Summary A method for the determination of low relative molecular mass carboxylic acids (C1–C4) in water is reported. The acids are converted to p-bromophenacyl esters prior to a glass-capillary gas chromatographic separation. By utilizing electron-caputre detection the detectability is substantially improved compared to flame-ionization detection. A comparison of three different ways to treat the water samples and to produce the derivatives is made. It is shown that the , p-dibromoacetophenone reagent decomposes to a small extent which limits the utility of the reagent. Nevertheless a detection limit for formic acid of approximately 2.5 mgl–1 is obtained. The method is applied to the determination of formic and acetic acids in a paper kraft water sample.  相似文献

A new colorimetric method for the determination of carboxylic acids using bromocresol green reagent is established. The formed color follows Beer's Lambert law and is stable for 24 hr. The method is successfully applied for measuring the disappearance of volatile fatty acids in P. oxalicum culture media. The method is reproducable and is used also for the quantitative determination of carboxylic acids in Dowex 1-X8 eluates. A linear relationship between the pK1 of different carboxylic acids and the calculated molar extinction coefficients of the color formed at 440 nm is demonstrated.  相似文献

The determination of the degree of substitution (DS) of fatty acid cellulose esters, representing a broad range of substituents (C6, C12, C18 and C22), was performed by alkaline hydrolysis of the ester groups and the quantification of fatty acids by gas chromatography-mass spectrometry (GC-MS) as their trimethylsilyl derivatives. The method was optimized and compared with established techniques for the DS determination (elemental analysis and alkaline hydrolysis/titrimetry). The results demonstrated that alkaline hydrolysis/GC-MS is a rapid, reliable and powerful method for analysis of fatty acid cellulose esters, particularly when different acyl substituents are present.  相似文献

The interaction of bleomycinA5 with nucleic acids has been investigated by using resonance Rayleigh scattering (RRS), molecular absorption and fluorescence spectra. The result shows that in near pH 2.2 buffer medium and absence of any metal ions, nucleic acids are capable of binding with bleomycinA5 (BLMA5) to form complexes which can remarkably enhance the RRS intensity and result in bathochromic and hyperchromic molecular absorption of nucleic acids and fluorescence quenching of bleomycinA5. The RRS spectral characteristics for the binding products of bleomycinA5 with various DNA and RNA are similar, and the maximum RRS peaks are at 301 nm for ctDNA and sDNA, 370 nm for hsDNA, 310 nm for RNAtypeVI and RNAtypeIII, respectively. The increments of RRS intensity are greatly different in which DNA enhances greatly and RNA enhances lightly. In this work, the optimum conditions of the interaction and some influencing factors have been investigated. The reaction mechanism and a binding model for the interaction of BLMA5 with the nucleic acids are discussed. In addition, a highly sensitive, simple and rapid new method for the determination of DNA has been developed. The detection limits (3σ) are 5.7 ng/mL for ctDNA, 7.4 ng/mL for sDNA and 9.2 ng/mL for hsDNA, respectively. The method can be applied to determination of trace amounts of DNA.  相似文献

A new ion chromatographic (IC) system, in which n-tetradecylphosphocholine (TDPC, a phosphobetaine type of zwitterionic surfactant) was used as the stationary phase, pure water as the mobile phase, and conductivity as the method of detection, has been developed for the determination of inorganic acids. Five model acids, HCl, HNO3, HClO4, H2SO4, and H3PO4, were separated to baseline and eluted in the order H3PO4 > HCl > HNO3 > H2SO4 > HClO4. When peak areas were plotted against the concentrations of the acids in samples, linear calibration curves were obtained. Ultimate determination limits were approximately 1 mmol L–1, but the discrimination of the method between solutions of different concentration was better than 10 μmol L–1 for those model analytes. Salts of divalent cations could also be separated, but they were eluted faster than the acids. No separation was observed for the salts of monovalent cations. This newly proposed approach is applicable to the simultaneous determination of the inorganic acids (produced by reactions of NOx, SOx, and HCl with water) in aerosols.  相似文献

