Two new hydrative carbocyclizations of oxa-alkyne-nitrile functionalities are reported to produce distinct nitrogen-containing heterocycles. Protracted heating of oxoalkynyl nitrile substrates with PtCl(2)/CO/H(2)O in hot 1,4-dioxane gave 2,3-dihydro-1H-pyrido[1.2-b]-isoquinolin-4(6H)-ones. In this hydration reaction, dicarbonyl nitrile intermediates were isolated efficiently after a brief period, and they were subjected to an NHC-based crossed benzoin coupling to give spiro alcohols that further reacted with TfOH to give spiro[indene-2,2'-piperidine]-1,6'(3H)-diones.  相似文献

The thermal and photochemical ring-opening of spiro(3H-naphtho[2,1-b]pyran-3,9'-thioxanthene-10,10-dioxide) results in the facile ring-contraction to 9-(naphtho[2,1-b]furan-2-yl)-9H-thioxanthene-10,10-dioxide . Similar behaviour is displayed by the isomeric spiro(2H-naphtho[1,2-b]pyran-2,9'-thioxanthene-10,10-dioxide) affording 9-(naphtho[1,2-b]furan-2-yl)-9H-thioxanthene-10,10-dioxide , though more severe reaction conditions were required. The comparative ease of this rearrangement for the isomers and was rationalised on the basis of the relative isomer populations of the ring-opened naphthopyrans. The rearrangement of simple mono- and bis-methylsulfonylphenyl substituted photochromic naphthopyrans , was examined; the former failed to rearrange whereas the latter could be induced to rearrange only under prolonged UV irradiation. The photochromism of diastereoisomerically pure sulfoxides derived from the oxidation of spiro(3H-naphtho[2,1-b]pyran-3,9'-thioxanthene) and spiro(2H-naphtho[1,2-b]pyran-2,9'-thioxanthene) resulted in conversion to the most thermodynamically stable trans-isomer in each case.  相似文献

Indan-1-one (1a), 1-tetralone (1b), fluorenone (1c), and anthrone (1d) reacted with mercaptoacetic acid in toluene in the presence of p-toluenesulfonic acid to give spiro[indan-1,2′-[1′,3′]oxathialan]-5′-one (2a), spiro[tetrahydro-naphthalene-1,2′-[1,3′]oxathialan]-5′-one (2b), spiro[fluorene9,2′-[1′,3′]-oxathialan]-5′-one (2c), and spiro[anthracene-9(10H)-2′-[1′,3′]-oxathialan]-5′-one (2d), respectively. Compounds 2a–d reacted with arenes in the presence of aluminum chloride to yield spiro[polycyclic-1′-isothiochroman]-4′-one derivatives 3a–t. The mechanisms of these reactions are discussed. All the synthesized spiroheterocycle derivatives were identified by conventional methods (IR, 1H-NMR spectroscopy) and elemental analyses. © 1996 John Wiley & Sons, Inc.  相似文献

A metal‐free trifluorosulfonate anhydride (TFAA)‐catalyzed strategy for the synthesis of spiro pyrrolo[1,2‐a]quinoxalines from 1‐(2‐aminophenyl)pyrroles and benzoquinones/ketones has been developed. With this general method, spiro pyrrolo[1,2‐a]quinoxalines have been accessed via nucleophilic addition and cyclization. This reaction exhibits good functional group tolerance, and a wide range of products are obtained in moderate to good yields.  相似文献

Condensation of N-substituted 3-azabicyclo[3.3.1]nonan-9-ones with difunctional N,N-, N,S-, and N,O-centered nucleophiles (o-phenylenediamine, 1,2-diphenylethane-1,2-diamine, 2-aminobenzenethiol, cysteine, 2-aminophenol, serine) gave the corresponding spiro heterocyclic compounds fused at the C9 atom. Treatment of N-tert-butoxycarbonyl-substituted spiro compounds with anhydrous hydrogen chloride resulted in elimination of the tert-butoxycarbonyl group with formation of spiro[3-azabicyclo[3.3.1]nonane-9,2′-azole] hydrochlorides.  相似文献

