An improved procedure for the synthesis of N-bromoacetyl--glycopyranosylamines from the corresponding -glycosylamines was developed. The procedure is applicable to a wide range of derivatives of monosaccharides (hexoses, 2-acetamido-2-deoxyhexoses, hexuronamides, and 6-deoxyhexoses) and some disaccharides. For the derivatives of pentoses and 2-deoxyhexoses, the use of the corresponding -glycosylammonium carbamates was found to be more convenient. N-Bromoacetyl--glycopyranosylamines derived from D-mannose, L-rhamnose, D-glucuronamide, 2-deoxy-D-arabino-hexose, 2-deoxy-D-lyxo-hexose, and melibiose were obtained for the first time.  相似文献

The action of ammonium carbamate in aqueous methanol in the presence of NH3 on three-, penta-, hexa-, octa-, and nonasaccharides of human milk and on decasaccharide (N-glycan from human immunoglobulin (IgG)) containing one or two a-l-fucose residues led to the corresponding β-glycopyranosylamines. After their N-acylation upon treatment with N-hydroxysuccinimide ester of N-Boc-glycine or N-Boc-glycine anhydride (Boc is the tert-butyloxycarbonyl) with subsequent removal of the Boc group, N-glycyl-β-glycopyranosylamines were obtained in up to 75% yield.  相似文献

The behaviour towards the retro-aldol or retro-Dieckmann reactions of several norbornene and norbornane amino acids functionalized with an oxygen atom at C-β and characterized by different features, such as the substitution pattern and the constraints, was investigated. By C2-C3 disconnection new differently functionalized cyclopentenyl- and cyclopentylglycines were prepared. This synthetic approach allows to control the stereochemistry from two to four stereocentres depending on the starting norbornane derivative and affords, for each derivative, a couple of epimeric compounds at amino acid stereocentre. Depending on the features of the starting reagent and of the reaction conditions, a partial control of the stereochemistry of the amino acid carbon atom is also possible.  相似文献

4-Hydroxymethylbutenolide 4 was transformed into its sulfamoyl derivative 5, which upon treatment with iodosobenzene diacetate and magnesium oxide in the presence of a rhodium catalyst afforded the product of intramolecular aziridination 6. Reaction of 6 with primary or secondary amines in DMA led to regioselective opening of the aziridine ring at C2 to give the corresponding bicyclic derivatives 7a-7g in good to excellent yields. Methanolysis of the lactone ring of the N-benzyl-N-methyl derivative 7c followed by protection of the resulting secondary hydroxy group and treatment of the product with Boc anhydride provided the activated cyclic sulfamates 13 and 14. The latter then reacted with a second nucleophile (azide or thiophenol) to give the corresponding difunctionalized α,β-diamino methyl esters 15-18, 20.  相似文献

It was demonstrated that, in reactions of the hydrolysis of model substrate 2-nitrophenyl-β-D-galactopyranoside (2-NPGP) monosaccharides D-fructose and D-xylose with hydroxyl substituents oppositely directed at the neighboring carbon atoms in the furan ring, as in D-glucose, act as noncompetitive inhibitors of β-galactosidase from E. coli; for mushroom, β-galactosidases from P. canescens and A. oryzae D-galactose is a stronger inhibitor. It was also found that the inhibition constant is the highest in the case of the most active enzyme (E. coli) and is the lowest for the least active one (P. canescens).  相似文献

An efficient protocol for the preparation of β,β-difluorinated 1-amino-1-cyclopentane carboxylic acid derivatives was developed. 2,2-Difluro-4-phenyl-3-butenoic acid 6 was used as substrate for the preparation of the starting vinyl difluoro imino esters 8. The key steps of this methodology rely on the chemo- and diastereoselective addition of allylzinc bromides over the iminic functionality of 8 and subsequent RCM reaction. This synthetic sequence was successfully applied to fluorous synthesis.  相似文献

We report an efficient and reliable method for the synthesis of α-chiral-β,γ-unsaturated carboxylic acid derivatives using chiral auxiliaries and vinylacetic acid. Two well-established chiral auxiliaries ((S,S)-pseudoephedrine and (R)-benzyl-oxazolidinone) were chosen to test the merits of this method. Six different electrophiles were examined in the diastereoselective alkylation with both auxiliaries. The pseudoephedrine auxiliary provided isolated yields between 61 and 85% and diastereomeric ratios all greater than 96:4. Employing the same reactions as with the pseudoephedrine derivative, the corresponding oxazolidinone auxiliary provided isolated yields between 0 and 80% with diastereomeric ratios from 80:20 to 93:7.  相似文献