An indirect method for the determination of ca. 10?4 M amino acids is described which is based on the reaction of the latter with 2,4,6-trinitrobenzene-1-sulphonic acid at pH 11.7 and the measurement of the S2 emission in a hydrogen-nitrogen flame that is produced from the resulting compound after the addition of hydrochloric acid. The optimum conditions for the determination of amino acids were investigated. The amino acids examined were glycine, alanine, valine, methionine, serine, threonine and histidine.  相似文献

A high‐speed separation method of capillary MEKC with LIF detection had been developed for separation and determination of amino acids in laver. The CE system comprised a manual slotted‐vial array (SVA) for sample introduction that could improve the separation efficiency by reducing injection volume. Using a capillary with 80 mm effective separation length, the separation conditions for amino acids were optimized. Applied with the separation electric field strength of 300 V/cm, the ten amino acids could be completely separated within 2.5 min with 10 mol/L Na2HPO4–NaOH buffer (pH = 11.5) including 30 mmol/L SDS. Theoretical plates for amino acids ranged from 72 000 to 40 000 (corresponding to 1.1–2.0 μm plate heights) and the detection limits were between 25 and 80 nmol/L. Finally, this method was applied to analyze the composition of amino acids in laver and eight known amino acids could be found in the sample. The contents of five amino acids, tyrosine, glutamic acid, glycine, lysine, and aspartic acid that could be completely separated in real sample were determined. The recoveries ranged from 82.3% to 123% that indicated the good reliability for this method in laver sample analysis.  相似文献

We developed a simple, rapid and reliable method for determination of 20 common amino acids based on derivatization with 9‐fluorenylmethyl chloroformate (FMOC‐Cl) and RP‐LC/UV, this method was first introduced into quantitative analysis of amino acids. The amino groups of amino acids were trapped with FMOC‐Cl to form amino acid‐FMOC‐Cl adducts which can be suitable for LC‐UV. Chromatographic separation was performed on a C18 column with a mobile phase gradient consisting of acetonitrile and sodium acetate solution. This method was shown to be sensitive for 20 common amino acids. In the intra‐day precisions assay, the range of RSDs was 3.21‐7.67% with accuracies of 92.34‐102.51%; for the inter‐day precisions assay, the range of RSDs was 5.82‐9.19% with accuracies of 90.25‐100.63%. The results also indicated that solutions of amino acids‐FMOC‐Cl can be kept at room temperature for at least 24 h without showing significant losses in the quantified values. The validated method was successfully applied to the determination of major four kinds of amino acids in R. isatidis samples (Arg, Pro, Met and Val). The total content of amino acids in different origin R. isatidis was 13.32‐19.16 mg/g. The differences between R. isatidis samples were large using HCA.  相似文献

Annihilation of the contribution of one chemical component from the original data matrix is a general method in rank annihilation factor analysis (RAFA). However, RAFA is not applicable for studying the protonation equilibria of multiprotic acids. In this work, a two-rank annihilation factor analysis (TRAFA) method was proposed for determination of the acidity constants of diprotic acids. After recording the electronic absorbance spectra of the acids at different pH, the contributions of both H2A and A2− were annihilated from the absorbance data, which made feasible the determination of two successive acidity constants. The method was validated by analysis of simulated data and its application to the determination of the acidity constants of calmagite, as a reference compound. A close agreement was obtained between the resulted values by TRAFA and the declared values. Indeed, the method was used for determination of the acidity constants of two new chromenone derivatives in binary solvents mixtures of methanol and water. The effects of changing solvent composition on acidity constant data were explained by linear solvation free energy relationships (LSFER) utilizing solvatochromic parameters.  相似文献

A new method for simultaneous determination of organic acids in red wine and must by liquid chromatography was studied. The determination of organic acids in wines can be achieved in less than 13 min, preceded only by a simple sample dilution and filtration step. With this method, the chromatographic separation of eight organic acids and interfering peaks present in red wine, required only one reversed phase column (Waters Atlantis dC18 column, 4.6 × 150 mm ID, 5 μm). As mobile phase, isocratic acetonitrile–0.01 mol L?1 KH2PO4 at pH 2.7 5:95 (v/v) at a flow rate of 0.8 mL min?1 was used. Detection wavelength was set at 210 nm except for ascorbic acid which was detected at 243 nm. Application to red wine and must confirmed good repeatability and showed a wide variation range for concentrations of organic acids.  相似文献