An efficient procedure for the synthesis of spiro[furan-3,3′-indolin]-2-ones and dispiro[cycloalkane-1,2′-furan-3′,3″-indolin]-2″-ones has been achieved in high yields and stereoselectivity by photoinduced electron transfer-catalyzed [3+2] reactions of substituted spiro[indoline-3,2′-oxiran]-2-ones with olefins. The reactions proceed by ring opening of spiro[indoline-3,2′-oxiran]-2-ones via Cβ–O bond cleavage and subsequent cycloaddition with olefins by using 2,4,6-triphenylpyrylium tetarfluoroborate (TPT) as a sensitizer.  相似文献

Treatment of 1,2,3,9a-tetrahydro-9H-imidazo- and 1,2,3,4,10,10a-hexahydropyrimido[1,2-a]indol-2-one derivatives with formic acid gave 1-carbamoylalkyl-2,3,3-trimethyl-2,3-dihydro-1H-indoles. 9-Carbamoylmethyl- and 9-(2-carbamoylethyl)-4a-methyl-2,3,4,4a,9,9a-hexahydro-1H-carbazoles were prepared from 5,6,7,7a-tetrahydro-1H,4H-imidazo- and 1,2,6,7,8,8a-hexahydro-5H-pyrimido[2,1-k]carbazolones in a similar manner. Synthesis of 2-(2-carbamoylpropyl)-2,3,3-trimethyl-2,3-dihydro-1H-indole was carried out by reduction of 1,3-dihydrospiro[2H-indolo-2,2'-piperidine] derivative with Zn in acetic acid solution.  相似文献

The five-membered ring of the indene system in 2,8-dioxo-3a-hydroxy-8a-R-1,2,3a,8a-tetrahydro-8H-indeno[1,2-d]imidazoles is hydrolytically cleaved in concentrated sulfuric acid solutions. A series of spiro[indoline‐3,2′‐[1,3,4]oxadiazol]‐2‐ones were prepared from the reaction of isatin derivatives and hydrazonoyl chlorides through the 1,3‐dipolar cycloaddition reaction. This method has some important aspects, such as mild reaction condition, easy purification, and high yield of products. Also, the synthesis of spiro[acenaphthylene‐1,2′‐[1,3,4]oxadiazol]‐2‐one and spiro[[1,3,4]oxadiazole‐2,9′‐phenanthren]‐10′‐one were studied under the same condition. The structures were confirmed spectroscopically (IR, 1H‐ and 13C‐NMR, and EI‐MS) and by elemental analyses. A plausible mechanism for this reaction is proposed.  相似文献

The first examples of sigmatropic rearrangements of ene-endo-spirocyclic, tetrahydropyridine-derived ammonium ylids are reported. Thus, spiro[6.7]-ylids rearrange primarily by a [2,3]-pathway, whereas the analogous [6.6]-ylids rearrange by [1,2]- and [2,3]-mechanisms in roughly equal proportions. This method serves as a rapid entry to the core of a range of alkaloids bearing a pyrrolo[1,2-a]azepine or octahydroindolizidine nucleus.  相似文献

New 1-cycloalkenyl-1-diazenes have been obtained in good yields from cyclic β-ketoesters and hydrazine derivatives. They furnished new cycloalkyl[d][1,3]thiazolines with thioamides or new spirocycloalkyl-thiazolinones with thioureas. Moreover they gave, with imidazolidine-2-thione and tetrahydropyrimidine-2-thione, new and interesting spiro[cycloalkyl-1,2′-imidazo[2,1-b][1,3]thiazole] or spiro[cycloalkyl-1,2′-[1,3]thiazolo[3,2-a]pyrimidine] derivatives, respectively. Cycloalkyl[d][1,3]thiazolines were useful for the further preparation of unknown thia-triaza-tricyclo derivatives. Novel hexahydro-1,3-benzothiazoles have been achieved by reaction of N,N′-dialkylthioureas on N-1-phenyl-2-(1-cyclohexenyl)-1-diazene-1-carboxyamide. The acidic hydrolysis of spirocycloalkyl-thiazolinones produced 2-imino-5-(ω-carboxyalkyl)-4-thiazolidinones.  相似文献

