Four‐ and five‐bond heteronuclear J‐couplings between the hydrogen H‐8 and carbons C‐6 and C‐2 in a series of 7‐ and 9‐benzyl substituted purine derivaties with variuous substituents in positions 2 and 6 were studied by coupled 13 C NMR and H,C‐HMBC experiments and by DFT calculations. We have found that for some of the derivatives, the five‐bond coupling H8‐C2 is higher than the four‐bond H8‐C6 coupling, which is also evidenced by a stronger crosspeak in the HMBC. This finding contradicts the generally accepted opinion that only strong three‐bond crosspeaks and one weak four‐bond H8‐C6 crosspeak can be observed in the HMBC spectra of purine derivatives. The misinterpretation of HMBC spectra may lead to an incorrect determination of the purine derivatives' structure. Copyright © 2012 John Wiley & Sons, Ltd.  相似文献

The thorough analysis of highly complex NMR spectra using pure shift NMR experiments is described. The enhanced spectral resolution obtained from modern 2D HOBS experiments incorporating spectral aliasing in the 13C indirect dimension enables the distinction of similar compounds exhibiting near‐identical 1H and 13C NMR spectra. It is shown that a complete set of extremely small Δδ(1H) and Δδ(13C) values, even below the natural line width (1 and 5 ppb, respectively), can be simultaneously determined and assigned.  相似文献

A detailed analysis of the proton high‐field (600 MHz) NMR spectra of tomato juice and pulp is reported for the first time. A combination of J‐resolved, COSY, TOCSY, DOSY, 1H–13C HSQC and 1H–13C HMBC 2D sequences was used to assign each spin system and to separate the components of the complex patterns in the 1D overlapped proton spectra. To obtain resolved proton spectra of tomato pulps the high‐resolution magic angle spinning technique was used; a comparison with the liquid‐state NMR spectra of the corresponding juices was accomplished. The addition reaction of 2,2‐bis‐[4‐(2,3‐epoxypropoxy)‐phenyl]‐propane (DGEBA) and preformed complexes of metal ions and disecondary diamines led to a large quantity of cyclic epoxide–amine oligomers. As shown by gel permeation chromatographic analysis, cycles of n = 1, 2, and 3 were formed. Functional epoxide end groups of the prepared oligomers were completely missing in the IR and 1H NMR and 13C NMR spectra. In the fast atom bombardment and matrix‐assisted laser desorption/ionization mass spectra, the molecular ions of the n = 1, 2, 3 cycles of DGEBA and N,N′‐dibenzyl‐5‐oxanonanediamine‐1,9 were detected at m/z = 680, 1361, and 2042. © 2003 Wiley Periodicals, Inc. (R)‐3‐(2′‐hydroxyprop‐1‐yl) adenine 4 was obtained by alkylation of adenine 1 with R‐propylene carbonate 2 in the presence of pulverized sodium hydroxide in the synthetic process of tenofovir. The elucidation of the structure of 4 was confirmed by single crystal X‐ray diffraction and NMR experiments such as 1D 1H, 13C, and DEPT, as well as 2D COSY, HSQC, and HMBC spectra.  相似文献