An amplified colorimetric method has been developed for the detection of protein and cancer cells based on the assembly of nucleic acids and proteins for the first time. In this process, the assembly of nucleic acids was triggered by a biotinylated DNA strand after a sandwich immunoreaction. The biotinylated DNA strand and sandwich immunocomplex were connected by streptavidin. Then, the assembly of biotinylated bovine serum albumin (Biotin-BSA) and streptavidin-horseradish peroxidase (SA-HRP) occurred at a node of the assembled products of nucleic acids through the biotin-streptavidin reaction. Under the catalysis of horseradish peroxidase, 3,3′,5,5′-tetramethylbenzidine (TMB) was oxidized by H2O2 and the oxidized product was analyzed by its UV–vis absorbance signal and sensitive colorimetric detection. This colorimetric sensor could not only achieve the quantitative determination of protein by UV–vis absorbance but could also be applied for semiquantitative determination by digital visualization. Using alpha-fetoprotein (AFP) as the model target, this proposed colorimetric method showed a wide linear range from 5 pg/mL to 1 ng/mL with a detection limit of 1.95 pg/mL by the instrument, and even 5 pg/mL target protein could be distinguished simply by the naked eye. This approach was then expanded to detect cancer cells based on the recognition of folic acid receptors that were over-expressed on the cancer cells by folic acid-tethered DNA. More importantly, this strategy can be further used as a universal colorimetric method for the determination of viruses or other proteins by changing the corresponding antibodies.  相似文献

Direct infusion mass spectrometry (DIMS) is a powerful technique in clinical diagnosis for screening neonatal amino acid metabolic disorders from dried blood spots (DBS). However, DIMS sometimes generated false-positive results for analysis of amino acids. In this work, we utilized a stable isotope derivatization method, combining with liquid chromatography tandem mass spectrometry (SID-LC-MS), to improve the specificity for screening amino acids in DBS specimens. A pair of isotope reagents, p-(dimethylamino)phenyl isothiocyanate (DMAP-NCS) and 4-isothiocyanato-N,N-bis(methyl-[2H2])aniline ([2H4]DMAP-NCS), was synthesized and used to label amino acids in DBS specimens. The [2H4]DMAP-NCS labelled amino acid standards were used as internal standards to compensate the matrix effect. This method was validated by measuring linearity, recovery and accuracy. The results showed that the developed SID-LC-MS method can be used for sensitive and selective determination of 12 diagnostically important amino acids in DBS specimens.  相似文献

A modification of a procedure by Hage [1] is proposed for the gas chromatographic evaluation of the content of free medium-chain fatty acids and related ethyl esters in beer. The method involves extraction of free fatty acids and ethyl esters by SPE using C18 bonded phase columns, derivatization of free fatty acids and related ethyl esters with diazomethane, and GC analysis using an SP-2340 capillary column. The results obtained have shown the method to be rapid and highly reproducible. The technique has been compared with other methods used for determination of free fatty acids.  相似文献

A simple and reliable co-electroosmotic capillary electrophoresis system for the fast determination of aromatic acids has been developed by employing poly (1-vinyl-3-butylimidazolium bromide) as the background electrolyte modifier. The polymeric ionic liquid was synthesized by the conventional radical polymerization. The reversed electroosmotic flow was obtained by adding a small amount of the polymeric ionic liquid (0.0006%, w/v) to the electrolyte. To further improve the resolution of aromatic acids, conditions including the concentration of polymeric ionic liquid and pH of background electrolytes were optimized. All eight aromatic acids were baseline resolved in one measurement in a short time (less than 3.5 min) under optimized conditions, 100 mM NaH2PO4 buffer containing 0.006% (w/v) polymeric ionic liquid, pH 6.0. Separation efficiencies were in the range from 355,000 to 943,000 (plates/m). Satisfactory reproducibility on the basis of the migration time of analytes was achieved. RSDs (n = 3) were less than 0.33% except the p-aminobenzoic acid (0.9%). The applicability of the present method has been demonstrated for the determination of water-soluble aromatic acids in a common drug for external use.  相似文献