1-(2-Bromomethylphenyl)-1-cyclopentanecarbonitrile has been synthesized by the cycloalkylation of (2-methylphenyl)acetonitrile with 1,4-dibromobutane with subsequent bromination of the intermediate product with N-bromosuccinimide. The product serves as a convenient intermediate in the synthesis of derivatives of a series of heterospiro systems. Condensation of it with primary amines leads to the hydrobromides of 1,2,3,4-tetrahydrospiro[isoquinoline-4,1'-cyclopentane]-3-imines, but with vicinal eneamino carbonyl compounds derivatives results in previously undescribed condensed spirocyclic systems, viz. spiro[5H-isoquino[2,3-a]quinazoline-7,1'-cyclopentane] and spiro[4H-thieno[3',2':5,6]pyrimido[1,2-b]isoquinoline-6,1'-cyclopentane].  相似文献

Diaminomethylene- and aminomethylthiomethylenehydrazones [2] of cyclic ketones 1–8 readily reacted with ethoxymethylenemalononitrile to give spiro[cycloalkane-1,2′-[1,2′,4′]triazolo[1,5′-c]pyrimidine-8′-carbonitrile] derivatives 12–19 through the electrocyclic reaction of the initially formed condensation products 26 in moderate to high yields. The spiro[cyclopentanetriazolopyrimidine] derivatives underwent ring-opening at the cycloalkane moiety upon heating in solution to give 2-alkyl-5-substituted-[1,2,4]triazolo[1,5-c]pyrimidine-8′-carbonitriles 20–23 . When an alkyl substituent was introduced into the cyclopentane ring, cleavage of the spiro compounds occurred preferentially at the cyclopentane moiety between the spiro carbon and the more branched one. In contrast, the cyclohexane ring, especially of spiro-5-amino-triazolopyrimidines 17 and 18 strongly resisted to ring-opening under similar conditions, but those of 5-methylthiotriazolopyrimidines 14 gave up to 17 percent of cleavage after prolonged heating in hot ethanol. 2-t-Butyl-5-methylthio-2,3-dihydro[1,2,4]triazolo[1,5-c]pyrimidine-8-carbonitrile 25 [R3 = C(CH3)3] was highly susceptible to the cleavage even at room temperature and produced the corresponding 2-unsubstituted triazolopyrimidine 24 with loss of the t-butyl group.  相似文献

An efficient, scaleable synthesis approach towards the spirocyclic oxindole analogue 1'-(tert-butoxycarbonyl)-2-oxospiro[indoline-3,4'-piperidine]-5-carboxylic acid (1) is described. The key steps are dianion alkylation and cyclization of ethyl 2- oxindoline-5-carboxylate (4) and demethylation of the resulting spirocyclic oxindole ethyl 1'-methyl-2-oxospiro[indoline-3,4'-piperidine]-5-carboxylate (5). The target compound was obtained in an overall yield of 35 % over eight steps without resorting to chromatographic purification.  相似文献

The arylidienes of fluorinated spiro thiazolidines (5) containing α,β-unsaturated function have been used as component of Micheal addition with equimolar amount of 2-aminopyridine (6a) to give novel fluorinated spiro [indole-3,2′-pyrido[1,2-a]thiazolo[5,4-e]pyrimidines] (7) in a single step under microwaves in presence of montmorillonite KSF as solid support. The new improved synthetic method for fluorinated spiro [indole-3,2′-thiazolo[4,5-d]pyrimidines] (8) has also been developed involving the reaction of (5) with thiourea under monomode microwave reactor. Comparison with conventional synthesis and multimode microwave oven indicated the enhanced yield with faster reactions under monomode microwave reactor. Structure-activity relationships between the chemical structures and the antimycobacterial, antifungal activity of the evaluated compounds are also discussed.  相似文献
