利用重组大肠杆菌E.coli HB101来进行直接生产羟基丁酸(HB)手性单体,研究了含pUCAB质粒的重组体E.coli HB101在各种条件下生长及积累HB单体的情况,研究了HB单体的积累随pH值变化的规律。结果表明,pH=6.8时,48 h内细菌可生产0.5 g/L以上的HB单体。  相似文献

本文通过同步辐射原位二维广角X射线(2D-WAXD)和原位红外光谱(FTIR)等手段,对聚(3-羟基丁酸酯-co-3-羟基戊酸酯)(PHBV)预取向膜二次拉伸过程及随后升温过程中的结构演化机制进行了系统研究。研究结果表明,二次拉伸过程中β晶的相对含量、取向程度和晶粒尺寸会随应变而升高;同时,体系的总α晶的取向度和晶粒尺寸变小,结晶度下降。这说明拉伸过程中,α晶会被破坏并转变为β晶。经过二次拉伸的PHBV膜,在升温过程中,α晶的取向、晶粒尺寸及总体结晶度会升高,而β晶的相对含量逐步下降到零。这说明在较高的温度下,β晶的分子链会重新排列,主要转变成了取向度高的α晶。β晶表现出从65℃到135℃的持续熔融转变。本研究初步阐释了PHBV薄膜中的β晶在拉伸和升温过程中的演化机制,为生物基聚酯薄膜的加工和性能调控提供了理论依据。  相似文献

An efficient process for the production of (R)‐hydroxycarboxylic acids (RHAs) from polyhydroxyalkanoates (PHAs) was developed. It involved the synthesis of PHA in bacteria, followed by bringing the culture broth directly to a pH optimal for in vivo PHA degradation, thus avoiding cell collection by centrifugation and pellet resuspension. The optimal pH was maintained to allow maximal release of RHAs. Using this process, cells having a dry weight (w) of 1.8 g · L−1 and 45% (w/w) PHA exhibited a linear PHA degradation rate of about 0.059 g · L−1 · h−1 in the first 9 h. Concomitantly, RHAs were released with a rate of 0.057 g · L−1 · h−1. Further incubation of up to 15 h resulted in almost 90% (w/w) degradation of PHA. Based on this approach in combination with chemostat and a plug flow reactor a continuous process for the production of RHAs could be achieved.

Experiments with diluted solutions of a customary water glas show that the rate of depolymerization depends not only upon pH and the SiO2 concentration, but also varies systematically as a function of the type and concentration of an additional electrolyte. Increasing cation activities of metal chlorides are causing a decrease of the rate constant in the order 1) Na+, K+, 2) Mn2+, Mg2+, Ca2+, Sr2+, 3) Zn2+, Ni2+, Ce3+, Cu2+. With respect to anions of sodium salts the rate constants are increasing with increasing activities in the order NO 3 , HCO 3 , Cl, SO 4 2– , whereas HPO 4 2– causes a decrease. The results permit to identify those components of water which are most responsible for a change of the depolymerization rate and may be used to evalute the properties of a water glass as a possible anticorrosive agent for water supply systems.  相似文献

设计合成了新型含有荧光基团的α-氰基丙烯酸酯单体,并与其它α-氰基丙烯酸酯单体共聚,得到产生荧光的聚氰基丙烯酸酯材料.将其包埋在小鼠背部肌肉层,可获得良好的荧光成像效果.通过对荧光强度的监测,初步研究了聚氰基丙烯酸酯材料中的侧链酯基在小鼠体内的降解情况.单体合成是以蒽合氰基丙烯酸和4,4'-二甲氧基三苯基-氨基己醇为原料,得到蒽合氰基丙烯酸(4,4'-二甲氧基三苯基-氨基己醇)酯,脱保护后再将异硫氰酸荧光素以化学键合的方式标记在末端氨基上,脱蒽还原烯双键后,得到可用于荧光标记的α-氰基丙烯酸(异硫氰酸荧光素-氨基己醇)酯单体.反应中间体及单体结构采用核磁共振氢谱和质谱进行表征.该单体及其高聚物均可在激发光(488 nm)和发射光(525 nm)条件下观察到明显荧光.  相似文献

正相条件下,在自制的涂敷型纤维素-三(3,5-二甲基苯基氨基甲酸酯)手性固定相上,首次直接拆分了几种新的外消旋过渡金属簇合物,考察了流动相中极性添加剂对手性拆分的影响.结果发现不仅不同立体结构的醇对手性簇合物的选择性不同,而且手性簇合物四面体骨架的四个顶点上的原子及与之配位的基团,对它们的色谱行为都有重要的影响.  相似文献

以咔唑-9-乙基氯甲酸酯(CEOC)作为柱前衍生试剂,采用毛细管电泳对14种二肽进行了手性拆分。以5种二肽为代表,考察了缓冲液种类、浓度、pH值、二元手性选择剂的组合配比等因素对二肽的拆分效果,优化了实验条件。在各自的优化条件下,14种二肽手性拆分的分离度均在3.63以上,最高分离度可达43.14(Gly-Ala)。  相似文献
