研究了Ru(bpy)2(dppx)2+-SDS-DNA(bpy=2,2′-联吡啶,dppx=7,8-二甲基-吡啶并[3,2-a:2′,3′-c]吩嗪)体系的共振光散射光谱。结果表明,在阴离子表面活性剂十二烷基硫酸钠(SDS)预胶束聚集体存在下,Ru(bpy)2(dppx)2+-SDS体系具有很强的共振光散射,DNA的加入使其共振散射光猝灭。探讨了反应机理。基于DNA对Ru(bpy)2(dppx)2+-SDS体系共振光散射的猝灭作用,建立了共振光散射法测定DNA的新方法。在最佳实验条件下,体系在393nm处的共振光散射猝灭程度与DNA的浓度呈线性关系,线性范围为0.01~1.2mg/L,检出限为1.5μg/L。 相似文献

Ru(phen)2dppx2+光谱探针法研究道诺霉素与脱氧核糖核酸的相互作用 总被引:2,自引:0,他引:2
以核酸分子“光开关”Ru(phen)2dppx2 为探针,分别采用紫外-可见吸收光谱法和荧光光谱法研究了抗癌药物道诺霉素(daunomycin,DNM)与小牛胸腺DNA之间的作用方式,结果表明:DNM是以插入的方式与DNA相互作用。Scatchard方程的研究表明,Ru(phen)2dppx2 与小牛胸腺DNA的结合比为1∶4~1∶5;表观结合常数为2.58×107L/mol。DNM加入后,表观结合常数大大降低,这也表明道诺霉素主要是以插入方式与DNA结合。作为研究核酸分子的荧光探针,与溴化乙锭相比,Ru(phen)2dppx2 具有灵敏度高、毒性低、稳定性好、选择性好、使用方便等优点。 相似文献

利用荧光和紫外可见吸收光谱研究了 Ru( bipy) 2 ( dppz) 2 +与 DNA之间的插入键合作用。结果表明 ,Ru( bipy) 2 ( dppz) 2 +是通过 dppz配体插入到 DNA的双螺旋结构中。而且 ,一定浓度的 Fe( CN) 4 - 6 和 Na Cl对 Ru( bipy) 2 ( dppz) 2 +- DNA复合物的荧光无猝灭作用 ,这一结果也证实了 Ru( bipy) 2 ( dppz) 2 +和 DNA之间的插入键合作用 相似文献

A new method for the study of triplex DNA is established according the fluorescence enhancement of molecular “Light Switch“ complex of Ru(phen)2(dppx)^2 when it intercalate into triplex DNA.Because the fluorescence intensity of Ru(phen)2(dppx)^2 bonded to triplex DNA is in ths case higher than that bonded to duplex DNA in certain range of DNA concentration,the method is much more sensitive than other methods reported previously. 相似文献

构建了一种新型、灵敏、便捷式流感病毒免疫传感器,通过在金电极表面键合抗-HA单克隆抗体,选择性捕获目标H1N1流感病毒。该方法基于吸附在病毒表面的辣根过氧化物酶(HRP)在亚甲基蓝(MB)溶液中可有效催化还原H2O2,利用方波伏安法(SWV)考察了还原峰电流的变化,从而实现了对抗原病毒的特异性识别。实验表明所构建的免疫传感器对H2O2-MB体系表现出快速的电流响应以及良好的稳定性,对流感病毒H1N1检测的动态响应范围为0.01~2.0μg/m L,检出限(S/N=3)为5.0 ng/m L。结果证明HRP可作为一种简单快速的探针用于流感病毒的实时监测。 相似文献

The working principle of a genosensor is based on the mechanism of ion‐channel mimetic sensors. The analytical signals generated upon hybridization processes were recorded by a redox active marker [Fe(CN)6]3?/4? present in the sample solution using voltammetric techniques. The developed genosensor was suitable for determination of 20‐mer complementary oligonucleotide sequence, and also of the PCR products containing the complementary 20‐mer sequence in various positions, with detection limits in the 10 pM range. The noncomplementary 20‐mer oligonucleotide sequence as well as the PCR product without complementary region generated very weak response. The good discrimination of the position of the complementary part in the PCR products was observed. 相似文献