NMR spectroscopy was applied for quantitative and qualitative characterization of the chemical composition and microstructure of a series of poly(3‐hydroxybutyrate‐co‐3‐hydoxyvalerate) copolymers, P(3HB‐co‐3HV), synthesized by mixed microbial cultures at several different feeding strategies. The monomer sequence distribution of the bacterially synthesized P(3HB‐co‐3HV) was defined by analysis of their high‐resolution 1D 13C NMR and 2D 1H/13C HSQC and 1H/13C HMBC NMR spectra. The results were verified by employment of statistical methods and suggest a block copolymer microstructure of the P(3HB‐co‐3HV) copolymers studied. Definitive distinction between block copolymers or a mixture of random copolymers could not be achieved. NMR spectral analysis indicates that the chemical composition and microstructure of the copolymers can be tuned by choosing a correct feeding strategy. We report the total assignments of the 13C and 1H NMR spectra of some 4‐methyl‐2‐oxo‐(2H)‐pyrido[1,2‐a]pyrimidine and 2‐methyl‐4‐oxo‐(4H)‐pyrido[1,2‐a]pyrimidine derivatives. The products were characterized by 1H and 13C NMR and reported data for similar compounds. No comparative data for the two sets of isomeric compounds with respect to 13C and 1H NMR have been reported to date. We made some detailed studies of the 2D NMR spectra of these compounds and observed that assignments made for non‐protonated carbon atoms by us and those reported in the literature for similar compounds need correction. The revised assignments were made on the basis of heteronuclear single quantum correlation (HSQC) and heteronuclear multiple bond correlation (HMBC) techniques. An unsymmetrical heterocyclic diamine, 1,2‐dihydro‐2‐(4‐aminophenyl)‐4‐[4‐(4‐aminophenoxy)‐4‐phenyl]‐(2H)phthalazin‐1‐one, was synthesized. Its 1H and 13C NMR spectra were completely assigned by utilizing the two‐dimensional heteronuclear 13C–1H multiple‐bond coherence (HMBC) spectroscopy, and heteronuclear 13C–1H one‐bond correlation spectroscopy, homonuclear shift correlation spectroscopy (H,H‐COSY) and rotating frame Overhauser enhancement spectroscopy (ROESY). The structure of the compound was shown to be the phthalazinone rather than the phthalazine ether from cross peaks and chemical shifts of the protons. 4‐Methyl‐6,8‐dihydroxy‐7H‐benz[de]anthracen‐7‐one was isolated from the sap of Aloe by column chromatography. Its 1H and 13C NMR spectra were completely assigned by utilizing two‐dimensional 1H‐detected heteronuclear one‐bond (HMQC) and multiple‐bond (HMBC) chemical shift correlation experiments together with 1H–1H COSY and DEPT techniques. These techniques were also valuable in assigning the protons and carbons of those benzanthrone compounds which were previously incompletely reported because of the overlap of proton signals. Two novel oligosaccharides, mono‐ and difructosyllactosucrose {[O‐β‐D ‐fructofuranosyl‐(2 → 1)]n‐β‐D ‐fructofuranosyl‐O‐[β‐D ‐galactopyranosyl‐(1 → 4)]‐α‐D ‐glucopyranoside, n = 1 and 2} were synthesized using 1F‐fructosyltransferase purified form roots of asparagus (Asparagus officinalis L.). Their 1H and 13C NMR spectra were assigned using several NMR techniques. The spectral analysis was started from two anomeric methines of aldose units, galactose and glucose, since they showed separate characteristic signals in their 1H and 13C NMR spectra. After assignments of all the 1H and 13C signals of two units of aldose, they were discriminated as galactose and glucose using proton–proton coupling constants. The HMBC spectrum revealed the galactose residue attached to C‐4 of glucose, fructose residue attached to the C‐1 of glucose, and further fructosyl fructose linkage extended from the glucosyl fructose residues. The1H and 13C NMR resonances for acridine derivatives 9‐substituted with chloro, allylamino and propargylamino groups were completely assigned using a concerted application of gs‐COSY, gs‐HMQC and gs‐HMBC experiments. 9‐(N‐Allyl)‐ and 9‐(N‐propargyl)acridinamine derivatives present amino–imino tautomerism including a large broadening of 1H and 13C NMR signals at room temperature. To obtain suitable resolution, therefore, these latter compounds were studied at 370 K in DMSO‐d6 solutions and showed a complete shift towards the imino tautomers. Unsymmetrical and generalized indirect covariance processing methods provide a means of mathematically combining pairs of 2D NMR spectra that share a common frequency domain to facilitate the extraction of correlation information. Previous reports have focused on the combination of HSQC spectra with 1,1‐, 1,n‐, and inverted 1JCC 1,n‐ADEQUATE spectra to afford carbon–carbon correlation spectra that allow the extraction of direct (1JCC), long‐range (nJCC, where n ≥ 2), and 1JCC‐edited long‐range correlation data, respectively. Covariance processing of HMBC and 1,1‐ADEQUATE spectra has also recently been reported, allowing convenient, high‐sensitivity access to nJCC correlation data equivalent to the much lower sensitivity n,1‐ADEQUATE experiment. Furthermore, HMBC‐1,1‐ADEQUATE correlations are observed in the F1 frequency domain at the intrinsic chemical shift of the 13C resonance in question rather than at the double‐quantum frequency of the pair of correlated carbons, as visualized by the n,1, and m,n‐ADEQUATE experiments, greatly simplifying data interpretation. In an extension of previous work, the covariance processing of HMBC and 1,n‐ADEQUATE spectra is now reported. The resulting HMBC‐1,n‐ADEQUATE spectrum affords long‐range carbon–carbon correlation data equivalent to the very low sensitivity m,n‐ADEQUATE experiment. In addition to the significantly higher sensitivity of the covariance calculated spectrum, correlations in the HMBC‐1,n‐ADEQUATE spectrum are again detected at the intrinsic 13C chemical shifts of the correlated carbons rather than at the double‐quantum frequency of the pair of correlated carbons. HMBC‐1,n‐ADEQUATE spectra can provide correlations ranging from diagonal (0JCC or diagonal correlations) to 4JCC under normal circumstances to as much as 6JCC in rare instances. The macrocyclic lactam alkaloid (±)‐(2R*,3R*)‐3‐hydroxycelacinnine ( 1 ) derived from spermidine was synthesized via stereoselective epoxide‐ring opening with magnesium azide and cesium carbonate promoted macrocyclization of the ditosylated diamino precursor 12 with 1,4‐dibromobutane in the two key steps (Scheme 2). 1H‐ and 13C‐NMR Signal assignments from COSY, HSQC, and HMBC 2D NMR data of the synthesized 1 were compared with the earlier‐described data of the natural 3‐hydroxycelacinnine. The similarity of their 13C‐NMR spectra point to the correctness of the proposed constitutional formula for natural 3‐hydroxycelacinnine; however, different 1H‐NMR chemical shifts and coupling constants (J(2,3)=9.0 vs. 1.2 Hz, resp.) in the α‐hydroxy‐β‐amino lactam moiety suggest that natural 3‐hydroxycelacinnine is the 2,3‐cis‐epimer of one synthetic (±)‐ 1 .  相似文献

