Abstract

The readily available methyl (methyl 3-deoxy-5,8:7,9-di-O-isopropylidene-β-D-glycero-D-galacto-2-nonulopyranosid)onate (7) was converted in five synthetic steps into methyl (methyl 4-acetamido-3,4-dideoxy-β-D-glycero-D-talo-2-nonulopyranosid)onate (11). Selective protection of the C-4, C-7, C-8 and C-9 hydroxy groups of methyl (methyl 3-deoxy-8,9-O-isopropylidene-β-D-glycero-D-galacto-2-nonulpyranosid)onate (2) followed by oxidation of the C-5 hydroxy group and then its oximination gave 5-hydroxyimino derivatives (15 and 16).

Methyl ester of abscisic acid (ABA), a plant hormone, gives a dehydrated ion at m/z 260 in electron ionization mass spectrometry (EI-MS). This dehydrated ion had been considered to be derived only from the elimination of the tertiary hydroxyl group at C-1'. We found that 34% of the dehydrated ion was formed by elimination of the oxygen atom at the 4'-carbonyl group, and the remaining 66% by elimination of the 1'-hydroxyl group. This unusual elimination of the carbonyl oxygen was shown with [4'-(18)O]ABA methyl ester. Involvement of the 4'-carbonyl oxygen in dehydration was observed in methyl ester of phaseic acid (PA), a natural metabolite of ABA, but not in 1'-deoxy-ABA methyl ester or isophorone. This suggested that the 1'-hydroxyl group was necessary for the elimination of the 4'-carbonyl oxygen. ABA methyl esters labeled with stable isotopes showed that hydrogen atoms at the 1'-hydroxyl group and at C-4 or -5 or -3' or - 5' or -7' were eliminated with the 4'-carbonyl oxygen. These results allow us to propose a formation mechanism of the dehydrated ion derived from the elimination of 4'-carbonyl oxygen and hydrogen atoms at C-4 and 1'-oxygen in ABA methyl ester as follows: first, ionization at the 1'-hydroxyl group occurs to give an ion radical, and the proton at the 1'-oxygen migrates to the 4'-carbonyl oxygen after the bond fission between C-1'-C-6'; second, migration of the proton at C-4 to the 1'-oxygen is followed by migration of the protons at C-5 and C-7' to C-4 and C-5, respectively; finally, the proton at the 1'-oxygen migrates to the 4'-hydroxyl group, and H(2)O at C-4' is eliminated to give the dehydrated ion. Our findings point out that a dehydrated ion is not always derived from the elimination of a hydroxyl group.  相似文献

Treatment of (R) methionine sulfoxide with NaOD led to exchange of the C-4 methylene and C-5 methyl protons; exchange of the chiral C-2 proton did not occur. Reducation with mercaptoacetic acid gave (R)-[4-2H2, 5-2H3] methionine. The latter was converted into its carbobenzyloxy methyl ester sulfoxide, pyrolysis of which followed by deprotection yielded (R)-[4-2H2] vinylglcine as the hydrochloride.  相似文献

A new entry to C-5 substituted 4-hydroxy-6-methyl-2-pyrones has been achieved. The best conditions to prepare the monobromo and the dibromo derivatives at C-3 and the C-6 methyl group of the title pyrone have been defined. The synthetic applicability of the phosphonium salts at CH3-C-6 of both 4-methoxy-6-methyl-2-pyrone, 5 , and dehydroacetic acid, 2 , has also been evaluated.  相似文献

Twenty-five different brominated derivatives of 4-hydroxy-6-methyl-2-pyrone (triacetic acid lactone) and 3-acetyl-4-hydroxy-6-methyl-2-pyrone (dehydroacetic acid) have been prepared. Fifteen derivatives have not been previously described and the preparations of a few known products have been improved. Bromination at C-3, C-5, methyl group at C-6 and deacetylations at C-3 have been the types of reactions used.  相似文献

