if exposed to prolonged light. Single light pulses are totally ineffective. The involvement of phytochrome can be shown by light pulse treatments following a prolonged light exposure. It is predominantly the action of blue/UV light which leads to a high responsiveness of anthocyanin synthesis towards phytochrome. Moreover, the data suggest a phytochrome-independent action of blue/UV light, in particular of UV-B, on anthocyanin synthesis.  相似文献

Abstract— Chloroplasts illuminated with light absorbed predominantly by Photosystem I can mediate the photoxidation of ascorbic acid to threonic and oxalic acids. Two micromoles of oxygen are consumed for each micromole of ascorbic acid added. The kinetics of this endogenous chloroplast system closely resemble the kinetics of an artificial flavin-manganese-catalase system in which flavin is the photosensitizing dye.  相似文献

Abstract Anthocyanin synthesis in the hypocotyl of tomato ( Lycopersicon esculentum ) seedlings responds strongly and specifically to blue/UV light while the response to red and far-red light, operating through phytochrome, is weak. The herbicide Norflurazon (SAN 9789) was used to inhibit synthesis of colored carotenoids almost completely without affecting growth and development measurably. Even though carotenoid content was reduced to less than 2% of normal and the fluence rate response function for blue and UV light was linear within the experimental range, Norflurazon treatment did not reduce seedling sensitivity toward blue/UV light. It was concluded that at least'bulk'carotenoids are not the photoreceptor chromophore of the blue/UV photoreceptor pigment.  相似文献

Abstract— Spectral sensitivities for C16 rranshexadecenoic acid accumulation in etiolated leaves of Vigna sinensis L. show two maxima, one in the blue and one in the far red, indicating a control by the high irradiance reaction. Abstract— As is known the chlorophyll fluorescence of photosynthetizing plants is strongly quenched by carotenoid triplet states if the exciting light intensity is high enough (> 10 kW/m2). This light-induced quenching process was studied by measuring the relative yield of chlorophyll fluoresccncc excited with a pulsed argon laser at 20 C in light adapted algae as function of time (within lo μ s ) and exciting light intensity (<400 kW/m2). The experimental yield against time and yield against intensity curves have been adequately explained by a statistical model of Photosystem 2 (PS 2) units based upon the existence of freely moving excitons according to which the carotenoid triplet quenchers are randomly distributed and are perfect traps for excitons. Accepting the hypothesis that carotenoid triplet quenching occurs only in PS 2 units. it could be concluded that the height of the true zero level of PS 2 fluorescence is somewhat lower than the half value of the fluorescence level of the dark adapted state.  相似文献

Abstract— The presence of light during exposure of intact pea leaves to high temperature (40°C) protects Photosystem II (PSII) against inactivation, as indicated by the preservation of the maximal variable 685 nm chlorophyll fluorescence and the photosynthetic oxygen evolution. This photoprotection was observed (i) to be saturated at low fluence rates ( ca 10 W m-2) and (ii) to be strongly dependent on the spectral characteristics of the light. It was specifically induced by red light (630–670 nm) whereas other wavelengths were much less protective. A strong antagonism between red and far-red lights was also observed, with PSII stabilization by red light being partially cancelled by additional far-red light.  相似文献

Abstract— Aninvestigation was made of the location, function and characteristics of production of carotenoid pigments of a strain of Mycobacterium marinum , an organism which produces its pigments only when exposed to light.
It was found that light-grown cells of this organism contain only one pigment. The intracellular location of this pigment was found to be the cell envelope. The pigment was found to be capable of protecting the cells against lethal photo-oxidations mediated by the dye, toluidine blue.
These studies also revealed that in dark-grown cells exposed to light, pigment production occurred after a short lag and then remained constant during logarithmic growth. Pigment was produced by starved cells, but to a lesser degree than son-starved cells; the addition of a carbon or nitrogen source to starved cells increased pigmentation to the level in non-starved cells. Pigmentation did not occur in the absence of oxygen. Cells exposed to light at 4°C were not pigmented immediately after exposure, but became so if incubated at 30°C. Production of pigment was found to obey the Bunsen-Roscoe reciprocity law. The determination of an action spectrum for the production of pigment indicates that maximum pigment productiion occurs in the vicinity of 410 and 439 mµ, with smaller maxima in the vicinity of 502 , 528 and 560 mµ.  相似文献

Abstract— The porphyrinogenic drug, 3,5-diethoxycarbonyl 1,4-dihydrocollidine (DDC) when administered orally to rats evoked large increases in hepatic δ-aminolevulinic acid synthetase (ALAS) activity and hepatic protoporphyrin levels. These increases varied markedly according to light exposure patterns of the animals. DDC-treated animals continuously exposed for one week to fluorescent Blacklight lamps (Westinghouse FS-40) demonstrated a greater than two-fold increase in hepatic ALAS and a greater than threefold increase in liver protoporphyrin levels as compared to DDC-treated animals exposed to ambient light-dark cycling. Furthermore, the skin of porphyric animals continuously exposed to light showed larger increases in porphyrin content as compared to rats exposed to ambient light. These studies indicate that light exposure patterns can profoundly alter the activity of the hepatic heme pathway in the rat and suggest that light exposure could play a role in the production of drug-induced porphyria in man.  相似文献

Under laser excitation at 457.9 and 514.5 nm, a frozen solution of chlorophyll a in n -octane displays fluorescence peak maxima at 2K that may be assigned to two distinct monomeric chlorophyll species. Using zero-field fluorescence-detected magnetic resonance the triplet state properties of the two chlorophyll species have been assigned to the monoligated and biligated chlorophyll monomer in which water serves as the ligand coordinated to the magnesium metal center. These triplet state properties for chlorophyll in solution are then utilized in interpreting triplet state results for in vivo chlorophylls associated with the light harvesting chlorophyll protein complex. It is shown that the triplet state data are consistent with attachment of the chlorophyll molecule to the protein site with a single ligand coordinated to the chlorophyll metal center.  相似文献