Three new nor‐neo‐clerodane diterpenes, named chamaedryoside A (1), B (2) and C (3), have been isolated from the organic extracts of Teucrium chamaedrys (L.) and their structural characterization has been accomplished by 1H and 13C‐NMR spectra, and DEPT, by COSY, TOCSY, HSQC, HSQC‐TOSCY and HMBC experiments, as well as by ESI‐MS/MS techniques. Irradiation of styryl‐4‐pyrones 1a‐1d or 2a‐2e (6‐9 × 10?3 M, methanol solution) with filtered (RAYONET photochemical reactor, 300 nm) or unfiltered uv‐light (high‐pressure mercury arc lamp) under aerobic conditions led mainly to dimeric products. Parent 5‐hydroxy‐substituted compounds 1a‐1d yielded exclusively “half‐cage” dimers 3a‐d characteristic for 4‐pyrone dimerization. 5‐Methoxy‐analogues 2a‐2e behave like typical stilbene structures and the mixture of tetrasubstituted cyclobutanes 4 and 5 accompanied with minor amount of phenanthrene‐like compound 6 were the only isolable products of the irradiation. The structure elucidation of products is based on spectral data obtained from MS, IR, 1H NMR and 13C NMR spectra applying COSY, APT, HETCOR, HMBC and NOESY techniques.  相似文献

3β‐(Stearyloxy)olean‐12‐ene was isolated from a hexane extract of Austroplenckia populnea Reiss (Celastraceae) leaves. The structure was solved by means of quantitative 13C‐NMR, HMBC, HMQC, COSY, NOESY, and NOE difference spectra. The mass spectrum showed an [M+1]+ ion peak at m/z 693, and the molecular formula C48H84O2 was confirmed by combustion analysis.  相似文献