Synthetic approaches to anthracycline antibiotic analogues in which the nitrogen atom of the carbohydrate portion is incorporated into a 1, 2, 3-triazolyl moiety were investigated. By using methyl 6-azido-2, 6-dideoxy-β-D--arabino-hexopyranoside and methyl 6-azido-2, 3, 6-tricdeoxy--α-L-arabino-hexopyranoside, the corresponding glycosides (16 a, b - 18 a, b) of carminomycinone and daunomycinone were prepared. The desired heterocyclic system was developed directly with the C-3′ and C-6′ azido anthracyclines by means of a cycloaddition process to give 7-0-[6′-(4, 5--dicarboethoxy-l, 2, 3-triazolyl)-2′, 6′-dideoxy-β-D-arabino--hexopyranosyl]-carminomycinone (23) and -daunomycinone (22), and 3′-(4, 5-dicarboethoxy - l, 2, 3-triazolyl)-4′ -epi-daunomycin (24).  相似文献

The 13C NMR chemical shifts for 1,3-dithiolane and 13 methyl substituted derivatives are reported. Substituent effects are derived and compared with those for cyclopentanes and 1,3-dioxolanes. The magnitude and variety of the substituent effects are best explained with the aid of a half-chair conformation where the S-1? C-2? S-3 plane passes between C-4 and C-5.  相似文献

Some simple rules for predicting the transfer of endo hydrogens in the mass spectra of norbornane carbonyl derivatives are proposed. Transfers from C-6 to C-2 generally occur, but are repressed if a methyl group is on C-1. Transfers from C-5 to C-2 can occur, but are less favorable than 1-3 transfers. Endo-2-acetylnorbornanes undergo a γ-hydrogen transfer from C-5 with ring fission.  相似文献

A comparative study of alkoxycarbonylation processes of both 19-nor-A-ring and A-ring stereoisomers of 1alpha,25-dihydroxyvitamin D3 analogues catalyzed by Candida antarctica lipase B (CAL-B) has been described. The presence of the methyl group in the A-ring at C-2, as in 3-6, has a determining role in the regioselectivity of the biocatalysis, mainly allowing the hydroxyl group at C-5 position to react. For the 19-nor-A-ring stereoisomers 7-10, which lack the C-2 methyl group, the configurations at C-3 and C-5 have a high influence in the selectivity exhibited by CAL-B. Thus, each couple of enantiomers showed opposing regioselectivities depending on the C-3 configuration. When C-3 possesses an (S)-configuration, enzymatic alkoxycarbonylations took place at the C-5-(R) or C-5-(S) hydroxyl groups. However, if the chiral centers at C-3 are (R), CAL-B alkoxycarbonylated the C-3-(R) hydroxyl group independently of the configuration at C-5. The corresponding carbonates are useful A-ring precursors of 1alpha,25-dihydroxyvitamin D3 analogues, selectively modified at the C-1 or C-3 positions. In addition, an improved synthesis of cis A-ring synthons 5 and 6 is described using a Mitsunobu methodology.  相似文献

A comparative study of reactions of 3-methyl-5-phenylisoxazole and 3-methyl-5-phenylisothiazole with electrophilic compounds in the presence of n-BuLi, LICA or LICA-TMEDA is reported. By using LICA-TMEDA, regioselective reactions of the heterocyclic compounds at the C-3 methyl group are obtained. With n-BuLi or LICA and the isoxazole derivative a product mixture at the C-4 position and the C-3 methyl group is found. In the case of isothiazole compound, only with methyl iodide and n-BuLi, the dialkylated product at both positions is formed.  相似文献

The reactivity of 3-methyl-5-phenylisoxazole against electrophilic compounds in the presence of different bases is studied. With n-BuLi, alkylated products at C-4 position and C-3 methyl group, and, in a few cases, dialkylated isoxazoles are obtained. When the reactions are carried out with LICA, the nature of the alkylated products depends on the alkyl halide used. By using LICA-TMEDA, as deprotonating system, regio-selective reaction at the C-3 methyl group is found.  相似文献

