Summary The separation of fullerenes with a monomeric octadecylsilica bonded phase using n-hexane or toluene/methanol mobile phase systems is described. Analytical and preparative separations, incorporating on-line UV/VIS spectral measurements, confirmed the existence of large fullerenes such as C76, C78 and C84. Summary This paper reports the use of liquid chromatography for the separation and determination of the major cannabinoids extracted from hashish samples. The direct coupling to the mass spectrometer enables the selective identification both of neutral and acidic cannabinoids. The developed method does not require any preliminary derivatization and should, therefore, be of interest in forensic analysis for simple and unequivocal determination of hashish constituents.Part of this work was presented at the 2nd Italian-Spanish Joint Meeting of Medicinal Chemistry, 30 August–3 September 1995, Ferrara, Italy.  相似文献

Multidimensional liquid chromatography (MDLC) may be used in either (i) the profiling mode where it is the objective to fractionate all components in a mixture or (ii) the targeted component mode in which it is the objective to determine specific analytes. This paper focuses on targeted component analysis from complex mixtures, addressing the critical operations of analyte selection and transport from the first to the second dimension. Although the physical operation of switching a component into the second dimension with computer controlled valving is simple, it is shown that changes in analyte retention time and peak width with column age and fouling are a serious problem. The analyte moves out of the preselected time window for valve switching and quantitation is compromised in the second dimension. It is proposed that a solution to the “drifting peak” phenomenon in targeted component analysis is to use binary mobility elution in the first dimension. Binary mobility refers to those systems, such as affinity chromatography, in which analyte mobility is generally either 0 or 1 relative to mobile phase velocity. Coupling these binary changes in analyte mobility in the first dimension with valve switching eliminates the “drifting peak” phenomenon. In addition, it is shown that a wide time window may be used in affinity separations without compromising the separation or accumulating contaminants. Several cases are described in which immunosorbents were used with reversed phase columns to provide quantitative targeted component analyses from complex mixtures.  相似文献

A rapid analytical method including pressurized liquid extraction (PLE) and liquid chromatography-electrospray ionisation-mass spectrometry (LC-ESI-MS) has been developed for the determination of tocopherols and tocotrienols in cereals. The pressurized liquid extraction parameters were optimized in order to maximize the extraction efficiency. The use of methanol as extraction solvent at a temperature of 50 °C and a pressure of 110 bar, using one cycle of extraction with a static time of 5 min, provided the best results. A good LC separation was achieved using a C18 column and a solution of 6.0 mM ammonia in methanol/water (97:3, v/v) as the mobile phase at a flow rate of 0.2 mL min−1. MS coupling with an ESI interface in the negative ion mode was used as the detection technique. In the present work, it is shown that the addition of a base to the mobile phase is required to enhance the ionization of tocopherols and tocotrienols in negative ion mode electrospray ionization. The applicability of the method to cereal samples was confirmed. The reproducibility of the procedure was good, with relative standard deviations in the 6-10% range. The recoveries of added tocopherols from cereal samples ranged from 91 to 109%.  相似文献

Summary The determination of selected pesticides and phenols in Portuguese river water samples was carried out from April to September, 1999. The method involved 200 mL samples taken by offline, solid phase extraction by OASIS polymeric cartridges followed by liquid chromatography-atmospheric pressure, chemical ionization-mass spectrometry (LC-APCI-MS). Recoveries of pesticides were 50–96% and 72–120% for the Platform and HP 1100 instruments, respectively. Chlorophenols gave recoveries of 60–91%. Triazines and transformation products like desethylatrazine (DEA) and desisopropylatrazine (DIA) and compounds such as diuron and chlorophenols were positively identified by LC-APCI-MS. The levels detected of the different compounds varied from 0.01–2.61 μg L−1, the most frequently detected compounds being, atrazine, simazine, terbuthylazine, alachlor, metolachlor, Irgarol, diuron, 2,4,6-trichlorophenol, desisopropylatrazine and desethylatrazine.  相似文献