Precolumn derivatization with the reagent o-phthalic aldehyde/N-acetyl-L-cysteine (OPA/NAC) was used for the determination of amino acid enantiomers by reversed-phase high-performance liquid chromatography. The influence of the composition and pH of the eluent on the separation of the resulting derivatives was studied with the example of four amino acids. It was found that the highest selectivity and efficiency of the separation of OPA/NAC derivatives of amino acids is attained with the use of the eluent methanol–0.01 M Na2HPO4 (pH 6.0). The optimum composition of the mobile phase and conditions of the gradient elution were selected for the separation of a mixture of 20 amino acid derivatives. A procedure was developed for the determination of amino acid enantiomers in parenteral nutrition preparations. The procedure was used for the determination of D-isomers of arginine, alanine, methionine, phenylalanine, and leucine in the preparation Polyamine.  相似文献

The use of ten fatty acid methyl ester reference standards coupled with a detailed quantification method was shown to significantly optimize the fatty acid determination of selected fish and microalgal oils when compared to methods that use only one reference standard (C19:0 or C23:0) as a relative response factor. When using the mixture of ten reference standards after transesterifying oils with NaOH/BF3, determination of total fatty acids, eicosapentaenoic acid and docosahexaenoic acid improved by an average of 7.3, 11.5 and 8.4%, respectively. Furthermore, improvements of 13.9, 18.9 and 6.8% of total fatty acids, EPA and DHA, respectively, were obtained when using the mixture of reference standards for fatty acid determination after directly extracting and transesterifying oil contained in microalgal cells with a mixture of methanol, HCl and chloroform. Fatty acid methyl ester standards dissolved in isooctane showed <5% variability throughout 130 days of stability testing when stored at ?20 °C. The optimized method can be used for improving the quantification of fatty acids in both oils (fish and microalgal oils) and dry microalgal cells.  相似文献

An ion-exchange ICP-AES method for the determination of 14 rare earth elements (REEs) and Y in geological materials is described. The separation of REEs from Ba using a Dowex 50W-X8 cation resin is especially considered since Ba is an excellent internal standard for REE determination by this technique. Although total recovery with either HCl or HNO3 may be achieved, it is advantageous to use both acids sequentially. Volume and concentration of the acids are optimized attaining a quantitative separation of REEs from Ba by the introduction of the sample solution in a 1.75 mol/l HCl medium, followed by elution with 2 mol/l HNO3 to remove matrix elements and with 7 mol/l HNO3 to elute the analytes. The total elution volume is significantly reduced without decreasing the efficiency. The behaviour of the matrix constituents under the selected conditions is also studied, evaluating their elution percentages in each step. The final solution obtained contains only the REEs and Y, with the bulk of Sc and minor amounts of Cr, Fe, Hf and Ta. Experimental data for 5 geological reference standards (NIM-G, GSP-1, AGV-1, NIM-L and NIM-S) are reported. Good agreement between the present results and previously accepted values by various analytical techniques is observed.  相似文献

Summary Atmospheric precipitation and aerosol samples are characterized by a complex mixture of several organic compounds. A simple method for the simultaneous determination of the main compound classes by GC/MS is presented. In detail, seasonal variations of C8–C32 fatty acids in precipitation in a semirural area have been studied. Total fatty acids concentrations of 7–53 g/l were detected. Summer rain is characterized by high amounts of fatty acids >C20 and lower amounts of C11–C20 species. Two effects may be responsible: larger emission rates of fatty acids during vegetation periods and an increased influence of vapour phase due to higher temperatures during summer. CPI values showed no general trend; lower CPI values for winter rain could not be observed. This would be expected if anthropogenic sources play an important role. Monounsaturated fatty acids (C16:1, C18:1) were more abundant during winter than in summer. High concentrations of polyunsaturated fatty acids (C18:x) could be detected during summer. High relative concentrations of the potential oxidation products of the unsaturated species, nonanedioic acid and w-oxononanoic acid, are associated with relatively low concentrations of their precursors.  相似文献