Diastereoselective conjugate addition of homochiral lithium (R)-N-allyl-N-alpha-methylbenzylamide to methyl (2E,5E)-hepatadienoate, followed by protecting group manipulation and subsequent iodocyclocarbamation allows a concise route to the core fragment, methyl (3R,5R,6R)-3,6-diamino-5-hydroxyheptanoate, of sperabillins B and D. Differentiation between the C-3 and C-6 primary amino groups of this core amino acid was readily achieved by treatment with acetone, giving the 5,6-isopropylidene and C-3-imine protected diamine, with subsequent regioselective acylation of the C-6-nitrogen facilitating the total synthesis of sperabillin D in 10.8% overall yield, and the first asymmetric synthesis of sperabillin B in 5.8% overall yield.  相似文献

Enantiomerically pure 4-methyl-tetrahydro-5-oxo-2-furancarboxylic acids and esters are prepared by enzymatic resolution of the chiral racemic esters. Their stereochemistry as well as their absolute configurations have been established by chemical correlation. The influence of the alkoxycarbonyl group at C-2 and that of the methyl group at C-4 on the sign of the Cotton effect in their CD spectra have been investigated. Formation of enantiomerically pure hydroxydiesters, precursors of the above-mentioned γ-lactones, by baker’s yeast reduction of the corresponding ketodiesters was unsatisfactory.  相似文献

A new series of 2-substituted 4-amino-N-[(4-benzyl-2-morpholinyl)methyl]-5-chlorobenzamides (4-39) including a few 4-fluorobenzyl analogues were prepared and evaluated for their gastrokinetic activity by determining their effects on the gastric emptying activity of phenol red semisolid meal in rats. The C-2 substituent comprises alkoxy and variously substituted alkoxy groups. Among the derivatives, 4-amino-N-[(4-benzyl-2-morpholinyl)methyl]-2-(n-butoxy)-5-chlorobenza mide (5), its 4-fluorobenzyl (6), and 3-methyl-2-butenyloxy analogues (22) were superior to cisapride and essentially equipotent to the 2-ethoxy analogue (1b, AS-4370 as its citrate) in gastrokinetic activity. These compounds, like AS-4370, had no dopamine D2 receptor antagonistic activity.  相似文献

Inter- and intramolecular hydrogen bonding of an N-H group in pyrazole complexes was studied using ligands with two different groups at pyrazole C-3 and C-5. At C-5, groups such as methyl, i-propyl, phenyl, or tert-butyl were present. At C-3, side chains L-CH(2)- and L-CH(2)CH(2)- (L = thioether or phosphine) ensured formation of chelates to a cis-dichloropalladium(II) fragment through side-chain atom L and the pyrazole nitrogen closest to the side chain. The significance of the ligands is that by placing a ligating side chain on a ring carbon (C-3), rather than on a ring nitrogen, the ring nitrogen not bound to the metal and its attached proton are available for hydrogen bonding. As desired, seven chelate complexes examined by X-ray diffraction all showed intramolecular hydrogen bonding between the pyrazole N-H and a chloride ligand in the cis position. In addition, however, intermolecular hydrogen bonding could be controlled by the substituent at C-5: complexes with either a methyl at C-5 or no substituent there showed significant intermolecular hydrogen bonding interactions, which were completely avoided by placing a tert-butyl group at C-5. The acidity of two complexes in acetonitrile solutions was estimated to be closer to that of pyridinium ion than those of imidazolium or triethylammonium ions.  相似文献

A new labdane diterpenoid, (E)-labda-8(17),12-dien-15,16-olide (1) together with eight known compounds, coronarin D (2), coronarin D methyl ether (3), coronarin D ethyl ether (4), isocoronarin D (5), coronarin B (6), labda-8(17),11,13-trien-15,16-olide (7), (E)-labda-8(17),12-diene-15,16-dial (8) and 16-hydroxylabda-8(17),11,13-trien-15,16-olide (9), are isolated from the rhizomes of Hedychium coronarium. Compounds 2-4, 5 and 9 are isolated as mixtures of C-15, C-14 and C-16 epimers, respectively. Their structures are determined on the basis of their spectroscopic data. The epimeric mixtures of 2 and 3 have not been reported before. Some of them were evaluated for their cytotoxicity.  相似文献