A sensitive and reliable method was developed and validated for detection and confirmation of melamine in egg based on gas chromatography-mass spectrometry (GC-MS) and ultra-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS). Trichloroacetic acid solution was used for sample extraction and precipitation of proteins. The aqueous extracts were subjected to solid-phase extraction by mixed-mode reversed-phase/strong cation-exchange cartridges. Using ultra-performance liquid chromatography and electrospray ionization in the positive ion mode, melamine was determined by LC-MS/MS, which was completed in 5 min for each injection. For the GC-MS analysis, extracted melamine was derivatized with N,O-bis(trimethylsilyl)trifluoracetamide prior to selected ion monitoring detection in electron impact mode. The average recovery of melamine from fortified samples ranged from 85.2% to 103.2%, with coefficients of variation lower than 12%. The limit of detection obtained by GC-MS and UPLC-MS/MS was 10 and 5 μg kg−1, respectively. This validated method was successfully applied to the determination of melamine in real samples from market.  相似文献

Summary A liquid chromatographic method has been developed for analysis of industrial polyglycerols, precursors of polyglycerol fatty esters, which are non-ionic surfacetants. The method utilizes two complementary chromatographic systems: porous graphitic carbon and an aminopropyl polymer with an acetonitrile-water mixture as mobile phase. Detection of these non-UV absorbing compounds was effected by means of evaporative light-scattering detection. Their structures were determined by comparison of their retention with that of synthesized standards, and by mass spectrometry.  相似文献

Summary A new analytical technique was established to improve the performance of DSP toxin determination in different sample matrices. High performance size exclusion chromatography (gel permeation chromatography, SEC) was applied for the clean-up of raw extracts from algal cells and mussel tissue prior to the determination of DSP toxins by LC/MS. The proposed protocol can be performed totally automatically and enables fast and sensitive analysis of large sample numbers. The recovery of the entire method protocol (consisting of extraction, clean-up and LC/MS determination) was approximately 70% with good repeatability (standard deviation ranging from 1.9% to 5.0% in the concentration range analyzed).  相似文献

Nonaqueous reversed-phase chromatography stabilizes the electrospray ionization, resulting in sensitivity that enables direct measurement of skin lipid extracts with no special sample preparation. Assignment of individual signals to the corresponding ceramide species is based on interpretation of the fragment spectra from MS-MS experiments. This enables much finer differentiation between ceramdies than that achievable by thin-layer chromatography. Summary Mass spectra of 12 triazines were obtained by electron impact (EI), positive-ion chemical ionization (PCI) and negative-ion chemical ionization (NCI) using methane and isobutane as reagent gases. EI mass spectrometry is more sensitive than PCI and NCI, although the chemical ionization modes increase selectivity markedly. A pre-column packed with polymer stationary phase was employed to preconcentrate surface and drinking water samples. After desorption of the analytes with ethyl acetate, an aliquot was injected directly into the GC-MS system. Atrazine and simizine were found in these samples at 10–80 ppt levels. The limits of detection for both herbicides were below 10 ppt in drinking water.  相似文献

Summary Liquid chromatography-mass spectrometry (LC-MS) with atmospheric pressure chemical ionization (APCI), and gas chromatography-mass spectrometry (GC-MS) with electron impact ionization (EI), are compared for the determination of eight pesticides in oranges. Seven of the selected pesticides, chlorpyriphos, chlorpyriphos-methyl, imazalil, α and β-endosulfan, endosulfan sulphate and dicofol, are commonly determined by GC whereas one, thiabendazole, can only be directly determined by LC. Primary ions [M-H] or [M-Cl+O] are obtained using LC-APCI-MS in negative ionization (NI) mode. In contrast, a high degree of fragmentation is reported with GC-MS. Both techniques were applied to oranges, which had been previously extracted with ethyl acetate and anhydrous sodium sulphate. The data indicate equivalent detection limits that range from 0.01 to 0.1 mgkg−1 and similar degree of specificity. Mean recoveries ranged from 82% for α-endosulfant to 96% for imazalil, with relative standard deviation ranging from 7 to 17%.  相似文献

Metabolite identification (Met ID) is important during the early stages of drug discovery and development, as the metabolic products may be pharmacologically active or toxic in nature. Liquid chromatography-mass spectrometry (LC-MS) has a towering role in metabolism research.This review discusses current approaches and recent advances in using LC-MS for Met ID. We critically assess and compare various mass spectrometers, highlighting their strengths and limitations. Citing appropriate examples, we cover recent LC and ion sources, isotopic-pattern matching, hydrogen/deuterium-exchange MS, data dependent analyses, MSE, mass defect filter, 2D and 3D approaches for the elucidation of molecular formula, polarity switching, and background-subtraction and noise-reduction algorithms. A flow chart outlines a comprehensive strategy for Met ID, including a focus on reactive metabolites.  相似文献