The chiral macrocycles 1 and 2 with multiple binding sites have been synthesized from D-phenylalanine as chiral solvating agents (CSAs) for the enantiomeric discrimination and determination of the enantiomeric excess of carboxylic acids and a-amino acids derivatives by the 1H NMR spectroscopy. With more and more organometallic compounds receiving attention for applications in medicinal organometallic chemistry, the need arises for stereoselective syntheses of more complicated structures containing organometallic moieties, for example as isosteric substitutes for organic drug candidates. Herein, the synthesis and characterization of both diastereomers of a planar chiral (η(6)-arene)Cr(CO)(3) containing carboxylic acid derivative, namely, 3-{η(6)-(1, 2, 3, 4-tetrahydro-1-endo/exo-methyl-2-oxonaphthalen-1-yl)-tricarbonylchromium(0)}propanoic acid (7 and 8) is reported. The molecular structures of both were confirmed by single crystal X-ray diffraction. The degree of diastereoselectivity in Cr(CO)(3) complexation with methyl/tert-butyl-3-(1,2,3,4-tetrahydro-1-methyl-2-oxonaphthalen-1-yl)propanoate (4a/4b) vs. the Michael addition of methyl/tert-butyl acrylate to (η(6)-1-methyl-2-tetralone)Cr(CO)(3) (9) was also examined. In the latter case the alkylation was found to be completely diastereoselective and gave methyl/tert-butyl-3-{η(6)-(1, 2, 3, 4-tetrahydro-1-endo-methyl-2-oxonaphthalen-1-yl)-tricarbonylchromium (0)}propanoate (5a and 5b) in excellent yield. Both the carboxylic acids 7 and 8 were coupled with the aminoresorcyclic acid core to achieve diastereomeric bioorganometallics 15a and 15b based on the naturally occurring antibiotic platensimycin lead structure (1a, see Fig. 1). The newly synthesized bioorganometallics were tested against various Gram-positive and Gram-negative bacterial strains but show no promising antibacterial activity.  相似文献

Some new mono-, bis-spiro- and dispiro-β-lactams have been synthesized from imines derived from 9H-fluoren-9-one and a ketene derived from 9H-xanthene-9-carboxylic acid or phenoxyacetic acid by a [2+2] cycloaddition reaction in good to excellent yields varying from 45 to 83%. The biological activity of these monocyclic β-lactams was successfully investigated against Plasmodium falciparum K14 resistant strain with excellent EC50 values up to 5 μM.  相似文献

Model compounds of some polychlorobiphenyl congeners, viz., 4,4´-dihalobiphenyls, were synthesized. These compounds can be successfully utilized by a chemical method for the in situ generation of arynes followed by the reaction with carbazole in order to prepare appropriate bis(carbazolyl)biphenyls as components for molecular electronics and OLED devices.

β-Keto carboxylic acids are key intermediates in organic syntheses, used for the development of fine chemicals, natural products, and various biologically relevant molecules. Their utilities stem from the structural features and facile bond formations, e.g., asymmetric reduction of carbonyl groups for the synthesis of β-hydroxy carboxylic acids and conjugated addition reactions through decarboxylative enolate nucleophiles, which utilize the amphiphilic reactivity of β-keto carboxylic acids. Despite their versatility and utilities, development of efficient and straightforward synthetic methods for β-keto carboxylic acids has not attracted considerable attention owing to their instability. As efficient synthetic strategies for β-keto carboxylic acids and their derivatives, reactions of α-diazoesters, acylation of malonate anions, cross-coupling reactions, and CO2 insertion reactions are summarized in this review.  相似文献

A new approach to the synthesis of 1,3-oxazolidine-2,4-diones, via a two-step reaction sequence, starting from the readily available α-ketols and isocyanates, is reported. The condensation of the latter led to the key precursors 4-methylene-2-oxazolidinones, which are converted into the diones by an oxidative cleavage of the exocyclic double bond. Thus, 5,5-disubstituted 1,3-oxazolidine-2,4-diones can be accessed in good yields from the appropriate functionalized α-ketols. Moreover, two alternative routes are also described either by functionalization of 4-oxazolin-2-ones or by alkylation of the 1,3-oxazolidine-2,4-dione core previously prepared. Upon hydrolysis of the 1,3-oxazolidine-2,4-diones, a series of α-hydroxyamides bearing a quaternary stereocenter were obtained.  相似文献
