Proton and 13C NMR chemical shifts and 1H? 1H scalar couplings for the two diastereomers of the potent vitamin K epoxide reductase (VKOR) inhibitor brodifacoum have been determined at 293 K from acetone solutions containing both diastereomers. To facilitate difficult assignments, homo‐ and heteronuclear correlation spectra were acquired at 750 and 900 MHz over 268–303 K temperature range. Conformations of both diastereomers inferred from the scalar couplings and 1‐D NOE measurements reveal that one diastereomer (SS/RR) adopts a strained geometry in the cyclohexene ring system of the tetralin group. The NMR spectra also show evidence of line broadening due to conformational exchange at room temperature for the SR/RS diastereomer. These assignments and conformational analyses may be useful in studies of biomolecular interactions of brodifacoum with target proteins such as VKOR and in source determination of brodifacoum. Two novel oligosaccharides, mono‐ and difructosyllactosucrose {[O‐β‐D ‐fructofuranosyl‐(2 → 1)]n‐β‐D ‐fructofuranosyl‐O‐[β‐D ‐galactopyranosyl‐(1 → 4)]‐α‐D ‐glucopyranoside, n = 1 and 2} were synthesized using 1F‐fructosyltransferase purified form roots of asparagus (Asparagus officinalis L.). Their 1H and 13C NMR spectra were assigned using several NMR techniques. The spectral analysis was started from two anomeric methines of aldose units, galactose and glucose, since they showed separate characteristic signals in their 1H and 13C NMR spectra. After assignments of all the 1H and 13C signals of two units of aldose, they were discriminated as galactose and glucose using proton–proton coupling constants. The HMBC spectrum revealed the galactose residue attached to C‐4 of glucose, fructose residue attached to the C‐1 of glucose, and further fructosyl fructose linkage extended from the glucosyl fructose residues. In this paper, a complete 1H and 13C NMR data assignment of ent-polyalthic acid, a biologically active labdane-type diterpene, is presented. The assignments were carried on the basis of spectroscopic data from 1H NMR, 13C{1H} NMR, gCOSY, gHMQC, and gHMBC experiments. Furthermore, a software-assisted methodology, using FOMSC3_rm_NB and NMR_MultSim programs, supported the detailed and unequivocal assignment of 1H and 13C signals, allowing all hydrogen coupling constants to be determined and thus clarifying all hydrogen signal multiplicities.  相似文献

New 2-substituted diazaphospholane-2-oxides (I-III, V-VIII) and diazaphosphorinane-2-oxide (IV) were synthesised and characterised by 1H, 13C, and 31P NMR, IR spectroscopy, and elemental analysis. The presence of chiral diamino groups in compounds II and V–VIII gives rise to various diastereomers so that the 31P{1H} NMR spectra demonstrated three and two peaks with different ratios, respectively. Also, the 1H and 13C{1H} NMR spectra of compounds II and V–VIII revealed three and two sets of signals for the related conformers (diastereomers). Interestingly, the 31P NMR spectrum of V in D2O indicated a great upfield shift (Δδ = 19.0) for 31P relative to the value obtained in DMSO-d6 (solvent effect). The two signals in V split further to three signals in the presence of β-cyclodextrin. Moreover, conformational analysis of diazaphospholane V was studied by ab initio calculations at the HF and B3LYP levels of theory using the Gaussian 98 program. Results indicated that among four suggested diastereomers (C1–C4) of V, C1 and C3 containing methyl group in the equatorial position are the most stable forms.  相似文献

The structure elucidations and complete 1H and 13C NMR assignments are reported for three new anthraquinone derivatives: Fusaquinon A (1), B (2), and C (3) isolated from the fermentation medium of the marine fungus Fusarium sp. (No. ZH‐210). HREIMS, Fourier transform infrared absorption spectrometry (FT‐IR), NMR experiments including gCOSY, gHMQC, gHMBC, and NOESY were used for the determination of the structures and NMR spectral assignments. An NMR study of one new and several known abietane diterpenes isolated from the roots of Aegiphila lhotzkyana is described. In addition to 1D NMR, several 2D shift‐correlated NMR pulse sequences (1H–1H‐COSY, NOESY, HMQC and HMBC) were used to establish all the structures, and unambiguously perform the 1H and 13C chemical shift assignments of the new natural diterpene and three derivatives, the NMR data for which have not been reported previously. We report the total assignments of the 13C and 1H NMR spectra of some 4‐methyl‐2‐oxo‐(2H)‐pyrido[1,2‐a]pyrimidine and 2‐methyl‐4‐oxo‐(4H)‐pyrido[1,2‐a]pyrimidine derivatives. The products were characterized by 1H and 13C NMR and reported data for similar compounds. No comparative data for the two sets of isomeric compounds with respect to 13C and 1H NMR have been reported to date. We made some detailed studies of the 2D NMR spectra of these compounds and observed that assignments made for non‐protonated carbon atoms by us and those reported in the literature for similar compounds need correction. The revised assignments were made on the basis of heteronuclear single quantum correlation (HSQC) and heteronuclear multiple bond correlation (HMBC) techniques. A nortriterpene glycoside, pyrocincholic acid 3β-O-β-6-deoxy-D-glucopyranoside-28-O-β-D-glucopyranoside, was isolated from the leaves of Isertia haenkeana and its structure established by 1H and 13C NMR spectral studies. The complete 1H and 13C NMR resonance assignments for this triterpene were confirmed by the conventional 1D NMR methods and 2D shift-correlated NMR techniques: DQF COSY, TOCSY, ROESY and HMQC.  相似文献

Structural characterization of vulcanized natural rubber was performed by high‐resolution latex‐state 13C NMR spectroscopy. The vulcanized natural rubber latex was prepared by vulcanization of high ammonia natural rubber latex with sulfur and sodium di‐n‐butyldithiocarbamate as vulcanizing agents. High resolution was attained for latex‐state 13C NMR spectroscopy even after vulcanization of the rubber latex, as is evident from no background in spectrum and narrow half width of signals, which were independent of vulcanization time. Small signals at 44 and 58 ppm in the carbon region were assigned by measurements of both distortionless enhancement by polarization transfer (DEPT) and attached proton test (APT) to secondary, tertiary, and quaternary carbons of crosslinking points. The assignment was proved by high‐resolution solution‐state NMR spectroscopy of vulcanized liquid cis‐1,4‐polyisoprene as a model, in which DEPT, APT, 2‐dimensional 1H‐13C correlation (HETCOR), and 2‐dimensional heteronuclear multiple bond correlation (HMBC) measurements were applied. © 2007 Wiley Periodicals, Inc. Meso- and Rac-Decaisopropylstannocene: Stereoisomerism Caused by Parallel Arrangement of Two Pentaisopropylcyclopentadienyl Ligands in Paddlewheel Conformation Decaisopropylstannocene 1 is the first Decaisopropylmetallocene, where X-ray diffraction data could be successfully refined and the third example of a group 14 Metallocene with a symmetric sandwich structure. Due to the directionality of the isopropyl substituents 1 occurs as a mixture of diastereomers in the crystalline state as well as in solution. The stereoisomers exhibit the expected signals in solution (1H and 13C NMR) and in the crystal (CPMAS 119Sn NMR).  相似文献