In this study the impact of solvent conditions on the performance of μLC/MS for the analysis of basic drugs was investigated. Our aim was to find experimental conditions that enable high-performance chromatographic separation particularly at overloading conditions paired with a minimal loss of mass spectrometric detection sensitivity. A focus was put on the evaluation of the usability of different kinds of acidic modifiers (acetic acid (HOAc), formic acid (FA), methansulfonic acid (CH3SO3H), trifluoroacetic acid (TFA), pentafluoropropionic acid (PFPA), and heptafluorobutyric acid (HFBA)). The test mixture consisted of eleven compounds (bunitrolol, caffeine, cocaine, codeine, diazepam, doxepin, haloperidol, 3,4-methylendioxyamphetamine, morphine, nicotine, and zolpidem). Best chromatographic performance was obtained with the perfluorinated acids. Particularly, 0.010–0.050% HFBA (v/v) was found to represent a good compromise in terms of chromatographic performance and mass spectrometric detection sensitivity. Compared to HOAc, on average a 50% reduction of the peak widths was observed. The use of HFBA was particularly advantageous for polar compounds such as nicotine; only with such a hydrophobic ion-pairing reagent chromatographic retention of nicotine was observed. Best mass spectrometric performance was obtained with HOAc and FA. Loss of detection sensitivity induced by HFBA, however, was moderate and ranged from 0 to 40%, which clearly demonstrates that improved chromatographic performance is able to compensate to a large extent the negative effect of reduced ionization efficiency on detection sensitivity. Applications of μLC/MS for the qualitative and quantitative analysis of clinical and forensic toxicological samples are presented.  相似文献

Summary Thermospray liquid chromatography-mass spectrometry has been applied to the identification of by-products in precolumn derivatization by phenylisocyanate. These byproducts are eluted early in the same chromatographic region as the low molecular weight derivatives and were located by chromatographic analysis of a blank sample. Their identification would offer further qualitative information in the use of phenylisocyanate as a derivatizing agent. Five compounds resulted from the reaction of phenylisocyanate and the reaction medium were identified: two from a reaction between phenylisocyanate and methanol, two from the reaction between phenylisocyanate and water, and one from the polymerisation of phenylisocyanate.  相似文献

Summary The major gene products of human orosomucoid, GP1 and GP2, were purified using immobilized copper affinity chromatography [Cu(II)-IMAC], with 3 mM imidazole as eluent. Both gene products bound to the Cu(II)-IMAC column in the presence of 1 M NaCl, but at different pHs. GP1 was not retained after treatment with diethylpyrocarbonate (DEPC). This modification was characterized using difference absorbance spectrophotometry and mass spectrometry. The latter provided unambiguous assignment of some of the modified residues. No correlation was observed between the modification of histidine/tyrosine and protein retention. Furthermore, removal of the carbethoxy groups of modified histidine and tyrosine by hydroxylamine treatment did not improve the retention. Therefore neither histidine nor tyrosine could be the critical residues in metal recognition. Results from mass spectrometric analysis of retained and unretained fractions of DEPC modified GP 1 indicated that the lysine residues 130/135 and 152 were modified significantly in both fractions, but to a relatively less extent in the retained one. We suggest that the retnetion of GP1 involves several residues including lysines, and that a critical number of these is necessary for retention.  相似文献

In this study, capillary electrophoresis and capillary liquid chromatography coupled to mass spectrometry (CE-TOF-MS and μLC-TOF-MS) were used to detect and characterise human transferrin (Tf) glycopeptide glycoforms obtained by tryptic digestion. After selecting μLC-TOF-MS because of improved performance in analysis of N413 and N611 glycopeptide glycoforms, the proposed methodology was applied to serum samples. Two immunoaffinity columns were employed to isolate Tf from serum samples. Both columns were activated with the same anti-Tf antibody but using two different bonding chemistries. After immunoaffinity purification and digestion, serum samples from a teetotal individual (as control) and from individuals with low and high alcohol dependence were analysed by μLC-TOF-MS. Relative abundance of each glycoform was useful to estimate the degree of alcohol dependence of each individual. Finally, the established methodology was used to analyse serum samples from specific individuals with an unknown degree of alcohol dependence.  相似文献
